Prevalence of hemoprotozoan parasites in small ruminants along a human-livestock-wildlife interface in western Uganda.

Small ruminants are important to community livelihood in developing countries; however information on the role of hemoprotozoan parasites is scanty. The objective of the study was to determine hemoprotozoan parasitic prevalence in western Uganda and identify major areas associated with these infections. This was a cross sectional study conducted at the edge of Budongo Conservation Forest in Masindi district of western Uganda in which 712 small ruminants were sampled. Blood from the jugular vein was collected from caprines and ovines and placed in an EDTA tube, and transported to the laboratory

Genetic diversity of human head lice and molecular detection of associated bacterial pathogens in Democratic Republic of Congo.

BACKGROUND: Head louse, Pediculus humanus capitis, is an obligatory blood-sucking ectoparasite, distributed worldwide. Phylogenetically, it occurs in five divergent mitochondrial clades (A-E); each exhibiting a particular geographical distribution. Recent studies suggest that, as in the case of body louse, head louse could be a disease vector. We aimed to study the genetic diversity of head lice collected in the Democratic Republic of the Congo (DR Congo) and to screen for louse-borne pathogens in these lice. METHODS: A total of 181 head lice were collected from 27 individuals at the Monkole

Molecular prevalence and ecoregion distribution of select tick-borne pathogens in Texas dogs.

Tick-borne diseases (TBD), caused by borrelial, rickettsial and babesial pathogens, are common across the United States and can cause severe clinical disease in susceptible hosts, such as domestic dogs. However, there are limited TBD molecular epidemiological reports for dogs in Texas, and none for the non-Lyme borrelial pathogen responsible for causing tick-borne relapsing fever (TBRF). Therefore, data to support the prevalence of TBRF in the canine population is inadequate. This study aimed to characterize the molecular prevalence of 11 causative agents responsible for three TBD groups withi

Development and validation of a multiplex, real-time PCR assay for Babesia rossi and Babesia vogeli.

Canine babesiosis is caused by tick-transmitted intraerythrocytic protozoan parasites occurring worldwide. In southern Africa, babesiosis is caused by Babesia rossi and B. vogeli and is one of the most common and important infectious diseases affecting dogs. There is no reliable, rapid and sensitive method for the detection of these parasites, especially when parasitaemia is low. The aim of this study was to develop a sensitive and specific multiplex TaqMan® MGB PCR assay for the diagnosis of canine babesiosis infections occurring in southern Africa, and to discriminate between Babesia rossi a

Prevalence, risk factors, and genetic diversity of veterinary important tick-borne pathogens in cattle from Rhipicephalus microplus-invaded and non-invaded areas of Benin.

Babesiosis, theileriosis, anaplasmosis, and heartwater are tick-borne diseases (TBD) that threaten livestock production in sub-Saharan Africa including Benin. This country has been faced with an invasion of Rhipicephalus microplus, a major vector for babesiosis, theileriosis, and anaplasmosis over the last decade. Yet, data on TBD and the impact of the invasive ticks are lacking, making risk level evaluation and disease control arduous. In this study, epidemiological features of Babesia bovis, B. bigemina, Theileria spp., Anaplasma marginale and Ehrlichia ruminantium infections in Benin cattle

Candidatus Neoehrlichia mikurensis and Borrelia burgdorferi sensu lato detected in the blood of Norwegian patients with erythema migrans.

The most common tick-borne human disease in Norway is Lyme borreliosis. Ticks in Norway also harbour less known disease-causing agents such as Candidatus Neoehrlichia mikurensis, Borrelia miyamotoi and Rickettsia helvetica. However, human infections caused by these pathogens have never been described in Norway. The main aims of the study were to evaluate the contribution of several tick-borne bacterial agents, other than Borrelia burgdorferi sensu lato, to zoonotic diseases in Norway and to determine their clinical pictures. Blood samples from 70 symptomatic tick-bitten adults from the Agder c

Trypanosoma vivax in water buffalo of the Venezuelan Llanos: An unusual outbreak of wasting disease in an endemic area of typically asymptomatic infections.

