Hippocampus and amygdala volumes in parents of children with autistic disorder.

OBJECTIVE: Structural and functional abnormalities in the medial temporal lobe, particularly the hippocampus and amygdala, have been described in people with autism. The authors hypothesized that parents of children with a diagnosis of autistic disorder would show similar changes in these structures. METHOD: Magnetic resonance imaging scans were performed in 17 biological parents of children with a diagnosis of DSM-IV autistic disorder. The scans were compared with scans from 15 adults with autistic disorder and 17 age-matched comparison subjects with no personal or familial history of autism

Dysregulation of arousal and amygdala-prefrontal systems in paranoid schizophrenia.

OBJECTIVE: The authors investigated impaired differentiation of limbic-prefrontal systems by autonomic arousal in schizophrenia. It was predicted that paranoid patients would be distinguished by a disjunction of hyperarousal but reduced amygdala and medial prefrontal activity relative to both healthy comparison subjects and patients with nonparanoid schizophrenia. METHOD: Pictures depicting facial expressions of fear were presented to 27 schizophrenia patients (13 paranoid, 14 nonparanoid) and 22 matched healthy comparison subjects in an implicit perception task to evoke limbic activity. Amygdala and hippocampal volumes in adolescents and adults with bipolar disorder.

BACKGROUND: The purported functions of medial temporal lobe structures suggest their involvement in the pathophysiology of bipolar disorder (BD). Previous reports of abnormalities in the volume of the amygdala and hippocampus in patients with BD have been inconsistent in their findings and limited to adult samples. Appreciation of whether volumetric abnormalities are early features of BD or whether the abnormalities represent neurodegenerative changes associated with illness duration is limited by the paucity of data in juvenile samples. OBJECTIVE: To investigate amygdala and hippocampal volu

Amygdala response to fearful faces in anxious and depressed children.

BACKGROUND: Alterations in amygdala function have been implicated in the pathophysiological characteristics of adult anxiety and depressive disorders. Studies with healthy adults and children, as well as with adults who have amygdala lesions, have found facial expressions of emotion to be useful probes of amygdala activity. Our study examined the amygdala response to fearful and neutral facial expressions in healthy, anxious, and depressed children. We hypothesized that children with anxiety and depression may show atypical amygdala responses to emotional stimuli. METHODS: Twelve children (8-

Effects of lesions of the perirhinal cortex on amygdala kindling in rats.

The perirhinal cortex (PRC), the region of temporal cortex adjacent to the rhinal sulcus, has been suggested as a critical substrate for the development and expression of generalized motor seizures in the late stages of kindling development. For further investigation of the role of the PRC in limbic kindling, excitotoxic lesions centered on PRC by microinjection of ibotenate were performed in rats 2 weeks before onset of amygdala kindling. Rats with large bilateral or unilateral PRC lesions showed the same rate and pattern of kindling development as sham-lesioned controls. The only significant

Laterality of dopamine function and neuroleptic action in the amygdala in the rat.

Rats selected according to turn preference in an open field were categorised as showing left or right hemispheric dominance (turning to right or left respectively). Dopamine was persistently infused into the central area of the left or right amygdala of animals showing left or right hemispheric dominance. Infusion was effected for 9 days by Alzet osmotic minipumps, implanted subcutaneously, which delivered 25 micrograms/24 hr of dopamine via chronically-indwelling, stereotaxically-located injection units. Dopamine caused marked and consistent hyperactivity only when infused into the left amygd

Role of the Amygdala in the Complex Relationship Between Sleep and Anxiety.

This review explores the multifaceted roles of the amygdala, focusing on its structural components, projections, physiological aspects, and key functions in emotional processing and fear conditioning. Beyond traditional roles, it examines the amygdala's involvement in sleep architecture during the NREM and REM phases. In NREM sleep, the amygdala is involved in memory replay and encoding. During REM sleep, it is highly active, contributing to the emotional content of dreams. This state is characterized by increased cerebral blood flow to the amygdaloid complexes and the generation of Ponto-Geni

Volumetric alterations of hippocampal and amygdala subfields in Azheimer's disease.

BACKGROUND: Numerous studies have shown that patients with Azheimer's disease (AD) or amnestic mild cognitive impairment (aMCI) have atrophy in the hippocampus and amygdala. However, there are few MRI studies that have jointly analyzed the volumes of the 19 hippocampal subfields and the 9 amygdala subfields. MATERIALS AND METHODS: In this cross-sectional study, we used the method of analysis of covariance (ANCOVA) to analyze the volume differences of hippocampal and amygdala subfields among three groups of 40 normal control (NC), 40 aMCI and 53 AD, and conducted correlation analysis between t

The case for hemispheric lateralization of the human amygdala in fear processing.

The amygdala has increasingly been proposed as a therapeutic target for patients with post-traumatic stress disorder (PTSD). However, the distinct contributions of the left and right amygdala to various aspects of fear processing remain inadequately understood. Here, we critically re-evaluate key findings from human functional neuroimaging and lesion studies on fear conditioning and extinction. We propose that while both amygdalae likely make critical contributions to fear processing, the right is more associated with sensory-mediated fear expression, and the left is associated with cognitive-

Cells and circuits for amygdala neuroplasticity in the transition to chronic pain.

