Biological control of Botrytis cinerea in tomato and Fusarium graminearum in wheat using the novel species Burkholderia mycopellens and Burkholderia crassaminum.

The use of beneficial microbes to reduce plant stress upon fungal pathogens is a promising strategy for sustainable crop protection. In this study, we evaluated five novel Burkholderia strains for their capacity to mitigate Botrytis cinerea infection in tomato (Solanum lycopersicum L.) and Fusarium graminearum infection in wheat (Triticum aestivum L.). All strains significantly suppressed B. cinerea infection symptoms in tomato plants, hallmarked by a smaller decrease in photosynthetic activity and chlorophyll content compared to infected control. Using a GFP-tagged F. graminearum strain, we s

Primary Mode of Action of the Novel Sulfonamide Fungicide against Botrytis cinerea and Field Control Effect on Tomato Gray Mold.

Botrytis cinerea is considered an important plant pathogen and is responsible for significant crop yield losses. With the frequent application of commercial fungicides, B. cinerea has developed resistance to many frequently used fungicides. Therefore, it is necessary to develop new kinds of fungicides with high activity and new modes of action to solve the increasingly serious problem of resistance. During our screening of fungicide candidates, one novel sulfonamide compound, N-(2-trifluoromethyl-4-chlorphenyl)-2-oxocyclohexyl sulfonamide (L13), has been found to exhibit good fungicidal activi

Sensitivity of Botrytis cinerea Isolates from Conventional and Organic Strawberry Fields in Brazil to Azoxystrobin, Iprodione, Pyrimethanil, and Thiophanate-Methyl.

Botrytis fruit rot, caused by Botrytis cinerea, is one of the most important strawberry diseases worldwide, and fungicide applications are often used to manage the disease in commercial production. Isolates of B. cinerea were collected from conventional and organic strawberry fields in four Brazilian States from 2013 to 2015 and their sensitivity to the main single-site mode-of action fungicides used in Brazil was tested. Resistance to azoxystrobin, iprodione, pyrimethanil, and thiophanate-methyl was found and values for effective concentration that inhibited mycelial growth by 50% were higher

Effects of crop development on the emission of volatiles in leaves of Lycopersicon esculentum and its inhibitory activity to Botrytis cinerea and Fusarium oxysporum.

Volatiles emitted from the leaves of Lycopersicon esculentum at the two-, ten-leaf and anthesis periods were collected by a gas absorbing method and analyzed by gas chromatography (GC)-mass spectrometry. In total, 33 compounds of volatiles emitted from three developmental stage plants were separated and identified, and quantitatively analyzed by the internal standard addition method. All of the samples of volatile were found to be rich in monoterpenes and sesquiterpenes. beta-phellandrene and caryophyllene predominated in the volatiles of the leaves of plants at the two- and ten-leaf stages. Yeast increases resistance in Arabidopsis against Pseudomonas syringae and Botrytis cinerea by salicylic acid-dependent as well as -independent mechanisms.

Cell-wall and glucopeptide components of yeast have been reported to exhibit elicitor activity. The mode of action of defense activation by yeast is not known so far. In this study, we used the model plant Arabidopsis to investigate the activation of defense responses by yeast, the effect on resistance against different pathogens, and the mode of action. Treatment of Arabidopsis plants with an autoclaved yeast suspension induced the expression of systemic acquired resistance-related genes and accumulation of the phytoalexin camalexin. Symptom development and bacterial growth after infection wi

Combined biocontrol to tomato gray mold and inhibition via multi-mechanistic antifungal activity to Botrytis cinerea by Trichoderma harzianum T4.

Tomato gray mold, caused by Botrytis cinerea, causes severe global economic losses. This study evaluates the biocontrol efficacy and underlying mechanisms of Trichoderma harzianum T4 which has application potential. Results showed that T. harzianum T4 significantly promoted tomato seed germination and growth while enhancing systemic resistance. T4 exhibited high ecological fitness, maintaining growth and sporulation under salinity, acid, and nutrient stress. Notably, both volatile organic compounds and cell-free culture filtrates of T4 markedly inhibited B. cinerea by disrupting physiological

Deep Learning-Guided Discovery of Celestolide as a Natural Allosteric Inhibitor Targeting Botrytis cinerea CYP51 and Its Application in Strawberry Preservation.

Most CYP51 inhibitors act competitively and are prone to resistance, whereas allosteric inhibitors hold promise but are difficult to develop. In this study, we employed the neural relational inference framework alongside the Gaussian network-based deep learning model CorrSite2.0 to proactively identify allosteric sites on B. cinerea CYP51. We confirmed celestolide as a natural allosteric inhibitor, targeting CYP51. Celestolide forms strong interactions with CYP51 at the identified allosteric site, inducing conformational changes in CYP51 that suppress its enzymatic activity. These findings wer

DHN melanin biosynthesis in the plant pathogenic fungus Botrytis cinerea is based on two developmentally regulated key enzyme (PKS)-encoding genes.

Botrytis cinerea is the causal agent of gray mold disease in various plant species and produces grayish macroconidia and/or black sclerotia at the end of the infection cycle. It has been suggested that the pigmentation is due to the accumulation of 1,8-dihydroxynaphthalene (DHN) melanin. To unravel its basis and regulation, the putative melanogenic and regulatory genes were identified and functionally characterized. Unlike other DHN melanin-producing fungi, B. cinerea and other Leotiomycetes contain two key enzyme (PKS)-encoding enzymes. Bcpks12 and bcpks13 are developmentally regulated and ar

Infection of Arabidopsis with a necrotrophic pathogen, Botrytis cinerea, elicits various defense responses but does not induce systemic acquired resistance (SAR).

