Valorization of bigeye tuna (Thunnus obesus) bone by-products via steam explosion: Novel umami peptides with synergistic taste enhancement and sodium reduction potential.

This study developed an integrated approach combining steam explosion pretreatment with enzymatic hydrolysis to isolate umami peptides from tuna (Thunnus obesus) bone by-products. Steam explosion enhanced the hydrolysis efficiency and increased the degree of hydrolysis from 9% to 18%. The ultrafiltration fraction below 3 kDa exhibited pronounced umami and saltiness-enhancing properties. Peptidomics and virtual screening identified three collagen-derived peptides (GPRGAPGPA, GPAGRPGPQ, GAQGPVGAPGPK) with umami detection thresholds between 0.15 and 0.45 mM. Molecular docking and dynamics simulat

Activity-Guided Isolation of sn-1,3-Dipalmitoyl-2-oleoylglycerol as Protein Tyrosine Phosphatase 1B and α-Glucosidase Inhibitor from Skipjack Tuna (Katsuwonus pelamis).

The skipjack tuna (Katsuwonus pelamis) is one of the least commercially exploited tuna globally. The possible medicinal benefits of K. pelamis extract have not been explored. We investigated the 70% ethanol (EtOH) soluble fractions of skipjack tuna heart for the inhibition of protein-tyrosine phosphatase 1B (PTP1B) and α-glucosidase. The dichloromethane (CH2Cl2) fraction significantly inhibited the activities of PTP1B and α-glucosidase. Repeated column chromatography of the active CH2Cl2 fraction based on bioactivity-guided fractionation yielded cholesterol, cholesteryl myristate, and sn-1,3-d

Rapid point-of-need blood fluke detection in Southern Bluefin Tuna samples using recombinase polymerase amplification coupled with lateral flow test (RPA-LF).

Aporocotylid blood flukes Cardicola forsteri and C. orientalis are considered one of the most significant health concerns for Southern Bluefin Tuna (SBT) Thunnus maccoyii ranched in Australia. There is a need for rapid point-of-need diagnostics to detect Cardicola spp. in SBT to allow the industry to make timely management decisions. Recombinase polymerase amplification (RPA) is an isothermal technique which operates at constant low temperature (25-42˚C), and when coupled with a lateral flow (LF) strip, makes an ideal diagnostic tool for rapid, specific, and sensitive identification of pathoge

Chemical Compositions and Characteristics of Biocalcium from Pre-Cooked Tuna Bone as Influenced by Sodium Chloride Pretreatment and Defatting by Asian Seabass Lipase.

Pre-cooked bone is a waste product generated during tuna processing and can serve as a potential source of biocalcium (BC). Generally, non-collagenous protein and fat must be removed properly from bone. A NaCl solution can be used to remove such proteins, while fish lipase can be used in a green process, instead of solvent, for fat removal. Thus, this study aimed to investigate the impact of NaCl pretreatment at different concentrations in combination with heat to eliminate non-collagenous proteins, and to implement fish lipase treatments at varying levels for fat removal, for BC production fr

Atlantic bluefin tuna tagged off Norway show extensive annual migrations, high site-fidelity and dynamic behaviour in the Atlantic Ocean and Mediterranean Sea.

Atlantic bluefin tuna (ABFT; Thunnus thynnus) is a highly migratory species. To investigate the migrations and vertical behaviours of ABFT migrating to Nordic waters, we deployed pop-up satellite archival transmitting tags on 25 ABFT off Norway (curved fork length: 228-292 cm). We obtained 16 full-year migrations, which differed between individuals, and physically recovered 13 tags, which provided 4699 days of archival depth and temperature data. ABFT occupied waters from the Arctic Circle to as far south as Cabo Verde, Africa, and occupied depths down to 1190 m and temperatures from 0.5 to 27

Immune reactivity in early life stages of sea-cage cultured Pacific bluefin tuna naturally infected with blood flukes from genus Cardicola (Trematoda: Aporocotylidae).

Pacific bluefin tuna (PBT), Thunnus orientalis, due to its high average price on the market is an economically valuable fish species. Infections by blood flukes from the genus Cardicola (Trematoda: Aporocotylidae) represent a growing concern for the cage culture of bluefin tuna in Japan, Australia and Southern Europe. The accumulation of numerous Cardicola eggs in the fish gills causes severe pathology that has been linked to mortality in PBT juveniles up to one year old. The only effective treatment used to mitigate the infection is the oral administration of the antihelminthic drug praziquan

Discovery of intermediate hosts for two species of blood flukes Cardicola orientalis and Cardicola forsteri (Trematoda: Aporocotylidae) infecting Pacific bluefin tuna in Japan.

