The light-dark forced swim test for simultaneous assessment of behavioral 'despair' and anxiety-like behavior in female mice.

Animal models are a valuable tool to study anxiety and depression, two common and severely debilitating brain disorders. Probing them experimentally typically relies on various rodent behavioral assays, such as the light-dark and the forced swim tests. However, the growing importance of testing novel CNS concepts and neuroactive drugs calls for further refinement of existing behavioral tests, as well as the development of new assays. One research strategy in this direction involves combining principles of several tests into one 'hybrid' assay. Using this approach, here we develop a novel 'hybr

Immunogenicity of antigens from outer membrane vesicles of Neisseria meningitidis associated with bilayer fragment of dioctadecyldimethylammonium in Swiss adult mice.

PURPOSE: In the present study, meningococcal serogroup B outer membrane vesicles (OMVs) were associated with bilayer fragments of a cationic lipid, dioctadecyldimethylammonium (DDA-BF), used as adjuvant, in an antigenic preparation tested in adult female outbred mice. This adjuvant was compared to the traditional adjuvant aluminum hydroxide. MATERIALS AND METHODS: The potential in generating humoral response was evaluated by enzyme-linked immunosorbent assay (ELISA). Individual serum was collected and immunoglobulin G (IgG), IgG1, IgG2a, and IgG2b were quantified. Analyses were carried out 15

Temporal Mapping of Tumor Development in Mice Reveals Late Onset and Benefits of Heterozygosity.

Despite decades of cancer research, the stage of life at which cancerous processes lead to tumor formation in various organs remains poorly understood. This uncertainty largely stems from the lack of systematic monitoring of organs across different life stages, resulting in tumors often being detected at various stages of development and making it difficult to determine when they initially emerged and began to grow. Moreover, individuals in real-world settings are exposed to diverse environmental factors that generate substantial inter-individual variability, as lifestyle undeniably contribute

Treatment with cationic liposome-DNA complexes (CLDCs) protects mice from lethal Western equine encephalitis virus (WEEV) challenge.

Having recently characterized a CD-1 outbred mouse model of pathogenesis for Western equine encephalitis virus, we examined the possible protective effects of cationic liposome-DNA complexes (CLDCs) against encephalitic arboviral infection. In this investigation, mice were pre-treated, co-treated, or post-treated with CLDC then challenged with a subcutaneous or aerosol dose of the highly virulent WEEV-McMillan strain, lethal in mice 4-5 days after inoculation. Pre-treatment with CLDCs provided a significant protective effect in mice, which was reflected in significantly increased survival rate

Evidence of immune system melatonin production by two pineal melatonin deficient mice, C57BL/6 and Swiss strains.

We evaluated two pineal melatonin deficient mice described in the literature, i.e., C57BL/6 and Swiss mice, as animal models for studying the immunomodulatory action of melatonin. Plasma melatonin levels in C57BL/6 and Swiss strains were detectable, but lower than levels in control C3H/HENHSD mice. Since these strains are suppose to be pineal melatonin deficient an extrapineal melatonin synthesis may contribute to plasma levels. Regarding cells and tissues from the immune system, all of them were found to synthesize melatonin although at low levels. N-acetyltransferase (AANAT) mRNA was also am

Humans and rhesus macaques share maturation pathways of HIV-1 envelope-reactive V3-glycan bnAb lineages.

Understanding broadly neutralizing antibody (bnAb) lineage development in rhesus macaques (RMs) infected with simian-human immunodeficiency virus (SHIV) may inform HIV-1 vaccine designs. We analyzed HIV-1 envelope (Env)-antibody coevolution in 18 RMs infected with SHIV.BG505 (subtype A) and found conserved patterns of antibody recognition and Env escape, including in three animals that developed V3-glycan-reactive bnAbs. From one RM with V3-glycan-targeted plasma Abs that neutralized heterologous HIV-1 strains, we isolated 203 members of a single clonal antibody lineage designated DH1030. DH10

Rearing conditions and domestication background determine regulation of hippocampal cell proliferation and survival in adulthood-laboratory CD1 and C57Bl/6 mice versus wild house mice.

