Delivery of potential natural disease-modifying osteoarthritis drugs: Beyond polyphenols and flavonoids.

Osteoarthritis (OA) is a whole-joint disorder in which cartilage, synovium, subchondral bone, meniscus, and the infrapatellar fat pad deteriorate under chronic inflammatory and mechanical stress. To date, no disease-modifying osteoarthritis drug (DMOAD) has reached approval, largely due to single-pathway targeting, insufficient joint-specific exposure, and heterogeneous OA phenotypes. Polyphenols and flavonoids are well-known natural compounds and have been extensively studied for their potential applications in OA and drug delivery. This review looks beyond commonly known polyphenols and flav

Withaferin A-mediated apoptosis in breast cancer cells is associated with alterations in mitochondrial dynamics.

Withaferin A (WA), a steroidal lactone derived from a medicinal plant (Withania somnifera), inhibits cancer development in transgenic and chemically-induced rodent models of breast cancer but the underlying mechanism is not fully grasped. We have shown previously that WA treatment causes apoptotic cell death in human breast cancer cells that is preceded by inhibition of complex III of the mitochondrial electron transport chain. This study extends these observations to now demonstrate alterations in mitochondrial dynamics in WA-induced apoptosis. Assembly of complex III was decreased in MCF-7 a

Plant-derived neuroprotective compounds and nanoformulations targeting Parkinson's disease: a semi-systematic review of mechanisms and therapeutic potential.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by aggregates of α-synuclein and the degeneration of dopaminergic neurons in the substantia nigra. Current pharmaceutical therapies mainly alleviate symptoms without halting disease progression. Evidence suggests that traditional plant-based interventions may serve as supplementary therapies by targeting oxidative stress, mitochondrial dysfunction, neuroinflammation, and apoptosis. This review explores the neuroprotective properties of ten medicinal plants commonly used in traditional medicine: Bacopa monnieri,

Withaferin A induced impaired autophagy and unfolded protein response in human breast cancer cell-lines MCF-7 and MDA-MB-231.

The autophagy-lysosome pathway and the ubiquitin-proteasome systems are the two major routes for eukaryotic intracellular protein clearance. Cancerous cells often display elevated protein synthesis and byproduct disposal, thus, inhibition of the protein degradation pathways became an emerging approach for cancer therapy. The present study revealed that withaferin-A (WA), the biologically active withanolide derived from Withania somnifera, initially induced formation of autophagosomes in human breast cancer cell-lines, MCF-7 and MDA-MB-231. WA treatment elevated the levels of autophagic substra
