CRISPR-Cas12-Based Diagnostic Applications in Infectious and Zoonotic Diseases.

Rapid detection of infectious and zoonotic diseases is very important for pathogen identification and infection control. Molecular diagnostic assays are well-known for high accuracy and sensitivity; however, conventional methods such as real-time PCR may require professional instruments and operations, preventing their wide applications in scenarios including animal quarantine. The recently developed CRISPR diagnostic (CRISPR-Dx) methods, employing the trans-cleavage activities of either Cas12 (e.g., HOLMES) or Cas13 (e.g., SHERLOCK), have shown great potential in rapid and convenient nucleic

Clinical endocrinology and metabolism. Receptors for gut peptides.

Most gut peptides exert their effects through G protein-coupled receptors, a family of about 700 membrane proteins, 87 of which are presently known to have peptide ligands. Three additional gut peptide receptors are not G protein-coupled receptors but regulate intracellular cyclic GMP accumulation. The aim of this review is to illustrate how the sequencing of the human genome and other recent advances in genomics has contributed to our understanding of the role of peptides and their receptors in gastrointestinal function. Recent discoveries include the identification of receptors for the pepti

Designing and evaluating risk-based surveillance systems: potential unwarranted effects of applying adjusted risk estimates.

Risk-based surveillance systems reveal occurrence of disease or infection in a sample of population units, which are selected on the basis of risk factors for the condition under study. The purpose of such systems for supporting practical animal disease policy formulations and management decisions are: A: to detect an emerging disease or infection, if it becomes introduced into a population; or B: to substantiate freedom from a condition in a population; or C: to detect cases and estimate the prevalence of an endemic condition in a population. In risk-based surveillance these aims should be me

Beyond Biomimetics: Pathology-Informed Engineering Rescues Reconstituted HDL From Inflammatory Dysfunction for Sepsis Immunotherapy.

Biomimetic therapies hold great promise for treating intractable diseases, yet their translation is often hindered by dysfunction-the loss of intended biological function upon exposure to pathological microenvironments. This challenge is well exemplified by reconstituted high-density lipoprotein (rHDL), a clinically advanced HDL mimetic and representative biomimetic drug delivery platform. Here, guided by a pathology-informed identification-design-evaluation-validation workflow, we identified representative sepsis-relevant dysfunction liabilities of HDL-based systems under pathology-relevant c

Nightly and seasonal movements, seasonal home range, and focal location photo-monitoring of urban striped skunks (Mephitis mephitis): implications for rabies transmission.

We followed radio-collared striped skunks (Mephitis mephitis) from January 2004-December 2005 in two urban areas of Flagstaff, Arizona, USA to determine seasonal patterns of movement and home-range size. We also used automated cameras to determine the potential for inter- and intraspecific interaction at skunks' diurnal resting sites and nocturnal focal locations. We found no difference between sexes in nightly rates of travel or in size of seasonal home range. Nightly rates of travel were greatest in the postbreeding months (May-July) and smallest from November to February, consistent with la

Rapid identification of porcine bacterial pathogens using an artificial intelligence Raman microbial analyzer for biology.

UNLABELLED: Rapid and accurate identification of bacterial pathogens is critical for effective disease control in pig farms. Conventional bacterial culture-based and molecular diagnostic methods are time-consuming and often impractical for clinical decision-making. Raman spectroscopy, combined with artificial intelligence, offers a label-free alternative and provides isolate-level identification within approximately 2 min. The aim of this study was to evaluate the performance of the commercial Artificial Intelligent Raman Microbial Analyzer for Biology (AIRMAB) for the rapid clinical identific

Research Techniques Made Simple: Parabiosis to Elucidate Humoral Factors in Skin Biology.