Brain development is sensitive to an individual's interaction with its environment. Deprivation of natural environmental stimulation especially in the phase after weaning has long-lasting consequences on neuroplasticity. However, previous findings concerning the effects of rearing environment on adult hippocampal cell proliferation and neurogenesis in rodents remain contradictory. To address the question, whether the variability of hippocampal plasticity in response to environmental conditions is a unique feature at least in mice, the present study examined the effects of social and physical d

Orf Virus-Based Therapeutic Vaccine for Treatment of Papillomavirus-Induced Tumors.

Orf virus (ORFV) represents a suitable vector for the generation of efficient, prophylactic antiviral vaccines against different pathogens. The present study investigated for the first time the therapeutic application of ORFV vector-based vaccines against tumors induced by cottontail rabbit papillomavirus (CRPV). ORFV-CRPV recombinants were constructed expressing the early CRPV gene E1, E2, E7, or LE6. In two independent experiments we used in total 23 rabbits which were immunized with a mixture of the four ORFV-CRPV recombinants or empty ORFV vector as a control 5 weeks after the appearance o

The Diversity Outbred Mouse Population Is an Improved Animal Model of Vaccination against Tuberculosis That Reflects Heterogeneity of Protection.

Many studies of Mycobacterium tuberculosis infection and immunity have used mouse models. However, outcomes of vaccination and challenge with M. tuberculosis in inbred mouse strains do not reflect the full range of outcomes seen in people. Previous studies indicated that the novel Diversity Outbred (DO) mouse population exhibited a spectrum of outcomes after primary aerosol infection with M. tuberculosis Here, we demonstrate the value of this novel mouse population for studies of vaccination against M. tuberculosis aerosol challenge. Using the only currently licensed tuberculosis vaccine, we f

Discordant susceptibility of inbred C57BL/6 versus outbred CD1 mice to experimental fungal sepsis.

Individual susceptibility differences to fungal infection following invasive and/or immunosuppressive medical interventions are an important clinical issue. In order to explore immune response-related factors that may be linked to fungal infection susceptibility, we have compared the response of inbred C57BL/6J and outbred CD1 mouse strains to different experimental models of fungal sepsis. The challenge of animals with the zymosan-induced generalised inflammation model revealed poorer survival rates in C57BL/6J, consistent with lower Th1 cytokine interferon (IFN)-γ serum levels, compared with

Rift Valley fever virus MP-12 vaccine encoding Toscana virus NSs retains neuroinvasiveness in mice.

Rift Valley fever is a mosquito-borne zoonotic disease endemic to sub-Saharan Africa. Rift Valley fever virus (RVFV; genus Phlebovirus, family Bunyaviridae) causes high rates of abortion and fetal malformation in pregnant ruminants, and haemorrhagic fever, neurological disorders or blindness in humans. The MP-12 strain is a highly efficacious and safe live-attenuated vaccine candidate for both humans and ruminants. However, MP-12 lacks a marker to differentiate infected from vaccinated animals. In this study, we originally aimed to characterize the efficacy of a recombinant RVFV MP-12 strain e

Therapeutic Vaccine Against Primate Papillomavirus Infections of the Cervix.

Currently available prophylactic vaccines have no therapeutic efficacy for preexisting human papillomavirus (HPVs) infections, do not target all oncogenic HPVs and are insufficient to eliminate the burden of HPV induced cancer. We aim to develop an alternative HPV vaccine which is broadly effective and capable of clearing preexisting infection. In an initial attempt to develop a broadly reactive therapeutic vaccine, we designed a putative papillomavirus (PV) ancestor antigen (circulating sequence derived antigenic sequences E1E2-CDSE1E2) based on the conserved E1 and E2 protein sequences from

Lung, ileum and heart are predilection sites for AApoAII amyloid deposition in CD-1 Swiss mice used for toxicity studies. Pulmonary amyloid indicates AApoAII.

Amyloid deposits represent frequent histological findings in SPF strains of mice mainly used for toxicological studies. Usually, these are deposits of reactive amyloid (AA-amyloid) derived from the acute phase protein serum amyloid A (SAA). The SAA is an apoprotein of high density lipoprotein (apoSAA). Senescence-accelerated amyloid (ASsam) occurs in a special strain of mice. This type of amyloid is derived from apolipoprotein-AII and, therefore, is called AApoAII. Recently, C57B1/Ka control mice not treated for long duration with immunosuppressive agents, were found to have developed AApoAII-

Morphological Changes in Articular Cartilage and Free-Radical Lipid Peroxidation in Rats with Posttraumatic Osteoarthrosis.

