Phages for biocontrol in foods: What opportunities for Salmonella sp. control along the dairy food chain?

Controlling the presence of pathogenic bacteria, such as Salmonella sp., in dairy products production is a burning issue since contamination with Salmonella can occur at any stage of the production chain. The use of Salmonella-phages applied as control agents has gained considerable interest. Nonetheless, Salmonella-phage applications specifically intended for ensuring the safety of dairy products are scarce. This review identifies recent advances in the use of Salmonella-phages that are or could be applied along the dairy food chain, in a farm-to-fork approach. Salmonella-phages can be promis

Variations in expression and release of botulinum neurotoxin in Clostridium botulinum type A strains.

In this study, we characterized the production and release of botulinum neurotoxin during growth of two Clostridium botulinum strains, Hall A and NCTC2916, of distinct gene organizations. The intra- and extracellular fractions of the bacterial cells harvested at various stages of growth were analyzed for the neurotoxin. Both strains exhibited a temporal neurotoxin gene expression; however, these two strains differ in their patterns of growth, toxin production, toxin release, and post-translational nicking. Comparing to the NCTC2916 strain, the Hall A strain showed an extended stationary phase,

Function of neisserial outer membrane phospholipase a in autolysis and assessment of its vaccine potential.

Outer membrane phospholipase A (OMPLA) is an outer membrane-localized enzyme, present in many gram-negative bacterial species. It is implicated in the virulence of several pathogens. Here, we investigated the presence, function, and vaccine potential of OMPLA in the human pathogen Neisseria meningitidis. Immunoblot analysis showed the presence of OMPLA in 28 out of 33 meningococcal strains investigated. The OMPLA-negative strains all contained a pldA gene, but these alleles contained premature stop codons. All six Neisseria gonorrhoeae strains tested, but only two out of seven commensal neisse

Studies of the relationship between the production of bacteriocines by group A streptococci and acute glomerulonephritis.

The previously reported relationship between nephritogenicity and bacteriocine production among Group A streptococci has been examined. Using techniques comparable to the earlier study, the present study did not reveal any such relationship. Among 73 strains tested, 53% produced bacteriocines; no significant difference was noted between bacteriocines from strains isolated from cases of nephritis and those from a group of strains recovered from patients with other conditions. In addition, no correlation between type and bacteriocine production was observed.

Characterization of Five Lytic Bacteriophages as New Members of the Genus Mosigvirus, Infecting Multidrug-Resistant Shiga Toxin-Producing Escherichia coli and Their Antibiofilm Activity.

The emergence of multidrug-resistant Shiga toxin-producing Escherichia coli (STEC) poses a major challenge to public health and necessitates the development of alternative antimicrobial strategies. This study aimed to isolate and characterize five lytic bacteriophages belonging to the genus Mosigvirus and evaluate their potential as biocontrol against MDR STEC strains and their biofilms. The five bacteriophages, designated vB_EcoM-pJBB (ΦB), vB_EcoM-pJBC (ΦC), vB_EcoM-pJBJ (ΦJ), vB_EcoM-pJBK (ΦK), and vB_EcoM-pJBL (ΦL), were isolated from sewage treatment plant samples using STEC ATCC 43895 as

Genetic characterization of ØVC8 lytic phage for Vibrio cholerae O1.

BACKGROUND: Epidemics and pandemics of cholera, a diarrheal disease, are attributed to Vibrio cholera serogroups O1 and O139. In recent years, specific lytic phages of V. cholera have been proposed to be important factors in the cyclic occurrence of cholera in endemic areas. However, the role and potential participation of lytic phages during long interepidemic periods of cholera in non-endemic regions have not yet been described. The purpose of this study was to isolate and characterize specific lytic phages of V. cholera O1 strains. METHODS: Sixteen phages were isolated from wastewater samp

Activated human monocytes inhibit the intracellular multiplication of Legionnaires' disease bacteria.

We have examined the interaction between virulent egg yolk-grown L. pneumophila, Philadelphia 1 strain, and in vitro-activated human monocytes, under antibiotic-free conditions. Freshly explanted human monocytes activated by incubation with concanavalin A (Con A) and human lymphocytes inhibited the intracellular multiplication of L. pneumophila. Both Con A and lymphocytes were required for activation. Con A was consistently maximally effective at greater than or equal to 4 mug/ml. Monocytes activated by incubation with cell-free filtered supernatant from Con A-sensitized mononuclear cell cultu

Evaluating Proteus mirabilis phage vB_PmiA_PM1 efficacy against catheter-associated urinary tract infections in artificial urine and laboratory media.

