Immunomodulatory and Anti-IBDV Activities of the Polysaccharide AEX from Coccomyxa gloeobotrydiformis.

A number of polysaccharides have been reported to show immunomodulatory and antiviral activities against various animal viruses. AEX is a polysaccharide extracted from the green algae, Coccomyxa gloeobotrydiformis. The aim of this study was to examine the function of AEX in regulating the immune response in chickens and its capacity to inhibit the infectious bursal disease virus (IBDV), to gain an understanding of its immunomodulatory and antiviral ability. Here, preliminary immunological tests in vitro showed that the polysaccharide AEX can activate the chicken peripheral blood molecular cell

In silico analysis of VP2 protein from infectious bursal disease virus isolate UPM1432/2019: structural dynamics and epitope prediction.

Infectious Bursal Disease Virus (IBDV) remains a significant threat to global poultry health due to its immunosuppressive effects and high mutation rate, which challenge existing vaccine strategies. This study presents a comprehensive computational analysis of the VP2 protein from Malaysian IBDV isolate UPM1432/2019 to elucidate its structural stability and antigenic potential. Phylogenetic analysis revealed evolutionary divergence among VP2 sequences, while physicochemical profiling demonstrated the protein's high thermostability (aliphatic index: 98.77) and cellular stability (instability in

Chitosan-coated PLGA nanoparticles as a delivery system for infectious bursal disease viral antigen in chickens.

In the present study, antigen-chitosan-PLGA nanoparticles were explored as the delivery system for inactivated whole infectious bursal disease virus (IBDV) antigens. The immunized birds showed the peak antibody level (ELISA titre 4095.65 ± 55.74) at the 3rd week post-immunization, which was significantly higher than that of the commercial inactivated vaccine (titre 2257 ± 30) (P < 0.0001). The virus-loaded chitosan-coated PLGA nanoparticles induced good cell-mediated immunity. The immunized birds showed better resistance against the challenge infection with a very virulent IBDV. Only one bird

The mutated very virulent IBDV (mvvIBDV) suppresses immune responses to avian influenza and newcastle disease immunization.

Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is one of the most devastating immunosuppressive diseases in poultry, posing persistent threats to global poutry farming. Over the past three decades, very virulent IBDV (vvIBDV) with high mortality has been an important dominant epidemic strain. To make matters worse, since 2023, outbreaks of atypical IBD attributable to a mutated vvIBDV (mvvIBDV) has been reported across multiple provinces in China. However, the harm of mvvIBDV has not yet been systematically recognized. In this study, an mvvIBDV strain of gen

Pair-epitopes vaccination: enabling offspring vaccination in the presence of maternal antibodies.

Maternally derived antibodies (MDAs) are critical for offspring protection during the first days of life. However, due to their short half-life (3-5 days), MDA levels decline rapidly and by 8-15 days post-hatch drop to below protective levels. In addition, MDAs against a specific pathogen often impede the efficient protective immune response to that pathogen by the new-born following vaccination. The combination of these two phenomena generates a gap in protection in the period between loss of MDA protection and development of vaccine-induced protective antibodies. Herein, a concept is present

Role of naturally occurring genome segment reassortment in the pathogenicity of IBDV field isolates in Three-Yellow chickens.

Reassortment among genome segments of infectious bursal disease virus (IBDV) field isolates was reported frequently worldwide, however the pathogenicity of the reassortant field IBDV is poorly understood. In this paper, a pathogenicity study on four representative IBDV field strains isolated from Southern China between 2005 and 2011 was conducted. Twenty-eight-day-old Three-Yellow chickens were divided into four groups and were inoculated intraocularly with one of the four field IBDV strains, namely NN1172, NN1005, GD10111 and JS7, respectively. The mortality and relative weight of bursa and t

Viral proteins expressed in the protozoan parasite Eimeria tenella are detected by the chicken immune system.

BACKGROUND: Eimeria species are parasitic protozoa that cause coccidiosis, an intestinal disease commonly characterised by malabsorption, diarrhoea and haemorrhage that is particularly important in chickens. Vaccination against chicken coccidiosis is effective using wild-type or attenuated live parasite lines. The development of protocols to express foreign proteins in Eimeria species has opened up the possibility of using Eimeria live vaccines to deliver heterologous antigens and function as multivalent vaccine vectors that could protect chickens against a range of pathogens. RESULTS: In thi

Multiplex gradient immunochip for detection of post-vaccinal antibodies in poultry.

Multiplex analysis as an immunochip-in-a well format for simultaneous detection of post-vaccinal antibodies to three poultry infections (Newcastle disease, infectious bronchitis and bursal disease) in one chicken sera was developed. The immunochip had a microarray format printed on the bottom of a standard microtiter plate well and consisted of 36 microspots (d = 400 μm each) with three lines of viral antigens absorbed in a gradient of five decreasing concentrations. Optimization of assay conditions revealed the necessity of careful choice of the reaction buffer due to the high tendency of chi

Evaluation of the immune responses of biological adjuvant bivalent vaccine with three different insertion modes for ND and IBD.

