Ameliorative effects and mechanisms of paeonol against rosacea through network pharmacology and experimental validation.

Rosacea is a long-term and relapsing inflammatory skin disorder resulting from dysfunction of the immune and/or neurovascular systems. Paeonol (Pae), the primary active component extracted from Moutan Cortex, exhibits various pharmacological effects, including anti-inflammatory and antioxidant properties. This study aimed to evaluate the therapeutic efficacy of Paeonol in treating rosacea and to investigate the underlying mechanisms of its action. Bioinformatics analysis was employed to assess the overlap between rosacea pathogenesis and the pharmacological targets of Pae. An LL-37-induced ros

Systemic Cytokines in Retinopathy of Prematurity.

Retinopathy of prematurity (ROP), a vasoproliferative vitreoretinal disorder, is the leading cause of childhood blindness worldwide. Although angiogenic pathways have been the main focus, cytokine-mediated inflammation is also involved in ROP etiology. Herein, we illustrate the characteristics and actions of all cytokines involved in ROP pathogenesis. The two-phase (vaso-obliteration followed by vasoproliferation) theory outlines the evaluation of cytokines in a time-dependent manner. Levels of cytokines may even differ between the blood and the vitreous. Data from animal models of oxygen-indu

Dental Pulp Stem Cell-Derived Secretome-Induced Reprogramming of Tongue Tumor Microenvironment.

Oral tongue cancer is among the most aggressive malignancies in the head and neck region, driven by a complex tumor microenvironment (TME) that fosters tumor progression, immune evasion, and therapy resistance. Conventional treatments often fail to address the stromal and immunological intricacies of the TME, highlighting the need for microenvironment-targeted therapies. One promising strategy involves the use of dental pulp stem cell-derived secretome (DPSC-Sec), which contains a broad range of bioactive molecules, cytokines, chemokines, growth factors, and extracellular vesicles enriched wit

Metabolic Effects on Wound Healing.

SIGNIFICANCE: Wound healing is an energetically demanding process that is easily disturbed by metabolic dysregulation. Metabolic dysregulation, induced by diabetes, obesity, insulin resistance, and various hormone imbalances, can disrupt each phase of wound repair, increasing the risk of post-operative complications, infection, wound dehiscence, and pathological scarring. We review the current evidence on the role of metabolic regulation in wound healing and highlight clinically relevant considerations for patient care. RECENT ADVANCES: Animal and human studies have advanced our understanding

Wireless Electromagnetic Generation of miRNA Sponges and Nerve Stimulation by an Adaptable Electrical Scaffold for Repair of Traumatic Brain Injury.

Bioelectronic transduction and microRNA (miRNA) regulation play crucial roles in shaping neuronal cell fate and supporting brain repair. However, clinical progress remains limited due to the lack of tools to perform spatiotemporal regulation of neuronal electrical activity and nonviral gene regulation in vivo. In this study, an adaptable electronic scaffold (AES) that functions both as an antenna and gene transfection agent was developed for neuron miRNA modulation in traumatic brain injury (TBI). The intrinsic properties of AES alleviate inflammation and glial scarring after TBI by suppressin

Functional and Mechanistic Insights of 3-Hydroxybutyrate (3-OBA) in Bladder Cancer.

Bladder cancer (BC), particularly muscle-invasive urothelial bladder carcinoma (UBC), remains a clinical challenge due to frequent recurrence, chemoresistance, and limited treatment options. This study investigates the functional and mechanistic insights of 3-hydroxybutyrate (3-OBA), a ketone body with known metabolic and epigenetic roles, in muscle-invasive UBC models. 3-OBA significantly inhibited cell viability, proliferation, migration, and invasion in T24 and HT1376 cell lines in a dose-dependent manner. In vivo, 3-OBA impaired tumor growth and angiogenesis in the chick chorioallantoic me

From pathogen to cure: exploring the antitumor potential of Toxoplasma gondii.

Toxoplasma gondii (T. gondii), an intracellular protozoan parasite, has attracted significant attention in recent years for its dual role in both promoting and inhibiting cancer. Although traditionally recognized as a potential risk factor for tumor development, particularly in immunocompromised individuals, emerging research suggests that T. gondii may possess anti-cancer properties. This paradox is rooted in the parasite’s ability to modulate the host’s immune system, triggering antitumor immune responses through the activation of immune cells and the secretion of cytokines such as TNF-α and

Gut homeostasis, injury, and healing: New therapeutic targets.

The integrity of the gastrointestinal mucosa plays a crucial role in gut homeostasis, which depends upon the balance between mucosal injury by destructive factors and healing via protective factors. The persistence of noxious agents such as acid, pepsin, nonsteroidal anti-inflammatory drugs, or Helicobacter pylori breaks down the mucosal barrier and injury occurs. Depending upon the size and site of the wound, it is healed by complex and overlapping processes involving membrane resealing, cell spreading, purse-string contraction, restitution, differentiation, angiogenesis, and vasculogenesis,

Insights into the diverse effects of nitric oxide on tumor biology.