Circulating factors in the blood and lymph support critical functions of living tissues. Parabiosis refers to the condition in which two entire living animals are conjoined and share a single circulatory system. This surgically created animal model was inspired by naturally occurring pairs of conjoined twins. Parabiosis experiments testing whether humoral factors from one animal affect the other have been performed for more than 150 years and have led to advances in endocrinology, neurology, musculoskeletal biology, and dermatology. The development of high-throughput genomics and proteomics ap

Characterizing cochlear mechano-electric transduction in ears damaged with pure tones.

Cochlear microphonics were recorded in response to Gaussian noise from the round window of Mongolian gerbils. A nonlinear systems identification procedure provided the frequency-domain parameters of a third-order polynomial equation describing cochlear mechano-electric transduction (MET). Exposure to an 8 kHz pure tone at 100 dB SPL for 20 min reduced the magnitude of the linear, quadratic, and cubic terms significantly. Animals exposed to a 1- or 4-kHz pure tone showed changes in the quadratic term. Differentiation of the polynomial equation and algebraic manipulations of the coefficients pro

Antibiotic susceptibility of Salmonella spp isolated from farm animals and environmental sources in Tripoli, Libya.

BACKGROUND: Nontyphoidal Salmonella (NTS) serovars remain a potential hazard to human and animal health. Food animals with Salmonella infections can either be cured or remain asymptomatic carriers, serving as a potential source of infection to humans and other healthy animals. Salmonella may also be a reservoir of resistance determinants that can be transferred to other commensal organisms, particularly in the gastrointestinal tract. AIM: This article addresses the prevalence of Salmonella serovars in important food animals and their environment. Antibiotics susceptibility profiling of Salmon

Abdominal perfusion in canine patients with pyometra and sepsis evaluated by Doppler and contrast-enhanced ultrasound.

BACKGROUND: Sepsis is a condition characterized by organic dysfunction, leading to hemodynamic instability and high morbidity and mortality rates in humans and animals. Early identification of perfusion changes and appropriate management of sepsis are crucial for improving patient prognosis. Currently, the Systemic Inflammatory Response Syndrome (SIRS) and Sequential Organ Failure Assessment (SOFA) scores are widely studied for sepsis identification and evaluation of organ dysfunction. However, these scores do not assess gastrointestinal involvement, which is common in this condition. Contrast

Rats, mice and men - models for immune effector mechanisms against schistosomiasis.

Experimental studies have demonstrated the diversity of immune effector mechanisms against schistosomes. Among the various animal models, the rat appears as an excellent experimental system for investigation of antibody-mediated immunity to Schistosoma mansoni. Rat monoclonal antibodies have allowed the identification of effector and regulatory mechanisms operating in human infection, together with the characterization of protective antigens, leading to promising approaches to vaccine development.

Polyphasic characterization of Bacillus species from anthrax outbreaks in animals from South Africa and Lesotho.

INTRODUCTION: Bacillus anthracis is the causative agent of anthrax, a disease endemic in regions of Northern Cape Province and Kruger National Park of South Africa. Accurate identification of virulent B. anthracis is essential but challenging due to its close relationship with other members of B. cereus group. This study characterized B. anthracis and Bacillus species that were recovered from animals and the environment where animals died of anthrax symptoms in southern Africa using a polyphasic approach. METHODOLOGY: For this purpose, 3 B. anthracis and 10 Bacillus isolates were subjected to

Sheep biometric identification based on multiple body parts.

Precision livestock farming technologies require individual animal identification to integrate specific animal data with farm processes, including health, milk recording, and feeding management. Identification methods, such as physical marks and ear tags, require manual handling and are prone to loss or wear. Radio Frequency Identification (RFID) tags enable contactless identification, but still face limitations under farm conditions, including tag loss, maintenance costs, and proximity to the sensor. Recent advances in computer vision and deep learning have led to alternative approaches for a

[Lactobacilli and their probiotic properties].