Infectious bursal disease (IBD) is a widespread problem in the poultry industry, and vaccination is the primary preventive method. However, moderately virulent vaccines may damage the bursa, necessitating the development of a safe and effective vaccine. The Newcastle disease virus (NDV) has been explored as a vector for vaccine development. In this study, reverse genetic technology was used to obtain three recombinant viruses, namely, rClone30-VP2L (P/M)-chGM-CSF (NP), rClone30-chGM-CSF (P/M)-VP2L (NP), and rClone30-VP2L-chGM-CSF (P/M). Animal experiments showed that the three biological adjuv

Identification of novel T-cell epitopes on viral protein VP4 of Infectious Bursal Disease Virus (IBDV) that play critical roles in eliciting cellular immune response.

Infectious bursal disease virus (IBDV) is a highly infectious RNA virus that causes severe damage to the bursa of Fabricius (BF), resulting in immunosuppression. Currently, IBDV vaccines mainly rely on the induction of neutralizing antibodies against VP2 for protection, and the role of cellular immunity against IBDV infection is unclear. Here, we show that IBDV VP4, a serine protease of the virion, is responsible for inducing specific T cell immunity against IBDV infection. Furthermore, we identified three specific T cell epitope peptides on VP4, among which, two epitopes (4/10 and 4/12) could

LEI0258 microsatellite variability and its association with humoral and cell mediated immune responses in broiler chickens.

Major histocompatibility complex (MHC) has a profound influence on disease resistance or susceptibility, productivity and important economic traits in chicken. Association of the MHC with a wide range of immune responses makes it a valuable predictive factor for the disease pathogenesis and outcome. The tandem repeat LEI0258 is a genetic marker which is located within the B locus of chicken MHC and strongly associated with serologically defined haplotypes. LEI0258 microsatellite marker was applied to investigate the MHC polymorphism in Ross 308 broiler chicken (N=104). Association of LEI0258 a

Infectious bursal disease virus influences the transcription of chicken γc and γc family cytokines during infection.

Infectious bursal disease virus (IBDV) infection causes immunodeficiency in chickens. To understand cell-mediated immunity during IBDV infection, this study perform a detailed analysis of chicken γc chain (chCD132) and γc family cytokines, including interleukins 2, 4, 7, 9, and 15. The mouse anti-chCD132 monoclonal antibody (mAb) was first generated by the E.coli-expressed γc protein. Immunofluorescence assay further showed that γc was a protein located with the anti-chCD132 mAb on the surface of chicken's splenic mononuclear cells. Real-time quantitative RT-PCR revealed that the chCD132 mRNA

Development of a recombinant VP2 vaccine for the prevention of novel variant strains of infectious bursal disease virus.

Since 2017, novel variant strains of infectious bursal disease virus (nvIBDV) have been detected in China, while the current vaccines on the market against very virulent IBDV have limited protection against this subtype virus. In this context, a strain of the virus has been isolated, and sequencing alignment and bird regression experiments showed that the virus was IBDV, belonging to the nvIBDV subtype (and named IBDV FJ-1812). Furthermore, the Escherichia coli expression system was used to successfully express soluble nvIBDV rVP2, which is specifically recognized by an anti-IBDV standard seru

Recombinant chicken interleukin-7 as a potent adjuvant increases the immunogenicity and protection of inactivated infectious bursal disease vaccine.

Our previous work showed that a plasmid-based chicken interleukin-7 (chIL-7) gene expression vector possessed potent adjuvant activity for a VP2 DNA vaccine against chicken infectious bursal disease virus (IBDV). Whether recombinant chIL-7 prepared in procaryotic expression system has the adjuvant activity for inactivated IBDV vaccine remains unknown. Here, we prepared recombinant chIL-7 using an E. coli expression system and analyzed its adjuvant activity for the inactivated IBDV vaccine. The results show that the recombinant chIL-7 was successfully prepared in E. coli using the pET20b vector

Genome analysis and pathogenicity of reticuloendotheliosis virus isolated from a contaminated vaccine seed against infectious bursal disease virus: first report in China.

Specific-pathogen-free (SPF) chickens were inoculated with the virus seed of an infectious bursal disease virus (IBDV)-attenuated vaccine, and positive reticuloendotheliosis virus (REV) antibody levels were subsequently detected in the chicken sera, indicating potential REV contamination of the vaccine. After neutralization with IBDV-positive blood serum, the vaccine was inoculated into DF-1 cells for REV isolation and identification. An REV strain, designated IBD-C1605, was identified using an immunofluorescence assay test. Three pairs of primers were employed for the amplification, cloning a
