Neuropeptides and astroglia in intracerebral hippocampal transplants: an immunohistochemical study in the rat.

The rat hippocampus and fascia dentata contain neurons and terminal fields that react immunohistochemically with antisera raised against the peptides somatostatin, cholecystokinin (CCK), and enkephalin. Neurons with overlapping distribution synthesize acetylcholinesterase (AChE) as shown histochemically after systemic administration of diisopropylfluorophosphate (DFP). We used these techniques to examine the organization of peptidergic neurons and fibers and AChE-positive neurons in transplants of late embryonic and early postnatal rat hippocampi inserted into the brain of immature and adult r

Injury-induced physiological events that may modulate gene expression in neurons and glia.

Damage to the brain triggers a host of reactive responses in neurons and glia which are seen at sites of focal injury as well as at sites that are at a distance from the injury. Although many of these responses have been studied extensively, the signals that initiate the different responses have not been fully characterized, and it is still not understood how focal injury affects neurons and glia in distant sites. The present review summarizes recent findings that suggest that physiological events that occur at the time of the injury or during the early postlesion period can play an important

Interaction between midazolam-induced anterograde amnesia and memory enhancement by treatments given hours later in hippocampus, entorrhinal cortex or posterior parietal cortex.

Rats were bilaterally implanted with indwelling cannulae in the CA1 region of the dorsal hippocampus, the entorrhinal cortex or the posterior parietal cortex. After recovery from surgery, they were trained in a one-trial step-down inhibitory avoidance task using a 0.3 mA footshock. The animals received i.p. 15 min before training either saline (1 ml/kg) or midazolam (1 mg/kg). Three hours after training they received, through the cannulae, infusions of saline, norepinephrine (0.3 microg/side), SKF38393 (7.5 microg/side), or 8-Br-cAMP (1.25 microg/side) into the brain regions mentioned. Animals

Posterior hippocampal sparing in Lewy body disorders with Alzheimer's copathology: An in vivo MRI study.

BACKGROUND: Lewy body disorders (LBD), encompassing Parkinson disease (PD), PD dementia (PDD), and dementia with Lewy bodies (DLB), are characterized by alpha-synuclein pathology but often are accompanied by Alzheimer's disease (AD) neuropathological change (ADNC). The medial temporal lobe (MTL) is a primary locus of tau accumulation and associated neurodegeneration in AD. However, it is unclear the extent to which AD copathology in LBD (LBD/AD+) contributes to MTL-specific patterns of degeneration. We employ a MTL subregional segmentation strategy of T1-weighted (T1w) MRI in biomarker-support

Magnetic resonance imaging and magnetic resonance spectroscopy in dementias.

This article reviews recent studies of magnetic resonance imaging and magnetic resonance spectroscopy in dementia, including Alzheimer's disease, frontotemporal dementia, dementia with Lewy bodies, idiopathic Parkinson's disease, Huntington's disease, and vascular dementia. Magnetic resonance imaging and magnetic resonance spectroscopy can detect structural alteration and biochemical abnormalities in the brain of demented subjects and may help in the differential diagnosis and early detection of affected individuals, monitoring disease progression, and evaluation of therapeutic effect.

Consolidation of object-discrimination memory is independent of the hippocampus in rats.

We examined whether retrograde amnesia would be more likely for object discriminations learned an hour before hippocampal damage than object discriminations learned days before. Specifically, rats were trained on two object-discrimination problems 72 h before surgery and another discrimination problem and the reversal of one of the previously learned problems 1 h before surgery. Importantly, novel procedures that minimized overtraining on the object discriminations were used to increase the possibility of the lesions causing amnesia. After either receiving sham or neurotoxic-induced hippocampa

Manual and automated analysis of atrophy patterns in dementia with Lewy bodies on MRI.

BACKGROUND: Dementia with Lewy bodies (DLB) is the second most common dementia type in patients older than 65 years. Its atrophy patterns remain unknown. Its similarities to Parkinson's disease and differences from Alzheimer's disease are subjects of current research. METHODS: The aim of our study was (i) to form a group of patients with DLB (and a control group) and create a 3D MRI data set (ii) to volumetrically analyze the entire brain in these groups, (iii) to evaluate visual and manual metric measurements of the innominate substance for real-time diagnosis, and (iv) to compare our groups

Experience-dependent changes in hippocampal spatial activity and hippocampal circuit function are disrupted in a rat model of Fragile X Syndrome.