Maladaptive plasticity is linked to the chronification of diseases such as pain, but the transition from acute to chronic pain is not well understood mechanistically. Neuroplasticity in the central nucleus of the amygdala (CeA) has emerged as a mechanism for sensory and emotional-affective aspects of injury-induced pain, although evidence comes from studies conducted almost exclusively in acute pain conditions and agnostic to cell type specificity. Here, we report time-dependent changes in genetically distinct and projection-specific CeA neurons in neuropathic pain. Hyperexcitability of CRF pr

Diverse processing of pharmacological and natural rewards by the central amygdala.

The central amygdala (CeA) with its medial (CeM) and lateral (CeL) nuclei is the brain hub for processing stimuli with emotional context. CeL nucleus gives a strong inhibitory input to the CeM, and this local circuitry assigns values (positive or negative) to incoming stimuli, guiding appropriate behavior (approach or avoid). However, the particular involvement of CeA in processing such emotionally relevant information and adaptations of the CeA circuitry are not yet well understood. In this study, we examined synaptic plasticity in the CeA after exposure to two types of rewards, pharmacologic

Serotonin transporter in the temporal lobe, hippocampus and amygdala in SUDEP.

Several lines of evidence link deficient serotonin function and SUDEP. Chronic treatment with serotonin reuptake inhibitors (SRIs) reduces ictal central apnoea, a risk factor for SUDEP. Reduced medullary serotonergic neurones, modulators of respiration in response to hypercapnia, were reported in a SUDEP post-mortem series. The amygdala and hippocampus have high serotonergic innervation and are functionally implicated in seizure-related respiratory dysregulation. We explored serotonergic networks in mesial temporal lobe structures in a surgical and post-mortem epilepsy series in relation to SU

Perceived stress modulates the activity between the amygdala and the cortex.

The significant link between stress and psychiatric disorders has prompted research on stress's impact on the brain. Interestingly, previous studies on healthy subjects have demonstrated an association between perceived stress and amygdala volume, although the mechanisms by which perceived stress can affect brain function remain unknown. To better understand what this association entails at a functional level, herein, we explore the association of perceived stress, measured by the PSS10 questionnaire, with disseminated functional connectivity between brain areas. Using resting-state fMRI from

Allocentric representation in the human amygdala and ventral visual stream.

The hippocampus and the entorhinal cortex are considered the main brain structures for allocentric representation of the external environment. Here, we show that the amygdala and the ventral visual stream are involved in allocentric representation. Thirty-one young men explored 35 virtual environments during high-resolution functional magnetic resonance imaging (fMRI) of the medial temporal lobe (MTL) and were subsequently tested on recall of the allocentric pattern of the objects in each environment-in other words, the positions of the objects relative to each other and to the outer perimeter

In Vivo Amygdala Nuclei Volumes in Schizophrenia and Bipolar Disorders.

Abnormalities in amygdala volume are well-established in schizophrenia and commonly reported in bipolar disorders. However, the specificity of volumetric differences in individual amygdala nuclei is largely unknown. Patients with schizophrenia disorders (SCZ, N = 452, mean age 30.7 ± 9.2 [SD] years, females 44.4%), bipolar disorders (BP, N = 316, 33.7 ± 11.4, 58.5%), and healthy controls (N = 753, 34.1 ± 9.1, 40.9%) underwent T1-weighted magnetic resonance imaging. Total amygdala, nuclei, and intracranial volume (ICV) were estimated with Freesurfer (v6.0.0). Analysis of covariance and multiple

Synaptic correlates of associative fear memory in the lateral amygdala.

Successful adaptation to the environment requires an accurate response to external threats by recalling specific memories. Memory formation and recall require engram cell activity and synaptic strengthening among activated neuronal ensembles. However, elucidation of the underlying neural substrates of associative fear memory has remained limited without a direct interrogation of extinction-induced changes of specific synapses that encode a specific auditory fear memory. Using dual-eGRASP (enhanced green fluorescent protein reconstitution across synaptic partners), we found that synapses among

Temporal dynamics of amygdala response to emotion- and action-relevance.

It has been proposed that the human amygdala may not only encode the emotional value of sensory events, but more generally mediate the appraisal of their relevance for the individual's goals, including relevance for action or task-based needs. However, emotional and non-emotional/action-relevance might drive amygdala activity through distinct neural signals, and the relative timing of both kinds of responses remains undetermined. Here, we recorded intracranial event-related potentials from nine amygdalae of patients undergoing epilepsy surgery, while they performed variants of a Go/NoGo task w

Sleep Regulation by Neurotensinergic Neurons in a Thalamo-Amygdala Circuit.

A crucial step in understanding the sleep-control mechanism is to identify sleep neurons. Through systematic anatomical screening followed by functional testing, we identified two sleep-promoting neuronal populations along a thalamo-amygdala pathway, both expressing neurotensin (NTS). Rabies-mediated monosynaptic retrograde tracing identified the central nucleus of amygdala (CeA) as a major source of GABAergic inputs to multiple wake-promoting populations; gene profiling revealed NTS as a prominent marker for these CeA neurons. Optogenetic activation and inactivation of NTS-expressing CeA neur
