Acrylamide-induced visceral neuropathy: evidence for the involvement of capsaicin-sensitive nerves of the rat urinary bladder.

The mechanisms underlying the severe urinary retention induced by acrylamide intoxication were studied in detail in the rat. Subcutaneous treatment with acrylamide monomer (50 mg/kg daily for 10 days) almost completely impaired the micturition reflex, resulting in urinary retention. In fact, the ability to eliminate an oral water load was virtually abolished, while bladder filling with saline (transvesical cystometrogram) failed to activate reflex micturition. Instead, a picture of overflow incontinence resulted in urethane-anaesthetized rats, which was not reversed by intravenous administrati

Chronic cerebroventricular infusion of hypertonic sodium chloride in rats reduces hypothalamic sympatho-inhibition and elevates blood pressure.

To determine whether or not salt loading restricted to the brain would elevate blood pressure, chronically implanted osmotic minipumps were used to infuse hypertonic sodium chloride solutions made in artificial cerebrospinal fluid into a jugular vein intravenously or the third cerebral ventricle, intracerebroventricularly, for 11 days in awake rats. During intracerebroventricular infusion of hypertonic sodium chloride, tail-cuff systolic pressures began to rise on day 4 and were significantly elevated by day 9. In contrast, infusions of either artificial cerebrospinal fluid alone, intracerebro

In Vivo Study on Mechanism Underlying Increased Pharmacological Effects of Phenobarbital in Rats with Glycerol-Induced Acute Renal Failure.

The mechanism underlying the increased pharmacological effects of phenobarbital in rats with glycerol-induced acute renal failure (ARF) was examined. In the experiments, a surgical cannula was inserted in the lateral ventricle of the rats for phenobarbital infusion, and the ARF induction was performed by intramuscular administration of 50% glycerol. The onset time of anesthesia by phenobarbital was determined with the tail flick method. In addition, cerebral microsomes were prepared from excised cerebral cortices of sham and ARF rats, and the cerebral expression of the γ-aminobutyric acid (GAB

Development of a flexible 3D printed scaffold with a cell-adhesive surface for artificial trachea.

Trachea stents are widely used to treat stenosis arising from various trachea injuries. However, they are associated with inflammation, re-stenosis, and tracheal obstruction. Seeking to overcome these problems, the development of an artificial trachea using tissue engineering has been explored. However, the artificial trachea did not mimic the natural rigidity and flexibility of the trachea and provide the micro-environment necessary for re-epithelialization. In this study, we developed a thermoplastic polyurethane (TPU) trachea scaffold that possesses a restoration characteristic, using flexi

Marginal bacterial leakage and pulp reactions in Class II composite resin restorations in vivo.

The presence of stainable bacteria under restorations and pulp reactions in 36 teeth restored in vivo with a modified Class II composite resin restoration with two different dentine treatment techniques were studied on three separate follow-up occasions (1-3, 7-10 and 28-32 days). Half of the cavities showed stainable bacteria at the cavity margins and bottoms. Teeth restored with method A (Gluma/Occlusin) showed significantly fewer restorations with stainable bacteria then teeth restored with method B (Life/Occlusin) (p less than 0.05). Significantly more restorations with detectable bacteria

Late UP-state bias in cortico-amygdalar coordination.

Neocortical slow oscillations temporally organize neuronal activity across distributed brain regions, but the coordination of basolateral amygdala (BLA) activity with these oscillations remains unclear. We investigated BLA neuronal activity relative to neocortical slow oscillations in male mice, primarily under urethane anesthesia. These oscillations are characterized by rhythmic alternation between active (UP) and inactive (DOWN) states at 0.3-3 Hz. Using unit recordings and calcium imaging, we found that BLA neuronal activity was sparse but preferentially associated with the later phase of t

In vitro evaluation of discoloration and surface alteration in self-adhesive resin cements using CIELAB and 3D profilometry.

BACKGROUND: Self-adhesive resin cements (SARCs) are widely used in the field of restorative dentistry due to their simplified application and adequate bonding properties. However, their long-term color and surface stability, particularly under the influence of pigment-rich dietary substances, remain a critical concern in aesthetic restorations. OBJECTIVES: The aim of the study was to evaluate the color stability and surface roughness of 4 contemporary SARCs after immersion in various staining media using digital photography. MATERIAL AND METHODS: A total of 160 disc-shaped specimens (n = 40

Effect of surface silica content on primer‑dependent resin bonding to nanoceramic‑filled polyetheretherketone.

OBJECTIVES: This study aimed to elucidate how silica layers on nanoceramic‑filled polyetheretherketone (CF-PEEK), derived from silica‑coated alumina airborne‑particle abrasion (SC‑APA) treatment, combined with clinically relevant cleaning and primer chemistries, influence the CF-PEEK-resin bonding performance and durability, thereby refining evidence‑based bonding strategies. METHODS: CF-PEEK specimens were treated using SC‑APA and subjected to various cleaning protocols. Their surface morphology, wettability, and roughness were characterized using field‑emission scanning electron microscopy,

ATR-FTIR assessment of degree of conversion across curing-state transitions in Bis-EMA- and UDMA-based microfilled dental resins for additive manufacturing.

Assessing the degree of conversion is important for evaluating processing quality in additively manufactured dental methacrylates. However, the widely used attenuated total reflectance-Fourier transform infrared (ATR-FTIR) method, based on the 1637/1608 cm⁻¹ ratio, may be unstable in contemporary methacrylate systems because of spectral congestion and the formulation-dependent behaviour of the aromatic reference band. This study evaluated a low-variance ATR-FTIR protocol based on a polymerisation-sensitive, ester-associated band pair at 1320/1352 cm⁻¹ to estimate surface or near-surface conver

Effects of Melatonin Formulations on Caffeine-Evoked Cortical Activity and Pentobarbital-Induced Sleep in Mice.