BACKGROUND: Fragile X syndrome (FXS) is a common single gene cause of intellectual disability and autism spectrum disorder. Cognitive inflexibility is one of the hallmarks of FXS with affected individuals showing extreme difficulty adapting to novel or complex situations. To explore the neural correlates of this cognitive inflexibility, we used a rat model of FXS (Fmr1-/y). METHODS: We recorded from the CA1 in Fmr1-/y and WT littermates over six 10-min exploration sessions in a novel environment-three sessions per day (ITI 10 min). Our recordings yielded 288 and 246 putative pyramidal cells f

Place cells recorded in the parasubiculum of freely moving rats.

Previous studies have identified neurons in the hippocampus, subiculum, and entorhinal cortex which discharge as a function of the animal's location in the environment. In contrast, neurons in the postsubiculum and anterior thalamic nucleus discharge as a function of the animal's head direction in the horizontal plane, independent of its behavior and location in the environment. Because the parasubiculum (PaS) has extensive connections, either directly or indirectly, with these structures, it is centrally located to influence the neuronal activity in these areas. This study was therefore desig

The Bulb, the Brain and the Being: New Insights into Olfactory System Anatomy, Organization and Connectivity.

BACKGROUND/OBJECTIVES: Olfaction is in many ways the least understood sensory modality. Its organization and connectivity are still under debate. The aim of this study was to investigate the anatomy of the olfactory system by using a cadaver fiber dissection technique and in vivo tractography to attain a deeper understanding of the subcortical connectivity and organization. METHODS: Ten cerebral hemispheres were used in this study for white matter dissection according to Klingler's technique. Measurements of different cortical structures and interhemispheric symmetry were compared. Diffusion

Evidence supportive of a bacterial component in the etiology for Alzheimer's disease and for a temporal-spatial development of a pathogenic microbiome in the brain.

BACKGROUND: Over the last few decades, a growing body of evidence has suggested a role for various infectious agents in Alzheimer's disease (AD) pathogenesis. Despite diverse pathogens (virus, bacteria, fungi) being detected in AD subjects' brains, research has focused on individual pathogens and only a few studies investigated the hypothesis of a bacterial brain microbiome. We profiled the bacterial communities present in non-demented controls and AD subjects' brains. RESULTS: We obtained postmortem samples from the brains of 32 individual subjects, comprising 16 AD and 16 control age-matche

Synaptic Tau Seeding Precedes Tau Pathology in Human Alzheimer's Disease Brain.

Alzheimer's disease (AD) is defined by the presence of intraneuronal neurofibrillary tangles (NFTs) composed of hyperphosphorylated tau aggregates as well as extracellular amyloid-beta plaques. The presence and spread of tau pathology through the brain is classified by Braak stages and thought to correlate with the progression of AD. Several in vitro and in vivo studies have examined the ability of tau pathology to move from one neuron to the next, suggesting a "prion-like" spread of tau aggregates may be an underlying cause of Braak tau staging in AD. Using the HEK293 TauRD-P301S-CFP/YFP expr

Clinical, imaging, and pathological heterogeneity of the Alzheimer's disease syndrome.

With increasing knowledge of clinical in vivo biomarkers and the pathological intricacies of Alzheimer's disease (AD), nosology is evolving. Harmonized consensus criteria that emphasize prototypic illness continue to develop to achieve diagnostic clarity for treatment decisions and clinical trials. However, it is clear that AD is clinically heterogeneous in presentation and progression, demonstrating variable topographic distributions of atrophy and hypometabolism/hypoperfusion. AD furthermore often keeps company with other conditions that may further nuance clinical expression, such as synucl

Hippocampal activation during the recall of remote spatial memories in radial maze tasks.

