Characterization of Tumor Immune Microenvironment in Meningiomas: Correlation of Tumor-infiltrating Lymphocyte Aggregates With Tumor Grade.

BACKGROUND/AIM: Malignant meningiomas are aggressive intracranial tumors with high recurrence rates and limited treatment options. The tumor immune microenvironment (TIME) plays a pivotal role in the tumor progression and treatment response. However, its role in meningioma remains largely unknown. This study aimed to analyze tumor-infiltrating lymphocytes (TILs) within the meningioma TIME and investigate their correlation with clinical parameters. PATIENTS AND METHODS: Using tumor specimens from patients diagnosed with meningioma (grade 1, 12 cases; grade 2, 10 cases), the densities of CD4+ T

Application of tumor organoids simulating the tumor microenvironment in basic and clinical research of tumor immunotherapy.

Immunotherapy has led to groundbreaking advances in anti-tumor treatment, yet significant clinical challenges remain such as the low proportion of beneficiaries and the lack of effective platforms for predicting therapeutic response. Organoid technology provides a novel solution to these issues. Organoids are three-dimensional tissue cultures derived from adult stem cells or pluripotent stem cells that closely replicate the structural and biological characteristics of native organs, demonstrating particularly strong potential in modeling the tumor microenvironment (TME). Tumor organoids can si

Deciphering the Anti-Tumor Mechanisms of Metformin Through Reprogramming of the Tumor Microenvironment.

Metformin is a cornerstone medication for type 2 diabetes and exhibits anti-tumor activities. Previous studies have demonstrated that metformin suppresses tumor progression by regulating multiple signaling pathways, including the AMPK, PI3K/AKT/mTOR, and JNK pathways. However, most previous studies have focused on its direct effects on tumor cells, with limited attention to its effects in the TME. The TME constitutes a multifaceted ecosystem that drives tumor development and therapeutic resistance via physical barrier formation, immune evasion, and abnormal angiogenesis. In this review, we sys

Tumor-associated neutrophils crosstalk with non-cancerous components of the gastrointestinal tumor microenvironment.

Gastrointestinal cancer progression is dictated by a dynamic and complex interaction between malignant cells and the non-malignant components of the tumor microenvironment. The unifying axis of this intercellular network is the functional reprogramming of stromal and immune cells. While tumor-associated neutrophils are known to play pivotal roles in gastrointestinal tissue tumorigenesis, research has rarely looked beyond their isolated functions. There is a critical gap in understanding the broader crosstalk through which neutrophils and neighboring non-malignant cells mutually alter each othe

Regucalcin in the Tumor Microenvironment: From Intracellular Tumor Suppression to Putative Extracellular Signaling.

Regucalcin (RGN), also known as senescence marker protein-30, is a calcium-binding protein without an EF-hand motif that regulates intracellular calcium homeostasis and Ca2+-dependent enzyme activity. Its expression is reduced in tumor tissue relative to matched normal tissue across several cancer types, and restoring RGN in cancer cell lines suppresses proliferation, migration, and invasion through cell cycle arrest, reduced expression of matrix metalloproteinases and epithelial-mesenchymal transition regulators, and transcriptional reprogramming involving p53, p21, Rb, c-myc, and β-catenin.

Hydrogel-dependent strategies to modulate the tumor microenvironment for improved anti-tumor treatment.

Cancer remains a leading cause of death worldwide, and effective therapies are still urgently needed. Although chemotherapy, surgery, radiotherapy, and immunotherapy have advanced cancer treatment, their efficacy and safety are often limited by the antagonistic tumor microenvironment (TME). Drug delivery systems across nano-, micro-, and macroscale dimensions offer opportunities to remodel the TME by improving pharmacokinetics and coordinating therapeutic modalities. Among them, hydrogels are particularly attractive owing to their tunable microstructures, high loading capacity, and manufacturi

Immunophenotypic changes in the tumor and tumor microenvironment during progression to multiple myeloma.

Investigation of the cellular and molecular mechanisms of disease progression from precursor plasma cell disorders to active disease increases our understanding of multiple myeloma (MM) pathogenesis and supports the development of novel therapeutic strategies. In this analysis, single-cell RNA sequencing, surface protein profiling, and B lymphocyte antigen receptor profiling of unsorted, whole bone marrow (BM) mononuclear cell samples was used to study molecular changes in tumor cells and the tumor microenvironment (TME). A cell atlas of the BM microenvironment was generated from 123 subjects

Senescent Tumor Cells in the Peritoneal Carcinomatosis Drive Immunosenescence in the Tumor Microenvironment.

More than half of all patients with colorectal cancer (CRC) develop distant metastasis and, depending on the local stage of the primary tumor, up to 48% of patients present peritoneal carcinomatosis (PC). PC is often considered as a widespread metastatic disease, which is almost resistant to current systemic therapies like chemotherapeutic and immunotherapeutic regimens. Here we could show that tumor cells of PC besides being senescent also exhibit stem cell features. To investigate these surprising findings in more detail, we established a murine model based on tumor organoids that resembles

Tumor-permeable smart liposomes by modulating the tumor microenvironment to improve the chemotherapy.

Low levels of accumulation and permeability in tumors are two primary reasons for the limited efficacy of conventional antineoplastic nanodrugs. In the present study, based on an original corosolic acid liposome (CALP) carrier with the functions of cell penetration, tumor permeability and anti-inflammation developed by our previous work, a versatile PTX/CALP was achieved by CALP loading paclitaxel (PTX). Compared to conventional PTX liposomes (PTX/LP) prepared by cholesterol and phospholipid, PTX/CALP exhibited extremely increasing cellular uptake and cytotoxicity in vitro, and in vivo enhanci

Adenoviral delivery of an immunomodulatory protein to the tumor microenvironment controls tumor growth.