Trypanosoma vivax has been associated with asymptomatic infections in African and South American buffalo. In this study, T. vivax was analyzed in water buffalo (Bubalus bubalis) from Venezuela in a molecular survey involving 293 blood samples collected from 2006 to 2015 across the Llanos region. Results demonstrated constant infections (average 23%) during the years analyzed. In general, animals were healthy carriers of T. vivax with low levels of parasitemia and were diagnosed exclusively by TviCATL-PCR. However, an outbreak of severe acute infections mostly in dairy animals was reported duri

Co-infection of blacklegged ticks with Babesia microti and Borrelia burgdorferi is higher than expected and acquired from small mammal hosts.

Humans in the northeastern and midwestern United States are at increasing risk of acquiring tickborne diseases--not only Lyme disease, but also two emerging diseases, human granulocytic anaplasmosis and human babesiosis. Co-infection with two or more of these pathogens can increase the severity of health impacts. The risk of co-infection is intensified by the ecology of these three diseases because all three pathogens (Borrelia burgdorferi, Anaplasma phagocytophilum, and Babesia microti) are transmitted by the same vector, blacklegged ticks (Ixodes scapularis), and are carried by many of the s

Reciprocal regulation of NF-kB (Relish) and Subolesin in the tick vector, Ixodes scapularis.

BACKGROUND: Tick Subolesin and its ortholog in insects and vertebrates, Akirin, have been suggested to play a role in the immune response through regulation of nuclear factor-kappa B (NF-kB)-dependent and independent gene expression via interaction with intermediate proteins that interact with NF-kB and other regulatory proteins, bind DNA or remodel chromatin to regulate gene expression. The objective of this study was to characterize the structure and regulation of subolesin in Ixodes scapularis. I. scapularis is a vector of emerging pathogens such as Borrelia burgdorferi, Anaplasma phagocyto

Spatiotemporal dynamics of emerging pathogens in questing Ixodes ricinus.

Ixodes ricinus transmits Borrelia burgdorferi sensu lato, the etiological agent of Lyme disease. Previous studies have also detected Rickettsia helvetica, Anaplasma phagocytophilum, Neoehrlichia mikurensis, and several Babesia species in questing ticks in The Netherlands. In this study, we assessed the acarological risk of exposure to several tick-borne pathogens (TBPs), in The Netherlands. Questing ticks were collected monthly between 2006 and 2010 at 21 sites and between 2000 and 2009 at one other site. Nymphs and adults were analysed individually for the presence of TBPs using an array-appr

Effects of tick infestation and tick-borne disease infections (heartwater, anaplasmosis and babesiosis) on the lactation and weight gain of Mashona cattle in south-eastern Zimbabwe.

The effects of ticks and tick-borne disease infections on the lactation and weight gain of Mashona cattle were studied at Mbizi Quarantine Station in the south-eastern lowveld of Zimbabwe. Twenty-nine Mashona cows were allocated to 2 balanced groups and kept in separate paddocks at a stocking rate of one animal per 8 ha. One group received regular acaricide treatment to control bont (Amblyomma hebraeum) and other ticks. The other group was left untreated. The cows were artificially inseminated. The acaricide-treated cows and calves were essentially tick free throughout the experiment, while th

Diagnostic value of microscopic blood smear examination using a digital cell morphology system for human granulocytic anaplasmosis.

BACKGROUND: Human granulocytic anaplasmosis (HGA) is an emerging tick-borne zoonosis caused by Anaplasma phagocytophilum. HGA typically presents with flu-like symptoms and, in rare cases, can lead to life-threatening complications. Diagnosis relies on the polymerase chain reaction (PCR), which is the most sensitive, as well as serology or blood smear (BS) examination. However, PCR and serology are generally restricted to specialized laboratories and, depending on the organization and specialization of each laboratory, are often unsuitable for urgent diagnoses. Microscopic examination of BS can

Development and preliminary evaluation of a chimeric epitope-based antigen (TevISDY) for serological detection of surra.

