SARS-CoV-2 is associated with changes in brain structure in UK Biobank.

There is strong evidence of brain-related abnormalities in COVID-191-13. However, it remains unknown whether the impact of SARS-CoV-2 infection can be detected in milder cases, and whether this can reveal possible mechanisms contributing to brain pathology. Here we investigated brain changes in 785 participants of UK Biobank (aged 51-81 years) who were imaged twice using magnetic resonance imaging, including 401 cases who tested positive for infection with SARS-CoV-2 between their two scans-with 141 days on average separating their diagnosis and the second scan-as well as 384 controls. The ava

Alcohol use and cardiometabolic risk in the UK Biobank: A Mendelian randomization study.

Observational studies suggest alcohol use promotes the development of some adverse cardiometabolic traits but protects against others including outcomes related to coronary artery disease. We used Mendelian randomization (MR) to explore causal relationships between the degree of alcohol consumption and several cardiometabolic traits in the UK Biobank. Using the well-established ADH1B Arg47His variant (rs1229984) and up to 24 additional SNPs recently found to be associated with alcohol consumption in an independent dataset as instruments, we conducted two-stage least squares and inverse weighte

Association of SBP and BMI with cognitive and structural brain phenotypes in UK Biobank.

OBJECTIVE: To test for associations between SBP and BMI, with domain-specific cognitive abilities and examine which brain structural phenotypes mediate those associations. METHODS: Using cross-sectional UK Biobank data (final N = 28 412), we examined SBP/BMI vs. cognitive test scores of pairs-matching, matrix completion, trail making test A/B, digit symbol substitution, verbal-numerical reasoning, tower rearranging and simple reaction time. We adjusted for potential confounders of age, sex, deprivation, medication, apolipoprotein e4 genotype, smoking, population stratification and genotypic a

Lipoprotein(a) in Disaggregated South Asian Groups: An Analysis of MASALA and the UK Biobank.

BACKGROUND: South Asian individuals have a high prevalence of elevated lipoprotein(a) [Lp(a)], but little is known about the variation between disaggregated South Asian subgroups. OBJECTIVES: The authors aimed to evaluate Lp(a) levels, nongenetic influences, and the association with incident atherosclerotic cardiovascular disease (ASCVD) in South Asian populations using data from the Meditators of Atherosclerosis in South Asians Living in America (MASALA) and the UK Biobank (UKB) studies. METHODS: Lp(a) levels were assessed by South Asian country of origin and by state among Indian-origin pa

Multi-omics integration predicts the incidence of 17 diseases in the UK Biobank.

Multi-omics technologies, such as metabolomics and proteomics, offer deep molecular perspectives that could enhance risk prediction, but large-scale studies integrating both are scarce. Here we show the predictive values of these two omics across 17 incident diseases in 23,776 UK Biobank participants with complete baseline for 159 NMR-based metabolites and 2,923 Olink affinity-based proteins. We found that adding omics data significantly improved risk prediction for all 17 diseases compared to clinical predictors alone. Proteomics-only models generally outperformed metabolomics-only models for

Major depressive disorder (MDD) and the risks of mortality and hospitalization in the UK Biobank.

BACKGROUND: MDD is a leading cause of disability worldwide, yet its associations with mortality and hospitalization remain unclear. We aimed to quantify the risks of cause-specific mortality and hospitalization associated with a lifetime history of MDD, while accounting for potential reverse causality and residual confounding. METHODS: We conducted a retrospective cohort study using data from 113,196 UK Biobank participants (62,822 women and 50,374 men), aged 40–69 years at recruitment, who were assessed between 2008 and 2010. Participants with pre-existing physical health conditions or mental

Polygenic dissection of treatment-resistant depression with proxy phenotypes in the UK Biobank.

BACKGROUND: Treatment-resistant depression (TRD) affects one-third of major depressive disorder (MDD) patients. Previous pharmacogenetic studies suggest genetic variation may influence medication response but findings are heterogeneous. We conducted a comprehensive genetic investigation using proxy TRD phenotypes (TRDp) that mirror the treatment options of MDD from UK Biobank primary care records. METHODS: Among 15,125 White British MDD patients, we identified TRDp with medication changes (switching or receiving multiple antidepressants [AD]); augmentation therapy (antipsychotics; mood stabil

The brief resilience scale: a genome-wide association study in the UK Biobank.

BACKGROUND: Some individuals exposed to traumatic stressors develop psychiatric disorders while others remain resilient. The Brief Resilience Scale (BRS) assesses the ability to "bounce back" from stress and is a widely used measure of trait resilience. We performed the first genome-wide association study of BRS in the UK Biobank (UKB). METHODS: Beginning 2022, a subset of UKB participants completed an on-line mental-health questionnaire that included the six-item BRS. BRS data and genome-wide typing and imputation were available for 124,774 participants of European ancestry. Genome-wide line

Serum lipid levels and risk of lymphoid malignancies in the UK Biobank study.

