Inhibition of aconitase by alloxan and the differential modes of protection of glucose, 3-O-methylglucose, and mannoheptulose.

Alloxan inhibited aconitase with a half maximal inhibitory concentration of 0.5 mM in sonically disrupted and 2.3 mM in intact isolated liver mitochondria. For dialuric acid the half maximal inhibitory concentrations were 1.1 mM and 2.5 mM, respectively. Ninhydrin and N-ethylmaleimide (NEM) also inhibited aconitase with half maximal inhibitory concentrations in the submillimolar range and t-butylhydroperoxide (BuOOH) in the millimolar range, which, however, were not different for disrupted and intact mitochondria. Only the aconitase substrate citrate, but not glucose provided protection of the

Assessment of the diabetogenic drugs alloxan and streptozotocin as models for the study of immune defects in diabetic mice.

The efficacy of the diabetogenic drugs streptozotocin and alloxan were evaluated as models for the study of immune defects associated with diabetes. Streptozotocin- or alloxan-treated mice, with a stable hyperglycaemia of 25-33 mmol/l plasma glucose, were severely impaired in their ability to mount antibody forming, mitogenic, or delayed-type hypersensitivity responses in vivo. Treatment of alloxan-diabetic mice with insulin in vivo completely reversed all immune defects, while insulin treatment of streptozotocin-diabetic mice restored immune function to only 70-80% of normal levels. Results o

[Effect of prolonged administration of the adrenergic blockaders, fentolamin and obsidan, on the course of alloxan diabetes in rats].

Administration of phentholamine or obsidan to rats for 3 weeks sharply diminished the diabetogenic action of alloxan. The insular tissue of the pancreas became less affected, and the percentage of animals with signs of diabetes (polyuria, glucosuria, and hyperglycemia) decreased. Hyperglycemia (averaging 276 and 286 mg%) was revealed in the majority of the animals given alloxan and adrenoblockers. Phentholamine sharply suppressed the development of polyuria in rats with intact pancreas following water load, but obsidan produced no such effect. The mechanism of action of adrenoblockers used on

Determination of Hypoglycemic, Hypolipidemic and Nephroprotective Effects of Berberis Calliobotrys in Alloxan-Induced Diabetic Rats.

Many plants of the Berberis genus have been reported pharmacologically to possess anti-diabetic potential, and Berberis calliobotrys has been found to be an inhibitor of α-glucosidase, α-amylase and tyrosinase. Thus, this study investigated the hypoglycemic effects of Berberis calliobotrys methanol extract/fractions using in vitro and In vivo methods. Bovine serum albumin (BSA), BSA-methylglyoxal and BSA-glucose methods were used to assess anti-glycation activity in vitro, while in vivo hypoglycemic effects were determined by oral glucose tolerance test (OGTT). Moreover, the hypolipidemic and

Effects of Cymodocea nodosa extract on metabolic disorders and oxidative stress in alloxan-diabetic rats.

This new study aimed to evaluate for the first time the effect of Cymodocea nodosa extract (CNE) on α-amylase activity, hyperglycemia and diabetes complications in the alloxan-induced diabetic rats. The in vitro evaluation and oral administration of CNE to surviving diabetic rats inhibited key enzyme related to hyperglycemia as α-amylase, helped to protect the β cells of the rats from death and damage confirmed by oral glucose test tolerance (OGTT), which leads to decrease in blood glucose level by 49% as compared to untreated diabetic rats. The CNE also decreased the triglyceride, low density

In vitro effects of alloxan/copper combinations on lipid peroxidation, protein oxidation and antioxidant enzymes.

The in vitro effects of alloxan and the product of its reduction dialuric acid (alone or in combination with copper ions) on lipid peroxidation, carbonyl content, GSH level and antioxidant enzyme activities in rat liver and kidney have been studied. The effects of Cu2+/alloxan and Cu2+/dialuric acid were compared with those of Fe3+/alloxan and Fe3+/dialuric acid. Unlike alloxan, dialuric acid increased liver and kidney lipid peroxidation; similar effects were registered in the presence of Fe3+. In the presence of Cu2+/dialuric acid, the lipid peroxidation was strongly inhibited and vice versa-

[Vitamin D3 transport across the intestines and its metabolism in the liver of rats with alloxan diabetes].

