Protocol for generating protoplasts of Trichoderma koningiopsis and transformation of plasmid DNA.

Trichoderma koningiopsis is a filamentous fungus that produces numerous bioactive molecules and has the potential to be used as a biological control and plant growth-promoting agent. Here, we present a protocol for generating protoplasts of T. koningiopsis. We then describe four techniques for the stable transformation of plasmid DNA: polyethylene glycol (PEG)-mediated transformation of protoplasts, Agrobacterium-mediated transformation, electroporation of spores, and lipofection of mycelia. We then detail procedures for confirming the expression of a spCas9 gene using fluorescence microscopy.

An integrated green control strategy for plant viral co-infections: from multivalent vaccine construction to synergistically modified nanodelivery systems.

Plant viral co-infections pose a significant threat to global agricultural productivity. In this study, a novel multivalent attenuated vaccine (CMV-R2TP) was developed using the cucumber mosaic virus (CMV) as a backbone, together with a ROS-responsive vaccine nanodelivery system (pCMV-R2TP@PTP-TAT), offering a comprehensive and environmentally friendly strategy for managing plant viruses. Through the modification of the CMV 2b protein, a stable base vector (pR2-2bIII) capable of accommodating up to 400 bp foreign fragments was constructed. The resulting CMV-R2TP vaccine demonstrated protective

Biologically safe restoration of heavy metal-contaminated soils through plant-microbe synergy.

Abandoned mines have created extensive idle areas contaminated with heavy metals (HMs). Conventional remediation methods are often costly, environmentally disruptive, and pose risks to human health. As a sustainable alternative, a biological approach utilizing metal-tolerant plant growth-promoting bacteria (mPGPBs) was employed to remediate HM-contaminated soils and assess their biological safety. Five mPGPB strains-Pseudarthrobacter sp., Pseudomonas sp., and Agrobacterium sp.-were isolated from contaminated mine soils and screened for metal tolerance and plant growth-promoting capabilities. I

Structural and kinetic properties of lumazine synthase isoenzymes in the order Rhizobiales.

6,7-Dimethyl-8-ribityllumazine synthase (lumazine synthase; LS) catalyzes the penultimate step in the biosynthesis of riboflavin in plants and microorganisms. This protein is known to exhibit different quaternary assemblies between species, existing as free pentamers, decamers (dimers of pentamers) and icosahedrally arranged dodecamers of pentamers. A phylogenetic analysis on eubacterial, fungal and plant LSs allowed us to classify them into two categories: Type I LSs (pentameric or icosahedral) and Type II LSs (decameric). The Rhizobiales represent an order of alpha-proteobacteria that includ

Targeting microbial pathogens by expression of new recombinant dermaseptin peptides in tobacco.

Dermaseptin B1 (DrsB1), an antimicrobial cationic 31 amino acid peptide, is produced by Phyllomedusa bicolor. In an attempt to enhance the antimicrobial efficacy of DrsB1, the DrsB1 encoding 93 bp sequence was either fused to the N or C terminus of sequence encoding chitin-binding domain (CBD) of Avr4 gene from Cladosporium fulvum. Tobacco leaf disk explants were inoculated with Agrobacterium rhizogenes harboring pGSA/CBD-DrsB1 and pGSA/DrsB1-CBD expression vectors to produce hairy roots (HRs). Polymerase chain reaction (PCR) was employed to screen putative transgenic tobacco lines. Semi-quant

Development of Rhizobium rhizogenes-induced hairy root cultures of Vitex negundo for vitexin production.

Vitex negundo L. is a significant indigenous medicinal plant with diverse therapeutic potentials owing to its rich reservoir of bioactive compounds including sitosterol, vitexoside, agnuside, isovitexin and vitexin which are predominantly present in the root. To induce hairy roots, explants of V. negundo L. were transformed using two strains of Rhizobium rhizogenes: MTCC 532 and MTCC 2364. Among them, the MTCC 2364 strain exhibited the highest transformation efficiency (94.44%). Genetic transformation of the induced hairy roots was confirmed by PCR amplification using primers specific for the

Type IV transporters of pathogenic bacteria.

Type IV transporters are produced by several bacterial pathogens such as Agrobacterium tumefaciens, Bordetella pertussis, Brucella spp., Bartonella henselae, Helicobacter pylori and Legionella pneumophila. These transporters are critical for the pathogenic process in that they export important virulence factors across the membranes of the bacteria. Although the virulence factors that are exported by these transporters can be either nucleic acid or protein, the general mechanism of transport appears to be similar for members of this family. Recent findings have shed light on the architecture of

Insect-resistant tobacco plants expressing insect-specific neurotoxin AaIT.

The recombinant plant expression vector pNGY-2 with designed and synthesized AaIT gene had been constructed. The AaIT gene was fused behind the sequence of TMV and inserted into expression vector under the control of two linked 35s promoters. The recombinant plasmid pNGY-2 was transferred into tobacco NC89 by agrobacterium-mediated transfer system. The GUS activity analysis and Southern blotting of regenerated plants indicated that AaIT gene had been integrated into tobacco genome. Insect bioassays showed that some transgenic plants had notable insect-resistant activity.

