Gamma-aminobutyric acid and glutamic acid levels in the auditory pathway of rats with chronic tinnitus: a direct determination using high resolution point-resolved proton magnetic resonance spectroscopy (H-MRS).

Damage to the auditory system following high-level sound exposure reduces afferent input. Homeostatic mechanisms appear to compensate for the loss. Overcompensation may produce the sensation of sound without an objective physical correlate, i.e., tinnitus. Several potential compensatory neural processes have been identified, such as increased spontaneous activity. The cellular mechanisms enabling such compensatory processes may involve down-regulation of inhibitory neurotransmission mediated by γ-amino butyric acid (GABA), and/or up-regulation of excitatory neurotransmission, mediated by gluta

[Models of tinnitus development : From cochlea to cortex].

The development of subjective tinnitus is still not mechanistically understood and existing models are controversially discussed. In this overview, the authors discuss three of the main models, all of which propose damage to the cochlea as the initial step in tinnitus development. Based on these models, a possible manifestation of tinnitus-related neuronal activity at the perceptually relevant level of the auditory pathway, the auditory cortex, is presented. Furthermore, it is demonstrated that one of the models offers a new view on the phenomenon, which could potentially help patients to bett

Auditory processing control by the medial prefrontal cortex: A review of the rodent functional organisation.

Afferent inputs from the cochlea transmit auditory information to the central nervous system, where information is processed and passed up the hierarchy, ending in the auditory cortex. Through these brain pathways, spectral and temporal features of sounds are processed and sent to the cortex for perception. There are also many mechanisms in place for modulation of these inputs, with a major source of modulation being based in the medial prefrontal cortex (mPFC). Neurons of the rodent mPFC receive input from the auditory cortex and other regions such as thalamus, hippocampus and basal forebrain

Electrophysiological Evidence for the Sources of the Masking Level Difference.

PURPOSE: The purpose of this review article is to review evidence from auditory evoked potential studies to describe the contributions of the auditory brainstem and cortex to the generation of the masking level difference (MLD). METHOD: A literature review was performed, focusing on the auditory brainstem, middle, and late latency responses used in protocols similar to those used to generate the behavioral MLD. RESULTS: Temporal coding of the signals necessary for generating the MLD occurs in the auditory periphery and brainstem. Brainstem disorders up to wave III of the auditory brainstem r

Exposure to high-altitude hypobaric hypoxic environment induces low-frequency hearing loss in C57BL/6J mice: Mediated by slowing down the postsynaptic electrical signal transmission speed in the cochlear-inferior colliculus auditory signaling pathway.

OBJECTIVE: To reveal the pattern of effects of exposure to high-altitude hypobaric hypoxia environment on the auditory function of C57BL/6J mice, so as to provide a theoretical basis for the targeted prevention and treatment of hearing loss, tinnitus, and otogenic vertigo in high-altitude areas. METHODS: Forty male C57BL/6J mice were randomly divided into a control group and an experimental group. The experimental group was further subdivided into 9 subgroups according to the exposure duration (3, 5, 7, 10, 15, 20, 25, 30, and 35 days), with 4 mice in each subgroup. Mice in the experimental g

Cerebellum mediates modality-specific modulation of sensory responses of midbrain and forebrain in rat.

Evidence of a sensory role of the cerebellum, mediating a modulation of effectiveness of afferent input at other parts of the brain, has been reported previously for certain sense modalities but has not been evaluated across several in a mammal. After a conditioning train of stimuli applied to the cerebellar surface in unanesthetized rats, diffuse flashes, acoustic clicks, and shocks to the sciatic nerve evoked multiunit and field potential responses that were recorded at three levels: midbrain, thalamus, and cerebral cortex. At a best interval between end of conditioning train (cerebellar) an

The interhemispheric miscommunication theory of auditory verbal hallucinations in schizophrenia.

Auditory verbal hallucinations (AVH) are hallmark symptoms of schizophrenia and have been linked to abnormal activation, connectivity and integration within the auditory, language, and memory brain networks. The interhemispheric miscommunication theory of AVH is based on a steadily growing number of studies using a variety of modalities (EEG, fMRI, DTI) reporting that both altered integrity of the interhemispheric auditory pathways and disturbed functional gamma-band synchrony between right and left auditory cortices significantly contribute to abnormal auditory processing and the emergence of

Priming central sound processing circuits through induction of spontaneous activity in the cochlea before hearing onset.

Sensory systems experience a period of intrinsically generated neural activity before maturation is complete and sensory transduction occurs. Here we review evidence describing the mechanisms and functions of this 'spontaneous' activity in the auditory system. Both ex vivo and in vivo studies indicate that this correlated activity is initiated by non-sensory supporting cells within the developing cochlea, which induce depolarization and burst firing of groups of nearby hair cells in the sensory epithelium, activity that is conveyed to auditory neurons that will later process similar sound feat

The neglected neglect: auditory neglect.

Whereas visual and somatosensory forms of neglect are commonly recognized by clinicians, auditory neglect is often not assessed and therefore neglected. The auditory cortical processing system can be functionally classified into 2 distinct pathways. These 2 distinct functional pathways deal with recognition of sound ("what" pathway) and the directional attributes of the sound ("where" pathway). Lesions of higher auditory pathways produce distinct clinical features. Clinical bedside evaluation of auditory neglect is often difficult because of coexisting neurological deficits and the binaural na

Brainstem connections of the macula lagenae in the chicken.

