Comparative transcriptomic analysis of the thymus and other immune-related tissues using post-mortem samples from stranded cetaceans in South Korea.

Cetaceans are mammals that have adapted to aquatic environments, but their immunological characteristics are not well understood. In this study, we performed comparative transcriptomic analyses of major immune-related tissues (thymus, spleen, liver, and kidney) from post-mortem samples of stranded cetaceans in Korea, including the finless porpoise, fetal minke whale and Risso's dolphin. For cross-taxa comparisons, we retrieved publicly available transcriptomic datasets from terrestrial mammals (human and mouse), teleosts (zebrafish, common carp, and turbot), birds (chicken), reptiles (green se

The dual carbonyl reductase activities of 17β-HSD12a during ovarian development in Japanese eel (Anguilla japonica).

17β-hydroxysteroid dehydrogenase subtype 12 (17β-HSD12) is responsible for the reactions between carbonyl and hydroxyl groups at the C17 position of sex steroids. Our previous research revealed that hsd17b12a was expressed at a high level during follicular maturation in Anguilla japonica, peaking at the migrating nucleus stage. This indicated the potential of 17β-HSD12 may convert 17α-hydroxyprogesterone (17OHP) into 17α,20β-dihydroxy-4-pregnen-3-one (DHP), a maturation-inducing steroid. To elucidate the functions of hsd17b12a in A. japonica, the spatiotemporal expression patterns of hsd17b12a

circRNA03358-directed metabolic reprogramming confers Physarum flavicomum GD217 with specific resistance to TiO2 nanoparticles: Moving toward the path of combating nanoparticle pollution/exposure.

The increasing use of titanium dioxide (TiO2) nanoparticles (NPs) has raised concerns related to their environmental accumulation and the associated ecological risks. Understanding the key biomolecular responses of TiO₂ NP-tolerant organisms like Physarum flavicomum GD217 is essential for combating the pollution of and exposure to these NPs. In this study, we employed multi-omics approaches combined with molecular biology techniques to investigate the stress responses of GD217 to mixed-phase TiO₂ NPs (M-TiO₂ NPs). We found that GD217's response is primarily metabolism-related and identified ci

Adaptations to hypoxia in the vertebrate retina.

As part of the CNS, the retina maintains high metabolic activity yet is more tolerant to the adverse effects of low oxygen (hypoxia) than the brain. Tissue hypoxia is a component of ischaemia and has devastating effects on the CNS, including neuronal death. Differences in blood supply, oxygen consumption, and the expression of excitatory receptors and oxygen-binding proteins contribute to regional differences in the sensitivity of retinal neurons to hypoxia. Although highly variable and species-dependent, the mammalian retina has the capacity to withstand the effects of oxygen deprivation thro

amh alleles on XY sex chromosomes regulate male differentiation in amphidiploid Carassius auratus.

The amh (anti-Müllerian hormone) gene, residing on the sex chromosomes, has been hypothesized to be a potential master sex gene in amphidiploid Carassius auratus (crucian carp/goldfish). However, its role in male determination or differentiation remains unclear. Here, we identified the heteromorphic X and Y chromosomes and confirmed the localization of amh on the sex chromosome by fluorescence in situ hybridization in amphidiploid C. auratus. The transcriptional expressions of X-linked and Y-linked amh alleles showed no significant difference during the period of sex determination and differen

Development and Validation of a Double-Antibody Sandwich ELISA (DAS-ELISA) for Detection of Anguillid Herpesvirus.

Anguillid herpesvirus (AngHV) is a highly pathogenic agent that causes "Mucus sloughing and hemorrhagic septicemia disease" in eels, resulting in high morbidity and cumulative mortality rates. Therefore, accurate diagnosis of AngHV infection is essential for effective clinical management and epidemiological control. In this study, three monoclonal antibodies (mAbs) against AngHV were developed, and their specificity and affinity were validated using Western blotting and indirect immunofluorescence assays. Subsequently, a double-antibody sandwich enzyme-linked immunosorbent assay (DAS-ELISA) wa

The intestinal microbiome and Cetobacterium somerae inhibit viral infection through TLR2-type I IFN signaling axis in zebrafish.

BACKGROUND: Evidence has accumulated to demonstrate that intestinal microbiome can inhibit viral infection. However, our knowledge of the signaling pathways and identity of specific commensal microbes that mediate the antiviral response is limited. Zebrafish have emerged as a powerful animal model for study of vertebrate-microbiota interactions. Here, a rhabdoviral infection model in zebrafish allows us to investigate the modes of action of microbiome-mediated antiviral effect. RESULTS: We observed that oral antibiotics-treated and germ-free zebrafish exhibited greater spring viremia of carp

Zinc attenuates arsenic overdose-induced brain damage via PERK/ATF6 and TLR/MyD88/NF-κB pathways.

Exposure to arsenic (As), a widespread non-metallic toxicant in nature, often results in neurotoxicity, although the exact mechanism is unknown. Zinc (Zn) is a powerful nutrient often thought to be beneficial for growth, development and immunity. Whether Zn can rescue brain damage caused by As contamination remains to be demonstrated. Therefore, in this study, a 30-day model of As poisoning (2.83 mg/L) in carp was established and treated with Zn (1 mg/L) to investigate the detoxification mechanism involved. Histological observations showed that As induced the loosening of the molecular layer s

CRISPR/Cas9-mediated knock-in of alligator cathelicidin gene in a non-coding region of channel catfish genome.