Botrytis cinerea is a non-specific necrotrophic pathogen that attacks more than 200 plant species. In contrast to biotrophs, the necrotrophs obtain their nutrients by first killing the host cells. Many studies have shown that infection of plants by necrosis-causing pathogens induces a systemic acquired resistance (SAR), which provides protection against successive infections by a range of pathogenic organisms. We analyzed the role of SAR in B. cinerea infection of Arabidopsis. We show that although B. cinerea induced necrotic lesions and camalexin biosynthesis, it did not induce SAR-mediated p

An essential-oil blend delivers dual-action control of grey mold in pomegranate by suppressing Botrytis cinerea and reshaping the husk volatilome.

Pomegranate (Punica granatum L.) fruit is highly susceptible to postharvest grey mold caused by Botrytis cinerea, which develops during cold storage and causes substantial losses, while concern over fungicide residues and resistance has intensified the search for natural alternatives. The present study evaluated an essential-oil blend (EOB) composed of thymol, eugenol and 1,8-cineole, applied at three doses, for the control of B. cinerea on pomegranate husk during 28 days of cold storage, and combined decay assessment with headspace solid-phase microextraction-gas chromatography-mass spectrome

Arsenic Resistance Protein BcArs2 Modulates Endogenous ROS Homeostasis in Infection Cushions to Enhance the Virulence of Botrytis cinerea Toward Host Plants.

Botrytis cinerea triggers destructive grey mould with substantial economic losses, yet the functions of numerous pathogenicity genes remain largely unclear. Here, by screening a T-DNA insertion library, we identified BcARS2, a key component of the Microprocessor complex. BcARS2 is constitutively expressed, localizes to the nucleus, and modulates gene expression. The ΔBcars2 mutants exhibited impaired vegetative growth, enhanced conidiophore sporulation, increased conidiation, reduced sclerotial size, and increased sensitivity to multiple stresses. Pathogenicity assays showed that different ino

Dietary cauliflower (Brassica oleracea var. botrytis) mitigates benzo[a]pyrene-induced oxidative stress, immune dysfunction, and tissue damage in Nile tilapia.

Benzo[a]pyrene (BaP) is a ubiquitous polycyclic aromatic hydrocarbon that has been reported to induce immunotoxicity and oxidative stress in fish. In the current study, the potential protective efficacy of dietary cauliflower powder (CAF; Brassica oleracea var. botrytis) is investigated in Nile tilapia (Oreochromis niloticus) subjected to water-borne BaP exposure. Fish (initial body weight 35.45 ± 5.33 g; n = 280) were randomly assigned to seven groups (n = 40/group): control, acetone (12.25 µL/L), CAF0.5 (0.5%), CAF1 (1%), BaP (12.25 µg/L), CAF0.5 + BaP, and CAF1 + BaP, and maintained on thei

Inhibition of Botrytis cinerea in tomatoes by allyl-isothiocyanate release from black mustard (Brassica nigra) seeds and detection by E-nose.

The aim of this work is to delayed the postharvest development of Botrytis cinerea in tomatoes by releasing allyl-isothiocyanate (AITC) from mustard seeds at room temperature, and to discriminate the aromatic profile by using an electronic device (E-nose). Olfactory sensory analysis showed that tomatoes inoculated in the presence of AITC did not develop the microorganisms until the eighth day of storage. The highest inhibitory concentration of AITC was found in Day 3 (175.18 ppb). However, tomatoes inoculated without the presence of AITC developed a moldy aroma on the third day of storage. The

Bacillus-Secreted Oxalic Acid Induces Tomato Resistance Against Gray Mold Disease Caused by Botrytis cinerea by Activating the JA/ET Pathway.

Bacillus spp. are known for their ability to control plant diseases; however, the mechanism of disease control by Bacillus spp. is still unclear. Previously, bacterial organic acids have been implicated in the process of disease suppression. We extracted the total organic acid from Bacillus cereus AR156 culture filtrate and identified oxalic acid (OA) as the programmed cell death-inducing factor. OA strongly suppressed the lesion caused by Botrytis cinerea without significant antagonism against the fungus. Low concentration of OA produced by Bacillus spp. inhibited cell death caused by high co

Oxidation of chlortetracycline and its isomers by Botrytis aclada laccase in the absence of mediators: pH dependence and identification of transformation products by LC-MS.

Tetracyclines are antibiotics considered emerging pollutants and currently, wastewater treatment plants are not able to remove them efficiently. Laccases are promising enzymes for bioremediation because they can oxidize a wide variety of substrates. The aim of this study was to evaluate the Botrytis aclada laccase for the oxidation of chlortetracycline and its isomers in the absence of a mediator molecule, at a pH range between 3.0 to 7.0, and to characterize the transformation products by LC-MS. Chlortetracycline and three isomers were detected in both, controls and reaction mixtures at 0 h a

Production of reactive oxygen species and wound-induced resistance in Arabidopsis thaliana against Botrytis cinerea are preceded and depend on a burst of calcium.

BACKGROUND: Wounded leaves of Arabidopsis thaliana produce reactive oxygen species (ROS) within minutes after wounding and become resistant to the pathogenic fungus Botrytis cinerea at a local level. This fast response of the plants to the wound is called wound-induced resistance (WIR). However the molecular mechanisms of this response and the signal cascade between the wound and ROS production are still largely unknown. Calcium is a conserved signal and it is involved in many abiotic stress responses in plants, furthermore, calcium pathways act very fast. RESULTS: The results of this study s