Trypanosoma evansi remains a neglected yet economically significant pathogen in Latin America, where current serological diagnostic tools suffer from low specificity, cross-reactivity, and limited field applicability. This study presents the design and preliminary evaluation of a novel chimeric antigen derived from epitopes of the TevSTIB805.3.100 protein, a variant surface glycoprotein (VSG) of T. evansi, and explores its potential application as a serological marker for surra. An ISDY-based chimeric antigen was engineered, expressed, purified, and structurally characterized using integrated

Molecular Profiling of Ticks and Associated Pathogens: First Report of Rickettsia sibirica, Rickettsia slovaca, and Babesia microti in Ticks From Pakistan.

Global distribution of ticks and their associated tick-borne pathogens (TBPs) presents substantial health concerns for both humans and animals. The present study aimed to investigate the distribution, morpho-molecular identification, and associated TBPs of diverse tick species collected from Khyber Pakhtunkhwa and Punjab provinces of Pakistan. Morphological identifed ticks were molecularly confirmed via cytochrome oxidase I (COI) and 16S rRNA genes, which showed 15 different tick species. Among them, we found the highest prevalence rate of Rhipicephalus (Rh) microplus (404/1803; 22.41%), while

Ticks and Tick-Borne Pathogens Encountered by Dogs and Cats: A North European Perspective.

Climate change is increasing the worldwide burden of tick-borne diseases (TBDs). Dramatic increases in human cases of borreliosis have been reported during the past few decades, including from Finland, located in North Europe. As human exposure to ticks carrying pathogens is increasing, so likely is exposure of dogs and cats. However, feline or canine TBD cases are not notifiable. Likewise, no combined databases of cases exist in Finland, hindering assessment of related trends. Here, we utilize crowdsourced tick samples to reveal how commonly and to which species of TBPs dogs and cats are expo

Microbiota profile in organs of the horseflies (Diptera: Tabanidae) in Northeastern China.

Tabanids, commonly known as horseflies and belonging to the family Tabanidae, are blood-feeding arthropods (BFA) found worldwide. They are known for their ability to mechanically and biologically transmit various animal pathogens. Tabanids are potential vectors for diseases such as Francisella tularensis, Anaplasma marginale, Theileria spp., and contributors to lumpy skin diseases. Despite their involvement in common BFA studies, tabanids have not been extensively explored in microbiome research. In this study, the microbiota structure and composition in various organs of four distinct genera

Trends in Bacterial Pathogens of Bats: Global Distribution and Knowledge Gaps.

Bats have received considerable recent attention for infectious disease research because of their potential to host and transmit viruses, including Ebola, Hendra, Nipah, and multiple coronaviruses. These pathogens are occasionally transmitted from bats to wildlife, livestock, and to humans, directly or through other bridging (intermediate) hosts. Due to their public health relevance, zoonotic viruses are a primary focus of research attention. In contrast, other emerging pathogens of bats, such as bacteria, are vastly understudied despite their ubiquity and diversity. Here, we describe the curr

Metatranscriptomic profiling reveals diverse tick-borne bacteria, protozoans and viruses in ticks and wildlife from Australia.

Tick-borne zoonoses are emerging globally due to changes in climate and land use. While the zoonotic threats associated with ticks are well studied elsewhere, in Australia, the diversity of potentially zoonotic agents carried by ticks and their significance to human and animal health is not sufficiently understood. To this end, we used untargeted metatranscriptomics to audit the prokaryotic, eukaryotic and viral biomes of questing ticks and wildlife blood samples from two urban and rural sites in New South Wales, Australia. Ixodes holocyclus and Haemaphysalis bancrofti were the main tick speci
