Dose-dependent compensation responses of the hypothalamic-pituitary-gonadal-liver axis of zebrafish exposed to the fungicide prochloraz.

Compensation responses and adaptability of hypothalamic-pituitary-gonadal (HPG) axis have been reported in fish exposed to model chemicals, however due to its importance in predictive toxicology further study was needed to elucidate details of the integrated responses to model chemicals. Transcriptional profiles of the hypothalamic-pituitary-gonadal (HPG) axis and concentrations of 17β-estradiol (E2) in plasma were measured in male and female zebrafish that had been exposed to one of seven concentrations of the fungicide, prochloraz: low (1, 3 or 10μg/L), medium (30 or 100μg/L) or high concent

Contrasting Starvation Strategies in Congeneric Pentatomids: A Predator (Arma chinensis) Versus a Pest (Halyomorpha halys).

Closely related insect species with different feeding habits may exhibit divergent physiological responses to starvation. Here, we compared the predatory Arma chinensis and the phytophagous pest Halyomorpha halys under identical starvation conditions. A. chinensis survived significantly longer than H. halys (females: 39 vs. 9 days; males: 29 vs. 7 days), with slower body weight decline. Starvation severely suppressed reproduction in both species, associated with arrested ovarian development and downregulation of vitellogenin (Vg) genes. In carbohydrate, lipid, and amino acid metabolism, A. chi

Candidatus Liberibacter Asiaticus-Mediated Manipulation of the Host Long Noncoding RNA Dclnc5 Enhances Fecundity in Diaphorina citri.

Candidatus Liberibacter asiaticus (CLas) enhances the fecundity of Diaphorina citri by manipulating D. citri long noncoding RNA 5 (Dclnc5), revealing an lncRNA-mediated regulatory role in pathogen-vector interactions. We found that CLas infection significantly increased D. citri fecundity and markedly upregulated vitellogenin (Vg) at both the transcript and protein levels. Ovary lncRNA-seq comparing CLas-positive (CLas+) and CLas-negative (CLas-) identified differentially expressed lncRNAs whose predicted targets were enriched in cellular signaling, lipid metabolism, and fat digestion and abso

Calcitonin-like diuretic hormone influences reproduction in adult female Rhodnius prolixus.

Calcitonin-like diuretic hormone (CT/DH) is a pleiotropic neuropeptide in insects that plays a role in regulating diuresis in Rhodnius prolixus, a blood-feeding insect and a vector of Chagas' disease. Here, we examine the role of this neuropeptide in another feeding-dependent event, that of reproduction in female R. prolixus. CT/DH-like immunoreactivity is present in the female reproductive system, including cells of the tropharium and pedicel of the ovariole, and nerve processes over the oviducts and calyx; tissues involved in egg maturation, ovulation, and egg laying. qPCR analysis reveals t

Review: Hemolymph lipoproteins of crustaceans and chelicerates.

The hemolymph of crustaceans (decapods, isopods, amphipods, copepods, branchiopods, cirripedes) and chelicerates (ticks, mites, spiders, scorpions, horseshoe crabs) contain female-specific and sex-independent lipoproteins in the high-density lipoprotein (HDL, density: 1.063-1.21 g/ml) and very high-density lipoprotein (VHDL, density>1.21 g/ml) classes. The first part of this review includes the history of the important developments in the study of hemolymph lipoproteins, synthesis by different tissues, important functions and synthesis/utilization by developing embryos. The sex-independent and

Projected late-century climate change alters reproductive gene expression pathways in the arbovirus vector Aedes aegypti.

Climate change is expected to alter the ecology, persistence, and geographic distribution of mosquito vectors, yet the molecular traits underlying vector responses to future environmental conditions remain insufficiently understood. Aedes aegypti, a major vector of arboviruses, depends on tightly regulated reproductive and lipid-transfer pathways that are linked to population replacement and persistence. Here, we experimentally evaluated whether projected late-century increases in temperature and atmospheric CO₂ affect the transcriptional regulation of lipophorin- and vitellogenin-associated g

Identification and ovarian developmental regulation of ribosomal protein S6 kinase in Macrobrachium nipponense.

