Filamentous virions act as non-infectious interfering particles to modulate papillomavirus infection.

Genus beta (β) human papillomaviruses (HPVs) potentially contribute to the development of non-melanoma skin cancer. Yet, comparatively little is known about their biology. In particular, details about initial infection, i.e., host cell entry, remain mostly elusive. During initial characterization of β HPV5 pseudovirion (PsV) preparations, surprisingly large amounts of filamentous particles were found besides the prototypical icosahedral (T = 7) virions. Whether these filamentous particles actively contribute to or interfere with infectivity of the spherical viruses is unknown. Using a combinat

Neutralization of Crimean-Congo hemorrhagic fever pseudotyped virions with heavy chain antibodies.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a highly pathogenic tick-borne virus that causes severe hemorrhagic fever with high mortality rates in humans. No licensed vaccines or efficacious antiviral therapies are currently available. Here, we identified seven heavy chain antibodies targeting CCHFV Gc, which consist of heavy-chain variable domain (VHH) fused to human IgG1 Fc region (VHH-Fc). These VHH-Fc antibodies exhibited neutralizing activity against both recombinant vesicular stomatitis virus (VSV)-vectored CCHFV pseudoviruses and CCHFV transcription- and entry-competent virus-like

Detachment of Membrane Bound Virions by Competitive Ligand Binding Induced Receptor Depletion.

Multivalent receptor-mediated interactions between virions and a lipid membrane can be weakened using competitive nonpathogenic ligand binding. In particular, the subsequent binding of such ligands can induce detachment of bound virions, a phenomenon of crucial relevance for the development of new antiviral drugs. Focusing on the simian virus 40 (SV40) and recombinant cholera toxin B subunit (rCTB), and using (monosialotetrahexosyl)ganglioside (GM1) as their common receptor in a supported lipid bilayer (SLB), we present the first detailed investigation of this phenomenon by employing the quart

Exocytosis of Alphaherpesvirus Virions, Light Particles, and Glycoproteins Uses Constitutive Secretory Mechanisms.

UNLABELLED: Many molecular and cell biological details of the alphaherpesvirus assembly and egress pathway remain unclear. Recently we developed a live-cell fluorescence microscopy assay of pseudorabies virus (PRV) exocytosis, based on total internal reflection fluorescence (TIRF) microscopy and a virus-encoded pH-sensitive fluorescent probe. Here, we use this assay to distinguish three classes of viral exocytosis in a nonpolarized cell type: (i) trafficking of viral glycoproteins to the plasma membrane, (ii) exocytosis of viral light particles, and (iii) exocytosis of virions. We find that vi

SARS-CoV-2 membrane protein recruits PP2A to dephosphorylate the nucleocapsid and promote virion production.

BACKGROUND: The nucleocapsid (N) protein of coronavirus harbors a conserved serine/arginine (SR)-rich motif whose phosphorylation by GSK-3 is essential for viral transcription and replication. Our previous studies in Severe acute respiratory syndrome coronavirus 1 (SARS-CoV-1) and the JHM strain of mouse hepatitis virus revealed a phosphorylation-to-dephosphorylation transition during the viral life cycle, with newly synthesized N highly phosphorylated and virion-associated N hypophosphorylated. Here, we characterize this transition in SARS-CoV-2 and define its functional significance. METHOD

Marek's disease virus UL16 tegument protein is essential for secondary envelopment and infectious virion formation.

Marek's disease virus (MDV) is a highly oncogenic alphaherpesvirus whose productive infection and cell-to-cell spread rely on the formation of infectious mature virions, a process mediated by tegument proteins. UL16 is a conserved herpesviral tegument protein, in other herpesviruses, its deletion causes severe replication defects. However, its role in MDV remains unknown. In this study, a UL16-null MDV mutant was generated using BAC-based reverse genetics to investigate UL16's function. Deletion of UL16 completely abrogated the recovery of infectious virus from BAC DNA in chicken embryonic fib

Optimized dual-AAV base editor delivery system with enhanced editing efficiency and virion production titer.

