Disruption of target interactions prevents the development of enkephalin immunoreactivity in sympathetic neurons.

We compared the development of enkephalin (Enk) expression in normal rats and rats in which target contact was transiently disrupted with 6-hydroxydopamine (6-OHDA). During the first 3 postnatal weeks, there was a striking increase in Enk immunoreactivity (-IR) in superior cervical ganglia (SCG) assayed by radioimmunoassay (RIA). This increase was correlated with the appearance of Enk-IR in postganglionic neurons. In the caudal region of the SCG, the proportion of Enk-IR neurons and their immunoreactivity increased until one-third of the neurons possessed Enk-IR between postnatal days (P) 14 a

Behavioral asymmetries and recovery in rats with different degrees of unilateral striatal dopamine depletion.

A detailed behavioral analysis during the first postoperative week was performed in rats which had sustained various degrees of unilateral neostriatal dopamine (DA) lesions by administration of the neurotoxin 6-hydroxydopamine into the substantia nigra. These animals were assigned to different groups according to their residual DA levels in the damaged neostriatum (as percentage of the intact side). On the first day after toxin injection into the substantia nigra, turning asymmetries (tight turns) toward the side of the lesion were observed in animals with a mean residual DA level of 32% or le

Recovery of brain noradrenaline after 5,7-dihydroxytryptamine-induced axonal lesions in the rat.

Time-dependent changes in regional CNS noradrenaline (NA) concentration, 3H-NA uptake and fluorescence morphology of CNS NA neurons were analysed in the adult rat up to 6 months after intraventricular injection of 5,7-dihydroxytryptamine (5,7-DHT), and compared with the time-course of changes in brain and spinal cord indolamine neurons. Following a substantial depletion of both amines in all CNS regions (telodiencephalon, brainstem and spinal cord) at 10 days after 150 mug 5,7-DHT, brain NA--but not 5-HT--levels recovered to near-normal values in brainstem and forebrain (35% below the age-matc

The biochemical basis of the behavioral disorder in the Lesch-Nyhan syndrome.

An inherited complete deficiency of hypoxanthine-guanine phosphoribosyltransferase in male children is associated with a severe neurological disorder characterized by chloroform and athetoid movements, hypertonicity, mental retardation, and self-injurious behavior. In the review that follows several possible mechanisms by which the enzyme defect may cause the CNS disorder are discussed. Current evidence suggests that the primary neural deficit in the Lesch-Nyhan syndrome is a deficiency of dopamine in the basal ganglia. It is argued that this neurochemical lesion results from a deficiency of p

Transplantation of Nurr1-overexpressing neural stem cells and microglia for treating parkinsonian rats.

BACKGROUND: Neural stem cells (NSCs) transplantation is considered a promising treatment for Parkinson's disease. But most NSCs are differentiated into glial cells rather than neurons, and only a few of them survive after transplantation due to the inflammatory environment. METHODS: In this study, neural stem cells (NSCs) and microglial cells both forced with the Nurr1 gene were transplanted into the striatum of the rat model of PD. The results were evaluated through reverse transcription polymerase chain reaction (RT-PCR), Western blot, and immunofluorescence analysis. RESULTS: The behavior

Repeated stimulation of D-1 dopamine receptors increases the circling response to bromocriptine in rats with a 6-OHDA lesion.

We have studied the effect of repeated injections of a specific D-1 agonist, (SKF 38393), on circling behaviour in response to a specific D-2 agonist (bromocriptine) and on striatal D-2 DA receptors in rats with a unilateral lesion of the nigrostriatal pathway. Two groups of rats were formed on the basis of their circling response to a first injection of SKF 38393: clear contralateral circling (SKF-positive) and no (or ipsiversive) circling (SKF-negative). A few days later, these rats were tested once for bromocriptine-induced circling. A repeated treatment of 8 injections of SKF 38393 over 4

Re-evaluating the neuroprotective landscape of Nigella sativa: Molecular mechanisms, nanomedicine, and clinical translation.

Nigella Sativa (NS) L., commonly known as black seed, and its major bioactive compound, thymoquinone (TQ), have long been used in traditional medicine and have recently gained increasing attention as potential neuroprotective agents. TQ exerts pleiotropic effects within the central nervous system through several interconnected mechanisms, including activation of the Nrf2/HO-1 antioxidant pathway, inhibition of NF-κB-mediated neuroinflammation and NLRP3 inflammasome signaling, and regulation of the mitochondrial Bax/Bcl-2/caspase-3 apoptotic pathway. These effects have been reported in diverse

Focused ultrasound enhances targeted curcumin delivery and alleviates behavioral and neuropathological deficits in a Parkinson's disease mouse model.

