Drug-induced liver injury (DILI) is a frequent adverse event associated with anti-tuberculosis therapy, and current clinical management options remain limited. Given the pivotal role of the gut-liver axis in liver pathophysiology, this study investigated the potential and underlying roles of Akkermansia muciniphila (A.muc) in ameliorating anti-tuberculosis drug-induced liver injury (ADLI). A reduction in the abundance of A. muc was observed during ADLI progression, suggesting its possible involvement in the modulation of liver injury. This study demonstrated that A. muc intervention ameliorate

Astaxanthin (ASX) gains wide interest in food and nutritional sciences due to its great antioxidant and anti-inflammatory properties. However, ASX faces challenges such as poor water solubility, low bioavailability, and sensitivity to environmental factors. In this study, we employ Akkermansia muciniphila (AKK) with mucoadhesive property as a carrier for ASX to enhance ASX stability and bioaccessibility. The complex multilayered cell envelope of AKK cell provided space for ASX loading. Immersion loading yielded the highest ASX encapsulation efficiency (10.80 ± 2.29 μg/mg in ASX@AKK group and 1

Preeclampsia (PE) is a severe metabolic disorder that occurs during pregnancy and is linked to dysbiosis of the gut microbiota, characterised by a decrease in Akkermansia muciniphila (AKK). Emerging evidence suggests that pasteurized Akkermansia muciniphila (pAKK) is a promising candidate for preventing or treating obesity-related metabolic disorders. However, the modulatory function and the underlying mechanisms of pAKK supplementation in PE remain to be fully elucidated. In this study, we examined the impact of pAKK oral administration on PE and its underlying mechanisms. Our findings demons

BACKGROUND: Hepatocellular carcinoma (HCC) mainly develops in cases of fibrosis and cirrhosis and is accompanied by intestinal flora disorder. HCC also affects CD8+ T cell immune function. 5,7-Dimethoxyflavone (DMF), an active flavonoid with anti-tumor effect, is found in Kaempferia parviflora. However, whether DMF can treat HCC remains unclear. This study aims to investigate the effect of DMF on HCC and to explore its possible mechanism, focusing on the gut microbiota regulation and the effect of CD8+ T cells in a murine model.
METHODS: The HCC mouse model was induced with diethylnitrosamine

The gut commensal bacterium Akkermansia muciniphila (AKK) has emerged as a candidate for treating liver disorders, yet its therapeutic potential in liver fibrosis remains poorly defined. Here, using a carbon tetrachloride (CCl4)-induced murine model, we show that AKK administration markedly attenuates collagen deposition, inflammation, and hepatic injury. AKK restored intestinal barrier integrity, reshaped microbial composition, and enhanced propionic acid transport from the gut to the liver, leading to suppression of hepatic stellate cell activation. Multi-omics profiling revealed that AKK en

BACKGROUND AND AIMS: Alcohol-perturbed gut immune homeostasis is associated with the development of alcoholic liver disease (ALD). However, the role of intestinal dendritic cells (DCs) in ALD progression is still unknown. This study aimed to investigate the cellular and molecular mechanisms through which intestinal DCs respond to alcohol exposure and contribute to the pathogenesis of ALD.
APPROACH AND RESULTS: After 8 weeks of alcohol consumption, the number of basic leucine zipper transcription factor ATF-like 3 ( Batf3 )-dependent conventional type 1 DCs (cDC1s) was dramatically decreased i

Nonalcoholic steatohepatitis (NASH) is one of the most prevalent diseases globally. A high-fat, high-cholesterol (HFHC) diet leads to an early NASH model. It has been suggested that gut microbiota mediates the effects of diet through the microbiota-gut-brain axis, modifying the host's brain metabolism and disrupting cognition. Here, we target NASH-induced cognitive damage by testing the impact of environmental enrichment (EE) and the administration of either Lacticaseibacillus rhamnosus GG (LGG) or Akkermansia muciniphila CIP107961 (AKK). EE and AKK, but not LGG, reverse the HFHC-induced cogni

Mucin is known to modulate the gut environment; however, its specific mechanisms and downstream metabolites in alleviating obesity and hepatic steatosis remain unclear. In this study, we investigated the beneficial effects of mucin in a high-fat diet (HFD) mouse model and explored the underlying mechanisms. Our results showed that mucin supplementation significantly reduced weight gain, improved glucose tolerance, and alleviated hepatic steatosis and fibrosis in HFD-fed mice. These benefits were abolished by antibiotic treatment, indicating a microbiota-dependent mechanism. Fecal 16S rRNA gene

Aging is associated with metabolic decline, brown adipose tissue (BAT) dysfunction, and remodelling of the gut microbiota. Akkermansia muciniphila (AKK) has been implicated in metabolic regulation; however, its relationship with BAT aging remains unclear. This study aims to investigate whether age-associated AKK depletion is linked to BAT aging and whether AKK supplementation attenuates aging phenotypes in BAT. Male mice aged 7 weeks, 7 months, and 11 months were analysed to characterise age-related changes in gut microbiota and BAT. 16S rRNA sequencing was performed to assess microbial compos

Akkermansia muciniphila (Akk) has been shown to improve obesity via gut microbiota, while its effects on modulating gut fungi remain underexplored. This study investigates the effects of Akk on obese mice, focusing on gut fungi, metabolites, and hepatic lipid metabolism. We found that Akk treatment significantly modulated gut fungal diversity, enhanced gut immune responses, and improved fatty liver. Specifically, the abundance of harmful fungi Fusarium decreased. Subsequently, Akk improved hepatic lipid metabolism via the PI3K/Akt pathway, as determined by proteomics analysis. Additionally, an

Type 2 diabetes mellitus (T2DM) and metabolic dysfunction-associated steatohepatitis (MASH) frequently coexist and are mechanistically linked via insulin resistance, lipotoxicity, and chronic inflammation. Cholic acid (CA), a primary bile acid (BA), modulates metabolic and immune pathways by influencing BA composition and the gut microbiota. In this study, we examined the role of dietary CA in the modulation of T2DM and MASH in Tsumura Suzuki obese diabetes (TSOD) mice, a model of spontaneous obesity and diabetes. Mice were fed a high-fat, high-cholesterol diet with or without CA (iHFC and CA(
