The duration of persistence of S. sonnei in the tissues of a guinea pig eye (Serény's model) has been studied. Bacteriological and histological investigations made with the use of fluorescent antibody techniques have revealed that S. sonnei can be preserved in macrophages (histiocytes) and intercellular space of the eye epithelium for a long time (up to 277 days). The injection of ACTH or Shigella vaccine into the animals may provoke the exacerbation of the infectious process and the release of the infective agent.

Brucellosis is a worldwide zoonotic disease transmitted to humans by consumption of contaminated milk and milk products. Brucellosis is endemic in Turkey, and Edirne has a high Brucella prevalence. Brucellosis is prevented by live-attenuated vaccines for animals and the vaccination program has been in place since 1984 in Turkey. Thrace is the pilot region for this vaccination program. The gold standard diagnostic technique for brucellosis is still the isolation of suspicious bacterial colonies followed by bacteriological identification, but it is very time consuming and laborious. Although long-term in vitro cultivation of the syphilis agent, Treponema pallidum subspecies pallidum, was introduced less than a decade ago, it has revolutionized and accelerated research with this difficult pathogen. Moreover, it is allowing for the exploration of topics that could not easily be pursued when propagation in the rabbit model was the only possible method for the expansion of treponemes. This report, based on talks presented at the Shanghai International Syphilis Symposium 2025, provides an overview of how in vitro cultivation is currently being applied to syphilis research.

Here, we describe the formulation and application of Hornsby-Alt-Nally (HAN) growth medium for the isolation and propagation of Leptospira species and serovars from host tissues. Cultures can be isolated directly from host samples by incubation at 29°C or 37°C using HAN liquid, semi-solid, or solid agar media.

Leptospires require specific conditions and nutrition to survive and thrive, essential for its ability to multiply or adapt to in vitro culture. This constraint on laboratory cultures limits the applicability of in vitro studies to the understanding of even simple pathogenic processes and diagnostics. Culture of leptospires in a laboratory setting using DMEM and EMEM media, common for culture of mammalian cells, is an alternative that mimics host environmental conditions, less complex, with high performance, and lower cost, which are imperative characteristics to improve routine laboratory act

Between September 2001 and March 2013, 62 bacterial cultures (37 aerobic and 25 anaerobic) were performed on 37 blood samples from 23 Antillean manatees ( Trichechus manatus manatus) that were kept in captivity at the Brazilian National Center for Research and Conservation of Aquatic Mammals (CMA) in Pernambuco (CMA-PE) and Alagoas (CMA-AL), Brazil. All of the animals sampled exhibited clinical signs at the time of sampling including abscesses (n = 8), debilitation and anorexia (n = 22), and profound lethargy-moribundity (n = 7). The 4 animals with profound lethargy-moribundity died shortly af

Leptospirosis is an emerging zoonosis with a worldwide distribution but is more commonly found in impoverished populations in developing countries and tropical regions with frequent flooding. The rapid detection of leptospirosis is a critical step to effectively manage the disease and to control outbreaks in both human and animal populations. Therefore, there is a need for accurate and rapid diagnostic tests and appropriate surveillance and alert systems to identify outbreaks. This review describes current in-house methods and commercialized tests for the rapid diagnosis of acute leptospirosis

Interferon-release assays (IGRAs) represent advances in tuberculosis immunology and evolutionary biology. IGRAs were designed to replace tuberculin skin test (TST) for the diagnosis of latent tuberculosis infection because of their logistical advantages and enhanced specificity over TST. Although IGRAs and TST have been useful in epidemiologic studies, they lack the sensitivity and reproducibility normally expected from diagnostic tests in clinical practice. In this review, we present an overview of the current recommendations and knowledge in the field and discuss practical approaches in area

For the diagnosis of urinary tract infection (UTI), besides the quantification of bacteria in the urine, cellular elements contained in the urine, the collection method used and the clinical syndrome should also be considered. Therefore, the microbiological diagnosis of UTI should be performed by an experienced person who takes into account the diversity of situations that may influence the result of each of the cultures. The processing of urine samples depends on the number of samples received daily. In laboratories with a high number, it is impossible to culture each of them, so negative uri

Frequent "contaminants" detected during mycobacterial culture of decontaminated samples are bacteria of the order Actinomycetales. These are usually bacteria classified as the family Corynebacterineae, genera Corynebacterium, Dietzia, Gordonia, Nocardia, Rhodococcus and Tsukamurella. These bacteria frequently colonize the airways and, under certain circumstances, they may cause life-threatening diseases. In severely immunocompromised patients, they regularly cause life-threatening infections with bacteria of the genus Nocardia. These filamentous bacteria, developing aerial mycelium in the cult

