CADM1 and CADM2 Trigger Neuropathogenic Measles Virus-Mediated Membrane Fusion by Acting in cis.

Measles virus (MeV), an enveloped RNA virus in the family Paramyxoviridae, is still an important cause of childhood morbidity and mortality worldwide. MeV usually causes acute febrile illness with skin rash, but in rare cases persists in the brain, causing a progressive neurological disorder, subacute sclerosing panencephalitis (SSPE). The disease is fatal, and no effective therapy is currently available. Although transsynaptic cell-to-cell transmission is thought to account for MeV propagation in the brain, neurons do not express the known receptors for MeV. Recent studies have shown that hyp

Inhibition of bovine and ovine capripoxviruses (Lumpy skin disease virus and Sheeppox virus) by ivermectin occurs at different stages of propagation in vitro.

Capripoxvirus diseases are listed as reportable diseases by World Organization for Animal Health (OIE). Lumpy skin disease virus (LSDV) and sheeppox virus (SPPV), which can only be distinguished by molecular analysis, cause moderately, severe, or sometimes fatal infections in cattle and sheep. Even though vaccines are the most effective way to control the infection, their effectiveness may decrease in some cases. Therefore, it is significant to explore antiviral drugs against these diseases along with the vaccine. This study aimed to investigate the antiviral efficiency of ivermectin (IVM) at

Differential binding efficiency between the envelope protein of Japanese encephalitis virus variants and heparan sulfate on the cell surface.

Japanese encephalitis (JE) virus infects a number of host cells, either mosquitoes or vertebrates, in nature. The viral envelope (E) protein is known to interact with molecule(s) on the cell membrane during the early stage of virus infection. In this study, two sets of virus variants including T1P1-L4/T1P1-S1 and CJN-L1/CJN-S1 derived from two strains (T1P1 and CJN) of the JE virus were used to evaluate the effects of genomic variations on virus entry. Each set of virus variant (T1P1-L4/T1P1-S1 or CJN-L1/CJN-S1) possessed a single amino acid variation in the E protein. The variation of Glu/Lys

First Impressions-the Potential of Altering Initial Host-Virus Interactions for Rational Design of Herpesvirus Vaccine Vectors.

PURPOSE: The earliest host-virus interactions occur during virus attachment and entry into cells. These initial steps in the virus lifecycle influence the outcome of infection beyond delivery of the viral genome into the cell. Herpesviruses alter host signaling pathways and processes during attachment and entry to facilitate virus infection and modulate innate immune responses. We suggest in this review that understanding these early signaling events may inform the rational design of therapeutic and prevention strategies for herpesvirus infection, as well as the engineering of viral vectors fo

Rabies virus binding to an acetylcholine receptor alpha-subunit peptide.

The binding of 125I-labeled rabies virus to a synthetic peptide comprising residues 173-204 of the alpha 1-subunit of the nicotinic acetylcholine receptor was investigated. Binding of rabies virus to the receptor peptide was dependent on pH, could be competed with by unlabeled homologous virus particles, and was saturable. Synthetic peptides of snake venom, curaremimetic neurotoxins and of the structurally similar segment of the rabies virus glycoprotein, were effective in competing with labeled virus binding to the receptor peptide at micromolar concentrations. Similarly, synthetic peptides o

Suramin Inhibits Chikungunya Virus Replication by Interacting with Virions and Blocking the Early Steps of Infection.

Chikungunya virus (CHIKV) is a mosquito-transmitted alphavirus that can cause a debilitating disease that is primarily characterized by persistent joint pain. CHIKV has been emerging globally, while neither a vaccine nor antiviral medication is available. The anti-parasitic drug suramin was previously shown to inhibit CHIKV replication. In this study we aimed to obtain more detailed insight into its mechanism of action. We found that suramin interacts with virions and can inhibit virus binding to cells. It also appeared to inhibit post-attachment steps of the infection process, likely by preve

Evaluation of medicinal herbs for Anti-CHIKV activity.

