Development and Certification of a Reference Material for Aflatoxins and Zearalenone in Corn/Peanut Blended Vegetable Oil.

A certified reference material (CRM) for aflatoxins (AFTB1, AFTB2, AFTG1, AFTG2) and zearalenone (ZEN) in corn/peanut blended vegetable oil (GBW(E)100863) was developed to address the critical need for matrix-specific reference materials in mycotoxin analysis. The CRM was prepared by blending naturally contaminated corn and peanut oils, followed by homogenization, sterilization, and sub-packing. Homogeneity and stability studies were conducted using high-performance liquid chromatography isotope-dilution tandem mass spectrometry with a dilute-and-shoot pretreatment process. The CRM demonstrate

Activated carbon co-modified by chitosan and SDS for targeted removal of aflatoxins from fragrant peanut oil.

The pore size structure and surface structure of activated carbon (AC) may be ameliorated using the hydrophobic groups and negative surface charge of sodium dodecyl sulfate (SDS), along with the abundant hydroxyl and amine groups of chitosan (CS). In this study, a composite adsorbent (CS/SDS/AC) created through co-modification of AC with CS and SDS was employed to remove aflatoxins from fragrant peanut oil. CS/SDS/AC exhibited a superior adsorption capacity (89.9 %) for adsorbing aflatoxins. More significantly, CS/SDS/AC improved the retention of sterols (64-99.3 %), tocopherols (61.4-82.7 %),

Aflatoxins screening in non-dairy beverages by Mn-doped ZnS quantum dots - Molecularly imprinted polymer fluorescent probe.

Synthesized molecularly imprinted polymer (MIP) materials have been anchored on the surface of PEG-Mn-doped ZnS quantum dots (QDs) to develop a fluorescence probe for aflatoxins (AFs) recognition/determination in non-dairy beverages. MIP synthesis used 5,7-dimethoxycoumarin (DMC) as a dummy template molecule, and methacrylic acid (MAA) as a functional monomer. Under optimized conditions (1.25 mL of 5 mg L-1 MIP-PEG-ZnS QDs solution, pH 5.0, and 12 min delay time before scanning), the prepared MIP-QDs composite was found to offer high affinity and selectivity for AFs (AFB1, AFB2, AFG1 and AFG2)

From Toxicity to Therapy: Probiotics and Novel Adsorbents in Aflatoxin Detoxification.

Aflatoxins (AFs), toxic secondary metabolites primarily produced by Aspergillus flavus and A. parasiticus, remain a serious global health challenge due to both acute and particularly chronic toxicity. Unlike acute toxicity, chronic low-dose exposure to AFs is insidious and cumulative, leading to long-term effects such as immune dysfunction, growth retardation, and carcinogenesis. These outcomes are often underestimated, underscoring the importance of preventive strategies over therapeutic interventions. Although pharmacological agents have been investigated to inhibit CYP450-mediated bioactiva

Aflatoxin Exposure and Human Health with a Focus on Northern Latin America.

Aflatoxins, mycotoxins produced by Aspergillus flavus and Aspergillus parasiticus, were discovered sixty-five years ago and remain a significant public health threat, particularly amid increasing instances of extreme weather events. Of the four principal forms of aflatoxins found in foods (B1, B2, G1, and G2), aflatoxin B1 is the most potent carcinogen. Aflatoxins commonly contaminate a variety of foodstuffs, with maize being among the most susceptible. Chronic exposure to aflatoxins has been linked to liver cancer, childhood stunting, gallbladder cancer, and other adverse health effects. Due

Aflatoxin Exposure in Immunocompromised Patients: Current State and Future Perspectives.

Aflatoxins (AFs), harmful secondary metabolites produced by the genus Aspergillus, particularly Aspergillus flavus and Aspergillus parasiticus, are one of the best-known potent mycotoxins, posing a significant risk to public health. The primary type, especially aflatoxin B1 (AFB1), is a potent carcinogen associated with liver cancer, immunosuppression, and other health problems. Environmental factors such as high temperatures, humidity, and inadequate storage conditions promote the formation of aflatoxin in staple foods such as maize, peanuts, and rice. Immunocompromised individuals, including

Aflatoxin profiles of Aspergillus flavus isolates in Sudanese fungal rhinosinusitis.

Aspergillus flavus is a commonly encountered pathogen responsible for fungal rhinosinusitis (FRS) in arid regions. The species is known to produce aflatoxins, posing a significant risk to human health. This study aimed to investigate the aflatoxin profiles of A. flavus isolates causing FRS in Sudan. A total of 93 clinical and 34 environmental A. flavus isolates were studied. Aflatoxin profiles were evaluated by phenotypic (thin-layer and high-performance chromatography) and genotypic methods at various temperatures and substrates. Gene expression of aflD and aflR was also analyzed. A total of

Effects of aflatoxin B1 on the cell cycle distribution of splenocytes in chickens.

Congenital generalized lipodystrophy type 4 (CGL4) is a rare disease caused by mutations in the gene polymerase I and transcript release factor (PTRF), the main symptoms of which are systemic reductions in adipose tissue and muscular dystrophy. The strategy of treating CGL4 is to improve the insulin resistance and hypertriglyceridemia that result from systemic reductions in adipose tissue. Metreleptin, a synthetic analog of human leptin, is effective against generalized lipodystrophies; however, there are no reports of the use of metreleptin in the treatment of CGL4. Herein, we discuss the tre

Aflatoxin B₁-Induced Developmental and DNA Damage in Caenorhabditis elegans.