CRISPR/Cas9-based gene knockout in animal cells, particularly in teleosts, has proven to be very efficient with regards to mutation rates, but the precise insertion of exogenous DNA or gene knock-in via the homology-directed repair (HDR) pathway has seldom been achieved outside of the model organisms. Here, we succeeded in integrating with high efficiency an exogenous alligator cathelicidin gene into a targeted non-coding region of channel catfish (Ictalurus punctatus) chromosome 1 using two different donor templates (synthesized linear dsDNA and cloned plasmid DNA constructs). We also tested

Genera and Species of the Anisakidae Family and Their Geographical Distribution.

Nematodes of the Anisakidae family have the ability to infest a wide variety of aquatic hosts during the development of their larval stages, mainly marine mammals, aquatic birds, such as pelicans, and freshwater fish, such crucian carp, these being the hosts where the life cycle is completed. The participation of intermediate hosts such as cephalopods, shrimp, crustaceans and marine fish, is an important part of this cycle. Due to morphological changes and updates to the genetic information of the different members of the family, the purpose of this review was to carry out a bibliographic sear

Functional analysis of hisJ in Aeromonas veronii reveals a key role in virulence.

Aeromonas veronii is an important aquatic zoonotic pathogen in humans and animals. In recent years, extracellular proteins from bacteria have been found to be the major pathogenic factors for aquatic animals. The aim of this study was to systematically analyze the extracellular proteins of nine sources of A. veronii and the effects of hisJ on virulence. We screened only the common proteins from nine different sources of A. veronii by liquid chromatography-tandem mass spectrometry and identified the gene hisJ. We then constructed ΔhisJ (deleted) and C-hisJ (complemented) variants of A. veronii

Gonadotropin receptors of Labeo rohita: Cloning and characterization of full-length cDNAs and their expression analysis during annual reproductive cycle.

Follicle-stimulating hormone (Fsh) and luteinizing hormone (Lh), secreted from pituitary, stimulate gonadal function by binding to their cognate receptors FSH receptor (FSHR), and LH/choriogonadotropin receptor (LHCGR). Rohu (Labeo rohita) is a commercially important seasonal breeder freshwater fish species, but till date, the regulation of expression of gonadotropins and their receptors gene during different phases of annual reproductive cycle has not been investigated. We envisaged the critical role of these molecules during seasonal gonadal development in this carp species. We cloned full-

Transbrachioradialis Approach to the Radial Tunnel: An Anatomic Study of 5 Potential Compression Sites.

BACKGROUND: Recent anatomic studies have failed to demonstrate a single utilitarian approach to intraoperative identification and surgical release of all 5 potential sites of posterior interosseous nerve (PIN) compression in the radial tunnel. This study examines if a single incision brachioradialis-splitting approach without the use of additional anatomic windows is capable of adequately exposing the entire length of the radial tunnel, including all 5 sites of PIN compression to allow for adequate release. METHODS: Ten fresh frozen cadaver forearms (6 female, 4 male) were dissected utilizing

Occurrence of Contracaecum bancrofti larvae in fish in the Murray-Darling Basin.

There is a paucity of information on the diversity and occurrence of freshwater fish parasites in Australia. This study investigates the distribution and occurrence of a parasitic nematode of the genus Contracaecum in freshwater fish from south-eastern Australia. Fish (n = 508) belonging to nine species and eight families were collected from eight wetlands associated with the Murrumbidgee River floodplain in the southern Murray-Darling Basin and subjected to laboratory examination. Third-stage nematode larvae were found in eight of the nine fish species. The exception was the fly-specked harde

Alternative Splicing Transcripts of Zebrafish LGP2 Gene Differentially Contribute to IFN Antiviral Response.

In mammals, RIG-I like receptors (RLRs) RIG-I and melanoma differentiation-associated gene 5 (MDA5) sense cytosolic viral RNA, leading to IFN antiviral response; however, LGP2 exhibits controversial functions. The same happens to fish LGP2. In this study we report that three zebrafish LGP2 splicing transcripts, a full-length LGP2, and two truncating variants, LGP2v1 and LGP2v2, play distinct roles during IFN antiviral response. Overexpression of the full-length LGP2 not only potentiates IFN response through the RLR pathway, in the absence or presence of poly(I:C) at limited concentrations, but

Molecular cloning of IgZ heavy chain isotype in Catla catla and comparative expression profile of IgZ and IgM following pathogenic infection.

Immunoglobulins serve as a crucial arm of the adaptive immune system against detrimental pathogenic threats in teleosts. However, whether the novel Ig isotype IgZ is present in the Indian major carp, Catla catla, has not yet been elucidated. The present study reports the presence of IgZ ortholog in C. catla (CcIgZ) and further demonstrates its comparative tissue specific expression with IgM (CcIgM) in response to bacterial and parasitic stimulation. The putative 139 amino acid sequence of IgZ heavy chain cDNA of C. catla showed homology with IgZ constant domains of other teleosts. Phylogenetic

Increased myocardial stiffness due to cardiac titin isoform switching in a mouse model of volume overload limits eccentric remodeling.

We investigated the cellular and molecular mechanisms of diastolic dysfunction in pure volume overload induced by aortocaval fistula (ACF) surgery in the mouse. Four weeks of volume overload resulted in significant biventricular hypertrophy; protein expression analysis in left ventricular (LV) tissue showed a marked decrease in titin's N2BA/N2B ratio with no change in phosphorylation of titin's spring region. Titin-based passive tensions were significantly increased; a result of the decreased N2BA/N2B ratio. Conscious echocardiography in ACF mice revealed eccentric remodeling and pressure volu
