High variability of the electron carrier plastocyanin in microalgae.

1. The plastocyanin content of Scenedesmus can be dramatically varied with the copper content of the culture medium. Figures as high as 7 mmol plastocyanin/mol chlorophyll are possible. Electron paramagnetic resonance (EPR) spectroscopy has been used to determine this physiological response quantitatively in intact cells having different amounts of plastocyanin. The results obtained by the EPR technique were compared with data on isolated plastocyanin determined either by spectrophotometry or immunoelectrophoresis. Agreement was found for the amount of plastocyanin detected by the first two me

Glycolipids slow interfacial proton migration while preserving surface proton retention.

Proton gradients power diverse biological processes, yet how interfacial proton migration is regulated remains unclear. Here we quantify how membrane composition controls interfacial proton migration using an approach that releases protons directly at the surface of a membrane patch via an embedded ionophore. Fluorometrically monitoring proton arrival at a distant patch across neutral, negatively charged, and positively charged membranes, we confirm that both the lateral surface diffusion coefficient and the activation barrier for proton release into the bulk vary rather modestly. In contrast

Arabidopsis chloroplastic ascorbate peroxidase isoenzymes play a dual role in photoprotection and gene regulation under photooxidative stress.

Though two types of chloroplastic ascorbate peroxidase (APX) located in the thylakoid membrane (tAPX) and stroma (sAPX) have been thought to be key regulators of intracellular levels of H(2)O(2), their physiological significance in the response to photooxidative stress is still under discussion. Here we characterized single mutants lacking either tAPX (KO-tAPX) or sAPX (KO-sAPX). Under exposure to high light or treatment with methylviologen under light, H(2)O(2) and oxidized proteins accumulated to higher levels in both mutant plants than in the wild-type plants. On the other hand, the absence

Chloroplast-to-chromoplast transition envisions provitamin A biofortification in green vegetables.

The carotenoids available in food are vital dietary micronutrients for human health. Plants synthesize and accumulate different carotenoids in plastids in a tissue-specific manner. The level of β-carotene (provitamin A) and other nutritionally important carotenoids is substantially low in the green tissues such as leaves compared to the fruits and roots. In photosynthetic tissues, chloroplasts can accumulate a moderate level of carotenoids, mainly to facilitate photosynthesis and environmental stress tolerance. However, chromoplasts from the storage tissues such as tomato fruit and carrot root

Improved response of triploid citrus varieties to water deficit is related to anatomical and cytological properties.

Polyploidy plays a major role in citrus plant breeding to improve the adaptation of polyploid rootstocks as well as scions to adverse conditions and to enhance agronomic characteristics. In Citrus breeding programs, triploidy could be a useful tool to react to environmental issues and consumer demands because the produced fruits are seedless. In this study, we compared the physiological, biochemical, morphological, and ultrastructural responses to water deficit of triploid and diploid citrus varieties obtained from 'Fortune' mandarin and 'Ellendale' tangor hybridization. One diploid clementine

Suppression of both ELIP1 and ELIP2 in Arabidopsis does not affect tolerance to photoinhibition and photooxidative stress.

ELIPs (early light-induced proteins) are thylakoid proteins transiently induced during greening of etiolated seedlings and during exposure to high light stress conditions. This expression pattern suggests that these proteins may be involved in the protection of the photosynthetic apparatus against photooxidative damage. To test this hypothesis, we have generated Arabidopsis (Arabidopsis thaliana) mutant plants null for both elip genes (Elip1 and Elip2) and have analyzed their sensitivity to light during greening of seedlings and to high light and cold in mature plants. In particular, we have e

Unraveling design principles of protein landscapes in photosynthetic membranes in plant chloroplasts.

The supramolecular organization of proteins within photosynthetic membranes is crucial for energy conversion in plants. Here, we introduce an analytical and computational pipeline that integrates high-resolution cryo-scanning electron microscopy, biochemical quantification, advanced Monte Carlo computer simulations, and statistical methods to elucidate the elusive protein landscapes of grana membranes in intact Arabidopsis leaves. Our integrated analysis challenges the prevailing view that particles on the exoplasmic fracture faces in freeze-fracture samples represent photosystem II exclusivel

Chloroplasts balance energy production and stress resilience via a cold-induced signaling pathway.