Ribosomal protein S6 kinase (RPS6K) is a member of the Ser/Thr kinase family. To elucidate the role of RPS6K from Macrobrachium nipponense (MnRPS6K) in ovarian development, we cloned the full-length cDNA sequence of MnRPS6K gene using rapid amplification of cDNA ends (RACE)-PCR and analyzed its expression pattern using quantitative real-time PCR (qRT-PCR). Immunohistochemistry (IHC) was used to determine the subcellular localization of the MnRPS6K protein in ovarian tissues. Furthermore, the function of MnRPS6K in ovarian development was investigated via recombinant protein and neurotransmitte

Impact of acetyl tributyl citrate on gonadal sex differentiation and expression of biomarker genes for endocrine disruption in Japanese medaka.

Plasticizers are broadly classified as phthalate or nonphthalate. Recently, acetyl tributyl citrate (ATBC), an environmentally friendly nonphthalate plasticizer, was revealed to have the ability to disrupt thyroid hormone activity in fish species. Therefore, we aimed to assess whether ATBC exhibits any sex hormone (i.e., androgenic or estrogenic) activities. First, we examined the effects of ATBC on gonadal sex differentiation. Subsequently, we analyzed the different expression of biomarker genes that respond to endocrine-disrupting chemicals (EDCs) with sexual hormone activity in the liver. W

The chaperone BiP promotes dengue virus replication and mosquito vitellogenesis in Aedes aegypti.

Binding immunoglobulin protein (BiP, also known as GRP78), a chaperone and master regulator of the unfolded protein response (UPR) pathway, plays an essential role in several flavivirus infections, but its functional role in regulating dengue virus replication in the mosquito remains largely unknown. We here demonstrated the interaction between a dengue virus serotype 2 (DENV2) and BiP in Aedes aegypti and report the discovery of a novel functional role of BiP in mosquito vitellogenesis. Silencing Ae. aegypti BiP (AaBiP) expression resulted in the significant inhibition of DENV2 viral genome r

A case study: temporal trends of environmental stressors and reproductive health of smallmouth bass (Micropterus dolomieu) from a site in the Potomac River Watershed, Maryland, USA.

Decades of poor reproductive success and young-of-the-year survival, combined with adult mortality events, have led to a decline in the smallmouth bass (SMB; Micropterus dolomieu) population in sections of the Potomac River. Previous studies have identified numerous biologic and environmental stressors associated with negative effects on SMB health. To better understand the impact of these stressors, this study was conducted at the confluence of Antietam Creek and the Potomac River from 2013 to 2019 to identify temporal changes associated with SMB reproductive health. Surface water samples wer

Identification and structural characterization of the factors involved in vitellogenesis and its regulation in the African Osteoglossiforme of aquacultural interest Heterotis niloticus (Cuvier, 1829).

The African bonytongue (Heterotis niloticus) is an excellent candidate for fish farming because it has outstanding biological characteristics and zootechnical performances. However, the absence of sexual dimorphism does not favor its reproduction in captivity or the understanding of its reproductive behavior. Moreover, no molecular data related to its reproduction is yet available. This study therefore focuses on the structural identification of the different molecular actors of vitellogenesis expressed in the pituitary gland, the liver and the ovary of H. niloticus. A transcriptomic approach

Development of the Larval Amphibian Growth and Development Assay: Effects of benzophenone-2 exposure in Xenopus laevis from embryo to juvenile.

The Larval Amphibian Growth and Development Assay (LAGDA) is a globally harmonized chemical testing guideline developed by the U.S. Environmental Protection Agency in collaboration with Japan's Ministry of Environment to support risk assessment. The assay is employed as a higher tiered approach to evaluate effects of chronic chemical exposure throughout multiple life stages in a model amphibian species, Xenopus laevis. To evaluate the utility of the initial LAGDA design, the assay was performed using a mixed mode of action endocrine disrupting chemical, benzophenone-2 (BP-2). X. laevis embryos

Effect of 17beta-estradiol and progesterone on vitellogenesis in the spotted ray Torpedo marmorata Risso 1810 (Elasmobranchii: Torpediniformes): studies on females and on estrogen-treated males.

