Influence of Light Wavelength and Optotype Size on Accommodative Response and Aberrometric Changes Across the Adult Lifespan.

PURPOSE: To analyse the effects of chromatic light (white, blue, red and green) and stimulus size (6/6 and 6/12) on pupil constriction, Zernike coefficients and the accommodative response curve using wavefront aberrometry across a wide age range of healthy subjects. METHODS: One hundred and sixty-four right eyes from participants aged 20-75 years were evaluated. All subjects showed normal near visual function for their age. Wavefront aberrations were measured under scotopic conditions using the IRX3 aberrometer. Accommodation was induced from 0 to 10 D in the younger group and from 0 to 5 D i

Multimodal circadian phenotyping reveals distinct mechanisms of photic entrainment failure in two sighted patients with non-24-hour sleep-wake disorder.

Non-24-h sleep-wake disorder (N24SWD) is a rare circadian rhythm disorder in sighted patients whose underlying pathophysiology remains poorly understood. Here, we aimed to investigate the mechanisms of impaired circadian entrainment by combining assessments of retinal and melanopsin function, circadian light responsiveness and endogenous circadian rhythmicity. Two sighted patients with confirmed N24SWD underwent comprehensive circadian phenotyping targeting three complementary components of the circadian system: retinal and melanopsin-mediated photoreception, endogenous circadian timing, and c

Integration of digital physicalomics and dual-fluid metabolomics flux ratios reveals tubular secretory dysfunction in early diabetic kidney disease.

BACKGROUND: Current screening for diabetic kidney disease (DKD) relies on the estimated glomerular filtration rate (eGFR) and albuminuria, which often fail to detect early tubular dysfunction and non-albuminuric phenotypes. The integration of macroscopic urine physical characteristics with metabolic signatures may offer a novel approach to precision stratification. METHODS: We conducted a multicenter, prospective-retrospective cohort study involving 364 participants with type 2 diabetes. We developed "FluxPro-DKD fusion model," that integrates "Digital Physicalomics" (computer-vision quantifi

Short-Term Axial Length Effects of Combining Paracentral Collimating Apertures with Near-Peripheral Myopic Defocus for Myopia Control.

Background/Objectives: Myopia is a growing global problem. Current preventative treatments are inadequate, and myopia control spectacles intentionally degrade peripheral optical quality over large retinal areas. Variable results have been related to near-peripheral retinal refractive variability and chromatic refractive spread. A narrow paracentral band of collimating apertures created by annuli with plus-add surrounding appropriately small distance-correcting areas is proposed to decrease defocus and dynamic accommodation cues at the most responsive retinal area for controlling axial elongati

Deep Learning-Based Semantic Segmentation and Classification of Otoscopic Images for Otitis Media Diagnosis and Health Promotion.

Background/Objectives: Otitis media (OM), including acute otitis media (AOM) and chronic otitis media (COM), is a common middle ear disease that can lead to significant morbidity if not accurately diagnosed. Otoscopic interpretation remains subjective and operator-dependent, underscoring the need for objective and reproducible diagnostic support. Recent advances in artificial intelligence (AI) offer promising solutions for automated otoscopic image analysis. Methods: We developed an AI-based diagnostic framework consisting of three sequential steps: (1) semi-supervised learning for automatic r

Color polymorphism, taxonomic confusion and cryptic diversity in the kukri snake Oligodon albocinctus (Cantor, 1839) (Squamata: Colubridae).

The kukri snake Oligodon albocinctus (Cantor, 1839) is a widespread element of the Indo-Himalayan and Indo-Burmese snake fauna with two distinct color patterns. The first and more typical morphotype has light bands with black edges, while individuals of the second morphotype have brown blotches or narrow crossbars. The polychromatic nature of this species has led to considerable taxonomic confusion between O. albocinctus and members of Oligodon occurring in sympatry or parapatry. In this study, we investigate patterns of genetic and morphological variation in O. albocinctus and explore whether

The influence of the environment and lifestyle on myopia.

BACKGROUND: Myopia, commonly known as near-sightedness, has emerged as a global epidemic, impacting almost one in three individuals across the world. The increasing prevalence of myopia during early childhood has heightened the risk of developing high myopia and related sight-threatening eye conditions in adulthood. This surge in myopia rates, occurring within a relatively stable genetic framework, underscores the profound influence of environmental and lifestyle factors on this condition. In this comprehensive narrative review, we shed light on both established and potential environmental and

Visual function and retinal thickness in children with type 1 diabetes mellitus.

CLINICAL RELEVANCE: The possibility that changes in blue-yellow visual thresholds and some retinal thickness measures in children with diabetes mellitus may be observed before any visible fundus changes points to the possibility of these measures being a useful predictor that the risks of diabetic retinopathy are higher in some children than in others. INTRODUCTION: Previous studies showed mixed results on chromatic and achromatic contrast sensitivity early in the course of diabetes mellitus, and the findings of these studies may have been influenced by a lack of experimental sensitivity to v

Visual Performance of People With Albinism Assessed With Generalizable and Adaptive AIM and FInD Methods.

PURPOSE: People with albinism (PwA) are known to have visual impairments; however, little is known about whether these functions are disrupted across earlier and later stages of the visual pathway. We investigated distinct perceptual functions and fixation stability within each observer and compared the data with age- (±5 years) and sex-matched controls. METHODS: Twenty-one self-reported PwA and twenty-one controls were recruited. Angular-indication measurement (AIM) and foraging-interactive-D-prime (FInD) psychophysical methods were deployed to measure OS, OD, and OU near visual acuity, spat

Choice of pre-processing pipeline influences clustering quality of scRNA-seq datasets.

