Meningococcal lipopolysaccharides: virulence factor and potential vaccine component.

Lipopolysaccharides (LPS) are surface components of the outer membrane of Neisseria meningitidis. Today, 12 different types of meningococcal LPS (immunotypes) are known, of which 3 are prevalent in the western world. The differences between these immunotypes are in the oligosaccharide part of the LPS molecule and consist of small differences in the oligosaccharide structure, the amount and location of phosphoethanolamine groups, and the degree of O acetylation of individual monosaccharides. Although the differences between the various immunotypes are small, they have a profound influence on th

Characterization of the role of putative Aeromonas caviae-specific virulence factor, flgB, in virulence and host-pathogen interactions.

Aeromonas caviae, Gram-negative bacteria ubiquitous in the environment, are an emerging human pathogen associated with various infectious diseases, particularly gastroenteritis. Despite recent studies demonstrating A. caviae is the most predominant Aeromonas species underlying human infection, A. caviae remains understudied, and no A. caviae-specific virulence factors associated with human disease have been identified. To identify A. caviae-specific putative virulence factors, we conducted comparative genomic analyses among clinical Aeromonas isolates (n = 431), which identified a variant of f

Virulence diversity among Porphyromonas gingivalis strains: a review of genetic factors.

BACKGROUND: Porphyromonas gingivalis (P. gingivalis) is a Gram-negative anaerobic bacterium and a keystone pathogen in chronic periodontitis. Beyond oral disease, it has been implicated in systemic conditions, including cardiovascular disease, type 2 diabetes, rheumatoid arthritis, and neurodegeneration. Marked strain-to-strain differences in tissue invasion, immune evasion, and pathogenicity suggest that virulence is shaped by complex genetic and regulatory interactions rather than a single determinant. OBJECTIVE: To summarize current knowledge of the genetic diversity of P. gingivalis virul

Streptococcus pneumoniae: Pathogenesis, virulence factors, antimicrobial resistance mechanisms, and therapeutic strategies.

BACKGROUND: Streptococcus pneumoniae is a Gram-positive, facultative anaerobic bacterium and a leading cause of community-acquired pneumonia, meningitis, bacteremia, and otitis media, responsible for an estimated 1.6 million deaths annually. Despite widespread polysaccharide and conjugate vaccine programs and decades of antibiotic therapy, pneumococcal disease continues to pose major public health challenges. These challenges are increasingly understood to reflect the organism's sophisticated capacity for adaptive recombination, allowing it to exploit every selective pressure applied through v

Analysis of virulence factors in extracellular vesicles secreted by Naegleria fowleri.

Naegleria fowleri is the etiological agent of primary amebic meningoencephalitis (PAM), a rapidly progressive acute and fulminant infection that affects the central nervous system, particularly of children and young adults, which has a mortality rate greater than 95%, and its symptomatologic similarity with other meningitis caused by virus or bacteria makes it difficult to make a quick and timely diagnosis that prevents the progression of the infection. It is necessary to know the antigenic determinants as well as the pathogenicity mechanisms of this amoeba to implement strategies that allow f

Assessing pH-dependent activities of virulence factors secreted by Candida albicans.

Candida albicans is an opportunistic pathogen that can thrive under adverse conditions including suboptimal pH, nutrient scarcity, and low levels of oxygen. Its pathogenicity is associated with the production of virulence factors such as extracellular hydrolytic enzymes and toxins. This study was aimed at determining the effect of external pH, substrate nature, and strain origin on protease, lipase, and hemolysin production. To achieve this objective, agar plate assays were performed at pH 5.0, 6.5, and 7.5 with substrates suitable for the detection of each family of enzymes. Moreover, the stu

Virulence factors and mechanisms of paediatric pneumonia caused by Enterococcus faecalis.

Paediatric pneumonia is a respiratory infection that affects infants and young children under the age of 3. This disease is the leading cause of infant and child mortality in developing countries because of the weak immune system of young children. The difficulty and length of time required to identify the pathogen and causative agent are the main reasons for this high mortality rate. In addition, the identification of certain causative agents is particularly important for the treatment of paediatric pneumonia. In this study, we explored the possible mechanisms by which pathogenic Enterococcus

Antibiotic resistance and virulence factors in lactobacilli: something to carefully consider.

Lactobacilli are a ubiquitous bacteria, that includes many species commonly found as part of the human microbiota, take part in the natural food fermentation processes, are used as probiotics, and in the food sector as starter cultures or bio-protectors. Their wide use is dictated by a long history of safe employ, which has allowed them to be classified as GRAS (General Recognized As Safe) microorganisms by the US Food and Drug Administration (FDA) and QPS (Qualified Presumption of Safety) by the European Food Safety Authority (EFSA, 2007; EFSA, 2021). Despite their classification as safe micr

Lytic Polysaccharide Monooxygenases as Chitin-Specific Virulence Factors in Crayfish Plague.

The oomycete pathogen Aphanomyces astaci, also known as "crayfish plague", is an obligate fungal-like parasite of freshwater crustaceans and is considered responsible for the ongoing decline of native European crayfish populations. A. astaci is thought to secrete a wide array of effectors and enzymes that facilitate infection, however their molecular mechanisms have been poorly characterized. Here, we report the identification of AA15 lytic polysaccharide monooxygenases (LPMOs) as a new group of secreted virulence factors in A. astaci. We show that this enzyme family has greatly expanded in A.

Perinatal Group B Streptococcal Infections: Virulence Factors, Immunity, and Prevention Strategies.

