Regain of sex determination system and sexual reproduction ability in a synthetic octoploid male fish.

Polyploids in vertebrates are generally associated with unisexual reproduction, but the direct consequences of polyploidy on sex determination system and reproduction mode remain unknown. Here, we synthesized a group of artificial octoploids between unisexual gynogenetic hexaploid Carassius gibelio and sexual tetraploid Carassius auratus. The synthetic octoploids were revealed to have more than 200 chromosomes, in which 50 chromosomes including the X/Y sex determination system were identified to transfer from sexual tetraploid C. auratus into the unisexual gynogenetic hexaploid C. gibelio. Environmental contaminants and biomarker responses in fish from the Rio Grande and its U.S. tributaries: spatial and temporal trends.

We collected, examined, and analyzed 368 fish of seven species from 10 sites on rivers of the Rio Grande Basin (RGB) during late 1997 and early 1998 to document temporal and geographic trends in the concentrations of accumulative contaminants and to assess contaminant effects on the fish. Sites were located on the mainstem of the Rio Grande and on the Arroyo Colorado and Pecos River in Texas (TX), New Mexico (NM), and Colorado. Common carp (Cyprinus carpio) and largemouth bass (Micropterus salmoides) were the targeted species. Fish were examined in the field for internal and external visible g

Postinjury changes in platelet-derived growth factor-like activity in fish and rat optic nerves.

The poor regenerative ability of the CNS of mammals has been attributed, at least in part, to the presence of mature oligodendrocytes, which have been shown to inhibit axonal growth. Proliferation of oligodendrocyte progenitor cells in the rat optic nerve during development, and thereby the timing of oligodendrocyte differentiation, has been shown to depend on a factor derived from type 1 astrocytes, later characterized as platelet-derived growth factor (PDGF). In the present study we examine whether injury to the optic nerve induces changes in the levels of PDGF in spontaneously regenerating

Zebrafish USP14 suppresses RIG-I-mediated innate antiviral immunity via targeted deubiquitination to facilitate SVCV replication.

Innate immunity serves as the host's primary defense against viral infection. Aberrant immune activation can trigger tissue damage and autoinflammatory disorders, which requires precise negative regulation to sustain immune homeostasis. RIG-I is a vital pattern recognition receptor that governs antiviral innate immune responses, and its function is tightly controlled by post-translational modifications. Ubiquitination directly modulates RIG-I activity, yet the regulatory mechanism underlying RIG-I inactivation in teleost fish remains poorly understood. Ubiquitin specific peptidase 14 (USP14) i

Exploring the mechanism of Acanthopanax in treating vertigo: A network pharmacology and molecular docking study.

Acanthopanax has therapeutic efficacy against vertigo; however, the underlying mechanism remains unclear. This study aimed to elucidate the mechanism by which Acanthopanax treats vertigo through integrated network pharmacology and molecular docking techniques, and retrieved all target genes of Acanthopanax for vertigo treatment from July to October 2025. Vertigo-related target genes were subsequently identified from public databases, including GeneCards and Online Mendelian Inheritance in Man. The intersection between Acanthopanax-derived targets and vertigo-related targets was analyzed to ide

Ellagic Acid Mitigates Lead (Pb)-Induced Toxicity in Cyprinus carpio: A Multi-Biomarker Assessment of Hematological, Immunological, and Oxidative Stress Responses Supported by Molecular Docking.

Lead (Pb) is a widespread environmental pollutant that induces systemic toxicity in aquatic organisms primarily through oxidative stress, hematological disruption, and immune dysfunction. This study investigated the protective effects of ellagic acid (EA) against Pb-induced toxicity in common carp (Cyprinus carpio) using a multi-biomarker approach and molecular docking. Fish were assigned to six experimental groups: control, EA-treated, Pb-I, Pb-I + EA, Pb-II, and Pb-II + EA. Fish in the Pb-I and Pb-II groups were exposed to 2.5 and 5 mg/L Pb, respectively, while EA was administered via diet a

Reactivation of cyprinid herpesvirus 2 (CyHV-2) in asymptomatic surviving goldfish Carassius auratus (L.) under immunosuppression.

Cyprinid herpesvirus 2 (CyHV-2) is a highly contagious pathogen of goldfish (Carassius auratus) and Prussian carp (Carassius auratus gibelio) causing herpesviral hematopoietic necrosis. Our previous study revealed that CyHV-2 can persistently infect the kidney and spleen of goldfish that recovered from a primary infection. In this study, we tried to identify the cells persistently infected with the virus in surviving fish and investigated virus reactivation in the survivors injected with immunosuppressants, namely dexamethasone (Dex) and cyclosporine A (CsA). Virus DNA was detected from the mo

Gonadotropins in the Russian Sturgeon: Their Role in Steroid Secretion and the Effect of Hormonal Treatment on Their Secretion.

