Continuous apomorphine infusion modulates cortical hyperexcitability and abnormal plasticity in Parkinson's disease.

BackgroundSeveral studies have demonstrated the clinical benefit of apomorphine subcutaneous infusion in Parkinson's disease (PD). However, it remains unclear whether apomorphine infusion also modulates the abnormal motor cortical excitability and plasticity that contribute to PD pathophysiology and progression.MethodsWe investigated the effects of continuous apomorphine infusion on cortical excitability and plasticity in patients with moderate-advanced PD, assessed at baseline and after 1, 12, and 24 weeks of treatment. Transcranial magnetic stimulation (TMS) was used to evaluate: i) motor co

Attenuation of age-related cognitive decline and memory deficits through apomorphine administration.

Oxidative stress, stemming from heightened production of reactive oxygen species and free radicals, significantly contributes to the aging process. Apomorphine emerges as a pivotal medication for managing Alzheimer's, Parkinson's and other age-related conditions. This study aims to explore the memory-enhancing and neuroprotective properties of apomorphine, utilizing male Albino Wistar rats aged 4 and 24 months as subjects. Rats were intraperitoneally injected with apomorphine for 6 days. Decreased glutathione peroxidase, superoxide dismutase and catalase activities with increased lipid peroxid

Apomorphine rescues reactive oxygen species-induced apoptosis of fibroblasts with mitochondrial disease.

Mitochondrial disease is a genetic disorder in which individuals suffer from energy insufficiency. The various clinical phenotypes of mitochondrial disease include Leigh syndrome (LS), myopathy encephalopathy lactic acidosis and stroke-like episodes (MELAS). Thus far, no curative treatment is available, and effective treatment options are eagerly awaited. We examined the cell protective effect of an existing commercially available chemical library on fibroblasts from four patients with LS and MELAS and identified apomorphine as a potential therapeutic drug for mitochondrial disease. We conduct

Dissociation between spontaneously emitted and apomorphine-induced stereotypy in Peromyscus maniculatus bairdii.

Stereotyped behavior is repetitive, topographically invariant motor activity that lacks an obvious function. We have previously characterized the spontaneous and persistent stereotypies that occur in deer mice housed in standard laboratory cages. Providing these animals with enriched environments markedly reduces their vulnerability to develop stereotypic behavioral repertoires, thus enabling us to generate behaviorally distinct (stereotypic and nonstereotypic) mice of the same species. As stereotypic behaviors are readily induced by systemic administration of a dopamine (DA) agonist, the pres

Therapeutic Spectrum of Intermittent Apomorphine in Parkinson's Disease: Neuropsychiatric, Behavioral, and Cognitive Analyses.

INTRODUCTION: Intermittent subcutaneous (SC) apomorphine is a fast-acting dopamine agonist used in the management of motor fluctuations in advanced Parkinson's disease (PD). While its motor benefits are well established the effects on non-motor domains and the modifying role of chronological age remain less certain. This prospective observational study aimed to evaluate the effectiveness of intermittent SC apomorphine on motor and non-motor outcomes in idiopathic PD patients with persistent motor fluctuations despite optimized oral therapy, and to examine whether age influenced therapeutic res

Risperidone on apomorphine-induced stereotyped behavior and auditory sensory gating in rhesus monkeys.

The ameliorating effect of risperidone on apomorphine-induced stereotyped behavior and inhibition of auditory sensory gating was investigated using rhesus monkeys. The total duration of the stereotyped behavior observed in the control group was 43.7 ± 23.0 s (n = 3) between 10 and 25 min after vehicle administration, whereas the duration in the apomorphine-treated (0.1 or 0.15 mg/kg i.m., n = 3) group was observed to be significantly prolonged to 413.0 ± 150.6 s. Administration of 0.01, 0.03, 0.1 mg/kg of risperidone 60 min before apomorphine, significantly reduced the duration of this apomorp

Lesions of the entopeduncular nucleus in rats prevent apomorphine-induced deficient sensorimotor gating.

Dopamine-induced hyperactivity and deficient sensorimotor gating, measured as prepulse inhibition (PPI) of the acoustic startle response (ASR), are used as animal models for neuropsychiatric disorders such as schizophrenia and Tourette's syndrome. We here investigated whether excitotoxic lesions of the rat entopeduncular nucleus (EPN), the equivalent to the human globus pallidus internus (GPi), would improve apomorphine-induced PPI-deficits and hyperactivity. Additionally, we investigated the effect of EPN lesions on cognition, motivation and motor skills. In male Sprague Dawley rats bilateral

Bell's palsy-induced blepharospasm relieved by passive eyelid closure and responsive to apomorphine.

