HBB contributes to individualized aconitine-induced cardiotoxicity in mice via interfering with ABHD5/AMPK/HDAC4 axis.

The root of Aconitum carmichaelii Debx. (Fuzi) is an herbal medicine used in China that exerts significant efficacy in rescuing patients from severe diseases. A key toxic compound in Fuzi, aconitine (AC), could trigger unpredictable cardiotoxicities with high-individualization, thus hinders safe application of Fuzi. In this study we investigated the individual differences of AC-induced cardiotoxicities, the biomarkers and underlying mechanisms. Diversity Outbred (DO) mice were used as a genetically heterogeneous model for mimicking individualization clinically. The mice were orally administere

Synthesis, Pharmacological Evaluation, and Molecular Modeling of Lappaconitine-1,5-Benzodiazepine Hybrids.

Diterpenoid alkaloids, originating from the amination of natural tetracyclic diterpenes, have long interested scientists due to their medicinal uses and infamous toxicity which has limited the clinical application of the native compound. Alkaloid lappaconitine extracted from various Aconitum and Delphinium species has displayed extensive bioactivities and active ongoing research to reduce its adverse effects. A convenient route to construct hybrid molecules containing diterpenoid alkaloid lappaconitine and 3H-1,5-benzodiazepine fragments was proposed. The key stage involved the formation of 5'

JI-CJ002 and Dabrafenib Combination Enhances Antitumor Activity in Melanoma Associated With the Downregulation of B7-H3.

Melanoma is the most lethal form of skin cancer, with a significantly poor prognosis after metastasis. The BRAF inhibitor dabrafenib increases response rates and overall survival (OS) in advanced-stage melanoma; however, most patients develop resistance rapidly, highlighting the need for improved combination regimens. This study investigated the therapeutic efficacy of JI-CJ002, an herbal extract consisting of Angelica gigas, Aconitum carmichaeli, and Zingiber officinale blended in a 2:1:3 (w/w) ratio, and assessed the combined antineoplastic effects of JI-CJ002 with dabrafenib in melanoma. Design, Synthesis, Antifeedant Activity, and Mechanistic Research of Aconitine Derivatives.

For decades, natural product-derived insect repellents have been a major focus of research. Aconitine, a diterpene alkaloid isolated from Aconitum plants, exhibits not only significant antifeedant activity but also potent toxicological effects against certain insects. In this study, 48 novel aconitine derivatives were synthesized via esterification of the C3-hydroxyl group. Among them, compound A4 demonstrated optimum antifeedant activity with an AFC50 value of 4.91 µg/cm2, surpassing that of the parent compound aconitine (AFC50 = 11.65 µg/cm2) and the positive control Azadirachtin (AFC50 = 5.

Aconitine induces mitochondrial energy metabolism dysfunction through inhibition of AMPK signaling and interference with mitochondrial dynamics in SH-SY5Y cells.

Aconitine, a highly toxic alkaloid derived from Aconitum L., affects the central nervous system and peripheral nervous system. However, the underlying mechanism of aconitine-induced neurotoxicity remains unclear. This study investigates the effects and mechanism of aconitine on mitochondrial energy metabolism in SH-SY5Y cells. Results demonstrated that aconitine exposure suppressed cell proliferation and led to an increase in reactive oxygen species (ROS) and excessive lactate dehydrogenase (LDH) release. Aconitine (400 μmol/L) induced abnormal mitochondrial energy metabolism that quantified b

Sexually concordant selection on floral traits despite greater opportunity for selection through male fitness.

Pollinators are important drivers of floral trait evolution, yet plant populations are not always perfectly adapted to their pollinators. Such apparent maladaptation may result from conflicting selection through male and female sexual functions in hermaphrodites. We studied sex-specific mating patterns and phenotypic selection on floral traits in Aconitum gymnandrum. After genotyping 1786 offspring, we partitioned individual fitness into sex-specific selfed and outcrossed components and estimated phenotypic selection acting through each. Relative fitness increased with increasing mate number,

Attenuation and Structural Transformation of Crassicauline A During Sand Frying Process and Antiarrhythmic Effects of its Transformed Products.

To ensure safety and efficacy, most Aconitum herbs should be processed before clinical application. The processing methods include boiling, steaming, and sand frying. Among these methods, the transformation pathways of diterpenoid alkaloids in the process of sand frying are more complicated. Therefore, crassicauline A, a natural product with two ester bonds, was chosen as the experimental object. Consequently, a known alkaloid, together with three new alkaloids, was derived from crassicauline A. Meanwhile, the cardiotoxicity of converted products was reduced compared with their parent compound
