Metallo beta lactamases in Pseudomonas aeruginosa and Acinetobacter species.

The multi drug resistant gram negative bacteria especially Pseudomonas aeruginosa and Acinetobacter species are on the rise. The major defense in these bacteria against beta-lactam antibiotics is production of metallo beta lactamases (MBLs) which degrade this group of antibiotics including carbapenems. Till now five main types of MBLs have been described throughout the World--IMP, VIM, SPM, GIM and SIM. A new MBL has been recently reported in P. aeruginosa from Australia--bla AIM-1. There are no standard guidelines by CLSI for detection of these enzymes in various bacteria. A number of phenoty

Genes involved in intrinsic antibiotic resistance of Acinetobacter baylyi.

Bacterial genes defining intrinsic resistance to antibiotics encode proteins that can be targeted by antibiotic potentiators. To find such genes, a transposon insertion library of Acinetobacter baylyi was screened with subinhibitory concentrations of various antibiotics to find supersusceptible mutants. A DNA microarray printer was used to replica plate 10,000 individual library clones to select mutants unable to grow at 1/10 the MICs of 12 different antibiotics. Transposon insertions in 11 genes were found to cause an eightfold or higher hypersusceptibility to at least one antibiotic. Most of

Experimental infection of human body lice with Acinetobacter baumannii.

The human body louse is currently recognized as a vector of Rickettsia prowazekii, Borrelia recurrentis, and Bartonella quintana. Previous studies have reported the isolation of Acinetobacter baumannii from the body lice of homeless patients. To study how the body louse acquires A. baumannii, we infected a rabbit by infusing 2 x 10(6) colony-forming units of the louse strain of A. baumannii. Two hundred body lice were infected by feeding on the bacteremic rabbit and compared with 200 uninfected lice and two groups of 200 lice feeding on rabbits infected either with another strain of A. baumann

Corneal perforation due to Acinetobacter junii: a case report.

Acinetobacter spp. is emerging as a common cause of nosocomial infections. Community acquired ocular infections due to Acinetobacter are rare. Only one case of perforation of cornea has been reported previously, where old nomenclature was used to describe the causal agent. We report a case of corneal perforation due to Acinetobacter junii for which a therapeutic penetrating keratoplasty was conducted and the patient eventually recovered.

Catechol 1,2-dioxygenase from Acinetobacter calcoaceticus: purification and properties.

Procedures for the purification of catechol 1,2-dioxygenase from extracts of Acinetobacter calcoaceticus strain ADP-96 are described. The purified enzyme was homogeneous as judged by ultracentrifugation and acrylamide gel electrophoresis. The enzyme contained 2 g-atoms of iron per mol of protein. The enzyme had a broad substrate specificity and catalyzed the oxidation of catechol, 4-methylcatechol, 3-methylcatechol, and 3-isopropyl catechol. The activity of the enzyme was inhibited by heavy metals, sulfhydryl inhibitors, and substrate analogues. The molecular weight of the enzyme was 85,000 as

In silico metabolic profiling of non-baumannii Acinetobacter species uncovers conserved functions and provides first evidence of siderophore biosynthetic genes in Acinetobacter junii.

The genus Acinetobacter is recognized for its metabolic versatility, which contributes to environmental persistence, virulence, and antimicrobial resistance. This study aimed to elucidate the metabolic pathways, particularly those associated with virulence and antimicrobial resistance (AMR), in non-baumannii Acinetobacter species. Genome-scale metabolic prediction of 19 Acinetobacter isolates identified 104 distinct pathways spanning 32 metabolic categories. Ninety-six pathways were conserved across all genomes, whereas a small subset exhibited species- or strain-specific distributions: four p

"In vitro evaluation of sulbactam combination therapies for Acinetobacter baumannii".

BACKGROUND AND OBJECTIVES: Acinetobacter baumannii, a leading cause of hospital-acquired infections in critically ill patients, is increasingly resistant to carbapenems, leading WHO to designate carbapenem-resistant Acinetobacter baumannii (CRAB) as a critical priority pathogen. While the 2024 IDSA guidelines recommend sulbactam-based regimens as the cornerstone of therapy, high-dose sulbactam is effective only against isolates with MICs up to 16-32 μg/mL. This study aimed to describe the sulbactam MIC distribution and to evaluate MIC reduction and synergy when combined with β-lactam antibioti

The glutathione pathway is required for biofilm formation in Acinetobacter baumannii.

