Activation of adrenal preganglionic neurons during autonomic dysreflexia in the chronic spinal cord-injured rat.

Autonomic dysreflexia (AD) occurs in a majority of high paraplegic and quadriplegic patients and is particularly characterized by a paroxysmal hypertension elicited by somatic or visceral stimuli. We have previously shown that plasma adrenaline and noradrenaline levels were significantly increased during episodes of AD in the 30-day spinal cord-injured (SCI) rats, suggesting the participation of adrenal catecholamines in the cardiovascular changes associated to AD. Thus, adrenal sympathetic preganglionic neurons (SPN) could be activated by visceral afferences leading to AD. The aim of this stu

Two types of parasympathetic preganglionic neurones in the superior salivatory nucleus characterized electrophysiologically in slice preparations of neonatal rats.

1. The electrophysiological properties of parasympathetic preganglionic neurones in the superior salivatory nucleus were studied in thin- and thick-slice preparations of rats aged 1 and 2 weeks using the whole-cell patch-clamp technique. 2. The superior salivatory neurones were identified by a retrograde tracing method with dextran-tetramethylrhodamine-lysine. The injection of the tracer into the chorda-lingual nerve labelled the neurones innervating the submandibular ganglia and those innervating the intra-lingual ganglia, while the injection into the tip of the tongue labelled the latter gro

RVLM C1 Neurons Innervate Sacral as well as Thoracolumbar Autonomic Preganglionic Neurons in the Rat.

We examined the distribution of axons throughout the spinal cord of the rat that were either immunoreactive for the adrenaline-synthesizing enzyme, phenylethanolamine N-methyltransferase (PNMT), or derived from medullary C1 neurons, one of the three groups of neurons in the brain that synthesize PNMT. We observed that PMNT-immunoreactive axons, as well as C1 axons labelled with GFP from viral transduction, innervate most, but not all, sympathetic preganglionic neurons in the thoracolumbar spinal cord. GFP-positive C1 axons provided innervation to sympathetic preganglionic neurons that expresse

A novel strategy for repairing preganglionic cervical root avulsion in brachial plexus injury by sural nerve grafting.

OBJECT: In this study, the authors evaluated the efficacy of a new surgical strategy for reconnecting the injured brachial plexus with the spinal cord using fibrin glue containing acidic fibroblast growth factor as an adhesive and neurotrophic agent. METHODS: Eighteen patients with preganglionic brachial plexus injuries, each with varying degrees of upper limb dysfunction, underwent cervical laminectomy with or without sural nerve grafting. The treatment of each avulsed root varied according to the severity of the injury. Some patients also underwent a second-stage operation involving supracl

The pupillary and ciliary components of the cat Edinger-Westphal nucleus: a transsynaptic transport investigation.

The distribution of preganglionic motoneurons supplying the ciliary ganglion in the cat was defined both qualitatively and quantitatively. These cells were retrogradely labeled directly, following injections of wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP) into the ciliary ganglion, or were transsynaptically labeled following injections of WGA into the vitreous chamber. Almost half of the cells are distributed rostral to the oculomotor nucleus, both in and lateral to the anteromedian nucleus. Of the remaining preganglionic motoneurons, roughly 20% of the total are locate

Location of parotid preganglionic neurons in the inferior salivatory nucleus and their relation to the superior salivatory nucleus of rat.

In major brain maps the location of the salivatory nuclei of the rat is depicted from the level of the root of the facial nerve to the level of the rostral tip of the nucleus of the solitary tract. Most published data deal with the superior salivatory nucleus (SSN). In the present study the topography of the parasympathetic preganglionic neurons of the inferior salivatory nucleus (ISN) that innervate the parotid gland through the otic ganglion was determined by means of a retrograde transneuronal labeling technique. Parasympathetic, sympathetic and sensory neurons were labeled following inject

A retrograde tracer strategy using True Blue to label the preganglionic parasympathetic innervation of the abdominal viscera.

A simple and reliable method for labeling the preganglionic neurons which innervate the abdominal viscera is described. Infusion of a True Blue (TB) suspension in several different concentrations directly into the peritoneal cavity consistently labeled the medullary vagal nuclei including the dorsal motor nucleus (DMN), nucleus ambiguus (NA), and retrofacial nucleus (RFN). Quantitative analysis of cell labeling after 30 microliters infusions of a large range of concentrations of TB (0.001-20.0%) showed that: (1) as little as 0.0075 mg of TB was sufficient to label DMN cells distinctly while on

Circulatory responses to ketamine: dependence on respiratory pattern and background anaesthesia in the rabbit.