BACKGROUND: Abnormal circulating lipid levels have been suggested in relation to lymphoid malignancy (LM) risk. METHODS: We studied UK Biobank participants (n = 403,625) with serum data for cholesterol (total [TC], high-density lipoprotein [HDL], direct low-density lipoprotein [LDL]), triglycerides (TG), and apolipoproteins A1 and B (ApoA1, ApoB). We conducted principal component (PC) analysis and multivariate Cox regression models to estimate hazard ratio (HR) overall, by lipid-lowering drug use and follow-up interval. RESULTS: During an average of 10.5 years of follow-up, 3006 incident LMs

Chondrocalcinosis and the haemochromatosis-linked HFE C282Y homozygous variant in the UK Biobank.

OBJECTIVES: C282Y genetic homozygosity is the main cause of the iron-overload disorder haemochromatosis. Musculoskeletal pain and arthropathy are common in haemochromatosis, but less is known about chondrocalcinosis (cartilage calcification) with the C282Y variant, especially in the community. We assessed knee chondrocalcinosis in intelligent dual-energy X-ray absorptiometer (iDXA) images from UK Biobank volunteers by HFE genotype. METHODS: Data were from 236 European genetic ancestry C282Y homozygotes and 236 age, sex, and body mass index (BMI)-matched controls with no C282Y alleles (48-80 y

Effects of vitamin D on COVID-19 risk and hospitalisation in the UK biobank.

BACKGROUND: Vitamin D (VitD) plays an important role in immune modulation. VitD deficiency is associated with increased susceptibility to acute respiratory syndrome as observed in COVID-19. We evaluated potential associations between serum VitD levels and risk of COVID-19 infection and hospitalisation, within the overall and cancer populations. METHODS: We performed a nested case-control study within the UK biobank cohort, among all individuals with at least one serum VitD level measurement at baseline (2006-2010) and a COVID-19 polymerase chain reaction (PCR) results recorded, and individual

Insights into the genetic architecture of cerebellar lobules derived from the UK Biobank.

In this work we endeavor to further understand the genetic architecture of the cerebellum by examining the genetic underpinnings of the different cerebellar lob(ul)es, identifying their genetic relation to cortical and subcortical regions, as well as to psychiatric disorders, as well as traces of their evolutionary trajectories. We confirm the moderate heritability of cerebellar volumes, and reveal genetic clustering and variability across their different substructures, which warranted a detailed analysis using this higher structural resolution. We replicated known genetic correlations with se

Discovery of runs-of-homozygosity diplotype clusters and their associations with diseases in UK Biobank.

Runs-of-homozygosity (ROH) segments, contiguous homozygous regions in a genome were traditionally linked to families and inbred populations. However, a growing literature suggests that ROHs are ubiquitous in outbred populations. Still, most existing genetic studies of ROH in populations are limited to aggregated ROH content across the genome, which does not offer the resolution for mapping causal loci. This limitation is mainly due to a lack of methods for the efficient identification of shared ROH diplotypes. Here, we present a new method, ROH-DICE (runs-of-homozygous diplotype cluster enumer

Multimorbidity and the risk of malnutrition, frailty and sarcopenia in adults with cancer in the UK Biobank.

BACKGROUND: Malnutrition, sarcopenia and frailty are distinct, albeit interrelated, conditions associated with adverse outcomes in adults with cancer, but whether they relate to multimorbidity, which affects up to 90% of people with cancer, is unknown. This study investigated the relationship between multimorbidity with malnutrition, sarcopenia and frailty in adults with cancer from the UK Biobank. METHODS: This was a cross-sectional study including 4122 adults with cancer (mean [SD] age 59.8 [7.1] years, 50.7% female). Malnutrition was determined using the Global Leadership Initiative on Mal

An epidemiological study of season of birth, mental health, and neuroimaging in the UK Biobank.

Environmental exposures during the perinatal period are known to have a long-term effect on adult physical and mental health. One such influential environmental exposure is the time of year of birth which affects the amount of daylight, nutrients, and viral load that an individual is exposed to within this key developmental period. Here, we investigate associations between season of birth (seasonality), four mental health traits (n = 137,588) and multi-modal neuroimaging measures (n = 33,212) within the UK Biobank. Summer births were associated with probable recurrent Major Depressive Disorder

Amount and intensity of physical activity and risk of incident cancer in the UK Biobank.

IMPORTANCE: The influence of total daily and light intensity activity on cancer risk remains unclear, as most existing knowledge is drawn from studies relying on self-reported leisure-time activities of moderate-vigorous intensity. OBJECTIVE: To investigate associations between total daily activity, including step counts, and activity intensity on incident cancer risk. DESIGN SETTING AND PARTICIPANTS: Prospective analysis of cancer-free UK Biobank participants who wore accelerometers for 7-days (between 2013-2015), followed for cancer incidence through national registries (mean follow-up 5.8