Base editors (BEs) are a promising tool for precise base conversion in human cells and animals, while the adeno-associated virus (AAV) is the major vector for human gene therapy. However, the size of the DNA cassette required for BE expression exceeds the 4.7 kb packing capacity of the AAV vector, making dual-AAV approaches based on trans-splicing intein necessary. Even with this approach, current split DNA cassettes are still larger than the AAV packing limit, posing a challenge for cellular production of AAV. Moreover, some split strategies yield variable editing results and target coverage.

A single residue in the yellow fever virus envelope protein modulates virion architecture and antigenicity.

Yellow fever virus (YFV) is a re-emerging flavivirus that causes severe hepatic disease and mortality in humans. Despite being researched for over a century, the structure of YFV has remained elusive. Here we use a chimeric virus platform to resolve the first high resolution cryo-EM structures of YFV. Stark differences in particle morphology and homogeneity are observed between vaccine and virulent strains of YFV, and these are found to have significant implications on antibody recognition and neutralisation. We identify a single residue (R380) in the YFV17D envelope protein that stabilises th

Virion Associated Importin Subunit Beta-1 Augments Nuclear Import of HIV-1 Pre-Integration Complex.

Import of proviral genome from the cytoplasm to the nucleus is a decisive step in the HIV-1 infection cycle, regulating which can decide the fate of HIV-1 infectivity. Exploring the heterogeneity in the replication potential of viruses emerging from various producer cells, we compared the infectivity dynamics of viruses produced by CD4+ T lymphocytes, a cell type that supports the HIV-1 propagation, with those from astrocytes that allow for limited viral replication. We found the viruses emerging from these two cell types not only differed in their infectivity, but also in the host proteins th

Getah virus capsid protein undergoes co-condensation with viral genomic RNA to facilitate virion assembly.

Getah virus (GETV) belongs to the Alphavirus genus in the Togaviridae family and is a zoonotic arbovirus causing disease in both humans and animals. The capsid protein (CP) of GETV regulates the viral core assembly, but the mechanism underlying this process is poorly understood. In this study, we demonstrate that CP undergoes liquid-liquid phase separation (LLPS) with the GETV genome RNA (gRNA) in vitro and forms cytoplasmic puncta in cells. Two regions of GETV gRNA (nucleotides 1-4000 and 5000-8000) enhance CP droplet formation in vitro and the lysine-rich Link region of CP is essential for i

A Virion-Based Combination Vaccine Protects against Influenza and SARS-CoV-2 Disease in Mice.

Vaccines targeting SARS-CoV-2 have been shown to be highly effective; however, the breadth against emerging variants and the longevity of protection remains unclear. Postimmunization boosting has been shown to be beneficial for disease protection, and as new variants continue to emerge, periodic (and perhaps annual) vaccination will likely be recommended. New seasonal influenza virus vaccines currently need to be developed every year due to continual antigenic drift, an undertaking made possible by a robust global vaccine production and distribution infrastructure. To create a seasonal combina

AS03-adjuvanted H7N9 inactivated split virion vaccines induce cross-reactive and protective responses in ferrets.

Human infections with avian H7N9 subtype influenza viruses are a major public health concern and vaccines against H7N9 are urgently needed for pandemic preparedness. In early 2013, novel H7N9 influenza viruses emerged in China that caused about 1600 human cases of infection with a high associated case fatality rate. In this study, two H7N9 split virion vaccines with or without AS03 adjuvant were tested in the naive ferret model. Serological analyses demonstrated that homologous hemagglutination inhibition and microneutralization antibody titers were detectable in the ferrets after the first im

A simple method for measuring porcine circovirus 2 whole virion particles and standardizing vaccine formulation.

Porcine Circovirus 2 (PCV2) is involved in porcine circovirus-associated disease, that causes great economic losses to the livestock industry worldwide. Vaccination against PCV2 proved to be very effective in reducing disease occurrence and it is currently performed on a large scale. Starting from a previous model concerning Foot-and Mouth Disease Virus antigens, we developed a rapid and simple method to quantify PCV2 whole virion particles in inactivated vaccines. This procedure, based on sucrose gradient analysis and fluorometric evaluation of viral genomic content, allows for a better stand

Analysis of herpes simplex virion tegument ICP4 derived from infected cells and ICP4-expressing cells.

