Acute acalculous cholecystitis due to Vibrio cholerae.

The case of a 57-year-old woman admitted with symptoms and signs suggesting an intestinal infection caused by Vibrio cholerae, and who also developed a clinical picture compatible with acute cholecystitis, is presented. Cholera was diagnosed by examining a fresh sample of stools and cultures. An abdominal sonogram disclosed signs of acute acalculous cholecystitis. She underwent cholecystectomy, and cultures of a clear fluid and a "milky" sediment found within the gallbladder were also positive for V. cholerae. This microorganism was seen at the gallbladder mucosa microscopically. The strain wa

Lower extremity manifestations of Vibrio vulnificus infection.

Vibrio vulnificus is a potentially lethal marine bacterium that has not been previously described in podiatric literature. A review of the microorganism's characteristics, susceptible patient population, and lower extremity manifestations of infection is presented. V. vulnificus is found as part of the normal flora of the Gulf of Mexico, Atlantic, and Pacific coastal waters and is often isolated from the filter feeding shellfish of these regions. Its pathogenicity is generally reserved for the immunocompromised host, and is specifically related to disease states which exhibit high serum iron l

[Mexican phenotype and genotype Vibrio cholerae 01].

This paper presents the phenotypical and genotypical characterization of 26922 Vibrio cholerae 01 strains isolated in Mexico from 1991 to 1993. All strains isolated were El Tor biovar. Strains were sensitive to antibiotics excluding furazolidone, streptomycin and sulfisoxasole to which we found resistance in 97% and we are using this characteristic as epidemiological markers. We detected a marked change in frequency of Inaba serotype from 1991, when it was dominant, with 99.5%, until 1992 when Ogawa serotype turned to be dominant with 95% of isolates. All Vibrio cholerae 01 strains, except one

Infections caused by halophilic marine Vibrio bacteria.

OBJECTIVE: The authors reviewed patients who developed sepsis or soft tissue infections caused by marine Vibrio bacteria in Florida. SUMMARY BACKGROUND DATA: Marine Vibrio bacteria are the most common bacteria found in seawater. They are concentrated in marine animals that feed by filtration such as oysters and clams. These bacteria can cause gastroenteritis, sepsis, cellulitis leading to necrotizing soft tissue infection after exposure to seawater or consumption of raw seafood. METHODS: The authors received 182 systemic infections that occurred in Florida between January 1, 1979, and Decemb

Addressing the Challenges of Identifying Vibrio Species; Genomic and Phenotypic Analyses of Misidentified Vibrio vulnificus Strains Reveal Vibrio campbellii and a Potentially Novel V. rotiferianus-like Species in New Zealand Oysters.

The genus Vibrio contains over 150 species of aquatic origin, some of which are important foodborne pathogens, while others cause diseases in finfish, shellfish and/or coral. Accurate identification of these bacteria is important, but highly challenging given their taxonomic diversity. Here we used a range of methods to identify strains from New Zealand oysters initially identified as Vibrio vulnificus. The API20E kit either misidentified, or failed to identify the strains depending on the database used; comparisons of the 16S rRNA gene sequences could not distinguish them from over 50 exempla

Vibrio repeats (VXRs) play a role in horizontal dissemination of sedentary chromosomal integron-associated antibiotic resistance genes across Vibrio species.

Sedentary chromosomal integrons (SCIs) are large integrons that harbor extensive arrays of gene cassettes and constitute major reservoirs of bacterial genetic diversity. In Vibrio, SCI cassettes are flanked by species-associated attC repeats, here referred to as Vibrio species repeats (VXRs). Although SCIs have been proposed as ancestral sources of antibiotic resistance genes (ARGs), the extent to which SCI-associated ARGs are exchanged among Vibrio species remains poorly understood. To investigate the dynamics of SCI-associated ARGs, we analyzed 37,812 genomes representing 181 Vibrio species.

Identification of Vibrio metschnikovii and Vibrio injensis Isolated from Leachate Ponds: Characterization of Their Antibiotic Resistance and Virulence-Associated Genes.

