A Modular Biosensor Design for Quantitative Measurement of Free Nedd8.

The objective of our study was to develop a genetically encoded biosensor for quantification of Nedd8, a post-translational modifier that regulates cellular signals through conjugation to other proteins. Perturbations in the balance of free (i.e., unconjugated) and conjugated Nedd8 caused by defects in Nedd8 enzymes or cellular stress are implicated in various diseases. Despite the biological and biomedical importance of Nedd8 dynamics, no method exists for direct quantification of free Nedd8, hindering the study of Nedd8 and activities of its associated enzymes. Genetically encoded biosensors

Biosensors for the detection of celiac disease.

Celiac disease (CeD) is an autoimmune disorder triggered by sensitivity to gluten, a protein complex found in wheat, barley, and rye. Gliadins, a component of gluten, are proteins that trigger an immune response in individuals with CeD, primarily affecting the small intestine's inner lining. Despite a 1-1.5% prevalence, only 24% of cases are diagnosed due to non-specific symptoms. Screening is advised for high-risk groups, including first-degree relatives and type 1 diabetes patients. The accurate diagnosis of this condition and the assessment of the patient's response to the current treatment

Highly sensitive surface acoustic wave biosensor for the detection of Hg2+ based on the thymine-Hg2+-thymine structure.

The detection of mercury ions (Hg2+) with an ultralow concentration is of great significance for the treatment of heavy metal pollution in industrial waste water and the monitoring of drinking water quality. In this study, a simple, sensitive, and portable surface acoustic wave (SAW) biosensor for the determination of Hg2+ concentration was developed. First, a DNA fragment with a specific binding capacity for Hg2+ was chemically adsorbed on to the sensitive region through forming Au-S bonds with the SAW biosensor. The DNA probe could then form a thymine-Hg2+-thymine (T-Hg2+-T) complex with str

Sensing Techniques for Organochlorides through Intermolecular Interaction with Bicyclic Amidines.

Toxic organochloride molecules are widely used in industry for various purposes. With their high volatility, the direct detection of organochlorides in environmental samples is challenging. Here, a new organochloride detection mechanism using 1,5-diazabicyclo[4.3.0]non-5-ene (DBN) is introduced to simplify a sensing method with higher detection sensitivity. Three types of organochloride compounds-trichloroethylene (TCE), dichloromethane (DCM), and dichlorodiphenyltrichloroethane (DDT)-were targeted to understand DCM conjugation chemistry by using nuclear magnetic resonance (NMR) and liquid chr

An "on-off-super on" photoelectrochemical sensor based on quenching by Cu-induced surface exciton trapping and signal amplification of copper sulfide/porous carbon nitride heterojunction.

In this work, we report an "on-off-super on" photoelectrochemical sensor for probing hydrogen sulfide due to its toxicity in water environment by using porous carbon nitride as photoelectric transducers. Synthesized by an alkaline-assisted hydrothermal method, the porous carbon nitride photoanode exhibited a remarkable photocurrent on the initial "on" state. Cu2+ immobilized on the surfaces of porous carbon nitride could significantly decrease the charge transfer efficiency and quench the photoelectrochemical signal in the "off" state. In addition, the introduction of S2- ions could eliminate

Fabrication and Characterization of Simple Structure Fluidic-Based Memristor for Immunosensing of NS1 Protein Application.

Non-structural protein 1 (NS1 protein) is becoming a commonplace biomarker for the diagnostic of early detection of dengue. In this study, we sought to use a label-free approach of detecting NS1 protein by harnessing fluidic-based memristor sensor. The sensor was fabricated using sol-gel spin coating technique, by which TiO2 thin film is coated on the surface of Indium tin oxide (ITO) and a glass substrate. The sensor was then functionalized with glycidoxypropyl-trimethoxysilane (GPTS), acting as antibody for NS1. The addition of the target NS1 formed an antibody-antigen complex which altered

Sensitive Detection of Nucleic Acids Using Subzyme Feedback Cascades.

The development of Subzymes demonstrates how the catalytic activity of DNAzymes can be controlled for detecting nucleic acids; however, Subzymes alone lack the sensitivity required to detect low target concentrations. To improve sensitivity, we developed a feedback system using a pair of cross-catalytic Subzymes. These were individually tethered to microparticles (MP) and separated by a porous membrane rendering them unable to interact. In the presence of a target, active PlexZymes® cleave a first Subzyme, which separates a first DNAzyme from its MP, allowing the DNAzyme to migrate through the

Recent trends and perspectives in enzyme based biosensor development for the screening of triglycerides: a comprehensive review.

Clinical manifestations of the elevated plasma triacylglycerol (TG) include a greater prevalence of atherosclerotic heart disease, acute pancreatitis, diabetes mellitus, hypertension, and ischemic vascular disease. Hence, these significant health troubles have attracted scientific attention for the precise detection of TG in biological samples. Numerous techniques have been employed to quantify TG over many decades, but biosensors hold the leading position owing to their superior traits such as highly specific recognition for target molecules, accuracy, minituarization, small sample requiremen

Development of a luminescence-based biosensor for detection of methylmercury.

