Analysis of the subproteomes of proteinases and heparin-binding toxins of eight Bothrops venoms.

Viperid snakes show the most complex snake-venom proteomes and offer an intriguing challenge in terms of understanding the nature of their components and the pathological outcomes of envenomation characterized by local and systemic effects. In this work, the venom complexity of eight Bothrops species was analyzed by 2-DE, and their subproteomes of proteinases were explored by 2-D immunostaining and 2-D gelatin zymography, demonstrating the diversity of their profiles. Heparin, a highly sulfated glycosaminoglycan released from mast cells, is involved in anti-coagulant and anti-inflammatory proc

Biochemistry and toxicology of toxins purified from the venom of the snake Bothrops asper.

The isolation and study of individual snake venom components paves the way for a deeper understanding of the pathophysiology of envenomings--thus potentially contributing to improved therapeutic modalities in the clinical setting--and also opens possibilities for the discovery of novel toxins that might be useful as tools for dissecting cellular and molecular processes of biomedical importance. This review provides a summary of the different toxins that have been isolated and characterized from the venom of Bothrops asper, the snake species causing the majority of human envenomings in Central

Control of venom production and secretion by sympathetic outflow in the snake Bothrops jararaca.

Many studies have examined the morphological and biochemical changes in the secretory epithelium of snake venom glands after a bite or milking. However, the mechanisms of venom production and secretion are not yet well understood. The present study was undertaken to evaluate the role of the sympathetic nervous system in the control of venom production and secretion. Venom glands were obtained from Bothrops jararaca (Viperidae) snakes, either unmilked previously or milked 4, 7 or 15 days before they were killed. Levels of tyrosine-hydroxylase-like immunoreactivity were higher in venom glands co

Properties of fibrinogen cleaved by Jararhagin, a metalloproteinase from the venom of Bothrops jararaca.

Haemorrhagic metalloproteinases from Bothrops jararaca and other venoms degrade vessel-wall and plasma proteins involved in platelet plug and fibrin clot formation. These enzymes also cause proteolytic digestion of fibrinogen which has been suggested to cause defective platelet function. Fibrinogen degradation by jararhagin, a metalloproteinase from B. jararaca, and the effect of jararhagin fibrinogenolysis on both platelet aggregation and fibrin clot formation were investigated. Jararhagin was found to cleave human fibrinogen in the C-terminal region of the A alpha-chain giving rise to a 285-

The humoral immune responses of patients bitten by the snake Bothrops jararaca (jararaca).

The isotype and specificity of antibodies produced by patients bitten by B. jararaca and submitted to serum therapy were studied. The IgG anti-B. jararaca antibodies have large individual dispersion, starting to appear 10 days after the first bite and increasing to at least 80 days after the bite. IgM antibodies appeared sooner than IgG antibodies but disappeared about 20 days after the bite. Secondary responses induced by an additional bite were characterized by a fast and higher IgG antibody response with no apparent change in the IgM antibody. The immunoblotting tests showed that the specif

Studies on blood coagulation and fibrinolysis in patients bitten by Bothrops jararaca (jararaca).

The blood coagulation and the fibrinolytic systems of nine patients envenomed by Bothrops jararaca in São Paulo (Brazil) were studied. Five of the accidents were caused by young snakes (less than 50 cm). On admission, four patients had non-clotting and three partially-clotting blood. Fibrinogen levels were decreased due to the thrombin-like activity of the venom as expected. Consequent secondary activation of the fibrinolytic system was evident from the low levels of alpha-2-antiplasmin and the high titres of fibrin(ogen) degradation products. High titres of cross-linked fibrin fragment D (D-d

Intracerebral hemorrhage (hemorrhagic stroke) secondary to Bothrops spp. snakebite envenomation in a horse.

This report describes the first documented case of intracerebral hemorrhage (ICH) in a horse following Bothrops snakebite envenomation. A 3-year-old mare developed acute kidney injury, systemic hemorrhage, and neurological signs before euthanasia. Postmortem findings confirmed massive ICH with intralesional venom identified by immunohistochemistry. Delayed treatment and insufficient antivenom likely contributed to the outcome. This case highlights the potential for cerebrovascular complications following Bothrops snakebite envenomation in horses, which may present as subtle neurological signs.

N-Glycoproteomic Portraits of Bothrops Snake Venoms Reveal Evolutionarily Conserved and Divergent Phenotypes.

Glycosylation is a major protein post-translational modification in snake venom proteins and contributes to the diversification of proteomes. In this study, we carried out an in-depth analysis of the glycosylation profile of seven Bothrops venoms, including neutral sugar quantification, glycoprotein profiling by lectin blot, and determination of N-glycosylation sites in proteins and their N-glycan compositions, by direct, intact glycopeptide analysis by mass spectrometry. Interestingly, all identified N-glycosylated peptides were from enzymatic venom components, mainly proteolytic enzymes that

From Forest to Intensive Care Unit: Managing Severe Complications of Bothrops bilineatus Envenomation.

