Carbonylative coupling of simple alkanes and alkenes enabled by organic photoredox catalysis.

A visible-light-driven direct carbonylative coupling of simple alkanes and alkenes via the combination of a hydrogen atom transfer process and photoredox catalysis has been demonstrated. Employing the N-alkoxyazinium salt as the oxidant and the precursor of an oxygen radical, a variety of α,β-unsaturated ketones could be obtained in a metal-free fashion.

Copper-Catalyzed Multicomponent Trifluoromethylphosphorothiolation of Alkenes: Access to CF3-Containing Alkyl Phosphorothioates.

An unprecedented copper-catalyzed multicomponent radical-based reaction involving alkenes, P(O)H compounds, sulfur powder, and Togni reagent II at room temperature has been developed. A variety of highly functionalized CF3-containing S-alkyl phosphorothioates can be directly prepared from a wide range of activated and unactivated alkenes. Moreover, this protocol highlights its potential in the late-stage functionalization of complex molecules and opens up a new avenue for the construction of C(sp3)-S-P bonds.

Intermolecular oxidative amination of unactivated alkenes by dual photoredox and copper catalysis.

Oxidative amination of alkenes via amidyl radical addition is potentially an efficient method to generate allylic amines, which are versatile synthetic intermediates to bioactive compounds and organic materials. Here by combining photochemical generation of amidyl radicals with Cu-mediated β-H elimination of alkyl radicals, we have developed an intermolecular oxidative amination of unactivated alkenes. The reaction relies on tandem photoredox and copper catalysis, and works for both terminal and internal alkenes. The radical nature of the reaction and the mild conditions lead to high functiona

Photoredox-catalyzed sulfonylation of alkyl iodides, sulfur dioxide, and electron-deficient alkenes.

A photoredox-catalyzed sulfonylation of alkyl iodides, sulfur dioxide, and electron-deficient alkenes under mild conditions is achieved. This reaction proceeds through alkyl radicals formed in situ from alkyl iodides under visible light irradiation in the presence of a photoredox catalyst. The alkyl radical intermediates would react with sulfur dioxide leading to alkylsulfonyl radicals, which would be trapped by electron-deficient alkenes giving rise to alkyl sulfones. Various functional groups including nitro, halo, acetyl, sufonyl, and pyridinyl are all tolerated under the photoredox conditi

Direct oxidative isoperfluoropropylation of terminal alkenes via hexafluoropropylene (HFP) and silver fluoride.

A copper-mediated oxidative isoperfluoropropylation of unactivated terminal alkenes with commercially available hexafluoropropylene (HFP) has been developed. With operational simplicity of the procedure and broad substrate applicability, this strategy provides a general and straightforward way for the construction of C(sp3)-C3F7-i bonds. A variety of allylic isoperfluoropropylated compounds have been efficiently synthesized using this strategy.

Cascade arylalkylation of activated alkenes: synthesis of chloro- and cyano-containing oxindoles.

The general method for the oxidative cyclization of arylacrylamides with dichloromethane or acetonitrile has been developed. The reactions described provide novel access to chloro- and cyano-containing oxindoles in good to moderate yields that allow the direct formation of a C-C bond and the construction of an oxindole ring in one reaction. The use of a cheap and easily prepared Mn(OAc)3 represents an added advantage of this method.

Copper-Photocatalyzed Selective 1,6-Thiosulfonylation of Alkenes via 1,5-Hydrogen Atom Transfer.

There are substantial difficulties in constructing universal, selective access towards alkene remote thiosulfonylation relying on classic synthetic pathways. In this work, a photocatalytic copper-catalyzed protocol has been devised for the mild regioselective thionification of remote aliphatic C-H bonds and modular anti-Markovnikov alkene sulfonylation. The transformation hinges on activation of thiosulfonates by the BINAP-Cu(I) catalyst to form sulfonyl radicals and a subsequent 1,5-HAT that furnishes transposed alkyl radicals. A broad spectrum of alkenes and thiosulfonates exhibited good com

A Dual Photoredox/Cobalt Catalytic System Enables Markovnikov-Selective Hydroaminoalkylation of Alkenes.

We report a visible light-driven dual photoredox/cobalt catalytic system that enables Markovnikov-selective hydroaminoalkylation of alkenes. This cooperative catalysis generates and couples two radical species derived from alkylamines and alkenes via sequential single-electron and proton transfers (SET/PT) from the α-amino C-(sp3)-H bonds of amines to alkenes. The method enables the installation of secondary or tertiary alkyl groups at α-amino positions of α-amino acid derivatives, peptides, and benzylaniline derivatives using a broad range of alkene partners. This strategy represents a concep

Reprogramming ThDP Enzymes for Z-Alkenes: Overriding Thermodynamic Preference via Noncovalent Controls.

