Berberine, a traditional Chinese medicine, was found to exhibit anticoccidial activity. However, its mechanism is unclear. Trace metals such as copper and zinc are extremely low (less than 0.01% of the total weight of the body) but play a vital role in organisms. In the present study, we investigated the effect of berberine on copper and zinc levels in chickens infected with Eimeria tenella. Firstly, our data confirmed that infected chickens with E. tenella exhibited classic impairment on the 8th day of post infection, such as weight loss and increased feed conversion. Further study showed tha

Riemerella anatipestifer, an important infectious bacterium affecting the duck industry, has 5-75% mortality, depending on strain virulence. We previously demonstrated that proinflammatory cytokines are involved in inflammation during, and regulating susceptibility to, R. anatipestifer infection. We investigated the effects of the anti-inflammatory compound berberine in duck splenic lymphocytes stimulated with killed R. anatipestifer, and in R. anatipestifer-infected ducks. IL-17A, IL-17F, and IL-1β transcripts were downregulated, and IFN-γ and IL-10 transcripts enhanced, in berberine-treated

BACKGROUND: In the central nervous system regions of the sporadic and familial FTLD and ALS patients, TDP-43 has been identified as the major component of UBIs inclusions which is abnormally hyperphosphorylated, ubiquitinated, and cleaved into C-terminal fragments to form detergent-insoluble aggregates. So far, the effective drugs for FTLD and ALS neurodegenerative diseases are yet to be developed. Autophagy has been demonstrated as the major metabolism route of the pathological TDP-43 inclusions, hence activation of autophagy is a potential therapeutic strategy for TDP-43 pathogenesis in FTLD

Our previous study demonstrated that a combination of alternative medicine berberine and conventional 5-aminosalicylic acid (5-ASA) showed promise to be a novel therapeutic strategy for ulcerative colitis (UC). This present study aims to sketch the pre-clinical toxicity profile of this combination (1:10 dose ratio) on mice. In acute toxicity test, the determined median lethal dose (LD50) was 278.7 mg/kg berberine plus 2787 mg/kg 5-ASA. The results from subacute toxicity test demonstrated that no toxic signs of clinical symptoms, no significant changes in hematological or biochemical parameters

Chikungunya virus (CHIKV) is an arthritogenic arbovirus of the Alphavirus genus, which has infected millions of people after its re-emergence in the last decade. In this study, a BHK cell line containing a stable CHIKV replicon with a luciferase reporter was used in a high-throughput platform to screen approximately 3000 compounds. Following initial validation, 25 compounds were chosen as primary hits for secondary validation with wild type and reporter CHIKV infection, which identified three promising compounds. Abamectin (EC50 = 1.5 μM) and ivermectin (EC50 = 0.6 μM) are fermentation product

INTRODUCTION: alkaloids are a group of naturally occurring chemical compounds that contain mostly basic nitrogen atoms. In particular berberine is a quaternary ammonium salt from the protoberberine group of isoquinoline alkaloids with anti-lipogenic and hypoglycemic effects. On the other hand, mate plant has reported to have high antioxidant potential, including an inhibitory ability against plasma and LDL oxidation.
AREAS COVERED: Most of the studies reported in literature about alkaloids are pre-clinical trials, and we have already discussed them in a previous review we conducted. This time

OBJECTIVE: Previous studies have indicated that berberine is an effective insulin sensitizer with comparable activity to metformin (Diabetes 2006, 55, 2256). Reduced insulin sensitivity is reportedly a factor adversely affecting the outcome of IVF in patients with polycystic ovary syndrome (PCOS) (Human Reproduction 2006, 21, 1416). Our objective was to evaluate the clinical, metabolic and endocrine effects of berberine vs metformin in PCOS women scheduled for IVF treatment and to explore the potential benefits to the IVF process.
DESIGN: We performed a prospective study in 150 infertile wome

Berberine, an isoquinoline alkaloid component of Coptidis Rhizoma (goldenthread) extract, has been reported to have therapeutic potential for central nervous system disorders such as Alzheimer's disease, cerebral ischemia, and schizophrenia. We have previously shown that berberine promotes the survival and differentiation of hippocampal precursor cells. In a memory-impaired rat model induced by ibotenic acid injection, the survival of pyramidal and granular cells was greatly increased in the hippocampus by berberine administration. In the present study, we investigated the effects of berberine

