Microwave-assisted digestion combined with silica-based spin column for DNA isolation from human bones.

A protocol for the extraction of DNA from ancient skeletal material was developed. Bone specimen samples (powder or slice), buffer, pretreatment, and extraction methodologies were compared to investigate the best conditions yielding the highest concentration of DNA. The degree of extract contamination by polymerase chain reaction (PCR) inhibitors was compared as well. Pretreatment was carried out using agitation in an incubator shaker and microwave digestion. Subsequently, DNA from bones was isolated by the classical organic phenol-chloroform extraction and silica-based spin columns. Decalcifi

In vivo performance of combinations of autograft, demineralized bone matrix, and tricalcium phosphate in a rabbit femoral defect model.

Large bone defects may be treated with autologous or allogeneic bone preparations. Each treatment has advantages and disadvantages; therefore, a clinically viable option for treating large (e.g., gap) bone defects may be a combination of the two. In the present study, bone repair was determined with combinations of autografts, allografts, and synthetic bone grafts using an established rabbit femoral defect model. Bilateral unicortical femoral defects were surgically prepared and treated with combinatorial bone grafts according to one of seven treatment groups. Recipient sites were retrieved at

Monitoring Demineralization and Subsequent Remineralization of Human Teeth at the Dentin-Enamel Junction with Atomic Force Microscopy.

Using atomic force microscopy, we monitored the nanoscale surface morphology of human teeth at the dentin-enamel junction after performing successive demineralization steps with an acidic soft drink. Subsequently, we studied the remineralization process with a paste containing calcium and phosphate ions. Repeated atomic force microscopy imaging of the same sample areas on the sample allowed us to draw detailed conclusions regarding the specific mechanism of the demineralization process and the subsequent remineralization process. The about 1-μm-deep grooves that are caused by the demineralizat

Hydrogen Peroxide Head Preparation: Enabling Cuttings and Anatomic Studies of Skull Base Dura Mater and Arachnoid Without Use of Drilling.

INTRODUCTION: Anatomic pieces that are preserved using formaldehyde allow us to undertake high-quality skull base studies. However, extensive drilling is often necessary, which can lead to damages to the dura mater and thus arachnoid. Formaldehyde and hydrogen peroxide can soften the bone, which, in turn, can be easily cut with a scalpel or removed with a curette. After having discovered this technique by chance, our aim was to establish a study protocol of the skull base dura mater without the use of the drill. METHODS: Ten heads were set with a 10% formalin solution and then injected with c

A prospective randomized comparison of PEEK cage containing calcium sulphate or demineralized bone matrix with autograft in anterior cervical interbody fusion.

PURPOSE: A variety of bone substitutes have been successfully used to fill PEEK cages in cervical interbody fusion in order to avoid the complications related to bone harvesting from the donor site. However, no controlled study has previously been conducted to compare the effectiveness of PEEK interbody cages containing calcium sulphate/ demineralized bone matrix (CS/DBM) with autogenous cancellous bone for the treatment of cervical spondylosis. The objective of this prospective, randomized clinical study was to evaluate the effectiveness of implanting PEEK cages containing CS/DBM for the trea

Efficacy of sinus crestal elevation with demineralized dentin matrix at the apex of the implants.

BACKGROUND: This study evaluated the clinical and radiographic outcomes of using demineralized dentin matrix (DDM) as a graft material at the implant apex during crestal sinus floor elevation. We hypothesized that DDM would promote predictable bone regeneration at the sinus floor and facilitate implant stability with minimal surgical morbidity. METHODS: A retrospective analysis was performed on 55 implants placed with transcrestal sinus floor elevation and DDM grafting at the implant apex in the posterior maxilla. Radiographic outcomes were assessed with CBCT. The primary variable was vertica

Skeletal analysis and comparison of bog bodies from Northern European peat bogs.

Although numerous bodies were deposited in Western European bogs in the past centuries, few were found and underwent archeological analysis. No studies comparing skeletal structure and mineralization of bog bodies from different ages have been performed to this day. Therefore, the aim of this study was to analyze and compare skeletal features and specifics of the human remains of three bog bodies from the Iron and Middle Ages found in Northern European peat bogs. Demineralization due to the acidic environment in peat bogs was comparably pronounced in all three bodies. Still, the macroscopic st

Reconstruction of distal tibia bone loss by intercalary distal tibial allograft and stabilization using anterograde tibiotalar intramedullary nailing : technical note and case report.

INTRODUCTION: The difficulties of managing bone loss in the distal tibia are well known in the literature. The various therapeutic options available to us include custom prosthetic replacement, talocrural arthrodesis with allograft, vascularised or non-vascularised autograft, bone transfer according to Illizarov and insertion of a metal augment. In the case of non-conservation of the talocrural joint, osteosynthesis is performed using adapted plates and screws or, more conventionally, transplanted centromedullary nailing. We report on a clinical case of bone loss in the distal tibia in an infe

The role of the combination of Moringa oleifera leaf extract and demineralized freeze-dried bovine bone xenograft (xenograft) as tooth extraction socket preservation materials on osteocalcin and transforming growth factor-beta 1 expressions in alveolar bone of Cavia cobaya.

AIM: Alveolar bone resorption, often occurring after tooth extraction, can be minimized through socket preservation. This process uses a combination of Moringa leaf extract and demineralized freeze-dried bovine bone xenograft (DFDBBX) that is expected to generate both transforming growth factor-beta 1 (TGF-β1) expressions as a transcription factor associated with osteoblast differentiation and osteocalcin accelerating alveolar bone formation. This research aimed to analyze the role of the combination of Moringa leaf extract and DFDBBX induced in socket preservation when generating TGF-β1 and o

Proteoglycan-collagen interactions.

Among the most important events in connective tissue physiology are the nucleation, growth and calcification of collagen fibrils. It has been speculated that all are associated with, or even controlled by collagen-proteoglycan interactions. We therefore developed methods for investigating these associations in tissues, particularly for understanding their significance for type I collagen, the commonest form of collagen in the body, especially predominant in bone. Using an electron-dense dye, Cupromeronic blue, in the 'critical electrolyte concentration' mode, and digestion by hyaluronidase, ch