ICP4 is the major transcriptional regulatory protein of herpes simplex virus (HSV). It is expressed in infected cells with immediate early kinetics and is essential for viral growth. ICP4 is also a structural component of the virion tegument layer. Herpesviral tegument proteins exert regulatory functions important for takeover of the host cell. Tegument ICP4 has not been well characterized. We examined the ICP4 present in HSV-1 virions that were either derived from wild type infected cells or from ICP4-expressing (E5) cells infected with ICP4 deletion virus d120. Limited proteolysis demonstrat

Agnogene deletion in a novel pathogenic JC virus isolate impairs VP1 expression and virion production.

Infection of glial cells by the human polyomavirus JC (JCV) causes progressive multifocal leukoencephalopathy (PML). JCV Encephalopathy (JCVE) is a newly identified disease characterized by JCV infection of cortical pyramidal neurons. The virus JCVCPN associated with JCVE contains a unique 143 base pair deletion in the agnogene. Contrary to most JCV brain isolates, JCVCPN has an archetype-like regulatory region (RR) usually found in kidney strains. This provided us with the unique opportunity to determine for the first time how each of these regions contributed to the phenotype of JCVCPN. We c

Multiple ways of targeting APOBEC3-virion infectivity factor interactions for anti-HIV-1 drug development.

HIV-1 infections and the resulting AIDS pandemic remain a global challenge in the absence of a protective vaccine and because of rapid selection of drug-resistant viral variants in response to all currently available antiviral therapies. The development of new and highly active antiviral agents would greatly facilitate effective clinical management of HIV-1 infections and delay the onset of AIDS. Recent advances in our understanding of intracellular immunity conferred by host cytidine deaminases APOBEC3G (A3G) and APOBEC3F (A3F) and the mechanism by which the virally encoded virion infectivity

Association of vesicular stomatitis virus glycoprotein with virion membrane: characterization of the lipophilic tail fragment.

The proteolytic enzyme, thermolysin, degraded the external segment of the membrane glycoprotein of intact vesicular stomatitis (VS) virions but left behind a small nonglycosylated fragment, presumably embedded in the virion membrane. Other proteases generated membrane-associated glycoprotein fragments differing somewhat in molecular weight. The thermolysin-resistant, virion-associated fragment, which can be selectively solubilized by either Triton X-100 or chloroform/methanol, has a molecular weight of 5,200. Amino acid analysis of the glycoprotein fragment reveals a preponderance of hydrophob

Hepatitis C virus p7 regulates Core protein processing via signal peptide peptidase to promote virion assembly.

The hepatitis C virus (HCV) p7 protein is a viroporin essential for the production of infectious virus particles and has been implicated in regulating Core protein localization. Here, we identify a previously unrecognized role of p7 in modulating Core maturation through interaction with host signal peptide peptidase (SPP), a mechanism that remains intact even in ion-channel deficient mutants. Using a transient expression system, we show that p7 from genotype 1a (H77S) delays SPP-mediated cleavage of full-length Core (fl-Core) to mature Core (m-Core), while genotype 2a (JFH1) p7 exerts a weaker

Indolylurea SST21 Targets Turnip Mosaic Virus Coat Protein to Disrupt Virion Assembly and Activate Host Defense.

The turnip mosaic virus (TuMV) is an RNA virus that poses a significant threat to crops. The coat protein (CP) plays a crucial role in the viral life cycle, including mediating host infection and cell-to-cell movement, which makes it an ideal target for antiviral drug development. In this study, a series of structurally diverse synthetic indolylureas were screened against TuMV CP using high-throughput methods such as isothermal titration calorimetry (ITC) and microscale thermophoresis (MST). This led to the identification of compound SST21, which exhibits a high affinity for TuMV CP. Further e
