Isolation and characterization of a major allergen from the fish parasite Anisakis simplex.

BACKGROUND: Ingestion of raw or undercooked fish can lead to infection of human subjects by the fish parasite Anisakis simplex, a disease known as anisakiasis or anisakidosis. Patients sensitized to this fish parasite show high levels of total and specific IgE. Cross-reactions seem to explain the fact that specific IgE antibodies are also found in a high number of normal subjects, as reported in other parasitoses. OBJECTIVE: We sought to purify and characterize a major IgE-binding protein from the parasite. METHODS: A protein was purified from the crude parasite extract by means of ethanol p

Cellular responses in rainbow trout Oncorhynchus mykiss to experimental Anisakis simplex infection.

Fish in wild and cultured populations may be infected with numerous types of pathogens but the host responses vary dependent on both host and parasite species. The present study demonstrates how an experimental infection with endoparasitic nematode larvae (Anisakis simplex) induces cellular and humoral immune responses in rainbow trout Oncorhynchus mykiss. The nematode larvae invaded the peritoneal cavity of the fish following oral administration and became encapsulated by a range of host cells including macrophages, neutrophils, mast cells, fibroblasts, and lymphocytes. The main part (92.7%)

Anisakis larvae (Nematoda: Anisakidae): retrospective morphological, morphometric, biogeography, and taxonomic status analysis.

The family Anisakidae includes parasite genera that are important for public health due to their zoonotic potential. Among these, the genus Anisakis contains some of the most prevalent parasites found in fish that are consumed and commercially exploited in Brazil. Thus, this study aimed to investigate records of the presence of third-stage larvae of Anisakis spp. fish parasites found in Brazilian territory, focusing on their morphological, morphometric, biogeographic, and prevalence aspects over a period of 40 years. This analysis identified the presence of Anisakis larvae in 18 orders, 2 grou

Leukocyte Nucleolus and Anisakis pegreffii-When Falling Apart Means Falling in Place.

The view of the nucleolus as a mere ribosomal factory has been recently expanded, highlighting its essential role in immune and stress-related signalling and orchestrating. It has been shown that the nucleolus structure, formed around nucleolus organiser regions (NORs) and attributed Cajal bodies, is prone to disassembly and reassembly correlated to various physiological and pathological stimuli. To evaluate the effect of parasite stimulus on the structure of the leukocyte nucleolus, we exposed rat peripheral blood mononuclear cells (PBMC) to the crude extract of the nematode A. pegreffii (Ani

Ani s 10, a new Anisakis simplex allergen: cloning and heterologous expression.

Anisakiasis is a human disease caused by accidental ingestion of larval nematodes belonging to the Anisakidae family. Anisakiasis is often associated with a strong allergic response. Diagnosis of A. simplex allergy is currently carried out by test based on the IgE reactivity to a complete extract of L3 Anisakis larvae although the specificity of these diagnostic tests is poor. Improving the specificity of the diagnostic test is possible using purified recombinant allergens. A new Anisakis allergen, named Ani s 10, was detected by immunoscreening an expression cDNA library constructed from L3 A

Anisakis simplex: analysis of expressed sequence tags (ESTs) of third-stage larva.

This study analyzed the expressed sequence tags (ESTs) of the third-stage larvae of Anisakis simplex, in an attempt to gain further insight into its genomic expression patterns. An A. simplex cDNA library was constructed using the Uni-ZAP XR expression vector. A total of 493 clones (insert DNA>400 bp) were sequenced out of 580 clones selected randomly from a cDNA library of the A. simplex third-stage larva. After BLAST search analyses, 154 (31.2%) ESTs were found to have very low similarity, or no match at all to any of the proteins and gene sequences in the published databases. Most matched c

Cross-reactivity between IgE-binding proteins from Anisakis, German cockroach, and chironomids.

Anisakis simplex larvae parasitize animals used as seafood and can produce a specific immune response in man. The ingestion of seafood contaminated with stage three of A. simplex larvae can induce a specific IgE response with clinical symptoms, usually urticaria, even if the fish is cooked before ingestion and the invasive infestation power destroyed by heating. Our preliminary studies showed a strong association of A. simplex sensitization with Ascaris lumbricoides, Daphnia, chironomid spp., Atlantic shrimp (Pandalus borealis), and German cockroach (Blattella germanica). We conducted the cros

Anisakis Contamination in Fillets of Chub Mackerel and Blue Mackerel Sold in Japan.

