Proteomics-based characterization of apigenin-mediated anti-inflammatory effects in TNF-α-stimulated ARPE-19 cells.

Tumor necrosis factor-α (TNF-α)-induced inflammation in retinal pigment epithelial (RPE) cells contributes to retinal degenerative disorders. Here, we investigated the anti-inflammatory effects of apigenin, a flavonoid compound, in human ARPE-19 cells and explored the underlying mechanisms. Apigenin cytotoxicity was evaluated using the MTT assay, and ARPE-19 cells were then pretreated with apigenin prior to TNF-α stimulation. Apigenin significantly and dose-dependently suppressed TNF-α-induced secretion of interleukin-6 (IL-6), interleukin-8 (IL-8), monocyte chemoattractant protein-1 (MCP-1),

Discovery of natural apigenin analogues as lysine-specific demethylase 1 inhibitors against tumoral testicular germ cells.

Testicular germ cell tumors (TGCT) pose a threat to men's health, discovery of small molecule compounds with potent antiproliferative activity against testicular germ cells is of great significance. Lysine-specific demethylase 1 (LSD1) is overexpressed in tumoral testicular germ cell lines, and it is a promising target for developing agents against TGCT. A series of natural apigenin analogues were evaluated for their inhibitory activity against LSD1, and their structure activity relationships (SARs) were explored. Among them, 8,3'-diprenylapigenin exhibited the most potent inhibitory activity

Natural Dietary Flavonoid Apigenin Mitigates Ulcerative Colitis via Modulating the AMPK/NF-κB/NLRP3 Signaling Axis.

Ulcerative colitis (UC) is a challenging inflammatory disease with higher relapse and lower remission rates, urgently requiring effective and safe complementary treatment options. Apigenin (Api), a natural flavonoid from parsley and celery, exhibits potent antioxidant and anti-inflammatory activities. In mice with 2.5% DSS-induced acute colitis, Api markedly alleviated weight loss, colon shortening, and elevated DAI scores, while restoring mucosal integrity and reducing oxidative stress and inflammation. Mechanistically, Api can directly bind to AMPK to activate it, thereby alleviating oxidati

Apigenin ameliorates inflamed ulcerative colitis by regulating mast cell degranulation via the PAMP-MRGPRX2 feedback loop.

PURPOSE: The aim of this study was to investigate the therapeutic effect of API on UC via the regulation of PAMP-MRGPRX2-mediated mast cells (MCs) degranulation. BACKGROUND: The pro-inflammatory positive feedback loop mediated by Mas-related G-protein-coupled receptor X2 (MRGPRX2) and its endogenous ligand, PAMP-12, is associated with ulcerative colitis (UC) progression. However, the therapeutic strategies that target MRGPRX2 in the treatment of UC are less reported. Apigenin (API), a natural flavonoid, can relieve inflammation. METHOD: A dextran sodium sulfate (DSS)-induced mouse UC model w

Apigenin attenuates depressive-like behavior via modulating monoamine oxidase A enzyme activity in chronically stressed mice.

Chronic stress is a risk factor for depression and is characterized by elevated levels of brain monoamine oxidase A (MAOA). Mounting evidence has shown that MAOA is a biochemical link between stress and depression. Apigenin (API), a natural flavonoid, as demonstrated in vitro inhibitory effect on MAOA, is suggestive of antidepressant-like activity. However, the in vivo inhibitory effect of API on MAOA and how it affects depression still remain unclear. Here, we report the probable mechanisms of action of API in chronic unpredictable mild stress (CUMS)-induced depression in mice. Treatment with

Apigenin, a novel candidate involving herb-drug interaction (HDI), interacts with organic anion transporter 1 (OAT1).

BACKGROUND: Apigenin is a flavonoid compound, widely distributed in natural plants. Various studies have suggested that apigenin has inhibitory effects towards several drug transporters, such as the organic anion transporting (OAT) polypeptides, 1B1 and 1B3 (OATP1B1 and OATP1B3). However, the mechanism by which apigenin interacts with OAT1 has not been well studied. METHODS: MDCK cells stably-expressing OAT1 were used to examine the inhibitory effects of apigenin on OAT1. UPLC-MS/MS was used to evaluate the in vitro and in vivo effects of apigenin on the uptake of acyclovir by OAT1. Cytotoxic

Activation of TRPV4 by dietary apigenin antagonizes renal fibrosis in deoxycorticosterone acetate (DOCA)-salt-induced hypertension.