BACKGROUND: Single-cell RNA sequencing (scRNA-seq) has quickly become one of the most dominant techniques in modern transcriptome assessment. In particular, 10X Genomics' Chromium system, with its high throughput approach, turn key and thorough user guide made this cutting-edge technique accessible to many laboratories using diverse animal models. However, standard pre-processing, including the alignment and cell filtering pipelines might not be ideal for every organism or tissue. Here we applied an alternative strategy, based on the pseudoaligner kallisto, on twenty-two publicly available sin

Interaction of aberrations, diffraction, and quantal fluctuations determine the impact of pupil size on visual quality.

Our purpose is to develop a computational approach that jointly assesses the impact of stimulus luminance and pupil size on visual quality. We compared traditional optical measures of image quality and those that incorporate the impact of retinal illuminance dependent neural contrast sensitivity. Visually weighted image quality was calculated for a presbyopic model eye with representative levels of chromatic and monochromatic aberrations as pupil diameter was varied from 7 to 1 mm, stimulus luminance varied from 2000 to 0.1  cd/m2, and defocus varied from 0 to -2 diopters. The model included t

Highly Predictive Model for a Protective Immune Response to the A(H1N1)pdm2009 Influenza Strain after Seasonal Vaccination.

Understanding the immune response after vaccination against new influenza strains is highly important in case of an imminent influenza pandemic and for optimization of seasonal vaccination strategies in high risk population groups, especially the elderly. Models predicting the best sero-conversion response among the three strains in the seasonal vaccine were recently suggested. However, these models use a large number of variables and/or information post- vaccination. Here in an exploratory pilot study, we analyzed the baseline immune status in young (<31 years, N = 17) versus elderly (≥50 yea

Different ATM Signaling in Response to Chromium(VI) Metabolism via Ascorbate and Nonascorbate Reduction: Implications for in Vitro Models and Toxicogenomics.

BACKGROUND: Carcinogenic hexavalent chromium [Cr(VI)] requires cellular reduction to generate DNA damage. Metabolism of Cr(VI) by its principal reducer ascorbate (Asc) lacks a Cr(V) intermediate, which is abundant in reactions with a minor reducing agent, glutathione. Cultured cells are widely used in mechanistic studies of Cr(VI) toxicity; however, they typically contain < 1% of normal Asc levels. Asc deficiency is also expected to diminish protection against reactive oxygen species. OBJECTIVES: We assessed how the presence of Asc in cells affects their stress signaling and survival response

Effects of androgen on the expression of brain aromatase cytopigment and nerve growth factor in neonatal rats with hypoxic-ischemic brain damage.

OBJECTIVE: To study the effects of androgen on the expression of aromatase cytopigment P450 (AROM) and nerve growth factor (NGF) in the brain and brain ultrastructure in neonatal rats with hypoxic-ischemic brain damage (HIBD) in order to investigate the mechanism underlying the protective effect of androgen against HIBD. METHODS: Ninety-six seven-day-old Sprague-Dawley rats were randomly divided into three groups: sham-operation, HIBD and androgen treatment (n=32 each). HIBD was induced by the ligation of left common carotid artery and hypoxia exposure. The rats in the androgen treatment and

Effects of vibration and hyaluronic acid on activation of three-dimensional cultured chondrocytes.

OBJECTIVE: To investigate the effects of vibration (Vib) and hyaluronic acid (HA) on 3-dimensional cultured cartilage. METHODS: Chondrocytes were obtained from metatarsophalangeal joints of freshly killed 6-month-old pigs. Twenty-four-well plates containing type I collagen sponge disks were used to culture samples. The frequency and the amplitude of the vibration of the well plate were 100 Hz and 0.5 nm, respectively. We produced 3-dimensional cartilage tissue using HA and vibration with collagen sponge as a carrier. Four different culture conditions were examined: a control HA-Vib- group, an

DNA-protein cross-links produced by various chemicals in cultured human lymphoma cells.

Chemicals such as cis-platinum, formaldehyde, chromate, copper, and certain arsenic compounds have been shown to produce DNA-protein cross-links in human in vitro cell systems at high doses, such as those in the cytotoxic range. Thus far there have only been a limited number of other chemicals evaluated for their ability to produce cross-links. The purpose of the work described here was to evaluate whether select industrial chemicals can form DNA-protein cross-links in human cells in vitro. We evaluated acetaldehyde, acrolein, diepoxybutane, paraformaldehyde, 2-furaldehyde, propionaldehyde, ch

G1 arrest and cell-cycle-dependent clastogenesis in UV-irradiated human fibroblasts.

The demonstrations of frequent allelic deletions in lung and colon cancers have reemphasized the importance of clastogenesis in carcinogenesis. We have investigated the mechanisms of induction of chromosome aberrations in ultraviolet-irradiated diploid human fibroblasts. Cells were irradiated with UV at various times during a parasynchronous wave of cell proliferation and then harvested during the first mitosis that followed irradiation. Metaphase spreads were stained with Geimsa and the yields of chromosome aberrations were quantified. Ultraviolet irradiation induced primarily chromatid-type