Most conventional alkene synthesis reactions (e.g., elimination et al.) inherently favor the formation of thermodynamically more stable E-isomers, posing a long-standing challenge for direct access to Z-alkenes. Here, we report the reprogramming of a thiamine diphosphate (ThDP)-dependent enzyme to catalyze a formal dehalogenative elimination that overrides this intrinsic thermodynamic bias, enabling the direct and selective synthesis of Z-α,β-unsaturated carboxylic acids. In contrast to classical approaches that rely on substrate control, directing groups, or complex ligand architectures, our

Visible-light-enabled three-component carboamidation of alkenes with aryl thianthrenium salts.

Herein, we report a visible-light-enabled strategy for activating aryl thianthrenium salts that enables a three-component Ritter-type carboamidation of alkenes with nitriles/H2O. Under neutral and mild irradiation, C-C and C-N bonds are sequentially formed in a single operation to deliver diverse benzylamides with broad functional-group tolerance. A wide range of aryl thianthrenium salts as well as diverse styrenes are compatible, and late-stage diversification of pharmaceutically relevant motifs is readily achieved. Mechanistic studies suggest that a single-electron transfer (SET) pathway is

Scalable Iron-Catalyzed Radical Bromochlorination of Alkenes with HCl under Thermal Conditions.

Herein, an efficient and sustainable iron-catalyzed radical bromochlorination of alkenes is disclosed, proceeding under conventional thermal conditions with excellent chemo- and regioselectivity. The reaction employs exceptionally affordable and ubiquitous hydrochloric acid both to activate N-bromosuccinimide for bromine radical generation and to serve as the chlorine source, while Fe(III)-Cl acts as the chlorine-atom transfer mediator via a radical ligand transfer process. This method features operational simplicity, a remarkably short reaction time (just 1 min), and multigram scalability. Mo

DMSO-Modulated Divergent Fluoroalcohol Addition to Alkenes via Cooperative Iron/Disulfide Photocatalysis.

Fluoroalcohols and their ether counterparts are indispensable pharmacophores in modern medicinal chemistry. Despite the inherent appeal of constructing these motifs through the direct addition of fluoroalcohols across alkenes, the profound inductive withdrawal exerted by the fluorine atoms engenders a formidable electronic barrier: it drastically suppresses oxygen nucleophilicity, pushes oxidation potentials to prohibitive levels, and renders the α‑C─H bonds kinetically inert. Here, we report a cooperative iron/disulfide photocatalytic framework that transcends this electronic recalcitrance th

Synthesis through C(sp3)-C(sp2) Bond Scission in Alkenes and Ketones.

ConspectusThe homolytic cleavage of C-C bonds adjacent to functional groups has recently become a popular strategy for restructuring the skeletons of complex organic molecules. In contrast to the traditional reactivity profiles of polar bond disconnections, homolytic scission furnishes carbon-centered free radicals primed for controlled termination with a diverse range of radicophiles. Beyond standard radical capture, transition-metal catalysis facilitates sophisticated C-C and C-heteroatom bond-forming reactions. Intensive efforts have been focused over many years into the cleavage of the nei

Photoinduced [3 + 2] Cycloadditions of Aryl Cyclopropyl Ketones with Alkynes and Alkenes.

The five-membered ring skeleton is one of the most pivotal in the area of pharmaceutical and natural products. [3 + 2] cycloadditions of cyclopropyl and unsaturated compounds are a highly efficient and atom-economical way to build a five-member compound. The previous works about the kind of [3 + 2] cycloadditions usually utilized metal or organic small molecule catalysts. However, an ideal [3 + 2] cycloaddition reaction that smoothly happens without any additives and catalysts under mild conditions is underdeveloped. Hence, we report [3 + 2] cycloadditions of aryl cyclopropyl without any addit

Ligand-enabled Ni-catalysed dicarbofunctionalisation of alkenes with diverse native functional groups.

The transition metal-catalysed dicarbofunctionalisation of unactivated alkenes normally requires exogenous strong coordinated directing groups, thus reducing the overall reaction efficiency. Here, we report a ligand-enabled Ni(II)-catalysed dicarbofunctionalisation of unactivated alkenes with aryl/alkenyl boronic acids and alkyl halides as the coupling partners with a diverse range of native functional groups as the directing group. This dicarbofunctionalisation protocol provides an efficient and direct route towards vicinal 1,2-disubstituted alkanes using primary, secondary, tertiary amides,

Biosynthesis of chiral diols from alkenes using metabolically engineered type II methanotroph.

Methanotrophs are environmentally friendly microorganisms capable of converting gas to liquid using methane monooxygenases (MMOs). In addition to methane-to-methanol conversion, MMOs catalyze the conversion of alkanes to alcohols and alkenes to epoxides. Herein, the efficacy of epoxidation by type I and II methanotrophs was investigated, and type II methanotrophs were observed to be more efficient in converting alkenes to epoxides. Subsequently, three (Epoxide hydrolase) EHs of different origins were overexpressed in the type II methanotroph Methylosinus trichosporium OB3b to produce 1,2-diols