Group B streptococcus (GBS) or Streptococcus agalactiae is a β-hemolytic, Gram-positive bacterium that is a leading cause of neonatal infections. GBS commonly colonizes the lower gastrointestinal and genital tracts and, during pregnancy, neonates are at risk of infection. Although intrapartum antibiotic prophylaxis during labor and delivery has decreased the incidence of early-onset neonatal infection, these measures do not prevent ascending infection that can occur earlier in pregnancy leading to preterm births, stillbirths, or late-onset neonatal infections. Prevention of GBS infection in pr

Clindamycin Affects Group A Streptococcus Virulence Factors and Improves Clinical Outcome.

Group A Streptococcus (GAS) has acquired an arsenal of virulence factors, promoting life-threatening invasive infections such as necrotizing fasciitis. Current therapeutic regimens for necrotizing fasciitis include surgical debridement and treatment with cell wall-active antibiotics. Addition of clindamycin (CLI) is recommended, although clinical evidence is lacking. Reflecting the current clinical dilemma, an observational study showed that only 63% of the patients with severe invasive GAS infection received CLI. This work thus aimed to address whether CLI improves necrotizing fasciitis outco

Identification of New Virulence Factors and Vaccine Candidates for Yersinia pestis.

Earlier, we reported the identification of new virulence factors/mechanisms of Yersinia pestis using an in vivo signature-tagged mutagenesis (STM) screening approach. From this screen, the role of rbsA, which encodes an ATP-binding protein of ribose transport system, and vasK, an essential component of the type VI secretion system (T6SS), were evaluated in mouse models of plague and confirmed to be important during Y. pestis infection. However, many of the identified genes from the screen remained uncharacterized. In this study, in-frame deletion mutants of ypo0815, ypo2884, ypo3614-3168 (cyoA

Inactivation of the virulence factors from 2,3-butanediol-producing Klebsiella pneumoniae.

The microbiological production of 2,3-butanediol (2,3-BDO) has attracted considerable attention as an alternative way to produce high-value chemicals from renewable sources. Among the number of 2,3-BDO-producing microorganisms, Klebsiella pneumoniae has been studied most extensively and is known to produce large quantity of 2,3-BDO from a range of substrates. On the other hand, the pathogenic characteristics of the bacteria have limited its industrial applications. In this study, two major virulence traits, outer core LPS and fimbriae, were removed through homologous recombination from 2,3-BDO

Contribution of different pneumococcal virulence factors to experimental meningitis in mice.

BACKGROUND: Pneumococcal meningitis (PM) is a life-threatening disease with a high case-fatality rate and elevated risk for serious neurological sequelae. In this study, we investigated the contribution of three major virulence factors of Streptococcus pneumoniae, the capsule, pneumococcal surface protein A (PspA) and C (PspC), to the pathogenesis of experimental PM. METHODS: Mice were challenged by the intracranial route with the serotype 4 TIGR4 strain (wt) and three isogenic mutants devoid of PspA, PspC, and the capsule. Survival, bacterial counts, and brain histology were carried out. To

Transcriptome Analysis Reveals Cross-Kingdom Virulence Factors in Erwinia persicina Cp2.

Erwinia persicina is a well-documented plant pathogenic bacterium, causing soft rot in various plant hosts. There are rare previous reports that it is associated with animal diseases. In this study, we used an alfalfa root infection model and a mouse model via gavage administration of E. persicina strain Cp2. Strain Cp2 was pathogenic on both alfalfa and mice. On alfalfa, Cp2 primarily caused leaf spot and etiolation. Mice exhibited moderate lesions in their liver, stomach and kidney tissues, with inflammation in the liver and kidney after gavage. Furthermore, viable Cp2 cells were successfull

FunVFPred: Predicting fungal virulence factors using a unified representation learning model.

Fungal virulence factors (VFs) are proteins that support host colonization, tissue invasion, immune modulation, and disease progression. Identifying candidate VFs across pathogenic fungi can help characterize mechanisms underlying fungal-host interactions and prioritize proteins for experimental investigation. We developed FunVFPred, a machine learning framework that uses UniRep protein sequence embeddings, and multiple classifiers to predict candidate fungal VFs. Using experimentally validated VFs from human-pathogenic Candida species, the random forest model achieved 73.4% accuracy and a Mat

Therapeutic significance of surface-exposed virulence factors in Acinetobacter baumannii pathogenicity.

Acinetobacter baumannii is a rapidly evolving opportunistic pathogen that has developed strategies to resist multiple antimicrobials and evade the host immune system. Its global emergence as a multidrug-resistant (MDR) pathogen has severely limited treatment options and increased mortality in hospital settings. Virulence factors associated with the cell envelope and outer membrane vesicles play central roles in bacterial survival and pathogenicity. In this context, this review aims to provide a concise overview of the virulence apparatus present on the surface of A. baumannii and evaluate its

Microbial Trojan Horses: Virulence Factors as Key Players in Neurodegenerative Diseases.

Changes in population demographics indicate that the elderly population will reach 2.1 billion worldwide by 2050. In parallel, there will be an increase in neurodegenerative diseases such as Alzheimer's and Parkinson's. This review explores dysbiosis occurring in these pathologies and how virulence factors contribute to the worsening or development of clinical conditions, and it summarizes existing and potential ways to combat microorganisms related to these diseases. Microbiota imbalances can contribute to the progression of neurodegenerative diseases by increasing intestinal permeability, ex