The macula lagenae, an otolithic hair-cell organ with probable vestibular function, lies close to the apical end of the avian auditory hair-cell epithelium, the papilla basilaris. In an earlier study in the pigeon in which lesioning techniques were used, Boord and Rasmussen ([1963] J. Comp. Neurol. 120:463-473) reported finding a projection of lagenar fibers to parts of the cochlear nuclei (nucleus magnocellularis and nucleus angularis). Subsequent to this report, it has been generally assumed that at least part of the cochlear nuclei has a vestibular or a combined vestibular-auditory function

Auditory hallucinations.

Auditory hallucinations constitute a phenomenologically rich group of endogenously mediated percepts which are associated with psychiatric, neurologic, otologic, and other medical conditions, but which are also experienced by 10-15% of all healthy individuals in the general population. The group of phenomena is probably best known for its verbal auditory subtype, but it also includes musical hallucinations, echo of reading, exploding-head syndrome, and many other types. The subgroup of verbal auditory hallucinations has been studied extensively with the aid of neuroimaging techniques, and from

Hyperactivity in the medial olivocochlear efferent system is a common feature of tinnitus and hyperacusis in humans.

Tinnitus and hyperacusis are common, burdensome sources of morbidity with a high rate of co-occurrence. Knudson et al. (J Neurophysiol 112: 3197-3208, 2014) demonstrated that efferent suppression of cochlear activity by the medial olivocochlear system is enhanced in individuals with tinnitus and/or hyperacusis. Their findings stress that atypical activity in the efferent auditory pathway may represent a shared substrate, as well as a potential therapeutic target, in tinnitus and hyperacusis.

Road traffic noise exposure and its impact on health: evidence from animal and human studies-chronic stress, inflammation, and oxidative stress as key components of the complex downstream pathway underlying noise-induced non-auditory health effects.

In heavily urbanized world saturated with environmental pollutants, road traffic noise stands out as a significant factor contributing to widespread public health issues. It contributes in the development of a diverse range of non-communicable diseases, such as cardiovascular diseases, metabolic dysregulation, cognitive impairment, and neurodegenerative disorders. Although the exact mechanisms behind these non-auditory health effects remain unclear, the noise reaction model centres on the stress response to noise. When exposed to noise, the body activates the hypothalamic-pituitary-adrenal axi

Childhood autism and auditory system abnormalities.

Hearing disorders are common among children with autism, ranging from peripheral and sensorineural hearing deficit or loss to auditory hypersensitivity with bizarre reactions to sounds. The auditory abnormalities and consequent sensory deprivation exacerbate the communication deficit of autism, and early auditory assessment holds an important place in the planning of intervention and the overall prognosis of patients. Physiologic, pathologic, imaging, and neurochemical studies have revealed an array of aberrations in the perception and processing of the audiologic stimuli, including (among oth

Differential Myeloarchitecture of Subfields of the Mammalian Auditory Cortex: A Comparative Approach.

BACKGROUND: The mammalian auditory cortex can be parcellated into multiple functional subfields, with each subfield making a distinct contribution to sound processing. For example, primary fields consisting of primary auditory cortex and anterior auditory field are the first to receive information from the thalamus, and the neurons in each of these fields have different properties in terms of latency and response duration. Non-primary auditory fields consist of secondary auditory cortex, which is involved in object recognition and emotional conditioning, while dorsal auditory fields are more r

Review: Neural Mechanisms of Tinnitus and Hyperacusis in Acute Drug-Induced Ototoxicity.

Purpose Tinnitus and hyperacusis are debilitating conditions often associated with age-, noise-, and drug-induced hearing loss. Because of their subjective nature, the neural mechanisms that give rise to tinnitus and hyperacusis are poorly understood. Over the past few decades, considerable progress has been made in deciphering the biological bases for these disorders using animal models. Method Important advances in understanding the biological bases of tinnitus and hyperacusis have come from studies in which tinnitus and hyperacusis are consistently induced with a high dose of salicylate, th

The Neglect of Recent Research on the Influence of the Cerebellum on Auditory Processing.

Classic studies beginning in the 1940s started to demonstrate, in animals, activation of different cerebellar regions following the delivery of auditory stimuli. Stimulation of these cerebellar regions elicits responses in other brain regions known to be involved in auditory processing. Lesions produced in specific cerebellar regions and the use of molecular markers suggest bidirectional connectivity between the cerebellum and the cochlear nucleus. Although such evidence exists, the findings are highly heterogeneous with respect to the vermal and cerebellar hemispheric regions potentially invo

[Research progress in auditory center plasticity and tinnitus mechanism].

Tinnitus is one of the most common clinical symptoms of otology, and its pathogenesis is still unclear. The mechanism of tinnitus has been studied through a cognitive progress from the periphery (cochlea) to auditory center to the limbic system. Auditory peripheral lesions, such as damages to ribbon synapses, may form excitatory deafferentation, then it induces the auditory center to start the compensatory gain, leading to an increase in excitatory response; If the damage is further aggravated, it may cause continuous enhancement of central gain effect, hyperexcitability may occur and leading