In rabbits which were initially conscious or lightly anaesthetized with pentobarbitone, ketamine respectively increased or did not change arterial pressure, whereas in mechanically ventilated animals there was prolonged depression of both pressure and pregangkioinic sympathetic activity. Although respiratory rate slowed during spontaneous ventilation, blood-gas changes were not responsible for these differences. Following bilateral division of the carotid sinus and aortic nerves, a depressor response to ketamine occurred during spontaneous respiration. When respiratory rate was slowed coincide

Assessment of sudomotor function.

PURPOSE: To review the currently available literature on clinical autonomic tests of sudomotor function. METHODS: We searched PubMED/MEDLINE for articles on technical principles and clinical applications of sudomotor tests with a focus on their drawbacks and perspectives in order to provide a narrative review. RESULTS: The quantitative sudomotor axon reflex sweat test (QSART) is the most widely used test of sudomotor function. The technique captures pathology with low intra- and inter-subject variability but is limited by technical demands. The thermoregulatory sweat test comprises topograph

Partial restoration of cardiovascular function by embryonic neural stem cell grafts after complete spinal cord transection.

High-level spinal cord injury can lead to cardiovascular dysfunction, including disordered hemodynamics at rest and autonomic dysreflexia during noxious stimulation. To restore supraspinal control of sympathetic preganglionic neurons (SPNs), we grafted embryonic brainstem-derived neural stem cells (BS-NSCs) or spinal cord-derived neural stem cells (SC-NSCs) expressing green fluorescent protein into the T4 complete transection site of adult rats. Animals with injury alone served as controls. Implanting of BS-NSCs but not SC-NSCs resulted in recovery of basal cardiovascular parameters, whereas b

Synaptic connections between endomorphin 2-immunoreactive terminals and μ-opioid receptor-expressing neurons in the sacral parasympathetic nucleus of the rat.

The urinary bladder is innervated by parasympathetic preganglionic neurons (PPNs) that express μ-opioid receptors (MOR) in the sacral parasympathetic nucleus (SPN) at lumbosacral segments L6-S1. The SPN also contains endomorphin 2 (EM2)-immunoreactive (IR) fibers and terminals. EM2 is the endogenous ligand of MOR. In the present study, retrograde tract-tracing with cholera toxin subunit b (CTb) or wheat germ agglutinin-conjugated horseradish peroxidase (WGA-HRP) via the pelvic nerve combined with immunohistochemical staining for EM2 and MOR to identify PPNs within the SPN as well as synaptic c

Review of the relationship of restless legs syndrome and periodic limb movements in sleep to hypertension, heart disease, and stroke.

Evidence is reviewed documenting an intimate relationship among restless legs syndrome (RLS) / periodic limb movements in sleep (PLMS) and hypertension and cardiovascular and cerebrovascular disease. Sympathetic overactivity is associated with RLS/PLMS, as manifested by increased pulse rate and blood pressure coincident with PLMS. Causality is far from definitive. Mechanisms are explored as to how RLS/PLMS may lead to high blood pressure, heart disease, and stroke: (a) the sympathetic hyperactivity associated with RLS/PLMS may lead to daytime hypertension that in turn leads to heart disease an

Characterization of substance P release from the intermediate area of rat thoracic spinal cord.

Substance P (SP) nerve terminals innervate the intermediolateral cell column (IML) of the thoracic spinal cord, where SP coexists with serotonin (5-HT), neurokinin A (NKA) and thyrotropin-releasing hormone (TRH). Neither the depolarization-induced release of SP nor the presence of other neurochemicals in the regulation of SP release has been directly studied in this system. In the present study, basal and K(+)-stimulated release of SP from the microdissected intermediate area (including the IML, intercalated nucleus and central autonomic nucleus) of the rat thoracic spinal cord, and the regula

The origin of the preganglionic parasympathetic fibres to the mandibular and sublingual salivary glands in the rat: a horseradish peroxidase study.

Horseradish peroxidase was applied to a ligation of the chorda tympani nerves running alongside the submandibular duct in order to label the preganglionic parasympathetic salivatory neurons in the brain stem. Labelled neurons were found in the ipsilateral motor trigeminal nucleus, lateral reticular formation and in the facial motor nuclei of both sides. However, the labelling of neurons in the motor nuclei of the trigeminal and facial nucleus was caused by the uptake of horseradish peroxidase by axons in the musculature surrounding the injection site which were probably damaged by the dissecti

Simultaneous identification of two populations of sympathetic preganglionic neurons using recombinant herpes simplex virus type 1 expressing different reporter genes.

We generated neurotropic herpes simplex type 1 viruses expressing human placental alkaline phosphatase and studied the utility of this enzyme as a marker of infected neurons. The neurotropism of these viruses was assessed by their ability to infect sympathetic preganglionic neurons after adrenal injection in hamsters. The transneuronal transfer of these viruses was examined by their ability to cross the peripheral synapse from the kidney to renal preganglionic neurons or to cross the central synapse from the adrenal gland to the medulla oblongata. Finally, we injected an alkaline phosphatase-e