DC-Analyzer-facilitated combinatorial strategy for rapid directed evolution of functional enzymes with multiple mutagenesis sites.

Iterative saturation mutagenesis (ISM) has been shown to be a powerful method for directed evolution. In this study, the approach was modified (termed M-ISM) by combining the single-site saturation mutagenesis method with a DC-Analyzer-facilitated combinatorial strategy, aiming to evolve novel biocatalysts efficiently in the case where multiple sites are targeted simultaneously. Initially, all target sites were explored individually by constructing single-site saturation mutagenesis libraries. Next, the top two to four variants in each library were selected and combined using the DC-Analyzer-f

A New Plant Expression System for Producing Pharmaceutical Proteins.

In the past decade, interest in the production of recombinant pharmaceutical proteins in plants has tremendously progressed because plants do not harbor mammalian viruses, are economically competitive, easily scalable, and capable of carrying out complex post-translational modifications required for recombinant pharmaceutical proteins. Mucuna bracteata is an essential perennial cover crop species widely planted as an underground cover in oil palm and rubber plantations. As a legume, they have high biomass, thrive in its habitat, and can fix nitrogen. Thus, M. bracteata is a cost-efficient crop

Development of a plant-based vaccine against brucellosis: stable expression of Brucella abortus OMP25 in transgenic tobacco.

Brucellosis, caused by Brucella species, is a global threat to livestock farming, resulting in economic losses and socio-economic challenges, particularly in rural areas. Despite its impact, no licensed human vaccines are available. Animal vaccination remains the most cost-effective control method, but traditional vaccine production is expensive. Edible vaccines, using plants as bioreactors to produce immunogenic antigens, offer a low-cost alternative by eliminating complex purification processes. This study developed a transgenic plant by expressing the Brucella abortus outer membrane protein

Barcoded mutant library enables high-throughput functional genomics in a filamentous fungus.

Advances in sequencing technology enabling rapid and inexpensive whole-genome sequencing highlight how few genes are functionally characterized. This problem is particularly acute in filamentous fungi, where even in the best studied organisms upward of half of genes are poorly characterized or unannotated. High-throughput tools to identify gene function exist for single-celled organisms, like yeast and bacteria. However, filamentous fungi present challenges to high-throughput gene characterization, including low transformation efficiency and multinucleate cells. Filamentous fungi are critical

Elicitor-enhanced steroidal sapogenin accumulation in hairy root cultures of Trigonella foenum-graecum.

In current work, we studied hairy root induction in Trigonella foenum graecum, which is an important medicinal plant, and examined the impact of different elicitors on some phytochemical characteristics and metabolites production in hairy root cultures. Accordingly, some factors such as five strain types of Agrobacterium rhizogenes (1724, 15834, A4, A13 and MSU) and three different explants, namely leaf, cotyledon and hypocotyl were studied. The results showed that different A. rhizogenes strains exhibited different infection efficiency. MSU and 15834 had highest efficiency of hairy root induc

[ENHANCEMENT OF AGROBACTERIAL TRANSFORMATION OF PLANTS USING PROTEIN KINASE INHIBITORS TRIFLUOPERAZINE AND GENISTEIN].

The effect of different concentrations of protein tyrosine kinase inhibitor, genistein and serine/threonine protein kinase inhibitor, trifluoperazine, on the frequency of Agrobacterium-mediated transformation of leaf explants of N. tabacum was investigated. The influence of different concentrations of trifluoperazine in the range from 10 to 300 μM was investigated. It was found that 10 μM trifluoperazine provoked the increase of the frequency of agrobacterial transformation of tobacco leaf disks on 25%. In parallel, the influence of different concentrations of genistein in the range from 10 to

Phlorizin has antioxidant and anti-aging pharmacological activities, which render it a highly promising candidate for applications in functional foods and the pharmaceutical industry. However, the incomplete elucidation of phlorizin biosynthesis in plants has constrained its large-scale production and application. Lithocarpus litseifolius, which contains a notably higher level of phlorizin than most plant species, serves as an ideal model for identifying key enzymes involved in phlorizin biosynthesis. A rare agent of continuous ambulatory peritoneal dialysis peritonitis: Rhizobium Radiobacter.

Peritonitis is a common clinical problem in patients treated by continuous ambulatory peritoneal dialysis (CAPD). The most common microbiological factors causing peritonitis are Gram-positive (especially Staphylococcus spp.). Peritonitis with Rhizobium radiobacter (Agrobacterium radiobacter) is a rare infection in CAPD patients. Peritonitis due to R. radiobacter has been reported in our patient's dialysate culture who underwent CAPD for three years. We report the case of a 26-year-old female PD patient who had CAPD peritonitis due to R. radiobacter and successfully treated with intraperitoneal
