BACKGROUND: Advanced maternal age (AMA) is associated with adverse pregnancy outcomes, but the mechanisms linking AMA to fetal overgrowth remain poorly quantified. This retrospective cohort study aimed to estimate the potential statistical mediating roles of prenatal BMI and blood lipids in the association between AMA and large-for-gestational-age (LGA) infants.
METHODS: In this retrospective cohort study, 11,504 singleton pregnancies (28-41 weeks) in Changzhou, China, were analyzed. Women were classified as AMA (≥35 years, n=1,330) or non‑AMA (n=10,174). Multivariable regression and mediatio

BACKGROUND: The biologic implications of delayed parenthood have been blamed for a major public health crisis in the United States, that includes high rates of neonatal morbidity and mortality (NMM). The objective of this study was to evaluate the risk of parent age on NMM and to provide results that can serve as a starting point for more specific mediation modeling.
METHODS: Data containing approximately 15,000,000 birth records were obtained from the United States Natality database for the years 2014 to 2018. A Bayesian modeling approach was used to estimate the both the total effect and th

ABSTRACT: This single-centre retrospective study evaluated whether blastocyst morphology remains an independent predictor of euploidy and implantation rates in an advanced maternal age (AMA) population after controlling for chromosomal status. Analysing 1,067 biopsied blastocysts from 308 IVF cycles (269 patients) between October 2021 and December 2022, embryos were graded via the Gardner and Schoolcraft system and categorised into Excellent, Good, Average, and Poor groups. The overall euploidy rate was 33.62%, with blastocysts of the Excellent group exhibiting a significantly higher euploidy

Advanced maternal age (≥35 years) is increasingly common and associated with a higher risk of threatened abortion, yet its molecular basis remains unclear. To elucidate the underlying mechanisms of advanced maternal age-related threatened abortion (AMA-TA) by integrating metabolomics and transcriptomics. Untargeted serum metabolomics was performed in nine AMA-TA patients and seven age-matched healthy pregnant women. An AMA-TA mouse model was established using mifepristone (4 mg/kg) to assess embryo resorption, placental morphology, and serum hormone levels. Serum metabolomics and placental tra

INTRODUCTION: Pregnancies at very and extremely advanced maternal age (VAMA, 45-49 years and EAMA, ≥50 years) are increasingly common, particularly regarding the influence of assisted reproductive technology (ART), yet their obstetric and perinatal outcomes remain underexplored. This study aimed to investigate maternal and neonatal outcomes in singleton pregnancies among women aged ≥45 years, with a specific focus on ART conception and oocyte source.
MATERIAL AND METHODS: This multicenter retrospective cohort study included women aged ≥45 years with singleton pregnancies delivered ≥22 weeks'

BACKGROUND: In the entire population, an aberrant right subclavian artery (ARSA) is closely associated with chromosomal abnormalities. ARSA with additional ultrasonic findings would increase risk of chromosomal abnormalities. The risk of fetal chromosomal abnormalities increased exponentially with the maternal age. These risks in the advanced maternal age (AMA) group are uncertain. This study aimed to determine the incidence of ARSA in Chinese AMA and non-AMA women and the frequency of aneuploidy among AMA and non-AMA women with ARSA.
METHODS: This retrospective study included 13,690 singleto

BACKGROUND: Prenatal alcohol exposure (PAE) can result in developmental defects that include growth restriction, craniofacial anomalies, and cognitive behavioral deficits, though the presence and severity of these adverse outcomes can vary dramatically among exposed individuals. Preclinical animal models have demonstrated that the dose and timing of PAE account for much, but not all, of this phenotypic variation, suggesting that additional factors mitigate the effects of PAE. Here, we used a mouse model to investigate whether maternal age modulates the effects of PAE on the severity and variat

The aim of this study was to assess the effect of advanced maternal age on pregnancy and neonatal outcomes in patients attending a tertiary centre hospital. Between January 2013 and December 2016, the records of all patients who were referred for pregnancy follow-ups and delivery were retrospectively reviewed and were divided according to their parity and age. Patients over 35 years old were categorised as advanced maternal age; (1) 35-40 years old. (2) 40-45 years old. (3) 45 years and over. Most of the prenatal complications were found to increase in the advanced maternal age group. The caes