Fish blood flukes (Aporocotylidae) are important pathogens of farmed finfish around the world. Among them, Cardicola spp. infecting farmed tuna are considered to be serious threats to tuna farming and have received tremendous attention. We conducted periodical samplings at a tuna farming site in Japan between January and May, 2015 to determine the life cycle of Cardicola spp. We collected over 4700 terebellid polychaetes from ropes, floats and frames of tuna culture cages and found nearly 400 infected worms. Sporocysts and cercariae found in Nicolea gracilibranchis were genetically identified

Incidence of three Kudoa spp., K. neothunni, K. hexapunctata, and K. thunni (Myxosporea: Multivalvulida), in Thunnus tunas distributed in the western Pacific Ocean.

A variety of tunas of the genus Thunnus are consumed daily in Japan as sliced raw fish (sashimi and sushi). The consumption of fresh sliced raw fish, i.e., unfrozen or uncooked, can sometimes cause food poisoning that is manifested by transient diarrhea and vomiting for a single day. One of the causes of this type of food poisoning has been identified as live Kudoa septempunctata (Myxosporea: Multivalvulida) in the olive flounder (Paralichthys olivaceus). Furthermore, raw slices of fresh tunas are highly suspected to be a possible causative fish of similar food poisoning in Japan. In the prese

Use of commercially available antimicrobial compounds for prevention of Listeria monocytogenes growth in ready-to-eat minced tuna and salmon roe during shelf life.

Listeria monocytogenes found in minced tuna and fish roe can cause listeriosis. These products are classified in category B according to the Codex Alimentarius Commission, i.e., ready-to-eat foods in which L. monocytogenes growth can occur. We investigated the effectiveness of nisin and other commercially available antimicrobial compounds (lysozyme, ε-polylysine, and chitosan) for prevention of L. monocytogenes growth during the expected shelf life of raw minced tuna and salmon roe products. Food samples inoculated with L. monocytogenes were incubated with each antimicrobial at 10°C for 7 days

Quantitative studies and taste reconstitution experiments of the sour and lingering mouthful orosensation in a debittered extract of traditional Japanese dried and fermented skipjack tuna (hongarebushi).

Hongarebushi, Japanese dried skipjack tuna and a high quality ingredient of Japanese dashi, was investigated for its taste active composition. The recent investigation focused on a debittered fish fraction, which revealed a strong umami and salt impact accompanied with a pleasant and pronounced sourness. Whereas the umami and salt tastes could be correlated to monosodium glutamate (MSG), ribonucleotides, and mineral salts, the pleasant sourness was not exclusively induced by organic acids. The essential compound imparting the sour orosensation, persistence, and mouthfulness of the debittered s

Anisakid larvae in the skipjack tuna Katsuwonus pelamis captured in Japanese waters: Two-year monitoring of infection levels after the outbreak of human anisakiasis in 2018.

In 2018, human anisakiasis caused by the ingestion of the skipjack tuna Katsuwonus pelamis occurred frequently in Japan. This may be attributable to a heavy infection of A. simplex (s.s.) in the host's muscle tissue. In this study, we investigate infection levels of anisakid L3 larvae in skipjack tuna captured in Japanese waters afterward (2019-2020) to contribute to predict and prevent the outbreak of human anisakiasis. A total of 476 larvae were detected from 78 out of 85 skipjack tuna captured at 14 stations of the Pacific and East China Sea. The present parasitological survey suggests that

Cytotoxicity evaluation and antioxidant enzyme expression related to heavy metals found in tuna by-products meal: An in vitro study in human and rat liver cell lines.

Heavy metals can accumulate in organisms via various pathways, including respiration, adsorption and ingestion. They are known to generate free radicals and induce oxidative and/or nitrosative stress with depletion of anti-oxidants. Tuna by-product meal (TBM) is rich in proteins and can, therefore, offer an attractive protein source for animals. This study was undertaken to assess the effects of metals present in TBM, namely cadmium (Cd), lead (Pb), and mercury (Hg), separately or in combination with oxidative stress, on cell viability. Three cell models: rat liver FTO2B, human hepatoma HepG2,

Colorimetric Analysis and Determination of Histamine in Samples of Yellowfin Tuna (Thunnus albacares) Marketed in Sardinia (Italy) by a Combination of Rapid Screening Methods and LC-MS/MS.

The consumption of fishery products has been steadily increasing in recent decades. Among the quantitatively more important species, the yellowfin tuna (Thunnus albacares), is one of the main at-risk species as regards the possibility to present important levels of histamine and to be associated with the so-called “Scombroid Fish Poisoning”. The main aim of the present study was to evaluate the colorimetric parameters, the occurrence, and the quantification of histamine contamination in yellowfin tuna samples marketed in Sardinia (Italy) by a combination of rapid screening and official control