Chikungunya, a mosquito-borne viral disease is now a global public health problem. In tropical countries such as India, periodic chikungunya outbreaks can occur due the high prevalence of the mosquito vector, circulation of virus and the high population density. To curtail the virus in outbreak situation, a ready to use drug for chikungunya is necessary. Using the literature mentioned plant extracts, we used four assays to screen and identify indigenous plants with CHIKV inhibitory activity. Our results showed that the aqueous extract of five plant extracts exhibited anti-CHIKV activity by inh

The Diverse Roles of Heparan Sulfate in RNA Virus Infections: Insights From Enterovirus A71, Severe Acute Respiratory Syndrome Coronavirus 2, and Chikungunya Virus.

Heparan sulfate (HS), a ubiquitously expressed glycosaminoglycan, functions as an attachment and/or internalization factor for diverse RNA and DNA viruses. Its broad viral attachment capacity arises from structural heterogeneity in sulfation patterns. This review examines HS interactions in enterovirus A71 (EV-A71), severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and chikungunya virus (CHIKV), emphasizing shared features and virus-specific distinctions. HS mediates viral attachment in all three, and additionally promotes SARS-CoV-2 internalization when host receptor angiotensin-c

The shape of pleomorphic virions determines resistance to cell-entry pressure.

Many enveloped animal viruses produce a variety of particle shapes, ranging from small spherical to long filamentous types. Characterization of how the shape of the virion affects infectivity has been difficult because the shape is only partially genetically encoded, and most pleomorphic virus structures have no selective advantage in vitro. Here, we apply virus fractionation using low-force sedimentation, as well as antibody neutralization coupled with RNAScope, single-particle membrane fusion experiments and stochastic simulations to evaluate the effects of differently shaped influenza A vir

Antiviral activity of cuprous oxide nanoparticles against Hepatitis C Virus in vitro.

Small molecular inhibitors in combination with or without interferon have improved sustained antiviral responses against Hepatitis C Virus (HCV) infection. Nonetheless, resistance to these inhibitors is expected to emerge rapidly due to the high mutation rate of the virus. Thus, new antiviral drugs, in combination with currently available therapies, are urgently needed to treat HCV infection. In the present study, we evaluated the antiviral efficacy of cuprous oxide nanoparticles (CO-NPs) against HCV in the HCVcc/Huh7.5.1 cell culture system. CO-NPs were able to significantly inhibit the infec

[Research into the Pathogenicity of Enterovirus 71].

Enterovirus 71 (EV71) is a major causative agent of hand, foot and mouth disease (HFMD). belongs to family Picornaviridae, genus Enterovirus, species A. EV71 infection usually affects subjects aged <5 years. HFMD caused by EV71 infection is usually mild in children. However, in some cases EV71 infection can lead to severe neurogenic disease and even death. EV71 infection has caused epidemic worldwide (especially in the Asia Pacific). HFMD caused by EV71 has become a major public-health prol lem across the Asia Pacific. In EV71 infection, the pathogenesis is determined by viral and host factor,

Enterovirus 71 uses cell surface heparan sulfate glycosaminoglycan as an attachment receptor.

Enterovirus 71 (EV-71) infections are usually associated with mild hand, foot, and mouth disease in young children but have been reported to cause severe neurological complications with high mortality rates. To date, four EV-71 receptors have been identified, but inhibition of these receptors by antagonists did not completely abolish EV-71 infection, implying that there is an as yet undiscovered receptor(s). Since EV-71 has a wide range of tissue tropisms, we hypothesize that EV-71 infections may be facilitated by using receptors that are widely expressed in all cell types, such as heparan sul

Transfer of Enteric Viruses Adenovirus and Coxsackievirus and Bacteriophage MS2 from Liquid to Human Skin.

