Transcriptome profiling of zebrafish optic fissure fusion.

Incomplete fusion of the optic fissure leads to ocular coloboma, a congenital eye defect that affects up to 7.5 per 10,000 births and accounts for up to 10 percent of childhood blindness. The molecular and cellular mechanisms that facilitate optic fissure fusion remain elusive. We have profiled global gene expression during optic fissure morphogenesis by transcriptome analysis of tissue dissected from the margins of the zebrafish optic fissure and the opposing dorsal retina before (32 hours post fertilisation, hpf), during (48 hpf) and after (56 hpf) optic fissure fusion. Differential expressi

nox2/cybb Deficiency Affects Zebrafish Retinotectal Connectivity.

NADPH oxidase (Nox)-derived reactive oxygen species (ROS) have been linked to neuronal polarity, axonal outgrowth, cerebellar development, regeneration of sensory axons, and neuroplasticity. However, the specific roles that individual Nox isoforms play during nervous system development in vivo remain unclear. To address this problem, we investigated the role of Nox activity in the development of retinotectal connections in zebrafish embryos. Zebrafish broadly express four nox genes (nox1, nox2/cybb, nox5, and duox) throughout the CNS during early development. Application of a pan-Nox inhibitor

Modelling acrylamide acute neurotoxicity in zebrafish larvae.

Acrylamide (ACR), a type-2 alkene, may lead to a synaptopathy characterized by ataxia, skeletal muscles weakness and numbness of the extremities in exposed human and laboratory animals. Currently, only the mildly affected patients undergo complete recovery, and identification of new molecules with therapeutic bioactivity against ACR acute neurotoxicity is urgently needed. Here, we have generated a zebrafish model for ACR neurotoxicity by exposing 5 days post-fertilization zebrafish larvae to 1 mM ACR for 3 days. Our results show that zebrafish mimics most of the pathophysiological processes de

Zebrafish heart regeneration: 15 years of discoveries.

Cardiovascular disease is the leading cause of death worldwide. Compared to other organs such as the liver, the adult human heart lacks the capacity to regenerate on a macroscopic scale after injury. As a result, myocardial infarctions are responsible for approximately half of all cardiovascular related deaths. In contrast, the zebrafish heart regenerates efficiently upon injury through robust myocardial proliferation. Therefore, deciphering the mechanisms that underlie the zebrafish heart's endogenous regenerative capacity represents an exciting avenue to identify novel therapeutic strategies

Zebrafish embryo model of Bartonella henselae infection.

Bartonella henselae (Bh) is an emerging zoonotic pathogen that has been associated with a variety of human diseases, including bacillary angiomatosis that is characterized by vasoproliferative tumor-like lesions on the skin of some immunosuppressed individuals. The study of Bh pathogenesis has been limited to in vitro cell culture systems due to the lack of an animal model. Therefore, we wanted to investigate whether the zebrafish embryo could be used to model human infection with Bh. Our data showed that Tg(fli1:egfp)(y1) zebrafish embryos supported a sustained Bh infection for 7 days with >1

Evaluating human cancer cell metastasis in zebrafish.

BACKGROUND: In vivo metastasis assays have traditionally been performed in mice, but the process is inefficient and costly. However, since zebrafish do not develop an adaptive immune system until 14 days post-fertilization, human cancer cells can survive and metastasize when transplanted into zebrafish larvae. Despite isolated reports, there has been no systematic evaluation of the robustness of this system to date. METHODS: Individual cell lines were stained with CM-Dil and injected into the perivitelline space of 2-day old zebrafish larvae. After 2-4 days fish were imaged using confocal mic

Zebrafish models of collagen VI-related myopathies.

Collagen VI is an integral part of the skeletal muscle extracellular matrix, providing mechanical stability and facilitating matrix-dependent cell signaling. Mutations in collagen VI result in either Ullrich congenital muscular dystrophy (UCMD) or Bethlem myopathy (BM), with UCMD being clinically more severe. Recent studies demonstrating increased apoptosis and abnormal mitochondrial function in Col6a1 knockout mice and in human myoblasts have provided the first mechanistic insights into the pathophysiology of these diseases. However, how loss of collagen VI causes mitochondrial dysfunction re

Antipsychotics produce locomotor impairment in larval zebrafish.

Zebrafish has been a favored vertebrate genetic model organism for studying developmental processes. It also holds a great potential for understanding the genetic basis of behavior and associated behavioral disorders. Despite such potential, their use in the study of behavior is greatly under-explored. It is well known that multiple classes of drugs used to treat psychiatric diseases produce extrapyramidal side (EPS) effects and consequent movement disorders in humans. The underlying molecular causes of these drug-induced movement disorders are poorly understood. Here we report that zebrafish

Molecular dissection of craniofacial development using zebrafish.

The zebrafish, Danio rerio, is a small, freshwater teleost that only began to be used as a vertebrate genetic model by the late George Streisinger in the early 1980s. The strengths of the zebrafish complement genetic studies in mice and embryological studies in avians. Its advantages include high fecundity, externally fertilized eggs and transparent embryos that can be easily manipulated, inexpensive maintenance, and the fact that large-scale mutagenesis screens can be performed. Here we review studies that have used the zebrafish as a model for craniofacial development. Lineage studies in zeb

Early life stage and genetic toxicity of stannous chloride on zebrafish embryos and adults: toxic effects of tin on zebrafish.

