Oxidation of palmitate by Mycobacterium lepraemurium isolated from C3H mice lepromata (in vivo) and also grown on Ogawa egg-yolk medium (in vitro) was investigated. Palmitate was found to be oxidized, after a lag period of about 8 h, by both the in vivo and in vitro grown bacilli. Cell-free extracts prepared from in vivo and in vitro grown cells catalysed an active oxidation of palmitate after a lag period of 3-4 h. Absorption of sodium [1-14C]glycolate by rat intestine was studied by using the tissue accumulation technique with everted intestinal rings. Saturation kinetics was observed for the absorption of glycolate in the jejunoileal region, with a Km of 6.25 mM for glycolate and a Vmax of 5.56 mumole/30 min/g wet wt. Using an improved segmented flow technique the uptake of salicylate in low concentration (1-3 mM) by rat jejunum is found to be steady for ca. 30 min, and is proportional to lumen concentration. At higher lumen concentrations (6-8 mM) the rate of uptake falls from the beginning to very low levels. Cachectin/tumor necrosis factor (TNF) one of the cytokines secreted by reticuloendothelial cells and lymphocytes, evokes a multitude of biological effects in different cells. Although the effects of TNF on adipocytes generally appear rather slowly, we have found that TNF almost instantaneously stimulates respiration and respiration-coupled ATP synthesis in 30A-5 preadipocytes. This novel TNF effect might be one of the essential early steps in the cachectic mobilization of energy reserves associated with TNF action in vivo.