To evaluate the risk of anisakiasis, we investigated their prevalence in fillets of chub mackerel and blue mackerel in 2022 and 2023. The results indicated that the prevalence (percent of larvae-positive fillet samples) and abundance (number of larvae per 100 g fillet) of Anisakis larvae in fillets of chub mackerel were not significantly different between the Sea of Japan and the Pacific Ocean. The prevalence and abundance of Anisakis larvae in blue mackerel were markedly lower than those in the chub mackerel.

Serum IgA contributes to the comprehension of Anisakis simplex associated chronic urticaria.

The phenotype of allergic diseases associated with Anisakis determines the pattern of cytokines related to antibody production. However, the role of serum IgA and the immunomodulatory mechanisms exerted by active infection of L3 or passive mucosal contact with A. simplex specific antigens has not been studied before. We measured serum cytokine by flow cytometry (IL-2, IL-4, IL-6, IL-10, TNF-α, IFN-γ, IL-17A, TGF-β1) and antibody levels (IgE, IgG4, IgA) by ELISA against total and excretory-secretory (ES) antigens, Ani s 3,and the group of major allergens Ani s 1, Ani s 7, and Ani s 13 in sera f

An investigation of the prevalence and diversity of Anisakis in China: marine food safety implications.

Anisakis can cause Anisakiasis in humans if raw or undercooked fish is consumed. Symptoms of infection may include vomiting, acute abdominal symptoms, or allergies. In this study, we collected 187 commercially available marine fish from the Yellow Sea, East China Sea, and South China Sea. Among them, 79 were found positive containing 520 Anisakis worms. The average prevalence rate was found 42% in this investigation. Ninety-two worms from different sea areas were selected and analyzed for identification, revealing the presence of five different species, which are Anisakis pegreffii, Hysterothy

A case of hepatic anisakidosis caused by Anisakis pegreffii mimicking liver cancer.

Extra-gastrointestinal anisakidosis is rare. We herein report an Anisakis pegreffii infection in a patient with hepatic anisakidosis diagnosed based on its molecular identification. A 71-year-old male patient had a hepatic tumor presenting as a low-density area of 20 mm in diameter in segment 6 of the liver on abdominal ultrasonography, computed tomography, and magnetic resonance imaging. The surgically resected pathological specimen revealed a necrotizing eosinophilic granuloma containing nematode larvae, possibly an Anisakis larva. Molecular and phylogenetic analysis demonstrated Anisakis la

Inflammatory Response in Caco-2 Cells Stimulated with Anisakis Messengers of Pathogenicity.

Background: Anisakis spp. third-stage larvae (L3) are the causative agents of human zoonosis called anisakiasis. The accidental ingestion of L3 can cause acute and chronic inflammation at the gastric, intestinal, or ectopic levels. Despite its relevance in public health, studies on pathogenetic mechanisms and parasite-human interplay are scarce. The aim of this study was to investigate the human inflammatory response to different Anisakis vehicles of pathogenicity. Methods: Human colorectal adenocarcinoma (Caco-2) cells were exposed to Anisakis L3 (the initial contact with the host), extracell

Occurrence of Nematodes of the Genus Anisakis in Mediterranean and Atlantic Fish Marketed in Sardinia.

Anisakiasis is a gastrointestinal fish-borne zoonosis caused by the ingestion of third stage larvae of the genus Anisakis. Between January and December 2013, 1112 specimens of four commercial fish species (Engraulis encrasicolus, Merluccius merluccius, Scomber colias and Trachurus mediterraneus) marketed in Sardinia (Italy) were examined for Anisakis sp. The overall prevalence of Anisakis spp larvae was 39.9%, all morphologically identified as Type I. Scomber colias showed the highest prevalence (100%), followed by M. merluccius (Atlantic 91.0%, Mediterranean 71.2%), T. mediterraneus (32.7%) a

Occurrence of Anisakis (Nematoda: Anisakidae) larvae in unusual hosts in Southern hemisphere.

Nematodes belonging to the genus Anisakis are important parasites due to their abundance in seafood and health impacts on humans. In the present study Anisakis larvae were found in a number of uncommon hosts including the Grey petrel, Procellaria cinerea, the Little penguin, Eudyptula minor, Blue-lipped sea krait, Laticauda laticaudata and Spinner shark, Carcharhinus brevipinna. Morphological examination showed nematodes in these animals are Anisakis larval type I. Genetic characterisation suggested that the larva from one Grey petrel was Anisakis berlandi, whereas the other larvae from the se