Chloroplasts are plant-specific organelles for photosynthesis and play crucial roles in stress response. However, how chloroplasts respond to cold stress remains unclear. Here, we identify an rapidly accelerated fifibrosarcoma (RAF)-like bi-kinase module, RAF3/6, as key mediators in chloroplast cold-stress responses. RAF3/6 separately localize to the cytoplasm and plasma membrane under growth conditions. Upon cold stress, RAF3/6 translocate into chloroplasts mediated by the chaperone HSP70-1, where they phosphorylate the PSII components D1 and LHCB1, promoting their redistribution across thyla

Hydrogen Sulfide Primes bZIP68 via Persulfidation to Enhance Redox-Dependent Transcription and Adaptation to Osmotic Stress in Rice.

Osmotic stress limits rice productivity, yet the crosstalk between hydrogen sulfide signaling and redox regulation remains incompletely understood. We previously showed that redox-dependent oligomerization of the basic (region) leucine zippers transcription factor bZIP68 at Cys245 confers osmotic tolerance. However, the role of an adjacent cysteine, Cys171, was undefined. Here, we demonstrate that osmotic stress induces persulfidation of bZIP68 specifically at Cys171. This modification facilitates Cys245-mediated oxidation-dependent oligomerization, thereby enhancing bZIP68 transcriptional act

Time-resolved transcriptomic analysis suggests candidate hub gene modules and putative regulatory pathways in tobacco under dicamba stress.

INTRODUCTION: Widespread dicamba use poses challenges of resistance and phytotoxicity. To investigate the temporal molecular response mechanisms of tobacco, this study performed a time-resolved transcriptomic analysis of tobacco seedlings exposed to dicamba. METHODS: Samples were collected at 6, 24, and 72 h after treatment. RESULTS: Results indicated that tobacco exhibits a putative three-phase transcriptional adaptation pattern of "perception-defense-repair". In the early stage (6h), NtIAA genes were rapidly induced alongside activation of the glutathione system, potentially alleviating ox

Endoplasmic reticulum stress and unfolded protein response in renal lipid metabolism.

The endoplasmic reticulum (ER) is a crucial cellular organelle involved in protein synthesis, folding, modification, and transport. Exposure to internal and external stressors can induce endoplasmic reticulum stress (ERS), leading to abnormal protein folding and ER malfunction. This stress can disrupt lipid synthesis, metabolism, and transport processes. Fatty acid oxidation is the primary energy source for the renal system. When energy intake exceeds the storage capacity of adipose tissue, lipids accumulate abnormally in non-adipose tissues, including kidneys, liver, and pancreas. Lipids accu

Unveiling the Role of SlRNC1 in Chloroplast Development and Global Gene Regulation in Tomato Plants.

RNC1, a plant-specific gene, is known for its involvement in splicing group II introns within maize chloroplast. However, its role in chloroplast development and global gene expression remains poorly understood. This study aimed to investigate the role of RNC1 in chloroplast development and identify the genes that mediate its function in the development of entire tomato plants. Consistent with findings in maize, RNC1 silencing induced dwarfism and leaf whitening in tomato plants. Subcellular localization analysis revealed that the RNC1 protein is localized to both the nucleus and cytoplasm, in

Biomimetic monolayer films of monogalactosyldiacylglycerol incorporating plastoquinone.

Photosynthetic organisms use light to convert the inorganic matter in organic one. Photosynthetic process consists on several steps, and one of them involves plastoquinone (PQ) that acts as electron and proton shuttle between photosystem II and cytochrome. We prepared membranes that mimic the characteristics and composition of natural photosynthetic membranes and we characterized them using several techniques in order to obtain both the PQ molecules disposition in the membrane and their electrochemical behavior. The selected lipid was monogalactosyldiacylglycerol (MGDG) that represents the 50%

Relaxation of the non-photochemical chlorophyll fluorescence quenching in diatoms: kinetics, components and mechanisms.