In the reproduction process of male and female fish, pituitary derived gonadotropins (GTHs) play a key role. To be able to specifically investigate certain functions of Luteinizing (LH) and Follicle stimulating hormone (FSH) in Russian sturgeon (Acipenser gueldenstaedtii; st), we produced recombinant variants of the hormones using the yeast Pichia pastoris as a protein production system. We accomplished to create in vitro biologically active heterodimeric glycoproteins consisting of two associated α- and β-subunits in sufficient quantities. Three dimensional modelling of both GTHs was conducte

Immunomodulatory effect of arsenic on cytokine and HSP gene expression in Labeo rohita fingerlings.

Immune system is fundamental for survival of an organism against invading pathogens and other harmful agents. Cytokines, the signaling proteins that are produced transiently after cell activation and exert pleiotropic effects on cells of the immune system, are important mediators of cell mediated immune response. When expressed in a dysregulated fashion cytokines can underlie either immunodeficient or immunopathologic states. Heat shock proteins (stress proteins, HSPs) are also key proteins, which play important role in immunomodulation, apoptosis and influence the immune responses. Arsenic is

Eutrophic urban ponds suffer from cyanobacterial blooms: Dutch examples.

Ponds play an important role in urban areas. However, cyanobacterial blooms counteract the societal need for a good water quality and pose serious health risks for citizens and pets. To provide insight into the extent and possible causes of cyanobacterial problems in urban ponds, we conducted a survey on cyanobacterial blooms and studied three ponds in detail. Among 3,500 urban ponds in the urbanized Dutch province of North Brabant, 125 showed cyanobacterial blooms in the period 2009-2012. This covered 79% of all locations registered for cyanobacterial blooms, despite the fact that urban ponds

Neural pathway for aggressive display in Betta splendens: midbrain and hindbrain control of gill-cover erection behavior.

Horseradish peroxidase (HRP) was used to identify parts of the presumptive neural pathway for gill cover erection, a behavioral display pattern performed by Siamese fighting fish (Betta splendens) during aggressive interactions. Motor, motor integration and sensory areas were identified in the medulla and mesencephalon. Motor neurons of the dilator operculi muscle, the effector muscle for gill cover erection, are located in the lateral and medial parts of the caudal trigeminal motor nucleus. Iontophoretic injections of HRP into the lateral trigeminal motor nucleus resulted in labeled cell bodi

Effects of Spice Treatment Methods on the Flavor Modification and Quality Characteristics of Surimi/Pea Protein Mixed Gels.

To develop a flavor-modification strategy for silver carp surimi products, pea protein was used to partially replace surimi protein at a substitution level of 30% (w/w) to prepare surimi/pea protein mixed gels (SPGs). Two spice treatment methods were compared: deodorized SPG prepared by adding spice water during gel preparation (DSPG-A) and deodorized SPG incubated in spice water during the second-stage heating (DSPG-I). The effects of these treatments on odor characteristics, volatile flavor composition, gel quality, and processing energy consumption were investigated. Sensory evaluation show

Acute exposure to the conspecific alarm substance and its intergenerational consequences in Astyanax bimaculatus (Linnaeus, 1758).

The conspecific alarm substance (CAS), a chemical compound likely based on glycosaminoglycans, purines, and nitrogen oxide-based pterins, is a natural stressor released by specialized epidermal cells damaged by injury or attack in fish, inducing hormonal and behavioral changes in conspecifics. Its effects in the reproductive context are poorly understood, but previous studies have demonstrated earlier spawning in broodstock exposed to CAS and an influence on the larval length of the offspring, potentially linked to increased cortisol transfer. In this study, 53 sexually mature specimens of Ast

In pursuit of saccade awareness: Limited volitional control and minimal conscious access to catch-up saccades during smooth pursuit eye movements.

Observers use smooth pursuit to track moving objects-like koi carp gliding through a pond. When positional errors accumulate, rapid catch-up saccades correct for them. Despite their abruptness, these saccades usually go unnoticed, creating the seamless experience of smooth tracking. We conducted three experiments to examine awareness and control of catch-up saccades (Experiment 1), the effect of training (Experiment 2), and of movement intention (Experiment 3). All experiments followed a similar protocol. On each trial, a target moved horizontally at one of three constant speeds (3-12 dva/s).

Molecular characterization and functional analysis of septin 8a in rohu (Labeo rohita): insights into immune regulation and developmental roles revealed by CRISPR/Cas9-mediated F0 mutagenesis.

Septins are conserved GTP-binding cytoskeletal proteins that regulate cytokinesis, membrane dynamics, and innate immunity. Here, we report the first molecular and functional characterization of septin 8a in rohu (Labeo rohita), an economically important Indian major carp. The full-length septin 8a cDNA encodes a 420-amino acid protein containing conserved GTP-binding motifs (G1-G5) belonging to the SEPT6 subgroup with a predicted molecular weight of ∼48.98 kDa, hydrophilic character and intracellular localization consistent with cytoskeletal scaffolding.