Vitamin D3 intestinal transport and liver metabolism were studied in rats with alloxan-induced diabetes. The condition was induced by i.v. alloxan injection in a dose of 40 mg/kg b.m. Diabetes development was monitored by blood serum glucose measurements, carried out for 30 days. [3H]-cholecalciferol absorption in the rat intestine was found inhibited in the diabetic animals as against the reference animals, which fact results in disordered entry of vitamin D3 to the body. [3H]-cholecalciferol absorption by the liver is reduced in the examined condition, and the time of its metabolism is incre

Physiological and morphological changes in islet B cells following treatment of the guinea pig with alloxan.

Alloxan injections (200 mg/kg i.v.) into guinea pigs led to a significant decline of serum insulin levels and a marked inhibition of normal weight gain during the first 96 hr following treatment. Both of these parameters tended to normalize between 96 and 120 hr after treatment. However, neither hyperglycemia nor glycosuria occurred at any time. Volume densities of islet B cells (= volume density of islet cells immunostained for insulin per islet volume density) decreased and pyknotic nuclei appeared in the islets during the initial 48 hr after alloxan treatment. Islet B-cell volume density th

Cardioprotection by Citrus grandis (L.) Peel Ethanolic Extract in Alloxan-Induced Cardiotoxicity in Diabetic Rats.

Diabetic cardiomyopathy (DCM) pathogenesis is multifarious, and there are insufficient therapeutic options to treat DCM. The present research explored the effects of Citrus grandis peel ethanolic extract (CGPE) in alloxan-induced DCM in rats. Diabetes was triggered by intraperitoneal (i.p.) injection of alloxan (150 mg/kg) in Wistar rats (200-250 g). CGPE (100, 200, and 400 mg/kg) or glibenclamide (Glib, 10 mg/kg) were administered orally for 2 weeks. After the treatment schedule, prooxidants (thiobarbituric acid reactive substances), antioxidants (glutathione, catalase, and superoxide dismuta

Islet amyloid polypeptide (amylin)-deficient mice develop a more severe form of alloxan-induced diabetes.

To examine whether islet amyloid polypeptide (IAPP), other than through amyloid formation, may be of importance in diabetes pathogenesis, IAPP-deficient mice (IAPP(-/-)) were challenged with alloxan (day 0). Diabetes in IAPP(-/-) mice was more severe at day 35, indicated by greater weight loss; glucose levels were higher in alloxan-treated IAPP(-/-) mice, whereas insulin levels were lower, indicating a greater impairment of islet function. Accordingly, glucose levels upon intravenous glucose challenges at days 7 and 35 were consistently higher in alloxan-treated IAPP(-/-) mice. At day 35, insu

Salivary glands in long-term alloxan-diabetic rats. A quantitative light and electron-microscopic study.

Fifty untreated diabetic animals were compared with 58 age-matched non-diabetic controls. Reduced salivary gland weight was evident after one month's diabetes and this was unchanged after 12 months of diabetes. Submandibular/sublingual gland weight was proportional to the reduced body weight in the diabetic rats. Parotid gland weight, however, was proportionally more reduced. Only diabetic rats had lipid inclusions in the acinar cells of their submandibular glands and the morphometrically estimated amount of inclusions was positively correlated to the blood glucose level. Acinar cell size was

Alloxan-induced diabetes exacerbates coronary atherosclerosis and calcification in Ossabaw miniature swine with metabolic syndrome.

BACKGROUND: There is a preponderance of evidence implicating diabetes with increased coronary artery disease (CAD) and calcification (CAC) in human patients with metabolic syndrome (MetS), but the effect of diabetes on CAD severity in animal models remains controversial. We investigated whether diabetes exacerbates CAD/CAC and intracellular free calcium ([Ca2+]i) dysregulation in the clinically relevant Ossabaw miniature swine model of MetS. METHODS: Sixteen swine, eight with alloxan-induced diabetes, were fed a hypercaloric, atherogenic diet for 6 months. Alloxan-induced pancreatic beta cell