This study aimed to evaluate the antibiotic resistance of 22 environmental Vibrio metschnikovii isolates and 1 Vibrio injensis isolate from landfill leachates in southwestern Colombia. Isolates were identified by Matrix-Assisted Laser Desorption/Ionization-Time-Of-Flight (MALDI-TOF), and 16S ribosomal RNA gene sequencing. Analysis of the susceptibility to six antibacterial agents by the Kirby-Bauer method showed susceptibility of all the isolates to ciprofloxacin and imipenem. We recorded resistance to beta-lactams and aminoglycosides, but no multidrug resistance was observed. The genome of on

Phage Endolysins as Potential Antimicrobials against Multidrug Resistant Vibrio alginolyticus and Vibrio parahaemolyticus: Current Status of Research and Challenges Ahead.

Vibrio alginolyticus and V. parahaemolyticus, the causative agents of Vibriosis in marine vertebrates and invertebrates, are also responsible for fatal illnesses such as gastroenteritis, septicemia, and necrotizing fasciitis in humans via the ingestion of contaminated seafood. Aquaculture farmers often rely on extensive prophylactic use of antibiotics in farmed fish to mitigate Vibrios and their biofilms. This has been postulated as being of serious concern in the escalation of antibiotic resistant Vibrios. For this reason, alternative strategies to combat aquaculture pathogens are in high dem

First detection of Vibrio vulnificus and other potentially pathogenic Vibrio species on different types of waste material in the Gulf of Gdańsk.

BACKGROUND: Vibrio bacteria, including pathogens like Vibrio vulnificus (V. vulnificus) and Vibrio cholerae (V. cholerae) (typical for tropical regions), are increasingly reported in the Baltic Sea, with rising infections linked to climate change. In our study, we examined the presence of Vibrio species on waste materials floating in the coastal zone. MATERIAL AND METHODS: Seventy-five waste items were collected. Samples were rinsed, homogenized, and cultured on CHROMagar Vibrio and thiosulfate-citrate-bile salts-sucrose (TCBS) agar. Vibrio isolates were tested for their tolerance to salinity

In situ dynamics of Vibrio parahaemolyticus and Vibrio vulnificus in water, sediment and triploid Crassostrea virginica oysters cultivated in floating gear.

AIMS: To understand spatial-temporal distribution of Vibrio parahaemolyticus and Vibrio vulnificus in triploid Crassostrea virginica in off-bottom aquaculture. METHODS AND RESULTS: Oysters, sediments and water were seasonally collected in Georgia, USA. V. parahaemolyticus and V. vulnificus were quantified with tlh/tdh/trh and vvhA genes, respectively. No tdh/trh genes were detected. Highest concentrations of tlh gene were observed in summer sediments > oysters > water (105 /g, 104 /g, 103 /ml). VvhA concentrations were similar in sediments and oysters but never exceeded ≥3 × 101 /ml in water.

Identification of a novel spätzle in Macrobrachium rosenbergii and its regulatory effects on the immune responses against Vibrio parahaemolyticus and Vibrio alginolyticus.

As a cytokine-like ligand, Spätzle activates the Toll signaling pathway through binding to Toll-like receptors (TLRs), thereby participating in the innate immune defense of crustaceans against diverse pathogens. A novel Spätzle (MrSpz5) was cloned from Macrobrachium rosenbergii in this study. The MrSpz5 cDNA is 1900 bp, comprising a 972-bp open reading frame (ORF) encoding 323 amino acids, a 662-bp 5' UTR, and a 266-bp 3' UTR. MrSpz5 was constitutively expressed in various tissues, with the highest expression in the gills. Knockdown of MrSpz5 via RNA interference in M. rosenbergii significantl

Vibrio parahaemolyticus quorum sensing controls phage VP882 transmission.

UNLABELLED: Quorum sensing is a cell-to-cell communication process bacteria use to orchestrate collective behaviors. Quorum sensing involves the production, release, and detection of extracellular signal molecules called autoinducers. Some temperate phages can monitor bacterial autoinducers, enabling them to track the abundance of potential host cells in the vicinity. Quorum-sensing-responsive phages can preferentially launch the transition from lysogeny to lytic replication at high cell density, presumably maximizing transmission. Once the phage lytic program is enacted, if nearby host cells

Lineage-specific defence systems of pandemic Vibrio cholerae.