Indirect exposure to waterborne viruses increases the risk of infection, especially among children with frequent hand-to-mouth contacts. Here, we quantified the transfer of one bacteriophage (MS2) and two enteric viruses (adenovirus and coxsackievirus) from liquid to skin. MS2, a commonly used enteric virus surrogate, was used to compare virus transfer rates in a volunteer trial to those obtained using human cadaver skin and synthetic skin. MS2 transfer to volunteer skin was similar to transfer to cadaver skin but significantly different from transfer to synthetic skin. The transfer of MS2, ad

The synergistic neutralization of Rift Valley fever virus by monoclonal antibodies to the envelope glycoproteins.

A panel of monoclonal antibodies (MAbs) mapping to different antigenic sites on the RVFV G1 and G2 proteins were used to examine the mechanisms involved in neutralization of the virus. Three types of synergistic neutralization of RVFV were observed on mixing various pairs of MAbs. Firstly, enhanced neutralization occurred for two MAb pairs that showed augmented binding for G2. These comprised a combination of a neutralizing MAb with a non-neutralizing antibody, as well as two antibodies which were non-neutralizing individually. In the second category, synergistic neutralization was observed be

A Computational Approach for Designing a Universal Epitope-Based Peptide Vaccine Against Nipah Virus.

Nipah virus (NiV) is highly pathogenic single-stranded negative sense RNA virus. It can cause severe encephalitis and respiratory disease in humans. In addition, NiV infects a large range of host including mammals. As a result of its higher zoonotic potential and pathogenicity for human, it has been rated as an alert in recent days. A therapeutic treatment or vaccines has become elusive to fight against this virus. In this study, the attachment (G) and fusion (F) glycoproteins of NiV, responsible for the viral attachment and entry to the host cell, were selected to develop epitope-based vaccin

Cellular receptors and host factors mediating orthoflavivirus entry.

Orthoflaviviruses are positive-sense, enveloped RNA viruses that include dengue virus (DENV), Zika virus (ZIKV), yellow fever virus (YFV), West Nile virus (WNV), and Japanese encephalitis virus (JEV). Viral entry is a multistep process governed by the envelope (E) glycoprotein, virion lipid composition, and host-cell molecules that act at distinct mechanistic stages. A major limitation of the current literature is that attachment factors, internalization receptors, immune-modulatory receptors, and membrane-associated cofactors are often collectively termed "receptors", which can obscure the st

Differential Use of the C-Type Lectins L-SIGN and DC-SIGN for Phlebovirus Endocytosis.

Bunyaviruses represent a growing threat to humans and livestock globally. The receptors, cellular factors and endocytic pathways used by these emerging pathogens to infect cells remain largely unidentified and poorly characterized. DC-SIGN is a C-type lectin highly expressed on dermal dendritic cells that has been found to act as an authentic entry receptor for many phleboviruses (Bunyaviridae), including Rift Valley fever virus (RVFV), Toscana virus (TOSV) and Uukuniemi virus (UUKV). We found that these phleboviruses can exploit another C-type lectin, L-SIGN, for infection. L-SIGN shares 77%

Mechanisms of pathogenic virus removal in a full-scale membrane bioreactor.

Four pathogenic virus removal mechanisms were investigated in a full-scale membrane bioreactor (MBR; nominal pore size 0.04 μm): (i) attachment of virus to mixed liquor solids; (ii) virus retention by a just backwashed membrane; (iii) virus retention by the membrane cake layer; and (iv) inactivation. We quantified adenovirus, norovirus genogroup II (GII), and F+ coliphage in the influent wastewater, the solid and liquid fractions of the mixed liquor, return flow, and permeate using quantitative PCR (adenovirus and norovirus GII) and infectivity assays (F+ coliphage). Exclusion of Superinfection or Enhancement of Superinfection in Pestiviruses-APPV Infection Is Not Dependent on ADAM17.

Some viruses can suppress superinfections of their host cells by related or different virus species. The phenomenon of superinfection exclusion can be caused by inhibiting virus attachment, receptor binding and entry, by replication interference, or competition for host cell resources. Blocking attachment and entry not only prevents unproductive double infections but also stops newly produced virions from re-entering the cell post-exocytosis. In this study, we investigated the exclusion of superinfections between the different pestivirus species. Bovine and porcine cells pre-infected with non-
