Metabolomics study of cat small intestine during the early stage of Toxoplasma gondii oocyst formation identifies potential biomarkers.

Toxoplasma gondii (T. gondii) is a zoonotic intracellular protozoan parasite that can invade, replicate and survive in almost all cells of warm-blooded animals. T. gondii infection threatens the life of the fetus or can cause morbidity in the infant. As the only definitive host of T. gondii, felids spread the pathogen mainly by forming oocysts in the small intestines and discharging the oocysts into the ambient environment, consequently polluting water, vegetables, and meat products. In this study, we used untargeted metabolomics technology to study the changes in metabolites that occurred dur

Characterization of rickettsial diseases in a hospital-based population in central Tunisia.

In Tunisia, 2 rickettsial groups, spotted fever group and typhus group, have been described since the beginning of the 20th century. Mediterranean spotted fever (MSF), also known as Boutonneuse fever, caused by Rickettsia conorii and transmitted by the dog tick Rhipicephalus sanguineus, is the most frequent rickettsial infection observed. Its seroprevalence in our region is 9% among blood donors and 23% in hospitalized febrile patients. Typhus group rickettsioses, caused by R. typhi and R. prowazekii, are less frequently reported than in the 1970s. Only sporadic cases of typhus were reported i

Beyond population size: long-term changes in free-roaming cat population structure during TNR implementation in Seoul.

BACKGROUND: Free-roaming cats (Felis catus) in urban environments are commonly managed through trap-neuter-return (TNR) programs, yet the long-term population effects of these measures remain inconsistently evaluated, particularly when considering both population size and structural dynamics. OBJECTIVES: This study aimed to assess long-term changes in free-roaming cat populations and population-structure patterns observed in the context of public TNR program in Seoul, Republic of Korea, using multiple demographic and structural indicators, specifically neutering rate, kitten proportion, and a

Development of a three-dimensional vitrification protocol for domestic cat cumulus-oocyte complexes and comparison with standard vitrification.

Three-dimensional (3D) systems may better mimic native tissue conditions and provide more physiologically relevant platforms for the study of cell function compared to traditional two-dimensional (2D) cultures. Cryopreservation of cells after encapsulation in 3D matrices is also likely to be a suitable alternative to traditional 2D approaches, as demonstrated with preantral ovarian follicles. Building on these findings, this study aimed to develop an alginate-based 3D vitrification protocol for domestic cat (Felis catus) cumulus-oocyte complexes (COCs). After determining the most suitable algi

The vaginal microbiota of healthy female cats.

The vaginal microbiota of the queen (i.e., female cat) has never been described using culture independent methods. The objectives of the present research were to describe the vaginal microbiota of healthy domestic shorthair queens using both 16S rRNA sequencing and culture, and to assess the effects of age, living environment, and reproductive season on its composition. Thirty queens undergoing elective ovariectomy were included in the study. The vaginal samples were collected just before surgery, from animals under general anaesthesia. Two consecutive mini-swabs were introduced in the queens'

Holobiont Urbanism: sampling urban beehives reveals cities' metagenomes.

BACKGROUND: Over half of the world's population lives in urban areas with, according to the United Nations, nearly 70% expected to live in cities by 2050. Our cities are built by and for humans, but are also complex, adaptive biological systems involving a diversity of other living species. The majority of these species are invisible and constitute the city's microbiome. Our design decisions for the built environment shape these invisible populations, and as inhabitants we interact with them on a constant basis. A growing body of evidence shows us that human health and well-being are dependent

Evidence on the prevalence of emerging and re-emerging tick- and flea-borne rickettsial agents in acute encephalitis syndrome endemic areas of northeast Uttar Pradesh, India.

Outbreaks of acute encephalitis syndrome (AES) with unknown aetiology are reported every year in Gorakhpur district, Uttar Pradesh, India, and Orientia tsutsugamushi, the rickettsial pathogen, responsible for scrub typhus has been attributed as the primary cause of AES problem. However, information on the prevalence of other rickettsial infections is lacking. Hence, this study was carried out to assess any occurrence of tick- and flea-borne rickettsial agents in villages reporting AES cases in this district. In total, 825 peridomestic small mammals were trapped, by setting 9254 Sherman traps i

Dermal microfilariae of dogs, jackals and cats in different regions of Iran.