Cholera remains a significant global health burden. The causative agent responsible for the ongoing cholera pandemic, which began in 1961, is the seventh pandemic El Tor (7PET) lineage of Vibrio cholerae. Over the past century, lineages of V. cholerae have been traced using phage typing schemes, DNA hybridization on microarrays and, more recently, comparative genomics enabled by next-generation sequencing. Such lineage tracing has provided essential insights into cholera transmission dynamics. Beyond their use as tools in typing schemes, phages have long been recognized as major players in cho

Synthesis and Assay of Vibrio Quorum Sensing Inhibitors.

Bacteria detect local population numbers using quorum sensing, a method of cell-cell communication broadly utilized to control bacterial behaviors. In Vibrio species, the master quorum sensing regulators LuxR/HapR control hundreds of quorum sensing genes, many of which influence virulence, metabolism, motility, and more. Thiophenesulfonamides are potent inhibitors of LuxR/HapR that bind the ligand pocket in these transcription factors and block downstream quorum sensing gene expression. This class of compounds served as the basis for the development of a set of simple, robust, and educational

Characterization of a Nonagglutinating Toxigenic Vibrio cholerae Isolate.

Toxigenic Vibrio cholerae serogroup O1 is the etiologic agent of the disease cholera, and strains of this serogroup are responsible for pandemics. A few other serogroups have been found to carry cholera toxin genes-most notably, O139, O75, and O141-and public health surveillance in the United States is focused on these four serogroups. A toxigenic isolate was recovered from a case of vibriosis from Texas in 2008. This isolate did not agglutinate with any of the four different serogroups' antisera (O1, O139, O75, or O141) routinely used in phenotypic testing and did not display a rough phenotyp

Bacteriophage ICP1: A Persistent Predator of Vibrio cholerae.

Bacteriophages or phages-viruses of bacteria-are abundant and considered to be highly diverse. Interestingly, a particular group of lytic Vibrio cholerae-specific phages (vibriophages) of the International Centre for Diarrheal Disease Research, Bangladesh cholera phage 1 (ICP1) lineage show high levels of genome conservation over large spans of time and geography, despite a constant coevolutionary arms race with their host. From a collection of 67 sequenced ICP1 isolates, mostly from clinical samples, we find these phages have mosaic genomes consisting of large, conserved modules disrupted by

Indole Inhibits ToxR Regulon Expression in Vibrio cholerae.

Indole is a degradation product of tryptophan that functions as a signaling molecule in many bacteria. This includes Vibrio cholerae, where indole was shown to regulate biofilm and type VI secretion in nontoxigenic environmental isolates. Indole is also produced by toxigenic V. cholerae strains in the human intestine, but its significance in the host is unknown. We investigated the effects of indole on toxigenic V. cholerae O1 El Tor during growth under virulence inducing conditions. The indole transcriptome was defined by RNA sequencing and showed widespread changes in the expression of genes

Atypical manifestation of Vibrio cholerae: fear the water!

BACKGROUND: In recent years, there has been an universal increase in number of reports of infections involving non-O1, non-O139 V. cholerae (NOVC). NOVC infection is only rarely complicated by septicaemia or invasive extra-intestinal infection. CASE SUMMARY: We report the first documented case of NOVC bacteraemia in a Belgian immunocompetent male after ingestion of water when paddling in a creek. CONCLUSION: Clinicians should include V. cholerae in their differential diagnosis in patients presenting with a self-limiting gastroenteritis after ingestion of raw and undercooked seafood or bathin

Laboratory Culturing Techniques and Maintenance of Vibrio cholerae.

Cholera is a severe diarrheal disease caused by the consumption of food or water contaminated with the aquatic gram-negative bacterium Vibrio cholerae. Infected hosts will experience vomiting and severe watery diarrhea and if not treated properly will ultimately succumb to death by dehydration. Due to the global prevalence and severity of cholera, V. cholerae has been extensively studied in both environmental and laboratory settings. Herein, we describe proper V. cholerae maintenance, in addition to classical and El Tor biotype culturing in a laboratory setting.

Fly Models of Vibrio cholerae Infection and Colonization.

Studies of Vibrio cholerae pathogenesis in the context of novel eukaryotic model systems have expanded our understanding of genes that underlie V. cholerae interactions with humans, as well as host organisms in the environment. These model systems have also helped uncover new functions for many gene products, revealing previously unknown virulence mechanisms. The Drosophila model for V. cholerae infection is a powerful tool for discovering new genetic pathways that govern bacterial physiology and colonization in the arthropod gastrointestinal tract. Assays to measure both virulence and coloniz