Proteus mirabilis is a leading cause of catheter-associated urinary tract infections (CAUTI) because of its capability to develop extensive crystalline biofilms that obstruct catheters. Phages can effectively eradicate P. mirabilis biofilms in vitro; however, the clinical trial outcomes have been controversial. The current study aimed to investigate phage efficacy against P. mirabilis CAUTI in artificial urine medium (AUM) and laboratory media. A novel lytic P. mirabilis phage, vB_PmiA_PM1, was isolated and characterized. The phage had a narrow host range, capable of infecting only its main ho

Genomic characterization of novel lytic phage vB_Sal_S6 with putative host FhuA interaction and its application for Salmonella biocontrol in milk.

Global foodborne Salmonella outbreaks pose a serious concern for food safety and public health underscoring the need for isolation of lytic phages with potential as biocontrol agents. A lytic bacteriophage vB_Sal_S6 was isolated which targets Salmonella serovars - Typhimurium, Bovismorbificans, Infantis, Typhi, Paratyphi, Weltevreden, Poona and as well as Escherichia coli. It exhibited strong bacteriolytic activity across a range of MOIs (0.01-10), with stability across pH 4-10 and temperature conditions (25-55 °C). In artificially contaminated milk, vB_Sal_S6 reduced Salmonella counts by 3.9l

Expression of heterologous O-antigen in Yersinia pestis KIM does not affect virulence by the intravenous route.

All strains of Yersinia pestis examined have been found to lack an O-antigen. In other members of the Enterobacteriaceae, the rough phenotype often results in attenuation. However, Y. pestis is the aetiological agent of bubonic plague. In evolving from the ancestral enteropathogenic Yersinia pseudotuberculosis, and with the development of an arthropod-vectored systemic pathogenesis, smooth LPS production is not necessary for Y. pestis virulence and the metabolic burden has been alleviated by inactivation of the O-antigen biosynthetic operon. To investigate this, Y. pestis strain KIM D27 was tr

Synergistic antibacterial activity between L-norvalyl-L-1-aminoethylphosphonic acid and nocardicin A.

The phosphonopeptide L- norvalyl -L-1- aminoethylphosphonic acid [ Nva -Ala(P)] has been studied in combination with 12 beta-lactam antibiotics for activity against Pseudomonas aeruginosa. Nocardicin A was found to give the most potent synergistic combination with Nva -Ala(P). This interaction was widely observed in clinical isolates of P. aeruginosa in vitro and in a mouse septicemia model. Synergy was also observed in vitro and in vivo in several other species, including Proteus mirabilis, indole-positive Proteus spp., and Serratia marcescens. The interaction between Nva -Ala(P) and nocardic

[Antibiotic sensitivity of the Salmonella isolated from humans ill with salmonellosis in 1967-1974].

Sensitivity to levomycetin, streptomycin, chlortetracycline, oxtetracycline, tetracycline, neomycin, monomycin and erythromycin of 548 fresh isolates of Salmonella of 32 serotypes of the main groups A, B, C, D and E was studied. The cultures were isolated from salmonellosis patients within 1967-1974. Sensitivity to neomycin, monomycin, levomycetin and streptomycin was found respectively in 96.9, 91.1, 76.5 and 74.8 per cent of the cultures. High sensitivity of the cultures was observed only to levomycetin (15.1 per cent). With respect to tetracyclines the isolates were mainly resistant (24.2-4

Comparative effects of anesthetics on the viability and integrity of Escherichia coli ML30.

Cells of Escherichia coli ML30 in a mineral salts medium were exposed to dichlorodifluoromethane (f-12), cyclopropane, halothane, or Ethrane at concentrations of 1.25, 0.2, 0.04, and 0.008 X saturation for times up to 1,200 min, and at temperatures in the range of 2 to 37 C. When any of these anesthetics were applied for 300 min at 1.25 X saturation, a substantial decrease in number of survivors occurred. Halothane was most bactericidal, cyclopropane and Ethrane were moderately bactericidal, and t-12 was least bactericidal. At saturation values of less than 1.0, none of the four anesthetics ha

Phenotypic and Genomic Comparison of Klebsiella pneumoniae Lytic Phages: vB_KpnM-VAC66 and vB_KpnM-VAC13.

Klebsiella pneumoniae is a human pathogen that worsens the prognosis of many immunocompromised patients. Here, we annotated and compared the genomes of two lytic phages that infect clinical strains of K. pneumoniae (vB_KpnM-VAC13 and vB_KpnM-VAC66) and phenotypically characterized vB_KpnM-VAC66 (time of adsorption of 12 min, burst size of 31.49 ± 0.61 PFU/infected cell, and a host range of 20.8% of the tested strains). Transmission electronic microscopy showed that vB_KpnM-VAC66 belongs to the Myoviridae family. The genomic analysis of the phage vB_KpnM-VAC66 revealed that its genome encoded 2