BACKGROUND: Due to the complexity of retrieving skin-dwelling microfilariae, filarioids of dogs presenting dermal microfilariae (e.g. Cercopithifilaria spp., Onchocerca lupi) are relatively unknown compared to Dirofilaria spp. and Acanthocheilonema spp. whose microfilariae circulate in the blood. Although Cercopithifilaria spp. and O. lupi filarioids are distributed worldwide, there is a paucity of information on their occurrence in Iran. The aim of this study was to investigate these filarioids in a large population of dogs from different regions of Iran. METHODS: From October 2018 to Septem

Surface proteome analysis and characterization of surface cell antigen (Sca) or autotransporter family of Rickettsia typhi.

Surface proteins of the obligate intracellular bacterium Rickettsia typhi, the agent of murine or endemic typhus fever, comprise an important interface for host-pathogen interactions including adherence, invasion and survival in the host cytoplasm. In this report, we present analyses of the surface exposed proteins of R. typhi based on a suite of predictive algorithms complemented by experimental surface-labeling with thiol-cleavable sulfo-NHS-SS-biotin and identification of labeled peptides by LC MS/MS. Further, we focus on proteins belonging to the surface cell antigen (Sca) autotransporter

Babesiosis in dogs and cats--expanding parasitological and clinical spectra.

Canine babesiosis caused by different Babesia species is a protozoal tick-borne disease with worldwide distribution and global significance. Historically, Babesia infection in dogs was identified based on the morphologic appearance of the parasite in the erythrocyte. All large forms of Babesia were designated Babesia canis, whereas all small forms of Babesia were considered to be Babesia gibsoni. However, the development of molecular methods has demonstrated that other Babesia species such as Babesia conradae, Babesia microti like piroplasm, Theileria spp. and a yet unnamed large form Babesia

Vector-borne pathogen profiling and phylogeny of ticks infesting domestic dogs in Chad, Africa, using targeted next-generation sequencing and DNA barcoding.

BACKGROUND: Understanding the range of vectors and pathogen diversity in sub-Saharan Africa is crucial for efficient disease prevention and control efforts. Domestic dogs are susceptible to many vector-borne pathogens (VBPs) and serve as sentinels for several human pathogens. Yet, critical surveillance gaps in canine VBPs persist in Central Africa, where the tropical climate and pastoral agro-livelihoods create unique transmission risks. METHODS: We conducted a cross-sectional survey from September to October 2021, collecting 4238 ticks from 1254 domestic dogs across 56 villages in Chad, Afri

Prevalence of selected infectious agents in Swedish cats with fever and/or anemia compared to cats without fever and/or anemia and to stable/stray cats.

BACKGROUND: There are multiple infectious agents of cats around the world; those transmitted by direct contact among cats, hunting, or exposure to fleas or ticks are frequently the most common. Some infectious disease agents have been reported in cats in Sweden; for example, Anaplasma phagocytophilum infection was first reported in a cat in this country. However, there has not been a study in Sweden that reported test results for agents with different transmission cycles in cats with and without signs of clinical disease. Thus, the aims of this study were to (i) investigate prevalence of expos

Molecular Detection and Distribution of Giardia duodenalis and Cryptosporidium spp. Infections in Wild and Domestic Animals in Portugal.

Enteric protozoan parasites Giardia duodenalis, Cryptosporidium spp., and, to a lesser extent, the ciliate Balantioides coli are responsible for severe human and animal intestinal disorders globally. However, limited information is available on the occurrence and epidemiology of these parasites in domestic, but especially wild species in Portugal. To fill this gap of knowledge, we have investigated G. duodenalis, Cryptosporidium spp., and B. coli occurrence, distribution, genetic diversity, and zoonotic potential by analyzing 756 fecal samples from several wild carnivores (n = 288), wild ungul