Lactobacilli rank among lactic acid bacteria, fermenting saccharides to produce lactic acid. The individual species and strains differ by the profiles of fermented saccharides, but this property is not sufficient for the identification at the species level. The species and strains differ in the 16S rDNA sequence and in the intergenic space between 16S and 23S rDNA, which make their differentiation and identification possible. Some representatives of the genus Lactobacillus are included in probiotic preparations with regard to their beneficial health effects on the human and animal organisms. F

Using rumination and activity data for early detection of anaplasmosis disease in dairy heifer calves.

Bovine anaplasmosis causes considerable economic losses in dairy cattle production systems worldwide, ranging from $300 million to $900 million annually. It is commonly detected through rectal temperature, blood smear microscopy, and packed cell volume (PCV). Such methodologies are laborious, costly, and difficult to systematically implement in large-scale operations. The objectives of this study were to evaluate (1) rumination and activity data collected by Hr-Tag sensors (SCR Engineers Ltd.) in heifer calves exposed to anaplasmosis; and (2) the predictive ability of recurrent neural networks

Promoting osteoblast-mediated bone formation: a more promising approach for natural products to treat osteoporosis.

Osteoporosis is a systemic skeletal disorder characterized by reduced bone mass, compromised bone microstructure, and an increased risk of fractures, primarily due to excessive osteoclast-mediated bone resorption relative to osteoblast-mediated bone formation. While current anti-osteoporosis drugs, such as bisphosphonates and denosumab, predominantly focus on reducing bone resorption, osteoanabolic approaches are essential for restoring bone microarchitecture and ultimately reducing fracture risk. Traditional Chinese medicines (TCMs) and their active ingredients have long been used in China fo

Gene therapy approaches for treating rheumatoid arthritis.

Current gene therapy approaches for treating rheumatoid arthritis have made use of gene transfer technology as an improved delivery system for emerging proteins and other biologicals whose activities may have therapeutic value. Preclinical research has focused on two primary directions, evaluation of methods of gene delivery and identification of gene products with antiarthritic potential. Although there are reports involving systemic gene delivery, the bulk of effort has focused on local, intraarticular administration using ex vivo and in vivo methods. Viral-based vectors, including adenoviru

Bat-associated leptospirosis.

Leptospirosis is a globally prevalent disease that affects humans, causing systemic illness that may lead to multi-organ involvement. Clinical signs include sudden fever, general malaise, muscular pain, conjunctival suffusion, and jaundice. Disease is caused by pathogenic bacteria including over 200 serologic variants. Most serologic variants have primary reservoirs in wild mammals, which continually infect and colonize domesticated animals. The organism has been recovered from rats, swine, dogs, cattle, and other animals, notably bats. Most studies have focused on domestic animals as reservoi

The application of a new microfluidic device for the simultaneous identification and quantitation of midazolam metabolites obtained from a single micro-litre of chimeric mice blood.

RATIONALE: Improvements in the design of low-flow highly sensitive chromatographic ion source interfaces allow the detection and characterisation of drugs and metabolites from smaller sample volumes. This in turn improves the ethical treatment of animals by reducing both the number of animals needed and the blood sampling volumes required. METHODS: A new microfluidic device combining an ultra-high pressure liquid chromatography (UHPLC) analytical column with a nano-flow electrospray source is described. All microfluidic, gas and electrical connections are automatically engaged when the cerami

Identification, genotyping, and genomic comparison of Streptococcus porcinus isolated from diseased swine in Brazil.

INTRODUCTION: Streptococcus porcinus is considered a zoonotic opportunistic pathogen for several animal species, including swine, and can cause systemic clinical conditions. There are morphological similarities between streptococcal species, leading to possible incorrect diagnosis and inappropriate treatment choice. Here, we describe the identification, and genotypic and genomic characterization of S. porcinus isolated from 5 adult sick pigs from Minas Gerais and São Paulo (Brazil) between 2010-2017. METHODOLOGY: Streptococcus spp. strains were isolated from vaginal discharge, joint abscess,