Diatoms are especially important microorganisms because they constitute the larger group of microalgae. To survive the constant variations of the light environment, diatoms have developed mechanisms aiming at the dissipation of excess energy, such as the xanthophyll cycle and the non-photochemical chlorophyll (Chl) fluorescence quenching. This contribution is dedicated to the relaxation of the latter process when the adverse conditions cease. An original nonlinear regression analysis of the relaxation of non-photochemical Chl fluorescence quenching, qN, in diatoms is presented. It was used to

β-Carbonic anhydrases integrate CO2 and light cues to coordinate photosynthesis in tomato.

Optimization of photosynthesis requires precise coordination between electron transport and CO2 assimilation, but how plants maintain this coordination under suboptimal CO2 and light conditions remains poorly understood. Here, we identify two tomato (Solanum lycopersicum) β-carbonic anhydrases (βCAs) responsive to deficient CO2. Although both are required for biomass accumulation and efficient photosynthesis, the chloroplast βCA1 does not directly participate in carbon assimilation but instead specifically coordinates photosystem performance and leaf internal structure. This role is distinct f

Temperature treatments of dark-grown pea seedlings cause an accelerated greening in the light at different levels of gene expression.

We have previously shown that heat-shock in the dark evokes photomorphogenesis-like effects and circadian rhythmicity at the level of mRNAs when applied to emerging pea plantlets during several consecutive days [15]. Here we extend these findings by showing that a temperature shift to 10 degrees C above average and a single heat-shock are sufficient for induction of circadian rhythmicity and changes in morphogenesis. The maximum response to a single heat-shock occurs at days 2 to 3 after sowing indicating additional developmental control of the response. An increasing number of heat-shock trea

The AtRNC1-AtWTF1 protein complex is essential for chloroplast group Ⅱ intron splicing, ribosome biogenesis, and embryo development in Arabidopsis.

The splicing of group Ⅱ introns in chloroplasts is essential for photosynthesis and plant development and relies on a series of nucleus-encoded RNA-binding proteins. Here, we demonstrate that two Arabidopsis proteins, ribonuclease Ⅲ-domain protein AtRNC1 and the Plant organelle RNA recognition (PORR) protein AtWTF1, are essential for chloroplast function. Null mutants of AtRNC1 or AtWTF1 are embryo lethal. We rescued the null mutant by expressing wild-type AtRNC1 under the control of the seed-specific ABSCISIC ACID-INSENSITIVE3 (ABI3) promoter. We also created atrnc1 knockdown plants using art

Survival without photosynthesis: physiological characterization of long-lived albino beech (Fagus sylvatica).

Albinism is typically lethal in autotrophic plants due to the absence of photosynthetic pigments and functioning chloroplasts. Yet, rare exceptions occur where achlorophyllous individuals persist in natural ecosystems. We investigated the physiological, anatomical and isotopic characteristics of naturally occurring albino European beech (Fagus sylvatica L.) trees in the Moravian Karst, Czechia. One albino individual, estimated to be ~30 years old, represents an unprecedented case of long-term survival without photosynthesis in a woody angiosperm. Using a multi-parameter approach-including stab

A laser ablation ICP-MS based method for multiplexed immunoblot analysis: applications to manganese-dependent protein dynamics of photosystem II in barley (Hordeum vulgare L.).

Manganese (Mn) constitutes an essential co-factor in the oxygen-evolving complex of photosystem II (PSII). Consequently, Mn deficiency reduces photosynthetic efficiency and leads to changes in PSII composition. In order to study these changes, multiplexed protein assays are advantageous. Here, we developed a multiplexed antibody-based assay and analysed selected PSII subunits in barley (Hordeum vulgare L.). A selection of antibodies were labelled with specific lanthanides and immunoreacted with thylakoids exposed to Mn deficiency after western blotting. Subsequently, western blot membranes wer

Cytological and Transcriptomic Analysis Provide Insights into the Formation of Variegated Leaves in Ilex × altaclerensis 'Belgica Aurea'.

Ilex × altaclerensis 'Belgica Aurea' is an attractive ornamental plant bearing yellow-green variegated leaves. However, the mechanisms underlying the formation of leaf variegation in this species are still unclear. Here, the juvenile yellow leaves and mature variegated leaves of I. altaclerensis 'Belgica Aurea' were compared in terms of leaf structure, pigment content and transcriptomics. The results showed that no obvious differences in histology were noticed between yellow and variegated leaves, however, ruptured thylakoid membranes and altered ultrastructure of chloroplasts were found in ye
