Current state and novel outlook on prevention and treatment of rising antibiotic resistance in urinary tract infections.

Antibiotic-resistant bacteria are currently an important public health concern posing a serious threat due to their resistance to the current arsenal of antibiotics. Uropathogens Escherichia coli (UPEC), Proteus mirabilis, Klebsiella pneumoniae and Enterococcus faecalis, antibiotic-resistant gram-negative bacteria, cause serious cases of prolonged UTIs, increasing healthcare costs and potentially even leading to the death of an affected patient. This review discusses current knowledge about the increasing resistance to currently recommended antibiotics for UTI therapy, as well as novel therape

Cranberry-Derived Proanthocyanidins Potentiate β-Lactam Antibiotics against Resistant Bacteria.

The emergence and spread of extended-spectrum β-lactamases (ESBLs), metallo-β-lactamases (MBLs), or variant low-affinity penicillin-binding proteins (PBPs) pose a major threat to our ability to treat bacterial infection using β-lactam antibiotics. Although combinations of β-lactamase inhibitors with β-lactam agents have been clinically successful, there are no MBL inhibitors in current therapeutic use. Furthermore, recent clinical use of new-generation cephalosporins targeting PBP2a, an altered PBP, has led to the emergence of resistance to these antimicrobial agents. Previous work shows that

Comparative in vitro fermentations of cranberry and grape seed polyphenols with colonic microbiota.

In this study, we have assessed the phenolic metabolism of a cranberry extract by microbiota obtained from the ascending colon and descending colon compartments of a dynamic gastrointestinal simulator (SHIME). For comparison, parallel fermentations with a grape seed extract were carried out. Extracts were used directly without previous intestinal digestion. Among the 60 phenolic compounds targeted, our results confirmed the formation of phenylacetic, phenylpropionic and benzoic acids as well as phenols such as catechol and its derivatives from the action of colonic microbiota on cranberry poly

Inhibition of proliferation by agricultural plant extracts in seven human adult T-cell leukaemia (ATL)-related cell lines.

Adult T-cell leukaemia (ATL) is caused by human T-cell leukaemia virus type I (HTLV-I) infection and is resistant to conventional chemotherapy. We evaluated the inhibitory effects of agricultural plants on the proliferation of seven ATL-related human leukaemia cells, using three ATL cell lines (ED, Su9T01 and S1T), two human T-cell lines transformed by HTLV-I infection (HUT-102 and MT-2) and two HTLV-I-negative human T-cell acute lymphoblastic leukaemia cell lines (Jurkat and MOLT-4). A total of 52 samples of 80% ethanol extracts obtained from 30 types of agricultural plants were examined. On

Species diversity of fungal endophytes across a stress gradient for plants.

Foliar fungal endophytes are one of the most diverse guilds of symbiotic fungi found in the photosynthetic tissues of every plant lineage, but it is unclear how plant environments and leaf resource availability shape their diversity. We explored correlations between leaf nutrient availability and endophyte diversity among Pinus muricata and Vaccinium ovatum plants growing across a soil nutrient gradient spanning a series of coastal terraces in Mendocino, California. Endophyte richness decreased in plants with higher leaf nitrogen-to-phosphorus ratios for both host species, but increased with s

Metabolic Activity of Anthocyanin Extracts Loaded into Non-ionic Niosomes in Diet-Induced Obese Mice.

PURPOSE: Anthocyanins (ACNs) are polyphenols that might reduce pathological processes associated with type 2 diabetes mellitus and other chronic diseases, but their bioavailability is still controversial. In this study, the metabolic activity of oral delivery of ACN-loaded niosomes was investigated and evaluated in a diet-induced obesity (DIO) mice model. METHODS: ACNs extracted from Vaccinium Meridionale by the supercritical fluid extraction method were loaded in niosomes. The niosomal formulation was physically characterized and further administrated in drinking water to obese, insulin resi

Impact of Moringa oleifera Leaf Powder Addition on the Properties of Cranberry‑Based Gummies and Marshmallows With Low-Glycemic Sweeteners.

Traditional sweets are associated with dental caries, high blood glucose, and obesity. This study evaluated the impact of incorporating Moringa oleifera leaf powder (MOLP) into soft candies (gummies and marshmallows) formulated with a fixed base matrix of cranberry juice and low-glycemic, noncariogenic sweeteners (isomaltulose, oligofructose, and tagatose). MOLP was incorporated at 0%, 2.5%, 5.0%, and 7.5% while keeping the proportions of cranberry juice and sweeteners constant, enabling isolation of the specific contribution of MOLP. The increase in MOLP levels produced statistically signific

Proteomic Profiling Informs Mechanisms of Esophageal Adenocarcinoma Inhibition by Cranberry Proanthocyanidins.

Cranberry proanthocyanidins (CPACs) exert potent antiinflammatory and antibacterial activities in humans and anticancer effects in preclinical models, including those targeting esophageal adenocarcinoma (EAC). This study applied proteomic profiling to investigate CPACs' inhibitory effects on reflux-induced EAC in a rat model. Tandem mass spectrometry was applied to protein isolated from water-, CPAC-, and reflux-exposed esophagi with and without CPAC treatment. Differentially expressed proteins were identified, followed by enrichment analyses to assess CPACs' capacity to ameliorate reflux-indu

The cranberry and the urinary tract.