In bacteria, multidrug-resistance pumps (MDRs) confer resistance to chemically unrelated amphipathic toxins. A major challenge in developing efficacious antibiotics is identifying antimicrobial compounds that are not rapidly pumped out of bacterial cells. The plant antimicrobial berberine, the active component of the medicinal plants echinacea and golden seal, is a cation that is readily extruded by bacterial MDRs, thereby rendering it relatively ineffective as a therapeutic agent. However, inhibition of MDR efflux causes a substantial increase in berberine antimicrobial activity, suggesting t

Prominent features of heliotrin-related chronic toxical disorders of the liver developing in hepatic cirrohosis include hyperfermentemia, hypoproteinemia, intensification of lipid peroxidation (LPO), as evidenced by decrease in the activity of the antioxidant defence. In chronic active liver disorders, hyperfermentemia and hyperlipoperoxidation are common, but it is later in the course of the experiment that hypoproteinemia and hypoalbuminemia and manifest depression of the antioxidant defence that come to be developing. Berberin experimental pharmacotherapy has been shown to attenuate the deg

Berberis aristata is an edible plant employed in the South Asian Traditional Medicine, particularly its fruits being used as a tonic remedy for liver and heart. In this investigation, berberine, a known compound from this plant, was studied for its possible antihepatotoxic action in rats. Pretreatment of animals with berberine (4 mg/kg; orally twice daily for 2 days) prevented the acetaminophen- or CCl4-induced rise in serum levels of alkaline phosphatase (ALP) and aminotransaminases (AST and ALT), suggestive of hepatoprotection. Post-treatment with three successive oral doses of berberine (4

A high-performance liquid chromatographic method for the determination of berberine in plasma, urine and bile samples is described. Plasma samples were pretreated by protein precipitation with acetonitrile and urine and bile samples were pretreated by organic solvent extraction using 5% 2-propanol in methylene chloride. Berberine was determined in all samples using an octyl reversed-phase column with a mobile phase of 60-63% acetonitrile in 0.1% phosphoric acid (pH 6.0) and with UV detection at 267 nm. The detection limits for berberine in plasma, urine and bile were 18.1 ng/ml, 2.3 ng/ml and

The effects of berberine on the contractions of the longitudinal muscle of the guinea-pig isolated ileum were investigated. Lower concentrations of berberine (less than or equal to 5 x 10(-5) M) induced a parallel rightward shift of the dose-response curve of acetylcholine, suggesting that berberine is antagonizing the actions of acetylcholine at the receptors competitively. At higher concentration, berberine (1 x 10(-4) M) facilitated a rightward shift of the dose-response curve of acetylcholine with a reduction of maximum response, indicating that the interactions of the two agents changed f

Chemotherapy-induced nephrotoxicity remains a major dose-limiting complication in cancer treatment. Accumulating evidence indicates that renal injury triggered by agents such as cisplatin, doxorubicin, methotrexate, and cyclophosphamide is primarily mediated by interconnected mechanisms including oxidative stress, mitochondrial dysfunction, inflammatory signaling, and apoptosis. Despite advances in supportive care, effective nephroprotective strategies remain limited. This review provides a comprehensive mechanistic synthesis of current preclinical evidence regarding the nephroprotective poten

Berberine, a natural isoquinoline alkaloid derived fromCoptidis rhizoma has long been used in traditional medicine and has recently attracted attention for its potential role in cancer therapy. Emerging evidence suggests that berberine can inhibit proprotein convertase subtilisin/kexin type 9 (PCSK9), a key regulator of lipid metabolism that has also been implicated in tumor biology. While the role of PCSK9 in cardiovascular diseases is well established, its contribution to cancer progression and therapeutic response remains incompletely understood. Here, we investigated whether berberine-medi

Candida albicans is ubiquitous on human mucosal surfaces and easy to cause infectious diseases. The widespread use of antifungal drugs has led to the increasing drug resistance of C. albicans, posing a significant challenge to clinical treatment. Berberine hydrochloride (BBH), a quaternary ammonium alkaloid isolated from Coptidis Rhizoma, exhibits broad-spectrum antifungal activity. This study explored the antifungal effect and mechanism of BBH combined with fluconazole (FLC) against drug-resistant C. albicans. Results showed that BBH could enhance the antifungal effect of FLC against the drug