INTRODUCTION: Curcumin exhibits potent neuroprotective properties but is limited by poor blood-brain barrier (BBB) permeability. This study aimed to evaluate whether focused ultrasound (FUS)-mediated BBB opening enhances curcumin delivery and therapeutic efficacy in a 6-hydroxydopamine (6-OHDA) mouse model of Parkinson's disease (PD). METHODS: Male C57BL/6 mice with unilateral 6-OHDA lesions received intravenous curcumin with or without FUS targeted to the lesioned striatum for 4 weeks. Behavioral performance was assessed using rotarod and open-field tests. Postmortem analyses included tyrosi

Roflupram Reduces 6-OHDA-Induced Cellular Damage in SH-SY5Y Cells by Inhibiting PDE4.

BACKGROUND: The main symptoms of Parkinson's disease (PD) include olfactory impairment and tremor. Current treatment methods for PD generally have limitations such as short duration and severe side effects. The novel phosphodiesterase 4 (PDE4) inhibitor Roflupram (Roflu) mitigates inflammatory responses and enhances cognitive functions in individuals with neurological conditions. However, it remains unknown whether Roflu provides neuroprotection in a PD model induced by 6-hydroxydopamine (6-OHDA). METHODS: Cell viability was assessed using a 3-(4,5-Dimethylthiazol-2-yl)-2,5-diphenyltetrazoliu

Treadmill exercise rescues motor deficits in parkinsonian mice by modulating striatal D2-MSN activity: evidence from calcium imaging and chemogenetics.

OBJECTIVE: The aim of this study is to investigate whether treadmill exercise alleviates motor dysfunction in a mouse model of Parkinson's disease (PD) by modulating the excitability of striatal medium spiny neurons expressing dopamine type 2 receptors (D2-MSNs). METHODS: A unilateral 6-hydroxydopamine (6-OHDA) injection was performed in the right striatum of D2-Cre mice to establish a hemi-lesioned PD model, with sham-operated mice serving as controls. PD mice were subjected to treadmill exercise (18 m/min, 40 min/day, 5 days/week for 4 weeks). Motor function was evaluated using the open-fie

Increased nerve density adversely affects outcome in colorectal cancer and denervation suppresses tumor growth.

BACKGROUND: The colon and rectum are highly innervated, with neural components within the tumor microenvironment playing a significant role in colorectal cancer (CRC) progression. While perineural invasion (PNI) is associated with poor prognosis in CRC, the impact of nerve density and diameter on tumor behavior remains unclear. This study aims to evaluate the prognostic value of nerve characteristics in CRC and to verify the impact of nerves on tumor growth. METHODS: Tissue samples from 129 CRC patients were stained with immunofluorescent markers NF-L and S100B to detect nerves. Nerve diamete

Neuroprotective effects of quercetin on motor impairments and anxiety-like behaviors in a rat model of Parkinson's disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by motor impairments such as bradykinesia and rigidity, and non-motor deficits including anxiety. These phenomena are closely associated with underlying pathological processes including oxidative stress(OS) and mitochondrial dysfunction that arise from the degeneration of dopaminergic neurons in the substantia nigra. Recent studies have shown that flavonoids like quercetin have neuroprotective effects through antioxidant and anti-inflammatory properties. In this study, we aimed to evaluate the impact of querceti

ML-based prediction to experimental validation: Development of dihydroquinazoline based multi-potent ligands as anti-Alzheimer's agents.

Alzheimer's disease (AD) is a multifactorial neurological disorder accounting for the cognitive decline in the patients. The disease is linked to numerous pathological factors including hyperactivation of acetylcholinesterase (AChE) and monoamine oxidase B (MAO-B), accumulation of amyloid-beta (Aβ) plaques and neurofibrillary tangles in the brain etc. The single-target medications already available in the market are found to be ineffective and the research focus is shifting towards the development of multitargeting agents. In order to find a multi-potent inhibitor against AChE, MAO-B and Aβ1-4

Airway sympathectomy attenuates inflammation, transcriptional ratios of Muc5ac and Muc5b, and airway mechanic deficits in mice delivered intranasal IL-13.

Excessive mucus in the airways is an underlying pathological feature of many airway diseases, including asthma. Therapeutic options to reduce mucus production in the airways remain limited. One possible therapeutic target is the airway sympathetic nerves. Although lung sympathetic innervation has been considered sparse, sympathetic nerves secrete neurotransmitters that act on adrenergic receptors, including β2-adrenergic receptor (β2AR). Interestingly, in experimental models, chronic use of β2AR agonists can augment mucus secretion. Thus, in the present study, we tested the hypothesis that air

HucMSCs-derived Exosomes Protect Against 6-hydroxydopamineinduced Parkinson's Disease in Rats by Inhibiting Caspase-3 Expression and Suppressing Apoptosis.

OBJECTIVE: Parkinson's disease (PD) is a progressive neurodegenerative disorder with symptoms including tremor and bradykinesia, while traditional dopamine replacement therapy and hypothalamic deep brain stimulation can temporarily relieve patients' symptoms, they cannot cure the disease. Hence, discovering new methods is crucial to designing more effective therapeutic approaches to address the condition. In our previous study, we found that exosomes (Exos) derived from human umbilical cord mesenchymal stem cells (hucMSCs) repaired a PD model by inducing dopaminergic neuron autophagy and inhib