Humans are exposed to stannous chloride (SnCl(2)), known as tin chloride, present in packaged food, soft drinks, biocides, dentifrices, etc. Health effects in children exposed to tin and tin compounds have not been investigated yet. Therefore, we evaluated the possible teratogenic effects and genotoxic of SnCl(2) in zebrafish (Danio rerio) adults and their embryos. In the embryo-larval study, SnCl(2) showed embryo toxicity and developmental delay after exposure to the various concentrations of 10-250 μM for 120 h. Teratogenic effects including morphological malformations of the embryos and lar

Dicofol induces neurotoxicity in zebrafish (Danio rerio).

Dicofol (DCF), a widely used organochlorine pesticide, is classified as a persistent organic pollutant (POP). However, its toxicological profile and the underlying mechanisms of action in vertebrates remain poorly understood. In this study, exposure of zebrafish (Danio rerio) to sublethal concentrations of DCF led to notable behavioral abnormalities in both larvae and adults, effects that were accompanied by increased apoptosis in brain tissues. Additionally, molecular docking analysis identified the α3-subunit containing Na+/K+-ATPase (ATP1A3A) as a potential target of DCF; this finding was c

Regulatory mechanism of Pabpc1a during zebrafish vasculogenesis.

Vascular development begins with vasculogenesis during early embryogenesis. This process is controlled by a highly integrated, multi-layered molecular network. However, the role of post-transcriptional regulation in this context remains poorly understood. Cytoplasmic poly(A) binding protein 1a (Pabpc1a) is a key RNA-binding protein that regulates gene expression at the post-transcriptional level and participates in many physiological and pathological processes. Its role in vascular development, however, is still unclear. In this study, we first found that pabpc1a is highly expressed in endothe

Nanoplastics amplify 6PPD ocular toxicity in zebrafish.

Nanoplastics and tire-derived chemicals are ubiquitous co-pollutants in aquatic environments, originating from road runoff and posing potential risks to vertebrate development through enhanced bioavailability and synergistic toxicity. Polystyrene nanoplastics (PS) can adsorb hydrophobic organics like the antioxidant N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine (6PPD), acting as vectors that increase tissue accumulation and exacerbate oxidative stress, while 6PPD alone disrupts mitochondrial function and induces sublethal effects in exposed organisms. The vertebrate eye, with its direct e

Tnfrsf1b Protects Zebrafish Against Klebsiella pneumoniae Infection.

OBJECTIVE: Klebsiella (K). pneumoniae is an opportunistic pathogen that causes severe infections in the lungs, urinary tract, and liver, yet its pathogenesis remains unclear. Tnfrsf1b, a member of the tumor necrosis factor receptor superfamily, is associated with inflammation and tissue damage. The objective of this study was to investigate the functional role of tnfrsf1b in zebrafish during K. pneumoniae infection and tnfrsf1b should be considered a candidate target for further investigation. METHODS: A tnfrsf1b-knockout zebrafish line was generated using CRISPR/Cas9 technology. Both wild-ty

Immune Cell Regulation of Zebrafish Heart Regeneration.

Zebrafish possess a remarkable capacity to regenerate cardiac tissues after injury, offering a powerful model to dissect the cellular and molecular mechanisms driving heart regeneration. Immune cells play distinct and context-dependent roles during regeneration, from debris clearance and inflammation resolution to modulation of cell proliferation and fibrosis. Here, we review the distinct contributions of neutrophils, macrophages, and lymphoid cells during zebrafish heart regeneration, with a focus on their temporal coordination and regulatory signaling pathways. Understanding proregenerative

LOXHD1b knockout alters swimming behavior in zebrafish.

Since human inner ear hair cells do not regenerate, the current treatments of hereditary deafness depend on hearing aids or cochlear implant. However, uncovering the functions of genes responsible for hereditary hearing loss is not only useful for their diagnosis but also for developing therapies. The pathogenetic mechanism of human non-syndromic deafness DFNB77 without morphological defects in the inner year caused by LOXHD1 mutations is not fully understood. We introduced zebrafish because the lateral line hair cells are structurally and physiologically similar to the human inner ear hair ce

Maternal thyroid hormones affect zebrafish embryo development.

Thyroid hormones (THs), including thyroxine (T4) and triiodothyronine (T3), regulate vertebrate development, influencing critical processes during embryogenesis and larval stages. However, their maternal transfer and developmental roles in fish remain poorly understood. This study investigates maternal THs' impact on zebrafish development, focusing on the somatotropic axis, a key regulator of growth and metabolism. Female zebrafish were fed diets with 12.5 or 25 μg T4/g body weight/day for five days, while controls received vehicle-only food. Plasma TH levels were measured, and females were br

ap4b1 -/- zebrafish demonstrate morphological and motor abnormalities.

OBJECTIVE: Hereditary spastic paraplegia type 47 (SPG47) is caused by biallelic loss-of-function variants in the AP4B1 gene, leading to neurodevelopmental and progressive motor impairment. This study aimed to generate and characterize a zebrafish (Danio rerio) model of SPG47 to investigate the role of ap4b1 in neurodevelopment and motor function. METHODS: We employed CRISPR/Cas9 gene-editing to generate a stable ap4b1-/- zebrafish line. Behavioral, morphological, and motor function analyses were performed, including survival under stress conditions, spontaneous locomotor activity, light-dark

Direct male development in chromosomally ZZ zebrafish.

The genetics of sex determination varies across taxa, sometimes even within a species. Major domesticated strains of zebrafish (Danio rerio), including AB and TU, lack a strong genetic sex determining locus, but strains more recently derived from nature, like Nadia (NA), possess a ZZ male/ZW female chromosomal sex-determination system. AB fish pass through a juvenile ovary stage, forming oocytes that survive in fish that become females but die in fish that become males. To understand mechanisms of gonad development in NA zebrafish, we studied histology and single cell transcriptomics in develo
