Trained immunity is a long-term increase in responsiveness of innate immune cells, induced by certain infections and vaccines. During the last 3 years of the COVID-19 pandemic, vaccines that induce trained immunity, such as BCG, MMR, OPV, and others, have been investigated for their capacity to protect against COVID-19. Further, trained immunity-inducing vaccines have been shown to improve B and T cell responsiveness to both mRNA- and adenovirus-based anti-COVID-19 vaccines. Moreover, SARS-CoV-2 infection itself induces inappropriately strong programs of trained immunity in some individuals, w

Previous urinary tract infections (UTIs) can predispose one to future infections; however, the underlying mechanisms affecting recurrence are poorly understood. We previously found that UTIs in mice cause differential bladder epithelial (urothelial) remodelling, depending on disease outcome, that impacts susceptibility to recurrent UTI. Here we compared urothelial stem cell (USC) lines isolated from mice with a history of either resolved or chronic uropathogenic Escherichia coli (UPEC) infection, elucidating evidence of molecular imprinting that involved epigenetic changes, including differenc

Protective variables for Coronavirus Disease 2019 (COVID-19) are unknown. "Trained immunity" of the populace as a result of Bacille Calmette-Guérin (BCG) vaccination policy implementation and coverage had been suggested to be one of the factors responsible for the differential impact of COVID-19 on different countries. Several trials are underway to evaluate the potential protective role of BCG vaccination in COVID-19. However, the lack of clarity on the use of appropriate controls concerning the measures of "trained immunity" or the heterologous cell-mediated immunity conferred by BCG vaccina

Trained immunity is a form of innate immune memory in which initial exposure to microbial components or vaccines leads to long-lasting functional reprogramming of innate immune cells through epigenetic and metabolic changes. Unlike adaptive immunity, it does not rely on B or T lymphocytes and provides broad, nonspecific protection against various pathogens. This phenomenon has been observed across a range of immune cells, including monocytes, macrophages, neutrophils, NK cells and even γδ T-cells, as well as at the level of haematopoietic stem and progenitor cells (HSPCs). Key inducers include

The connection between hypoxia and inflammation is well established at the local level, with HIF (hypoxia-inducible factor) playing a central role, which controls both the cellular response to oxygen deficiency and immune cell functioning. The literature contains extensive data regarding the roles of the transcription factors HIF, NF-κB (a key regulator of inflammatory processes) and NRF2 (a master regulator of antioxidant defense), as well as the roles of oxidative stress level and mitochondrial functional features, in the response to oxygen deficiency and the development of inflammation. Invertebrates rely exclusively on innate immunity but exhibit memory-like responses termed immune priming or trained immunity. In the commercially vital whiteleg shrimp (Litopenaeus vannamei), infection by Vibrio parahaemolyticus causes severe economic losses, yet the molecular networks driving secondary immune recall remain poorly understood. In this study, we established a two-step immune challenge model in L. vannamei using formaldehyde-inactivated V. parahaemolyticus and performed transcriptomic analysis on hemocytes to compare primary and secondary immune responses. BACKGROUND: Trained immunity provides a framework for understanding persistent innate immune reprogramming in chronic inflammatory and immune-mediated disorders. Its relationship with osteoimmunology and inflammatory bone disease, however, remains insufficiently characterized.
METHODS: We performed a bibliometric and literature-level text-mining analysis using WoSCC as the primary source for formal bibliometric analyses. A two-corpus design was used, with a core corpus for disease-focused bibliometric analysis and an extended corpus for semantic screening and sensitivity analyses. Scopus and

Although chronic psychosocial stress is a well-established independent risk factor for cardiovascular disease (CVD), the underlying mechanisms linking stress to CVD remain incompletely understood. Here, we review clinical and preclinical research documenting how stress can activate innate immune cells. Based on these findings, we propose that trained innate immunity is one possible mechanism responsible for the lasting effects of stress on the innate immune system, and hence contributes to the association between stress and CVD. Trained immunity is defined as a persistent hyperinflammatory phe

BACKGROUND: Despite successful reduction of bacterial load and clinical inflammation following periodontal therapy, high recurrence rates indicate that prior infections induce lasting host alterations, maintaining latent disease susceptibility AIM: This review elucidates the biological basis of periodontal recalcitrance through innate immune memory (trained immunity), examining durable functional reprogramming at both local and systemic levels.