Hypertension-induced renal fibrosis contributes to the progression of chronic kidney disease, and apigenin, an anti-hypertensive flavone that is abundant in celery, acts as an agonist of transient receptor potential vanilloid 4 (TRPV4). However, whether apigenin reduces hypertension-induced renal fibrosis, as well as the underlying mechanism, remains elusive. In the present study, the deoxycorticosterone acetate (DOCA)-salt hypertension model was established in male Sprague-Dawley rats that were treated with apigenin or vehicle for 4 weeks. Apigenin significantly attenuated the DOCA-salt-induc

Apigenin ameliorates hypertension-induced cardiac hypertrophy and down-regulates cardiac hypoxia inducible factor-lα in rats.

Apigenin is a natural flavonoid compound that can inhibit hypoxia-inducible factor (HIF)-1α expression in cultured tumor cells under hypoxic conditions. Hypertension-induced cardiac hypertrophy is always accompanied by abnormal myocardial glucolipid metabolism due to an increase of HIF-1α. However, whether or not apigenin may ameliorate the cardiac hypertrophy and abnormal myocardial glucolipid metabolism remains unknown. This study aimed to examine the effects of apigenin. Rats with cardiac hypertrophy induced by renovascular hypertension were treated with apigenin 50-100 mg kg(-1) (the doses

Apigenin Alleviates Ligamentum Flavum Ossification by Dual-Targeting CA9 and CDK6 and Modulating Calcium/PI3K-Akt Signaling.

While apigenin shows therapeutic potential for bone-related disorders like osteoporosis and osteoarthritis, its mechanisms in regulating ligamentum flavum ossification (OLF) are unclear. An integrated computational strategy combining network pharmacology, machine learning, and molecular docking was used to study apigenin's therapeutic potential for OLF. First, differential gene expression profiling was done using the GSE113212 dataset to find OLF-related differentially expressed genes. At the same time, apigenin's putative targets and bioactive components were collected from pharmacological da

Apigenin enhances Nrf2-induced chaperone-mediated autophagy and mitigates α-synuclein pathology: Implications for Parkinson's disease therapy.

BACKGROUND: The aggregation of α-synuclein (SNCA) in dopaminergic neurons of the substantia nigra is a key factor in the pathogenesis of Parkinson's disease (PD). Despite years of drug discovery efforts targeting SNCA aggregation, no disease-modifying drugs have been approved to date. The failure of numerous clinical trials can be attributed, at least in part, to the difficulty in identifying potent compounds during preclinical investigations. OBJECTIVE: Establish a screening approach based on molecular docking and autophagic flux detection to identify natural compounds from new perspectives

Multifunctional iron-apigenin nanocomplex conducting photothermal therapy and triggering augmented immune response for triple negative breast cancer.

Triple negative breast cancer (TNBC) presents a formidable challenge due to its low sensitivity to many chemotherapeutic drugs and a relatively low overall survival rate in clinical practice. Photothermal therapy has recently garnered substantial interest in cancer treatment, owing to its swift therapeutic effectiveness and minimal impact on normal cells. Metal-polyphenol nanostructures have recently garnered significant attention as photothermal transduction agents due to their facile preparation and favorable photothermal properties. In this study, we employed a coordinated approach involvin

β3-Adrenoreceptor Activity Limits Apigenin Efficacy in Ewing Sarcoma Cells: A Dual Approach to Prevent Cell Survival.

Ewing Sarcoma (ES) is an aggressive paediatric tumour where oxidative stress and antioxidants play a central role in cancer therapy response. Inhibiting antioxidants expression, while at the same time elevating intracellular reactive oxygen species (ROS) levels, have been proposed as a valid strategy to overcome ES cancer progression. Flavonoid intake can affect free radical and nutritional status in children receiving cancer treatment, but it is not clear if it can arrest cancer progression. In particular, apigenin may enhance the effect of cytotoxic chemotherapy by inducing cell growth arres