OBJECTIVE: We describe the case of a woman with Bell's Palsy-induced blepharospasm (BPIB) of the right eye that appeared simultaneously with a complete left facial nerve palsy. The involuntary spasm was relieved by passive lowering of the upper eyelid on the paretic side. METHODS: The recovery curve of the blink reflex was evaluated on the non-paretic side in baseline conditions, after subcutaneous apomorphine and placebo administration and 8 months later, at recovery from the palsy. RESULTS: We found increased recovery of the test-R2 responses at short interstimulus intervals at baseline, w

Blood pressure effects of intravenous apomorphine in conscious deoxycorticosterone-acetate salt-hypertensive rats.

The present study reports the effects of apomorphine (APO) on blood pressure and the principal site of action of this agonist in 4-week deoxycorticosterone-acetate (DOCA)-hypertensive conscious rats. In these preprations, intravenous (i.v.) administration of APO (0.50-1 mg/kg) induced short-lasting and dose-dependent decreases in mean arterial pressure. The hypotensive response to APO (0.3 mg/kg) was reversed into a significant pressor effect by i.v. hexamethonium (30 mg/kg), whereas it was enhanced by i.v. pretreatment with the vasopressor antagonist of arginine vasopressin (AVP) d(CH2)5Tyr(M

New dopamine D2 receptor polymorphisms in rats and association with apomorphine-induced stereotypies.

Adult Wistar rats injected with the dopamine receptor agonist apomorphine display different types of motility patterns with respect to oral stereotypes and locomotor activities. It was tested whether phenotypes exhibiting either 'sniffing' or 'non-sniffing' behaviour differed in gene structures of dopamine receptors D1 or D2. Forty-five Wistar rats of both genders were tested after a single dose of apomorphine (2 mg/kg s.c.) for stereotyped behaviour. Sequence analysis of the 5' flanking region, the 5' untranslated region and the coding region of the two genes revealed a new sequence for the 5

Differential responses in central dopaminergic activity induced by apomorphine in IPL nude rat.

The IPL nude rat, derived by spontaneous mutation from the Sprague-Dawley strain, presents alterations in the prolactin synthesis and secretion due to an increased dopaminergic inhibition. However, there are no reports concerned to central dopamine activity. The corpus striatum is a brain area involved in the development of stereotyped behavior after the activation of mesolimbic and/or nigro-striatal dopamine pathways. In order to identify possible mesolimbic and/or nigro-striatal dysfunctions in the IPL nude rat, we study the spontaneous oral behaviors and the effects of apomorphine-induced d

Involvement of dopamine D2 receptors in apomorphine-induced facilitation of forebrain serotonin output.

The effect of systemic administration of the nonselective dopamine receptor agonist apomorphine on efflux of serotonin (5-hydroxytryptamine, 5-HT) in striatum and hippocampus of freely moving rats was examined using in vivo microdialysis. 5-HT efflux was increased by a moderate dose of apomorphine sufficient for a postsynaptic dopaminergic effect (0.5 mg/kg, s.c.), but not by a lower dose (0.1 mg/kg, s.c.), that acts preferentially on presynaptic dopamine receptors. This effect was blocked by a dopamine D2 receptor antagonist raclopride, administered either systemically or locally into striatu

Postpartum exposure to triethyl tin produces long-term alterations in responsiveness to apomorphine.

Male and female rat pups were exposed to a single dose of triethyl tin (TET) or ethanol vehicle on day 5 postnatally and tested for responsiveness to apomorphine as adults. TET-exposed animals displayed an increased response to the stereotypic effects of 1 mg/kg of apomorphine. Female rats exposed neonatally to TET had decreased baseline rates of responding on a food reinforced variable interval (VI) 15 sec. schedule, while TET-exposed males were found to have increased baseline rates relative to controls. The rats maintained on a VI 15 sec. schedule were given various doses of apomorphine (0.

Identification of the efficacy and safety of apomorphine in the treatment of Parkinson's disease via meta-analysis.

BACKGROUND: The majority of Parkinson's disease (PD) patients experience motor fluctuations and dyskinesias as the disease progresses. Apomorphine has been used as an acute treatment for off episodes in PD in recent years. However, conclusions from clinical studies regarding the efficacy and safety of apomorphine have been inconsistent, particularly prior to the publication of larger pivotal trials. OBJECTIVES: To systematically evaluate the efficacy and safety of apomorphine in PD patients, a meta-analysis of randomized controlled trials (RCTs) and single-armed studies were performed. METHO

The prevalence of impulsive compulsive behaviors in patients treated with apomorphine infusion: a retrospective analysis.

BACKGROUND: Impulsive compulsive behaviors (ICBs) can affect a significant number of Parkinson's disease (PD) patients. OBJECTIVE: We have studied brain samples from a brain bank of PD patients who received apomorphine via continuous infusion in life to assess the prevalence and outcome of ICBs. METHODS: A search on the Queen Square Brain Bank (QSBB) database for cases donated from 2005 to 2016 with a pathological diagnosis of idiopathic PD was conducted. Notes of all donors who used apomorphine via continuous infusion for at least three months were reviewed. Clinical and demographic data we