Trained immunity, characterized by the long-term functional reprogramming of innate immune cells through epigenetic and metabolic modifications, has emerged as a pivotal concept bridging innate and adaptive immune responses. This review explores the dual role of trained immunity as both a protective mechanism in cancer and a pathogenic driver in autoimmune diseases. We first discuss the underlying mechanisms involving histone modifications, chromatin remodeling, and metabolic pathways such as glycolysis and the mTOR/HIF-1α axis, alongside key regulators including NOD2 and pattern recognition r

Trained immunity enables innate immune cells to acquire memory-like responses, offering a strategy to enhance antitumor immunity. However, the metabolic‒epigenetic mechanisms underlying this process remain poorly defined. Here, we show that lipopolysaccharide-induced macrophage training is encoded by a mitochondrial metabolic checkpoint. Integrated transcriptomic, metabolomic, and epigenomic profiling reveals that TLR4-NF-κB signaling represses SLC1A5_var, a mitochondrial glutamine transporter, limiting glutaminolysis and reducing α-ketoglutarate availability. This metabolic restriction limits

BACKGROUND: The prognosis of patients with cancer in which tumor-associated macrophages with a pro-tumoral phenotype are abundant is poor. This includes aggressive forms of non-medullary thyroid carcinoma (NMTC). Trained immunity describes long-term epigenetic and metabolic reprogramming in innate immune cells and their bone marrow progenitors, leading to improved responsiveness, which is currently being explored as a potential new treatment approach in cancer. We aimed to assess whether trained immunity can be induced in myeloid cells of patients with NMTC to enhance the anti-tumor immune res

Diabetes mellitus, a chronic metabolic condition, is marked by ongoing hyperglycemia and poses an increasing global health issue. Beyond its recognized contribution to the development of cardiovascular diseases and kidney problems, diabetes can profoundly impact immune system functions. Recent developments in immunology have revealed trained immunity as a mechanism through which innate immune cells experience enduring functional modifications following their first encounter with specific stimuli. This review compiles the latest evidence concerning the role of trained immunity in the developmen

Fungal skin infections are distributed worldwide and can be associated with economic and social traits. The immune response related to skin cells is complex and its understanding is essential to the comprehension of each cell's role and the discovery of treatment alternatives. The first studies of trained immunity (TI) described the ability of monocytes, macrophages and natural killer (NK) cells to develop a memory-like response. However, the duration of TI does not reflect the shorter lifespan of these cells. These conclusions supported later studies showing that TI can be observed in stem an

Antibiotic resistance and the surge of infectious diseases during the pandemic present significant threats to human health. Trained immunity emerges as a promising and innovative approach to address these infections. Synthetic or natural fungal, parasitic and viral components have been reported to induce trained immunity. However, it is not clear whether bacterial virulence proteins can induce protective trained immunity. Our research demonstrates Streptococcus pneumoniae virulence protein PepO, is a highly potent trained immunity inducer for combating broad-spectrum infection. Our findings sh

Disrupted immune response is an important feature of many neurodegenerative conditions, including sepsis-associated cognitive impairment. Accumulating evidence has demonstrated that immune memory occurs in microglia, which has a significant impact on pathological hallmarks of neurological diseases. However, it remains unclear whether immune memory can cause subsequent alterations in the brain immune response and affect neurobehavioral outcomes in sepsis survivors. In the present study, mice received daily intraperitoneal injection of low-dose lipopolysaccharide (LPS, 0.1 mg/kg) for three conse

BCG vaccination protects not only against tuberculosis but also against heterologous infections. This effect differs between individuals, yet the factors responsible for this variation are unknown. BCG-induced nonspecific protection is, at least partially, mediated by innate immune reprogramming (trained immunity), which can be induced by the muramyl dipeptide (MDP) component of peptidoglycans. We aimed to study whether differential release of MDP in healthy individuals may explain variability of their response to BCG vaccination. Circulating MDP concentrations were increased up to three month

Adaptive features of innate immunity, also termed 'trained immunity', have recently been shown to characterize monocytes of BCG vaccinated healthy volunteers. Trained immunity leads to increased cytokine production in response to non-related pathogens via epigenetic reprogramming of monocytes. Recently, memory-like properties were also observed in NK cells during viral infections, but it is unknown if memory properties of NK cells contribute to trained immunity due to BCG vaccination. BCG vaccination of healthy volunteers increased proinflammatory cytokine production following ex vivo stimulat

As early as 1931, bacillus Calmette-Guérin (BCG) vaccination was hypothesized to reduce neonatal mortality through nonspecific immune stimulation. The concept of trained immunity was formally proposed in the 21st century, and clinical observations during the COVID-19 pandemic further stimulated interest in the possibility that BCG vaccination might confer broad, non-specific protection against heterologous infections. Trained immunity represents a paradigm-shifting concept in immunology, defining the innate immune system's ability to develop a memory-like state through persistent epigenetic an

Breakthroughs in cancer immunotherapy have been overwhelmingly driven by advances in adaptive immunity, leaving the therapeutic potential of innate immunity largely untapped, a gap in the current treatment paradigm. Trained immunity, defined by the durable, mitotically maintained metabolic and epigenetic memory that innate immune cells acquire after transient stimulation, offers a new approach to addressing this deficit. Here we survey the triggering signals, cellular heterogeneity, and molecular circuitry that govern trained immunity within the tumor microenvironment. We dissect its dualistic
