In silico and structural investigation of sulfonamides targeting VraSR two component system in methicillin-resistant Staphylococcus aureus.

Drug-resistant Staphylococcus aureus strains are global health concerns. Several studies have shown that these strains can develop defences against cell wall antibiotics such as β-lactams, glycopeptides and daptomycin which target cell wall biosynthesis. The coordination of these responses have been associated with two component system (TCS) regulated by histidine kinase protein (VraS) and its cognate regulator VraR which influences the target DNA upon signal recognition. Computer-based screening methods, predictions and simulations have emerged as more efficient and quick ways to identify pro

The effect of lipidation and glycosylation on short cationic antimicrobial peptides.

The global health threat surrounding bacterial resistance has resulted in antibiotic researchers shifting their focus away from 'traditional' antibiotics and concentrating on other antimicrobial agents, including antimicrobial peptides. These low molecular weight "mini-proteins" exhibit broad-spectrum activity against bacteria, including multi-drug resistant strains, viruses, fungi and protozoa and constitute a major element of the innate-immune system of many multicellular organisms. Some naturally occurring antimicrobial peptides are lipidated and/or glycosylated and almost all antimicrobial

Impact of added copper, alone or in combination with chlortetracycline, on growth performance and antimicrobial resistance of fecal enterococci of weaned piglets.

Studies suggest a link between added copper (Cu) and co-selection of antimicrobial resistance (AMR) in Enterococcus spp., but data are inconsistent. This study aimed to assess the impact of added Cu, alone or with a feed-grade antimicrobial, on growth performance, transferable Cu resistance gene (tcrB) prevalence, abundance of tcrB in fecal community DNA, and AMR in fecal enterococci in weaned piglets. A total of 320 barrows (DNA 200 × 400, DNA Genetics) weaned at approximately 21 d of age with 7.4 kg (7.4 ± 0.06 kg) BW were used in a 28-d study. Piglets were fed a common non-medicated diet fo

A Six-Year Surveillance of Nasal Methicillin-Resistant Staphylococcus aureus Colonization on Intensive Care Unit Admission: Do We Need Screening?

Background: Methicillin-resistant Staphylococcus aureus (MRSA) colonization is a risk factor for potential staphylococcal infection and outbreaks. Although it is recommended to obtain a swab culture to detect nasal colonization its necessity in low-prevalence countries is debated. The aim of this study was to determine the prevalence of MRSA nasal colonization, the rate of invasive infection development, and the risk factors for invasive infections in patients admitted to the intensive care unit. Materials and Methods: This retrospective study included patients who were followed up in one of t

Comparative Effectiveness of Piperacillin-Tazobactam or Fluoroquinolones Versus Third-Generation Cephalosporins in Spontaneous Bacterial Peritonitis: A Systematic Review and Meta-Analysis.

Spontaneous bacterial peritonitis (SBP) remains a significant cause of morbidity and mortality among patients with cirrhosis. Third-generation cephalosporins (TGCs) have long been considered standard empiric therapy in major guidelines such as the British Society of Gastroenterology (BSG), the European Association for the Study of the Liver (EASL), and the American Association for the Study of Liver Diseases (AASLD). However, rising resistance patterns have led to increased use of piperacillin-tazobactam within the National Health Service (NHS). We aimed to evaluate the current evidence on the

Antimicrobial activity of a natural compound and analogs against multi-drug-resistant Gram-positive pathogens.

UNLABELLED: The increasing prevalence of methicillin-resistant Staphylococcus aureus (MRSA) has sparked global concern due to the dwindling availability of effective antibiotics. To increase our treatment options, researchers have investigated naturally occurring antimicrobial compounds and have identified MC21-A (C58), which has potent antimicrobial activity against MRSA. Recently, we have devised total synthesis schemes for C58 and its chloro-analog, C59. Here, we report that both compounds eradicate 90% of the 39 MRSA isolates tested [MIC90 and minimum bactericidal concentration (MBC90)] at

The avermectin, emamectin benzoate, kills gram-positive bacteria and targets the cell envelope of Bacillus subtilis.

The avermectin family of anthelmintics is largely considered to lack antibacterial activity against gram-positive and gram-negative bacteria. Here, we screened six avermectins (ivermectin, eprinomectin, doramectin, abamectin, selamectin, and emamectin benzoate) and a structurally related milbemycin (moxidectin) for antibacterial activity against a panel of representative gram-positive and gram-negative bacteria. We report that emamectin benzoate exhibited activity against several species of gram-positive bacteria, whereas selamectin was active against Staphylococcus aureus and Staphylococcus e

MRSA Profiles Reveal Age- and Gender-Specificity in a Tertiary Care Hospital: High Burden in ICU Elderly and Emerging Community Patterns in Youth.

Methicillin-resistant Staphylococcus aureus (MRSA) is a devastating global health concern. Hypervirulent strains are on the rise, causing morbidities and mortalities worldwide. In tertiary care hospitals, critically ill patients, those undergoing invasive procedures, and pediatric and geriatric patients are at risk. It is not fully clear how strains adapt and specialize in humans and emerge despite the well-established commonality of the S. aureus genome from humans and animals. This study investigates the influence of age-, gender-, and source-specific profiles (clinical, intensive care unit

Antimicrobial Susceptibility Profile of Bacterial Isolates Among Patients Admitted to the Neurosurgery Intensive Care Unit of a Tertiary Care Hospital: A Cross-Sectional Study.

BACKGROUND AND OBJECTIVES: Patients admitted to the neurosurgery intensive care unit (ICU) are often exposed to mechanical ventilation, invasive procedures, and polypharmacy. Prolonged hospital stays and multiple antibiotics facilitate the development of hospital-acquired infections (HAI). We planned this study to determine the bacteriological profile and antimicrobial susceptibility testing (AST) patterns among neurosurgery ICU patients. METHODS: This cross-sectional study was conducted from May 2024 to April 2025 at the Kalinga Institute of Medical Sciences (KIMS) in Bhubaneswar, India. Thi

Reduced glycopeptide and lipopeptide susceptibility in Staphylococcus aureus and the "seesaw effect": Taking advantage of the back door left open?

Methicillin-resistant S. aureus (MRSA) constitutes approximately 50% of clinical S. aureus isolates and is most commonly the result of production of a mutated pencillin-binding protein, PBP2a, which is able to carry out essential cell wall synthesis functions while maintaining a low-affinity for nearly all beta-lactam antibiotics. Decreased susceptibility to glycopeptides, typically considered first-line MRSA agents, has also been documented. Interestingly, among MRSA isolates, an increase in beta-lactam susceptibility has been documented in the presence of declining lipo- and glycopeptide sus