Melatonin has chronobiotic, antioxidant, anti-inflammatory, and neuromodulatory properties, and its oral effects may vary according to formulation. This study compared conventional melatonin with a lipid-multiparticulate (LMP) formulation in two acute pharmacological mouse paradigms. Eighty-four male BALB/c mice were assigned to caffeine-evoked cortical activity or pentobarbital-induced sedative-hypnotic experiments. Mice received caffeine (7.5 mg/kg, i.p.) alone or with standard or LMP-melatonin (10 or 20 mg/kg, oral). ECoG recorded cortical electrical activity under urethane anaesthesia. In

Gestational VPA Exposure Impairs the Facial Stimulation-Induced Cerebellar MF-GC LTP by Enhancing mGluR1/5-CB1 Receptor Cascade in Offspring Mice.

The abnormalities in cerebellar circuit function within the Crus I/II lobules in autism spectrum disorder (ASD) are widely recognized as critical contributors to impairments in motor behavior and social interaction. We here investigated the impact of prenatal valproic acid (VPA) exposure on facial stimulation-evoked synaptic plasticity at cerebellar mossy fiber-granule cell (MF-GC) synapses in urethane-anesthetized offspring mice in vivo. When the GABAA receptor was blocked, 20 Hz facial stimulation induced MF-GC LTP in control mice, whereas the same stimulation paradigm triggered MF-GC LTD in

A comparative study on the mechanical and antibacterial properties of BPA-free dental resin composites.

OBJECTIVE: The commonly used base monomer utilized in resinous commercial dental restorative products is bis-GMA which is derived from bisphenol-A (BPA) - a well-known compound which may disrupt endocrine functions. To address concerns about its leaching into the oral environment and to optimize the quality of dental composites, a BPA-free alternative base monomer, fluorinated urethane dimethacrylate (FUDMA), was designed by modifying a UDMA monomer system. METHODS: Nine groups of composites were prepared by mixing the base monomers and TEGDMA in a ratio of 70/30 wt% to which were added silan

TLR4 inhibitors through inhibiting (MYD88-TRIF) pathway, protect against experimentally-induced intestinal (I/R) injury.

Intestinal ischemia/reperfusion (I/R) injury is a serious condition that causes intestinal dysfunction and can be fatal. Previous research has shown that toll-like receptor 4 (TLR4) inhibitors have a protective effect against this injury. This study aimed to investigate the protective effects of TLR4 inhibitors, specifically cyclobenzaprine, ketotifen, amitriptyline, and naltrexone, in rats with intestinal (I/R) injury. Albino rats were divided into seven groups: vehicle control, sham-operated, I/R injury, I/R-cyclobenzaprine (10 mg/kg body weight), I/R-ketotifen (1 mg/kg body weight), I/R-ami

Impact of Activity-Based Training on Bowel Function in a Rat Model of Spinal Cord Injury.

Significant bowel-related issues after spinal cord injury (SCI) that affect morbidity and quality of life (QOL) include diminished bowel motility, loss of sphincter control, gastric ulcers, autonomic dysreflexia, pain, diarrhea, constipation, and fecal incontinence. Clinical diagnoses and research in humans have largely relied on anorectal manometry (ARM) procedures to increase understanding of the functional effects of SCI on colorectal motility and defecation physiology. Recent pre-clinical rodent studies have also used ARM to further our understanding of bowel-related dysfunctions post-SCI.

Understanding Neuromodulation Pathways in tDCS: Brain Stem Recordings in Rat During Trigeminal Nerve Direct Current Stimulation.

BACKGROUND: Recent evidence suggests that transcranial direct current stimulation (tDCS) indirectly influences brain activity through cranial nerve pathways, particularly the trigeminal nerve. However, the electrophysiological effects of direct current (DC) stimulation on the trigeminal nerve (DC-TNS) and its impact on trigeminal nuclei remain unknown. These nuclei exert control over brainstem centers regulating neurotransmitter release, such as serotonin and norepinephrine, potentially affecting global brain activity. OBJECTIVES: To investigate how DC-TNS impacts neuronal activity in the pri

In vivo electrophysiological study of the targeting of 5-HT3 receptor-expressing cortical interneurons by the multimodal antidepressant, vortioxetine.

The antidepressant vortioxetine has high affinity for the ionotropic 5-HT3 receptor (5-HT3 R) as well as other targets including the 5-HT transporter. The procognitive effects of vortioxetine have been linked to altered excitatory:inhibitory balance in cortex. Thus, vortioxetine purportedly inhibits cortical 5-HT3 R-expressing interneurons (5-HT3 R-INs) to disinhibit excitatory pyramidal neurons. The current study determined for the first time the effect of vortioxetine on the in vivo firing of putative 5-HT3 R-INs whilst simultaneously recording pyramidal neuron activity using cortical slow-w

Carotid body stimulation as a potential intervention in sudden death in epilepsy.

OBJECTIVE: To investigate carotid body (CB) mechanisms related to sudden death during seizure. Ictal activation of oxygen-conserving reflexes (OCRs) can trigger fatal cardiorespiratory collapse in seizing rats, which presents like human sudden unexpected death in epilepsy (SUDEP). The CB is strongly implicated in OCR pathways; we hypothesize that modulating CB activity will provide insight into these mechanisms of death. METHODS: Long-Evans rats were anesthetized with urethane. Recordings included: electrocorticography, electrocardiography, respiration via nasal thermocouple, and blood pressu