Among its many roles in cellular biology, nitric oxide (·NO) has long been associated with cancers both as a protumorigenic and as an antitumorigenic agent. The dual nature of this signaling molecule in varied settings is attributable to its temporal and concentration-dependent effects that produce different phenotypes. The steady-state ·NO concentration within the cell is a balance between its rate of enzymatic synthesis from the three nitric oxide synthase (NOS) isoforms and consumption via numerous metabolic pathways and demonstrates strong dependence on the tissue oxygen concentration. NOS

Development and partial characterization of fused human endostatin.

BACKGROUND AND OBJECTIVES: Angiogenesis plays a central role in the progression of numerous pathological conditions, making its modulation an important therapeutic strategy. Endostatin, a naturally occurring endogenous inhibitor of angiogenesis, exhibits potent anti-angiogenic activity however, its broader clinical application has been constrained by reported limitations such as structural instability and rapid systemic clearance. Fusion protein engineering represents a rational strategy to potentially enhance the therapeutic applicability of bioactive proteins. In this study, we aimed to desi

Metabolic Regulation of Angiogenesis in Pancreatic Ductal Adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) remains highly lethal, with a 5‑year survival of only 13%. Its aggressive biology and late diagnosis often lead to metastatic spread at presentations. This review examines how metabolic reprogramming regulates angiogenesis in PDAC and explores the therapeutic implications of this coupling. We systematically analyzed published studies on glucose, amino acid, and lipid metabolic alterations and their interconnection with neovascularization through hypoxia-inducible factor-1α (HIF-1α), oncogenic signaling, and metabolite-mediated pathways. Based on a synth

Single-cell sequencing systematically analyzed the mechanism of Emdogain on the restoration of delayed replantation periodontal membrane.

The repair of the periodontal membrane is essential for the successful management of periodontal disease and dental trauma. Emdogain® (EMD) is widely used in periodontal therapy due to its ability to promote repair. Despite substantial research, the cellular and molecular mechanisms underlying EMD's effects, particularly at the single-cell resolution, remain incompletely understood. This study established a delayed tooth replantation model in rats to investigate these aspects. Tooth loss rate and degree of loosening were evaluated at 4 and 8 weeks. Micro-CT, HE staining, TRAP staining, and imm

Celastrol inhibits LL37-induced rosacea by inhibiting Ca2+/CaMKII-mTOR-NF-κB activation.

Rosacea is a common chronic facial inflammatory disease that affects millions of people worldwide. Due to the unclear etiology of rosacea, effective treatments are limited. Celastrol, a plant-derived triterpene, has been reported to alleviate inflammation in various diseases. However, whether celastrol exerts protective effects in rosacea remains to be elucidated. In this study, weighted gene co-expression network analyses (WGCNA) were performed. Hub modules closely related to rosacea clinical characteristics were identified and found to be involved in inflammation- and angiogenesis-related si

Gender differences and pharmacological regulation of angiogenesis induced by synovial fluids in inflammatory arthritis.

Several mediators including cytokines, growth factors and metalloproteinases (MMP) modulate pathological angiogenesis associated with inflammatory arthritis. The biological factors underlying sex disparities in the incidence and severity of rheumatic musculoskeletal diseases are only partially understood. We hypothesized that synovial fluids (SFs) from rheumatoid arthritis (RA) and psoriatic arthritis (PsA) patients would impact on endothelial biology in a sexually dimorphic fashion. Immune cell counts and levels of pro-angiogenic cytokines found in SFs from RA and PsA patients (n = 17) were h

Breaking barriers in peritoneal fibrosis: extracellular vesicles as promising diagnostic and therapeutic strategy.

BACKGROUND: Peritoneal dialysis is an essential therapy for end-stage kidney disease, but its long-term efficacy is severely limited by peritoneal fibrosis. Driven by mesothelial-mesenchymal transition, inflammation and angiogenesis, this pathological process leads to treatment failure and increased mortality. The urgent need for effective therapies has shifted research focus to extracellular vesicles-phospholipid bilayer nanovesicles that shuttle bioactive cargoes to modulate pathogenic pathways, positioning them as key mediators in peritoneal fibrosis. DISCUSSION: This review synthesises pr

Traditional Chinese medicine derived exosome-like nanovesicles in wound repair and skin regeneration.

Skin injuries, including acute wounds, burns, and chronic ulcers, pose significant clinical challenges due to their potential to cause delayed healing and functional impairment. Exosome-like nanovesicles (ELNVs) derived from traditional Chinese medicinal (TCM) herbs have recently emerged as promising natural agents for skin repair and regeneration. These nanoscale vesicles combine the structural advantages of plant-derived delivery systems with the inherent pharmacological activities of TCM phytochemicals, offering dual roles as both bioactive agents and therapeutic carriers.