OBJECTIVE: Prenatal cytogenetic diagnostic methods for the diagnosis of fetal chromosomal anomalies have been used reliably over the last 40 years. Advanced maternal age has become a basic indication for amniocentesis.
METHODS: We examined the results of the chromosome analyses of 3485 women that had amniocentesis for any reason during their antenatal care in our perinatology clinic in 2007-2009. Amniocentesis was performed for advanced maternal age in 1456 women (41.8%) and for other reasons in the remaining 2029 women (58.2%). Chromosomal anomalies were examined numerically and structurally

BACKGROUND: As the proportions of older women giving birth increase, there is a growing body of evidence on the increased risks of poorer maternal and perinatal outcomes for this group. However, the associations are not completely understood. This study aimed to establish the prevalence of selected maternal morbidities and examine whether advanced maternal age is associated with a higher risk of morbidity for women giving birth in Victoria.
METHOD: Data on all births over 20 weeks‧ gestation for 2005 and 2006 were obtained from the Victorian Perinatal Data Collection. Unadjusted and adjusted

BACKGROUND: Preeclampsia is a frequent syndrome and its cause has been linked to multiple factors, making prevention of the syndrome a continuous challenge. One of the suggested risk factors for preeclampsia is advanced maternal age. In the Western countries, maternal age at first delivery has been steadily increasing, yet few studies have examined women of advanced maternal age with preeclampsia. The purpose of this registry-based study was to compare the obstetric outcomes in primiparous and preeclamptic women younger and older than 35 years.
METHODS: The registry-based study used data from

OBJECTIVE: To examine the impact of gestational diabetes mellitus(GDM) on perinatal outcome in a setting where influences of maternal age and obesity would be minimal.
RESEARCH DESIGN AND METHODS: A case-control study was done to compare the outcome of pregnancy in 65 women with GDM and 153 women with normal carbohydrate metabolism matched for age, height, and prepregnancy weight.
RESULTS: The frequencies of preeclampsia and primary cesarean sections were higher and delivery was earlier in pregnancies complicated by GDM. Birth weight, symmetry index, and chest circumference were greater, and

PURPOSE: Ubiquitination represents a promising therapeutic target for age-associated diseases, yet its functional role in ovarian aging remains poorly defined. This study systematically characterizes ubiquitinated proteins in luteinized granulosa cells (GCs) from women of advanced maternal age (AMA), aiming to provide molecular insights into the ubiquitination-dependent regulatory network underlying ovarian aging and further elucidate the mechanisms governing energy metabolism reprogramming in senescent GCs.
METHODS: A total of 60 patients undergoing their first in vitro fertilization/intracy

BACKGROUND: Advanced maternal age (AMA) threatens offspring brain development, yet its neurodevelopment-disrupting mechanisms remain poorly understood. Our prior work has suggested that AMA may impair myelin development in offspring, accompanied by increased Dnmt3a expression and DNA methylation levels in the hippocampus. This study explores whether Dnmt3a mediates myelination defects in offspring of AMA rats.
METHODS: Lentiviral vectors were intracerebroventricularly administered to modulate Dnmt3a expression in AMA offspring after birth. Protein expression was analyzed by western blotting a

Natural ovarian aging is one of the major causes for declining fertility in female animals, which has become an insurmountable issue in human reproduction clinics and assisted reproductive technology procedures. Nevertheless, the molecular basis of oocyte aging remains poorly understood, and feasible improvement strategies are unavailable. In the present study, in vivo supplementation of pyrroloquinoline-quinone effectively elevated the fecundity of reproductively aged mice by balancing hormonal secretion, harmonizing the estrus cycle, and eliminating ovarian fibrosis. Moreover, oocyte quality

The decision to postpone parenting has gained momentum in recent years, a shift driven by evolving social dynamics and improved access to fertility treatments. Despite their increasing prevalence, pregnancies at advanced maternal ages are associated with increased risks of adverse maternal and neonatal outcomes. This article addresses the association between advanced maternal age and hypertensive disorders of pregnancies (HDPs), which are more prevalent and a significant cause of maternal morbidity and mortality in this population. This review explores the biological mechanisms and age-related

We aimed to explore the association of BMI in pre-pregnant women with metabolic syndrome in pregnancy in advanced maternal age. A total of 229 maternal women and 536 maternal women participated in this study. Pregnancy women underwent a 75 g-oral glucose tolerance test and maternal lipid profile test between 24 and 28 weeks. Data about biological and sociodemographic characteristics were recorded for each case. The metabolic equivalent (Met) was 9.6% in the maternal age ≥35 group, 5.4% in the age 20-34 group (P = 0.027), and 6.7% in all pregnant women. Results also demonstrated that gestationa