Targeted modulation of the immune system against tumors can achieve responses in otherwise refractory cancers, which has spurred efforts aimed at optimizing such strategies. To this end, we have previously investigated cancer immunotherapy approaches using recombinant adenovirus vectors, as well as via modulation of the self-ligand receptor SLAMF7. Here, we present a gene transfer-based immunotherapy approach using targeted expression of a SLAMF7-Fc fusion construct directly into tumors at high concentrations via a recombinant adenoviral vector (Ad-SF7-Fc). Using multiple murine cancer models,

Phosphorylcholine-based stealthy nanocapsules enabling tumor microenvironment-responsive doxorubicin release for tumor suppression.

The efficient delivery of anticancer drugs into tumor tissues to improve therapeutic efficacy remains an urgent demand. To satisfy this demand, a drug delivery system based on a stealthy nanocapsule was developed. This nanocapsule was fabricated by encapsulating stealthy cross-linked poly(2-methacryloyloxyethyl phosphorylcholine) (PMPC) and benzaldehyde groups around the protein bovine serum albumin (BSA) followed by conjugation of doxorubicin (Dox) through a pH-responsive benzoic-imine bond. The in vitro results show that the Dox-conjugated nanocapsule (nBSA-Dox) released the drug under an ac

Mitochondrial transfer-driven immune evasion in the tumor microenvironment.

The tumor microenvironment (TME) is a complex landscape where metabolic interactions significantly dictate antitumor immunity. Immune evasion in cancer is typically discussed in terms of inhibitory receptors and ligands, suppressive cytokines, defective antigen presentation, and metabolic competition. However, recent evidence reveals that intercellular mitochondrial transfer adds a new mechanism of immune evasion in the TME. The mitochondrial fitness of T cells is central to sustained effector function, memory formation, and responsiveness to immune-checkpoint blockade. Tumor cells can act as

JAK inhibition in PD-1 immunotherapy and tumor microenvironment.

Targeting the programmed cell death 1 (PD-1)/PD-L1 axis has revolutionized cancer therapy; however, the durability of clinical responses is frequently compromised by chronic inflammation and an immunosuppressive tumor microenvironment (TME). The Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway serves as a central intracellular node integrating cytokine signals that drive these resistance mechanisms. While physiological JAK/STAT signaling is essential for antitumor immunity, its persistent aberrant activation promotes malignant progression, upregulates

Immunosuppressive tumor microenvironment and immunotherapy resistance of esophageal carcinoma.

Esophageal carcinoma (EC) remains one of the most lethal malignancies, with a five-year survival rate of only 15-25% in advanced stages. Although immunotherapy including immune checkpoint inhibitors (ICIs) and antibody-based therapies have improved clinical outcomes, durable responses are confined to a minority of patients because primary and acquired resistance affect the majority. The tumor microenvironment (TME)-composed of immune cells, stromal cells, and the extracellular matrix-has emerged as a central driver of the immune resistance, wherein infiltrating immune cells and stromal cells f

Microbial bile acids: Immunomodulatory signals in the tumor microenvironment.

Microbial metabolites can act as systemic signals that connect gut dysbiosis to tumor immune remodeling. Recently, Li et al. showed that gut microbiota-derived deoxycholic acid activates the farnesoid X receptor-nuclear factor κB-interleukin-6 axis in breast cancer cells, promoting the recruitment of granulocytic myeloid-derived suppressor cells, T helper polarization, and tumor progression. These findings offer new insights into microbial bile acid signaling in the tumor microenvironment.

Diversity within ovarian cancer subtypes and their tumor microenvironment.

Ovarian cancer is the deadliest gynecologic cancer in individuals assigned female at birth. Patients diagnosed with ovarian cancer experience poor survival rates due to frequent late-stage diagnosis and aggressive metastases. Ovarian cancer is often diagnosed at stages III or IV, where survival rates decline substantially. Ovarian cancer comprises several subtypes, each with distinct biological characteristics, clinical behaviors, and treatment responses, thereby complicating the identification of effective therapeutic targets. This review focuses primarily on the most prevalent subtype, high-

Polyamines in pancreatic cancer: reshaping the immunosuppressive tumor microenvironment.

Pancreatic ductal adenocarcinoma (PDAC) is a lethal malignancy, characterized by its aggressiveness and poor outcomes. Emerging evidence implicates dysregulated polyamine metabolism as a key driver of PDAC immunosuppression, yet the mechanisms underlying this metabolic-immune crosstalk remain poorly defined. This review summarizes recent findings demonstrating that PDAC is uniquely dependent on glutamine-derived ornithine for de novo polyamine synthesis, orchestrated by the KRAS-MYC axis. Through metabolic reprogramming of immune cells, polyamines polarize tumor-associated macrophages toward M

Tumor Microenvironment: Recent Advances in Immunotherapies of Pancreatic Cancer.

The progression of pancreatic cancer (PC) is significantly influenced by the immune system. In the United States, PC is the third leading cause of cancer-related mortality. The high lethality of PC is attributed to its immunological advantage, which is facilitated by an immunosuppressive microenvironment, a low mutational burden, and minimal T-cell infiltration. Although immunotherapies, such as checkpoint blockades or genetically engineered T cells, have not yet demonstrated viability, there is a growing body of evidence suggesting that innovative combinations of conventional therapies and va