Extended-Spectrum beta-Lactamase (ESBL)-producers in the family Enterobacteriaceae are recognized worldwide as nosocomial pathogens, however it is difficult to screen them in the routine laboratory processing. ChromID ESBL agar newly developed for screening ESBL-producing Enterobacteriaceae was released in Japan in April, 2007. We evaluated the clinical assessment of ChromID ESBL agar in routine microbiology laboratory. The 47 strains investigated were clinical isolates belonging to the family Enterobacteriaceae with the MICs of cefpodoxime greater than 2 mug/ml. The 27 ESBL-producers examined

AIMS: A rapid procedure was developed to screen for bacteria that are able to grow on polycyclic aromatic hydrocarbons (PAHs).
METHODS AND RESULTS: A drop of ethyl ether-dissolved PAH is spread on a sterilized cellulose acetate/nitrate filter lying on the top of a mineral salts agarose plate. After the evaporation of ethyl ether, a serially diluted sample is spread over the filter and incubated. Subsequently, the PAH degrading bacteria can be counted and isolated.
CONCLUSIONS: This procedure is a simple method for screening bacterial isolates for the ability to grow with PAHs.

We compared blood culture and in situ hybridization method (Hybrisep) to detect bacteria in blood samples. One thousand and two hundred and sixty nine blood culture samples were tested in 2003 in our hospital. One hundred and sixty seven samples (13.1%) were positive for bacteria. Total number of detected bacteria was 178. Of 178 bacteria, 49.4% was gram positive, 32.6% was gram negative, 13.5% was Candida spp., and 4.5% was anaerobic. Twenty five samples were tested with both blood culture and Hybrisep. Three samples were positive for both methods, and 13 samples were negative for both method

The diagnosis of leptospirosis is often made using the microscopic agglutination test (MAT), in which live antigens representing >20 serogroups undergo reaction with patient serum samples to detect agglutinating antibodies. Data derived from this assay are often used to infer the identity of the infecting leptospiral serovar or serogroup; however, paradoxical reactions and cross-reactions between serogroups are common. To evaluate the usefulness of this approach, data on culture-proven cases of leptospirosis that occurred in Barbados from January 1980 through December 1998 were reviewed. A tot

The identification of Staphylococcus aureus directly from blood cultures is clinically relevant, but it requires a test that is both rapid and reliable. Previously, biochemical, immunological, tube coagulase, and thermostable-endonuclease methods have shown variable sensitivity and specificity. Testing directly from blood culture broth has not been described for the latex kit Staphaurex Plus (Murex Diagnostics Ltd.), and the modified conventional tests have not been used with the newer, continuously monitored blood culture systems. In addition, the commercial RAPIDEC staph kit (bioMerieux Vite

The recognition of unusual, but important, pathogens such as Burkholderia pseudomallei is essential for the rapid implementation of appropriate antimicrobial therapy--delays can be fatal. Melioidosis should be considered as a potential diagnosis for any patient with exposure to areas of endemicity, and thus laboratories should be aware of the differential features of the disease and the causative organism. Isolation of B. pseudomallei is readily achieved using standard culture media such as blood, MacConkey or cystine-lactose-electrolyte-deficient (CLED) agars, and routine blood culture broths

During the period of August 16, 1981, through December 16, 1982, 15-mL blood culture specimens collected at the Seattle Veterans Administration Medical Center (SVAMC) were divided into two aliquots. The first 10-mL aliquot was inoculated directly into aerobic and anaerobic BACTEC (Johnston Laboratories, Towson, MD) vials (DIR); the remaining 5 mL was processed in an resin-containing Antimicrobial Removal Device (Marion Scientific, Kansas City, MO) before transfer to a second, identical set of aerobic and anaerobic vials (ARD). The final volume of inoculated blood from the ARD specimen was half

Since the summer of 1982, we have cultured patient specimens for Lyme disease spirochetes. Of 118 patients cultured, four specimens yielded spirochetes: two from blood, one from a skin biopsy specimen of erythema chronicum migrans (ECM), and one from cerebrospinal fluid. All four isolates appeared identical when examined with a monoclonal antibody. However, attempts to recover the spirochete from synovium or synovial fluid were unsuccessful. In addition, the organism could not be visualized in skin or synovial biopsy specimens using the avidin-biotin peroxidase complex detection system. Thus,