Aflatoxin B₁ (AFB₁) is a ubiquitous mycotoxin produced by toxicogenic Aspergillus species. AFB₁ has been reported to cause serious adverse health effects, such as cancers and abnormal development and reproduction, in animals and humans. AFB₁ is also a potent genotoxic mutagen that causes DNA damage in vitro and in vivo. However, the link between DNA damage and abnormal development and reproduction is unclear. To address this issue, we examined the DNA damage, germline apoptosis, growth, and reproductive toxicity following exposure to AFB₁, using Caenorhabditis elegans as a study model. Results

Impact of aflatoxin B1 on the pharmacokinetic disposition of enrofloxacin in broiler chickens.

The potential impact of subchronic exposure of aflatoxin B1 was investigated on the pharmacokinetic disposition of enrofloxacin in broiler chickens. Broiler chickens given either normal or aflatoxin B1 (750μg/kg diet) supplemented diet for 6 weeks received a single oral dose of enrofloxacin (10mg/kg body wt). Blood samples were drawn from the brachial vein at predetermined time intervals after drug administration. Enrofloxacin plasma concentrations analyzed by RP-HPLC were significantly lower in aflatoxin B1-exposed broiler chickens at 0.167, 0.5 and 1.0h after drug administration. In aflatoxi

Assessment of workers' exposure to aflatoxin B1 in a Portuguese waste industry.

Aflatoxin B1 (AFB1) is considered by different International Agencies as a genotoxic and potent hepatocarcinogen. However, despite the fact that the fungi producing this compound are detected in some work environments, AFB1 is rarely monitored in occupational settings. The aim of the present investigation was to assess exposure to AFB1 of workers from one Portuguese waste company located in the outskirt of Lisbon. Occupational exposure assessment to AFB1 was done with a biomarker of internal dose that measures AFB1 in the serum by enzyme-linked immunosorbent assay. Forty-one workers from the w

Global burden of aflatoxin-induced hepatocellular carcinoma: a risk assessment.

BACKGROUND: Hepatocellular carcinoma (HCC), or liver cancer, is the third leading cause of cancer deaths worldwide, with prevalence 16-32 times higher in developing countries than in developed countries. Aflatoxin, a contaminant produced by the fungi Aspergillus flavus and Aspergillus parasiticus in maize and nuts, is a known human liver carcinogen. OBJECTIVES: We sought to determine the global burden of HCC attributable to aflatoxin exposure. METHODS: We conducted a quantitative cancer risk assessment, for which we collected global data on food-borne aflatoxin levels, consumption of aflatox

Supercritical fluid extraction of aflatoxin B(1) from soil.

This research describes the development of a supercritical fluid extraction (SFE) method to recover aflatoxin B(1) from fortified soil. The effects of temperature, pressure, modifier (identity and percentage), and extraction type were assessed. Using the optimized SFE conditions, the mean recovery from air dried soil was 72%. The variables associated with changes in recovery of aflatoxin were co-solvents, static extraction, and temperature. Acetonitrile-2% acetic acid, used both in-cell and on-line, provided the most efficient recovery. The results indicate that desorption from the soil was th

Reducing human exposure to aflatoxin through the use of clay: a review.

Innovative sorption strategies for the detoxification of aflatoxins have been developed. NovaSil clay (NS) has been shown to prevent aflatoxicosis in a variety of animals when included in their diet. Results have shown that NS clay binds aflatoxins with high affinity and high capacity in the gastrointestinal tract, resulting in a notable reduction in the bioavailability of these toxins without interfering with the utilization of vitamins and other micronutrients. This strategy is being evaluated as a potential remedy for acute aflatoxicosis, and as a sustainable human intervention for aflatoxi

In vivo identification of aflatoxin-induced free radicals in rat bile.

Aflatoxin B1 (AFB1) is a potent hepatocarcinogen. We have recently detected [via electron spin resonance (ESR) spectroscopy] free radicals in vivo in rat bile following AFB1 metabolism using the spin trapping [alpha-(4-pyridyl-1-oxide)-N-tert-butyl nitrone (4-POBN)] technique. The aim of the present study was to identify the trapped free radical intermediates from the in vivo hepatic metabolism of AFB1. Rats were treated simultaneously with AFB1 (3 mg/kg i.p.) and the spin trapping agent 4-POBN (1 g/kg i.p.), and bile was collected over a period of 1 h at 20 min intervals. On-line high perform

Aflatoxin levels in corn available as wild turkey feed in Georgia.

Samples of corn available as wildlife feed from retailers throughout Georgia (USA) were collected during April 1997 and analyzed for aflatoxin to determine if levels harmful to wild turkeys (Meleagris gallopavo) were present. Three of 31 (10%) samples collected from a 40-country area were positive. An enzyme-linked immunosorbent assay qualitatively determined that two samples contained from 0 to 20 ppb aflatoxin. A chromatography analysis of a third sample measured 380 ppb total aflatoxin. A small percentage of our sample of wildlife feed collected during one season contained levels of aflatox

Systemic aspergillosis caused by an aflatoxin-producing strain of Aspergillus flavus.

The first case of human systemic infection by an Aspergillus flavus isolate demonstrated to produce aflatoxins in vitro and in vivo is described. The patient, a 41-year-old man with acute myelogenous leukaemia, developed a complication of suspected pulmonary Aspergillus infection during remission induction therapy. Antifungal chemotherapy brought about a considerable degree of improvement, but remission of the underlying disease was not attained. Bone marrow transplantation was also not effective. The patient showed recovery from neutropenia but died despite aggressive antifungal chemotherapy.