Acinetobacter baumannii is a Gram-negative nosocomial bacterium that is a member of the ESKAPE group of pathogens, notable for its virulence and intrinsic antibiotic resistance. It causes diverse infections, including respiratory and soft tissue disease, that are increasingly difficult to treat. Glutathione (GSH), the major intracellular redox buffer, is known in other bacteria to protect against stress and influence physiological processes such as biofilm formation. To investigate the roles of GSH in A. baumannii, we analyzed transposon mutant strains lacking gshA and gshB genes, which encode

Genomic epidemiology of carbapenem resistance in Acinetobacter baumannii in Chile.

BACKGROUND: Carbapenem-resistant Acinetobacter baumannii (CRAb) is a World Health Organization (WHO) critical priority pathogen, yet genomic data from Chile are scarce. Here, we investigated the genetic diversity of Chilean isolates, focusing on molecular epidemiology, carbapenemase genes, and mobile genetic elements. METHODS: We analysed 83 A. baumannii genomes (42 newly sequenced, 41 public). Newly sequenced isolates underwent antimicrobial susceptibility testing, and whole-genome sequencing was performed using Illumina, with hybrid assemblies for two CRAb strains. Genomes were assembled an

Roles of GacSA and DJ41_1407 in Acinetobacter baumannii ATCC 19606.

Two-component systems (TCSs) in bacteria are often involved in the global regulation of various physiological activities and behaviours. This study investigated the GacSA TCS and DJ41_1407 transcriptional sensor adjacent to GacA in Acinetobacter baumannii ATCC 19606. The relationship between GacS, GacA, and DJ41_1407 and their functions and signal transduction mechanisms are described. A. baumannii ATCC 19606 mutants, ∆gacS, ∆gacA, and ∆DJ41_1407, were generated using markerless mutation and cultured in LB medium, then collected for RNA sequencing. It was found that GacS, GacA, and DJ41_1407 r

Arginine catabolism is essential to polymyxin dependence in Acinetobacter baumannii.

Polymyxins are often the only effective antibiotics against the "Critical" pathogen Acinetobacter baumannii. Worryingly, highly polymyxin-resistant A. baumannii displaying dependence on polymyxins has emerged in the clinic, leading to diagnosis and treatment failures. Here, we report that arginine metabolism is essential for polymyxin-dependent A. baumannii. Specifically, the arginine degradation pathway was significantly altered in polymyxin-dependent strains compared to wild-type strains, with critical metabolites (e.g., L-arginine and L-glutamate) severely depleted and expression of the ast

Regulation of tricarboxylate transport and metabolism in Acinetobacter baylyi ADP1.

Despite the significant presence of plant-derived tricarboxylic acids in some environments, few studies detail the bacterial metabolism of trans-aconitic acid (Taa) and tricarballylic acid (Tcb). In a soil bacterium, Acinetobacter baylyi ADP1, we discovered interrelated pathways for the consumption of Taa and Tcb. An intricate regulatory scheme tightly controls the transport and catabolism of both compounds and may reflect that they can be toxic inhibitors of the tricarboxylic acid cycle. The genes encoding two similar LysR-type transcriptional regulators, TcuR and TclR, were clustered on the

An ionophore breaks the multi-drug-resistance of Acinetobacter baumannii.

Within intensive care units, multi-drug resistant Acinetobacter baumannii outbreaks are a frequent cause of ventilator-associated pneumonia. During the on-going COVID-19 pandemic, patients who receive ventilator support experience a 2-fold increased risk of mortality when they contract a secondary A. baumannii pulmonary infection. In our recent paper (De Oliveira et al. (2022), Mbio, doi: 10.1128/mbio.03517-21), we demonstrate that the 8-hydroxquinoline ionophore, PBT2 breaks the resistance of A. baumannii to tetracycline class antibiotics. In vitro, the combination of PBT2 and zinc with eithe

Vaccinomics to Design a Multi-Epitopes Vaccine for Acinetobacter baumannii.

Antibiotic resistance (AR) is the result of microbes' natural evolution to withstand the action of antibiotics used against them. AR is rising to a high level across the globe, and novel resistant strains are emerging and spreading very fast. Acinetobacter baumannii is a multidrug resistant Gram-negative bacteria, responsible for causing severe nosocomial infections that are treated with several broad spectrum antibiotics: carbapenems, β-lactam, aminoglycosides, tetracycline, gentamicin, impanel, piperacillin, and amikacin. The A. baumannii genome is superplastic to acquire new resistant mecha