Using the rat model of posttraumatic osteoarthrosis of the knee joint induced by surgical transection of their anterior cruciate ligaments, we showed that irreversible loss of hyaluronan by the extracellular matrix of the joint cartilage tissue against the background of oxidative stress accompanied by accumulation of intermediate LPO products in blood serum and formation of thiol system incompetence was one of the key patterns of dystrophic degeneration of the cartilage tissue. Considerable metabolic shifts were associated with structural modification of the articular hyaline cartilage: its th

[Chronotropic properties of adaptogenic drugs].

Adaptogenic agents of hormonal (melatonin) and plant origin (ginseng and bilobil) produce optimizing effect on circadian rhythm of locomotion and on the temporal dynamics of forced swimming in intact and false-operated rats. Melatonin exhibits the most expressed impact on behavior of animals. After the removal of the pineal gland, these effects of investigated drugs were significantly decreased. It is suggested that the pineal gland may take part in the chronotropic activity of adaptogenic drugs.

Sodium bicarbonate enhances the severity of infection in neutropenic mice orally inoculated with Listeria monocytogenes EGD.

Epidemiological studies have suggested an association between antacid therapy and development of listeriosis in humans. In this study we used a neutropenic mouse model to demonstrate that oral administration of sodium bicarbonate shortly before intragastric (i.g.) inoculation with Listeria monocytogenes EGD (serotype 1/2a) significantly increased the severity of the resulting systemic infection. An explanation for this observation is provided by evidence that L. monocytogenes EGD is rapidly inactivated in synthetic gastric fluid at pH below 5. A second strain of L. monocytogenes (CM [serotype

Hepatic toxicological and multi-omics alterations associated with short- and long-term nonylphenol exposure in mice.

Nonylphenol (NP) is an alkylphenol environmental contaminant with endocrine-disrupting activity and potential hepatotoxicity. However, the molecular responses associated with different NP exposure durations remain insufficiently characterized. In this study, male Kunming mice were exposed to NP through drinking water at 500 μg/L for 10 days (NP-S) or 90 days (NP-L) (n = 8/group), and hepatic toxicological alterations were evaluated using biochemical, histopathological, transcriptomic, metabolomic, and integrative analyses. Network toxicology analysis was first applied as a hypothesis-generatin

Hypoxia Promotes Gluconeogenesis Through PGC-1α in the Liver of Lasiopodomys brandtii.

Oxygen is a critical factor for the survival of most lifeforms, as inadequate availability disrupts internal metabolic balance. Hypoxia-induced disruptions in glucose metabolism can be fatal to many animals. However, there is currently limited research on the energy metabolism of species that inhabit environments with intermittently low oxygen levels. In this study, we investigated the gluconeogenic metabolic response patterns of adult Lasiopodomys brandtii (Brandt's vole) and Mus musculus (Kunming mice) under hypoxia (10% O2, 12 h), followed by 1 h reoxygenation period. Our results indicated

Effect and Mechanism of Tauroursodeoxycholic Acid in Blue Fox Bile on Acute Alcohol-Associated Liver Injury in Mice.

Animal bile, such as bear bile, have long been used in traditional medicine for its therapeutic benefits. Blue fox bile, similar to bear bile, is believed in certain traditional practices to possess hepatoprotective properties. This study examined the components and effects of blue fox bile on alcohol-associated liver injury in mice. Bile was collected from 30 blue foxes and processed into dried powder. The chemical composition of blue fox bile was analyzed using high-performance liquid chromatography and ultra-performance liquid chromatography coupled with quadrupole time-of-flight mass spect

Hepatotoxicity from long-term administration of hepatoprotective low doses of oleanolic acid in mice.

Oleanolic acid is a triterpenoid existed in many medicinal herbs/plants. Oleanolic acid at low doses are hepatoprotective but at high doses produce cholestasis. This study examined hepatotoxicity potential of low doses of oleanolic acid after log-term administration. Male Kunming mice were orally given oleanolic acid at 100, 200 and 300 μmol/kg daily for 14 weeks. Body weights were monitored, and liver injury was determined via blood biochemistry. Histopathology was examined via H&E, Masson, and Sirius red stains. Oleanolic acid accumulation in plasma and liver was determined by LC-MS and hepa