Temporally graded retrograde amnesia is observed in human patients with medial temporal lobe lesions as well as in animal models of medial temporal lobe lesions. A time-limited role for these structures in memory recall has also been suggested by the observation that the rodent hippocampus and entorhinal cortex are activated during the retrieval of recent but not of remote memories. One notable exception is the recall of remote memories for platform locations in the water maze, which requires an intact hippocampus and results in hippocampal activation irrespective of the age of the memory. The

Estrogen-receptor occurrence in the female mouse brain: effects of maternal experience, ovariectomy, estrogen and anosmia.

Maternal behavior (ultrasound-induced pup-searching and retrieving) was studied in eight groups of female house mice with various hormonal backgrounds, experience with pups and function of the olfactory system. In their brains, estrogen receptor immunoreactive (ER-IR) cells were localized and quantified. All animals of all groups had ER-IR cells in a 'reliable subset' of brain areas, the medial preoptic area (MPOA) and ventromedial (VMH) and arcuate nucleus (ARH) of the hypothalamus. In another subset of brain areas, the anterior hypothalamic area (AHA) and cortical (CA) and medial (MA) amygda

Association of Circulating Vitamin D in Midlife With Increased Tau-PET Burden in Dementia-Free Adults.

BACKGROUND AND OBJECTIVES: Low circulating vitamin D in later-life has been associated with increased risk of cognitive impairment and clinical dementia. However, whether serum vitamin D in early mid-life is associated with neuroimaging markers of preclinical dementia is unknown. We aimed to determine the association between early mid-life serum vitamin D and subsequent tau- and amyloid- burden on brain-PET, in a cohort of dementia-free adults. METHODS: This was a prospective cohort study of Framingham Heart Study Generation 3 cohort participants who were dementia-free at time of PET, had ser

Association of C-reactive protein with brain micro- and macro-structure among older adult men.

BACKGROUND: The process by which aging leads to increased risk for Alzheimer's disease and related dementias is not entirely understood, but one hypothesized contributor is the occurrence of low-grade inflammation in older age. Associations between peripheral C-reactive protein (CRP), a marker of systemic inflammation, and brain structure have been widely studied, but fewer studies have examined CRP in relation to diffusion measures, particularly using newer techniques such as restriction spectrum imaging (RSI). In the current study, we examined how high sensitivity CRP (hsCRP) relates to diff

Comparison of volumes of brain areas in patients with bilateral early high-tension and normal-tension glaucoma in 7 Tesla MRI.

BACKGROUND: Glaucoma is an optic neuropathy characterized by progressive retinal ganglion cells degeneration and associated visual field defects. Although elevated intraocular pressure is a major risk factor, glaucoma can also develop in individuals with statistically normal intraocular pressure, implying alternative mechanisms such as neurotrophins deprivation and primary lateral geniculate nucleus injury. These mechanisms may suggest a neurodegenerative aspect of the disease. METHODS: In this study, we evaluated 15 patients with bilateral early-stage normal-tension glaucoma, 10 with bilater

Voxel- and surface-based morphometry in the cortical thickness and cortical and subcortical gray matter volume in patients with mild-to-moderate Alzheimer's disease.

AIM: This study aimed to investigate alterations in whole-brain cortical thickness (CT) and cortical and subcortical gray matter volume (GMV) in patients with Alzheimer's disease (AD) compared with healthy controls (HC) using voxel-based morphometry (VBM) and surface-based morphometry (SBM). Furthermore, we sought to develop a combined predictive model based on these neuroimaging markers and assess their potential clinical utility for the early detection and diagnosis of AD. METHODS: A total of 42 patients diagnosed with mild-to-moderate AD and 49 demographically matched HC were recruited for

CSF Levels of NPTX2 Are Associated With Less Brain Atrophy Over Time in Cognitively Unimpaired Individuals.

INTRODUCTION: Neuronal pentraxin 2 (NPTX2) is a synaptic protein involved in synaptic plasticity and regulation of neuronal excitability. Lower baseline cerebrospinal fluid (CSF) NPTX2 levels have been shown to be associated with an earlier onset of mild cognitive impairment (MCI), a pre-dementia syndrome, even after CSF Alzheimer's Disease (AD) biomarkers (amyloid beta (Aβ42/40), and phosphorylated tau (p-tau181)) were considered. To date, however, it is not known whether CSF NPTX2 levels among cognitively unimpaired individuals are associated with longitudinal brain atrophy. OBJECTIVE(S): E