A recombinant whole-cell bacterial sensor for highly selective and sensitive detection of the bioavailable methylmercury in the environment was constructed. The biosensor carries luciferase gene, luxAB, from Vibrio harveyi as a reporter under the control of the mercury inducible regulatory part of mer-operon from Pseudomonas K-62 plasmid pMR26. In addition, a merB gene encoding organomercurial lyase which cleaves the C-Hg bond of methylmercury to give Hg(2+ )was coexpressed in the sensor. The resulting bacterial sensor responded specifically to methylmercury, and the lowest detectable concentr

Subnanomolar cyanide detection at polyphenol oxidase/clay biosensors.

A novel, inexpensive, and simple amperometric biosensor based on immobilization of polyphenol oxidase (PPO) into Zn-Al layered double hydroxides, also called anionic clays, is applied for determination of cyanide. The detection of cyanide was performed via its inhibiting action on the PPO electrode. Measurement was carried out with 3,4-dihydroxyphenylacetic acid as enzyme substrate, the enzymatically generated quinoid products being electroreduced at -0.2 V. An extremely sensitive detection limit (0.1 nM) was obtained for cyanide. Enzyme immobilization into an anionic exchanger clay seems to c

Liquid Crystals for Chemical and Biological Sensing: Emerging Transduction Strategies beyond Polarized Optical Microscopy.

Liquid crystal (LC) sensors exploit the ability of these anisotropic soft materials to amplify molecular-scale interfacial events into macroscopic optical responses. Although classical LC sensing platforms have demonstrated high sensitivity toward chemical and biological analytes, their practical implementation remains limited by the predominance of polarized-light optical microscopy (POM), which is often qualitative, instrument-intensive, and difficult to standardize. This Spotlight examines emerging transduction strategies that move LC-based sensing beyond conventional birefringent texture a

Integration of photonic crystals with terahertz for next-generation analytical imaging and biosensing: A review.

Photonic crystals (PCs), which are periodic dielectric structures that control light propagation, represent a transformative approach for overcoming long-standing challenges in terahertz (THz) technology. Although the THz spectrum (0.1-10 THz) offers unique advantages for spectroscopy, imaging, and biosensing through non-ionizing radiation and molecular sensitivity, its practical implementation has been hindered by difficulties in THz generation and manipulation. Recent advances in PC-based THz waveguides, filters, and sensors have demonstrated how tailored photonic bandgaps can dramatically e

Nanozyme-based biosensing strategies for aflatoxin detection: Recent advances and perspectives.

Aflatoxins (AFs) are among the most toxic mycotoxins, with a profound impact on food safety and health. Consequently, trace-level detection is required, as exemplified by 2 μg/kg for aflatoxin B1 (AFB1) in cereals under European Union guidelines. Although traditional chromatography and immunoassay-based detection methods are reliable, they are often time-consuming and require specialized expertise. Nanozyme-based biosensors have attracted increasing attention because nanozymes provide stable, engineerable, enzyme-mimicking catalytic amplification. In this article, we introduce major AFs, diffe

Single Microbead Floating/Sinking-Enabled Digital Biosensing.

Digital bioassays have emerged as cutting-edge techniques for highly sensitive biomarker analysis. However, the requirement of sophisticated operation and equipment, fully sealed microchambers, and well-trained professionals confines its application to central laboratories. Herein, we present dMSink, a new conceptual mix-and-read digital biosensing platform based on the single microbead (MB) floating/sinking mechanism that is compatible in general biolabs and even in-field testing. Specifically, MBs will float on the surface of a liquid with a finely tuned critical density to ensure that even

Ultra-Broadband Metasurface Absorber Enabled by a Central-Bar-Coupled Split-Disk Dimer.

A hybrid metasurface absorber (MSA) based on a central-bar-coupled split-disk dimer is proposed and numerically investigated for high-resolution refractive-index sensing in the near-infrared spectral region. The metasurface consists of silicon nitride dielectric resonators integrated with a gold plasmonic layer, enabling strong electromagnetic confinement, enhanced light-matter interaction, and ultra-narrow resonant features within the 1000-1400 nm wavelength range. The optimized structure supports multiple resonant modes under both x- and y-polarized excitation, producing sharp reflection dip

Convenient and sensitive detection of viable Escherichia coli employing a sequential reaction between antibody-enzyme complexes.

We developed a rapid, convenient, and viable cell-selective sensing system for Escherichia coli (E. coli) based on a sequential enzymatic reaction on an intact cell surface. In food safety and infectious disease diagnostics, it is crucial to selectively detect viable cells that represent actual infection risk. However, conventional methods such as culture-based method and polymerase chain reaction are time-consuming and require specialized equipment, limiting their applicability for on-site testing. The proposed system is based on a cooperative reaction of antibody-enzyme complexes, in which c

A Label-Free Cell-Based Biosensor Method for Ethanol Quantification Using Temperature-Induced Spontaneous Cell Detachment.

Rapid, low-cost ethanol quantification is vital for beverage quality control, biofuel production, and pharmaceutical applications, yet current approaches are costly, reagent- or label-dependent, or rely on spectroscopy with substantial sample preparation. We introduce a purely cell-based, label-free biosensor that exploits temperature-gradient-induced spontaneous detachment of Saccharomyces cerevisiae from a chip surface. The readout is the detachment half-time, td50, derived from time-resolved changes in interfacial thermal resistance, Rth, at the solid-liquid interface. Cells were pre-expose