The influence of 17beta-estradiol (E(2)) on vertebrate vitellogenesis is well ascertained. The aim of the present paper is to study the involvement of E(2) and progesterone (P) in the induction and regulation of vitellogenesis in females and experimental E(2)-treated males of Torpedo marmorata. We analyzed females in various stages of the reproductive cycle and E(2) experimentally treated males. The presence of vitellogenin was investigated in the plasma and in the liver by western blot and immunohistochemistry; its site of synthesis was investigated by in situ hybridization. Effect of GnRH analogue on steroidogenesis in the teleost fish striped murrel (Channa striata).

The striped murrel (Channa striata) is a high-value food fish in Southeast Asia; however, captive breeding success has remained limited due to a lack of knowledge about its reproductive physiology. This study examined the impact of synthetic gonadotropin-releasing hormone analogue (sGnRHa) on the reproductive biology of females. Brooders (450-650 g) were administered sGnRHa at 0.4 mL/Kg for male and 0.6 mL/Kg for female brooders (n = 18) and 0.9% saline as a control (n = 6). To assess temporal steroidogenic responses, samples were collected at 0, 6, and 12 h post-injection. qPCR analysis was u

The VBP and a1/EBP leucine zipper factors bind overlapping subsets of avian retroviral long terminal repeat CCAAT/enhancer elements.

Two long terminal repeat (LTR) enhancer-binding proteins which may regulate high rates of avian leukosis virus (ALV) LTR-enhanced c-myc transcription during bursal lymphomagenesis have been identified (A. Ruddell, M. Linial, and M. Groudine, Mol. Cell. Biol. 9:5660-5668, 1989). The genes encoding the a1/EBP and a3/EBP binding factors were cloned by expression screening of a lambda gt11 cDNA library from chicken bursal lymphoma cells. The a1/EBP cDNA encodes a novel leucine zipper transcription factor (W. Bowers and A. Ruddell, J. Virol. 66:6578-6586, 1992). The partial a3/EBP cDNA clone encode

Role of Activin A in regulating ovarian development during vitellogenesis in Japanese eel (Anguilla japonica).

Activin A, a member of the TGF-β superfamily, plays pivotal roles in vertebrate reproductive regulation. This research investigated the biological functions and potential regulatory mechanisms of Activin A during vitellogenesis in Japanese eel. Analysis of the structural characteristics and evolutionary relationships confirmed that Japanese eel Activin A is highly conserved, providing a molecular basis for elucidating its role in ovarian development. Using a prokaryotic expression system, recombinant Japanese eel Activin A protein was successfully produced, and a series of in vitro functional

The impact of triclosan exposure during the growth and development period on ovarian differentiation in female zebrafish and its mechanisms.

Triclosan (TCS), a common antimicrobial agent, has been identified as an environmental estrogenic compound. Our previous studies have demonstrated that post-embryonic exposure to TCS disrupts the reproductive endocrine system in male zebrafish. Given that estrogenic compounds may differentially affect gonadal development in male and female fish, this study aims to investigate reproductive toxicity of post-embryonic TCS exposure in female zebrafish from endocrine perspective. Zebrafish were exposed to TCS from fertilization until adulthood (120 day post-fertilization). We measured growth parame

Myosuppressin signaling deficiency affects ovarian development via repression of 20-hydroxyecdysone biosynthesis in Grapholita molesta.

The steroid 20-hydroxyecdysone (20E) is crucial in regulating ovarian development. However, the neuropeptidergic mechanisms underlying ovarian development via 20E are underexplored. In this study, we investigated myosuppressin (MS) signaling in the dominant fruit pest Grapholita molesta and revealed that MS signaling is necessary for 20E biosynthesis during ovarian maturation. Pharmacological and molecular docking analyses confirmed that the GmMS mature peptide could activate its receptor GmMSR. Additionally, transcript expression analyses of GmMS and GmMSR showed different distribution patter