Snakebites are a critical health issue in remote regions of the Amazon basin. I report a case of Bothrops bilineatus envenomation in a 29-y-old biologist conducting fieldwork in Parque Nacional del Manu, Peru. Following significant logistic delays-including an initial suboptimal antivenom dose and limited access to additional vials-the patient developed progressive edema, necrosis, coagulopathy, and signs of compartment syndrome. Management involved 2 partial doses of antivenom (1 at 1600 and the other at 2100), a subsequent transfer to a tertiary care center, antibiotics, analgesia, and event

Plant-Derived Lapachol Analogs as Selective Metalloprotease Inhibitors Against Bothrops Venom: A Review.

Plant compounds that inhibit snake venom activities are relevant and can provide active molecules to counteract snake venom effects. Numerous studies on snake viperid venoms found that metalloproteinases play a significant role in the pathophysiology of hemorrhage that occurs on envenomation. Preclinical studies using vitro and in vivo protocols investigated natural compounds and viperid snake venoms, evaluating the enzymatic, procoagulant, hemorrhagic, edematogenic, myotoxic, and lethal activities. Many studies focused on Bothrops venoms and ascribed that angiorrhexis and hemorrhage resulted

Brain death due to intracranial hemorrhage in a child following suspected Bothrops snakebite.

Snakebites from the genus Bothrops are common and are responsible for the highest mortality rate in Brazil. Factors related to the species, treatment, and patient influence the clinical manifestations and prognosis of the condition. Young patients without comorbidities have better prognoses and rarely develop severe systemic complications. This report discusses a case of brain death of an 11-year-old boy due to intracranial hypertension following possible Bothrops snakebite. Despite receiving antivenom therapy, the patient experienced seizures, mental confusion, and decreased consciousness. Au

Antiophidic potential of chlorogenic acid and rosmarinic acid against Bothrops leucurus snake venom.

Bothrops leucurus is responsible for most cases of snakebite in Northeast Brazil; however, this species is not included in the pool of venoms used in antivenom production in Brazil. The serotherapy has logistical and effectiveness limitations, which stimulates the search for therapeutic alternatives. Chlorogenic acid and rosmarinic acid present several biological activities, but their antiophidic potential has been poorly explored. Thus, the aim of this approach was to evaluate the potential inhibitory effects of these compounds on B. leucurus venom. Initially, the enzymatic inhibition of toxi

Observation of Bothrops atrox Snake Envenoming Blister Formation from Five Patients: Pathophysiological Insights.

In the Brazilian Amazon, Bothrops atrox snakebites are frequent, and patients develop tissue damage with blisters sometimes observed in the proximity of the wound. Antivenoms do not seem to impact blister formation, raising questions regarding the mechanisms underlying blister formation. Here, we launched a clinical and laboratory-based study including five patients who followed and were treated by the standard clinical protocols. Blister fluids were collected for proteomic analyses and molecular assessment of the presence of venom and antivenom. Although this was a small patient sample, there

Acute kidney injury induced by thrombotic microangiopathy in two cases of Bothrops envenomation.

CONTEXT: Bothrops snakes are the most frequent agents of snakebites in South and Central America. Acute kidney injury (AKI) is one of its complications and has multifactorial origin. Thrombotic microangiopathy (TMA)-induced AKI in snakebites is uncommon and is not described in Bothrops envenomation. CASE DETAILS: We report two cases of patients bitten by young Bothrops jararaca who developed AKI induced by TMA. Both patients evolved with mild envenomation and received the specific antivenom within 4 h after the snakebite. None of them had hypotension or shock, bleeding or secondary infection.

Action of two phospholipases A2 purified from Bothrops alternatus snake venom on macrophages.

The in vitro effects of BaltTX-I, a catalytically inactive Lys49 variant of phospholipase A2 (PLA2), and BaltTX-II, an Asp49 catalytically active PLA2 isolated from Bothrops alternatus snake venom, on thioglycollate-elicited macrophages (TG-macrophages) were investigated. At non-cytotoxic concentrations, the secretory PLA2 BaltTX-I but not BaltTX-II stimulated complement receptor-mediated phagocytosis. Pharmacological treatment of TG-macrophages with staurosporine, a protein kinase C (PKC) inhibitor, showed that this kinase is involved in the increase of serum-opsonized zymosan phagocytosis in

Activation of Bothrops jararaca snake venom gland and venom production: a proteomic approach.

UNLABELLED: Viperidae venom glands have a basal-central lumen where the venom produced by secretory cells is stored. We have shown that the protein composition of venom gland changes during the venom production cycle. Here, we analyzed the venom gland proteins during the venom production cycle by proteomic approach. We identified specific proteins in each stage of the cycle. Protein species from endoplasmic reticulum (PDI and GPR78) and cytoplasm (actin, vimentin, tropomyosin, proteasome subunit alpha type-1, thioredoxin, and 40S ribosomal protein) are more abundant in the activated stage, pro

Hypericum brasiliense plant extract neutralizes some biological effects of Bothrops jararaca snake venom.

Alternative treatments for snake bite are currently being extensively studied, and plant metabolites are considered good candidates for such purpose. Here, the ability of a crude ethanolic extract of Hypericum brasiliense plant in neutralizing Bothrops jararaca snake venom was investigated by in vitro (coagulation, hemolysis or proteolysis) and in vivo (hemorrhage, lethality and edema) biological assays. We describe for the first time the ability of H. brasiliense extracts to inhibit some pharmacological effects of a Brazilian snake venom. Inhibitory assays were performed by incubating B. jara