Cranberry products have been heralded as natural treatments for urinary tract infections (UTIs) and have been widely used for this purpose. Current evidence favours an antibacterial role for the cranberry's natural polyphenols or tannins. Although limited species- and strain-specific direct inhibition has been determined in vitro, it has been suggested that a key mechanism of inhibition, especially for the abundant uropathogenic E. coli, relies on anti-adhesion properties. Many studies of prevention have been complicated due to the enrollment of patients who have had complicated urinary tracts

A comprehensive overview of strategies to improve blueberry fruit quality.

Blueberry (Vaccinium spp.) is one of the most economically and nutritionally important crops worldwide due to its sensory attributes and high levels of bioactive compounds, which contribute to human health. Fruit quality is defined by a combination of biometric, chromatic, organoleptic, and textural aspects, as well as biochemical properties such as soluble sugar levels, organic acids, polyphenols, vitamin C content, and antioxidant capacity. In recent years, numerous sustainable preharvest methods have been developed to improve plant physiological development, increase nutrient-use efficiency

Natural α-Amylase Inhibitors from Medicinal Herbs: In Vitro Evaluation of Extracts Prepared with Food-Compatible Solvents.

Medicinal plants represent a promising source of bioactive compounds with potential antidiabetic activity, while the efficacy of plant extracts depends on both plant matrix and extraction conditions. This study aimed to systematically compare selected medicinal plants and extraction solvents to evaluate their impact on extracts' in vitro α-amylase inhibitory activity, total polyphenol content (TPC), antioxidant capacity, and antimicrobial properties. Extracts of sage (Salvia officinalis), blueberry leaf (Vaccinium myrtillus), nettle (Urtica dioica), wormwood (Artemisia absinthium), and green a

Ficus carica L. (Fig) promotes nerve regeneration in a mouse model of sciatic nerve crush.

Peripheral nerve injuries result in significant loss of motor and sensory function, and the slow rate of nerve regeneration can prolong recovery time. Thus, approaches that promote axonal regeneration are critical to improve the outcomes for patients with peripheral nerve injuries. In this study, we investigated the effects of Ficus carica L. (fig) and Vaccinium macrocarpon Ait. (cranberry), which are rich in phytochemicals with demonstrable and diverse medicinal properties, on nerve regeneration in a mouse model of sciatic nerve crush. Our investigation revealed that fig extract, but not cran

Screening of plant extracts for human tyrosinase inhibiting effects.

Screening for tyrosinase (TYR) inhibitors potentially useful for control of skin pigmentation has been hampered by the limited availability of human TYR. To overcome this hurdle, we have established human embryonic kidney (HEK293)-TYR cells that constitutively express human TYR. In the current study, we assayed human TYR inhibition activities of 50 plant extracts using the lysates of transformed HEK293-TYR cells. The strongest inhibition of human TYR was shown by the extract of Vaccinium bracteatum Thunberg, followed by the extract of Morus bombycis Koidzumi. The former extract did not inhibit

Evidence-Based Anti-Diabetic Properties of Plant from the Occitan Valleys of the Piedmont Alps.

Data on urban and rural diabetes prevalence ratios show a significantly lower presence of diabetes in rural areas. Several bioactive compounds of plant origin are known to exert anti-diabetic properties. Interestingly, most of them naturally occur in different plants present in mountainous areas and are linked to traditions of herbal use. This review will aim to evaluate the last 10 years of evidence-based data on the potential anti-diabetic properties of 9 plants used in the Piedmont Alps (North-Western Italy) and identified through an ethnobotanical approach, based on the Occitan language mi

Anti-Adhesion Activity of A2-type Proanthocyanidins (a Cranberry Major Component) on Uropathogenic E. coli and P. mirabilis Strains.

Urinary tract infections (UTIs) are relatively common in women and may be classified as uncomplicated or complicated, depending upon the urinary tract anatomy and physiology. Acute uncomplicated cystitis (AUC) occurs when urinary pathogens from the bowel or vagina colonize the periurethral mucosa and reach the bladder. The vast majority of episodes in healthy women involving the same bacterial strain that caused the initial infection are thought to be reinfections. About 90% of AUC are caused by uropathogenic Escherichia coli (UPEC), but Proteus mirabilis also plays an important role. Several

Updates to Recurrent Uncomplicated Urinary Tract Infections in Women: AUA/CUA/SUFU Guideline (2025).

PURPOSE: Our perceptions of recurrent UTI (rUTI) have evolved due to additional insights into rUTI pathophysiology, an appreciation for the adverse effects of repetitive antimicrobials ("collateral damage"), rising rates of bacterial antimicrobial resistance, and better reporting of the natural history of localized cystitis and rUTI. This document seeks to guide the evaluation and management of patients with rUTIs to prevent inappropriate antibiotic use, decrease the risk of antibiotic resistance, reduce adverse effects of antibiotics, provide guidance on strategies for rUTI prevention, and im
