Withania somnifera osmotin (WsOsm) confers stress tolerance in tobacco and establishes novel interactions with the defensin protein (WsDF).

Pathogenesis-related proteins (PR), including osmotins, play a vital role in plant defense, being activated in response to diverse biotic and abiotic stresses. Despite their significance, the mechanistic insights into the role of osmotins in plant defense have not been extensively explored. The present study explores the cloning and characterization of the osmotin gene (WsOsm) from Withania somnifera, aiming to illuminate its role in plant defense mechanisms. Quantitative real-time PCR analysis revealed significant induction of WsOsm in response to various phytohormones e.g. abscisic acid, sal

Evaluating the effects of azelaic acid in the metabolism of Arabidopsis thaliana seedlings through untargeted metabolomics and ionomics approaches.

The present study demonstrates that low concentrations of azelaic acid (AZA) significantly impact the metabolism of Arabidopsis thaliana seedlings, leading to imbalances in numerous minerals and metabolites due to AZA-induced stress. Untargeted metabolomic analyses were conducted on untreated and AZA-treated seedlings at two time points: 7 and 14 days after treatment initiation. The results revealed a general accumulation of sugars (e.g., glucose, mannose, xylose), amino acids (e.g., lysine, GABA, threonine, glutamine), and organic acids (e.g., glutaric acid, shikimic acid, succinic acid) in A

The BZR1-EDS1 module regulates plant growth-defense coordination.

Plants have developed sophisticated strategies to coordinate growth and immunity, but our understanding of the underlying mechanism remains limited. In this study, we identified a novel molecular module that regulates plant growth and defense in both compatible and incompatible infections. This module consisted of BZR1, a key transcription factor in brassinosteroid (BR) signaling, and EDS1, an essential positive regulator of plant innate immunity. We found that EDS1 interacts with BZR1 and suppresses its transcriptional activities. Consistently, upregulation of EDS1 function by a virulent Pseu

Transcriptional Analysis Reveals Different Fertilizer Treatments Improve the Yield and Quality of Tomato Fruits by Postponing Age-Triggered Senescence.

Inappropriate fertilization has led to adverse environmental effects and food-safety issues. Biostimulants have been widely used to improve the growth, yield, and quality of crops. However, the effects of biostimulants on age-triggered senescence in plants, which affect the later yield and quality, have scarcely been reported. In this study, the fertilization formulas that were modified according to the requirements of plants in different growth stages, and adding yeast extract agents or microorganisms were respectively used in soilless tomato culture as three fertilization treatments. The Yam

Cell-type specific early perception of nine phytohormones revealed by single-nucleus transcriptomics in Arabidopsis.

Plant development and stress responses are coordinated through phytohormone-mediated gene networks, yet resolving their spatiotemporal crosstalk remains challenging. Here, we generate a single-nucleus transcriptomic atlas of Arabidopsis seedlings, capturing early (0.5 h, 3 h) responses to 9 hormones across ~500,000 nuclei, including auxin, cytokinin, ABA, gibberellin, strigolactone, brassinosteroid, ethylene, JA, and SA. Within this window, most hormones showed rapid and cell-type-specific responses, whereas JA, SA, ABA showed more sustained and convergent responses by 3 h. Co-directional tran

GhSQS1-mediated squalene accumulation coordinates cotton development and stress resilience.

Squalene is a high-value triterpenoid. It is increasingly recognized for its regulatory roles in plant signaling. However, the functions of squalene in cotton remain unexplored. Here, we show that exogenous squalene can promote cotton developmental traits (root elongation and ovule development) and identify GhSQS1 as the predominant functional squalene synthase. Overexpression of GhSQS1 promotes root development, along with metabolic and hormonal reprogramming, including the activation of brassinosteroid (BR)-associated signaling. Furthermore, stable overexpression of GhSQS1 markedly elevates

Hormone-modulated transcription factors orchestrating the root nodule symbiosis.

Transcription factors play essential roles modulating gene expression during plant development and the adaptation to environmental cues through the control of morphogenetic programs. In the root nodule symbiosis between legumes and rhizobia, two coordinated morphogenetic programs are activated by the perception of bacterial signals: the organogenesis of the nodule, a lateral root organ specialized in nitrogen fixation, and the infection process that allows the bacteria to colonize the nodule. These programs are influenced by the action of phytohormones, mainly auxin, cytokinin, ethylene, gibbe

PcXTH22 as a core regulator enhancing aluminum toxicity tolerance in Populus cathayana.

Aluminum (Al) toxicity in acidic soils restricts plant growth. Xyloglucan endotransglucosylase/hydrolases (XTHs) modulate Al responses, but their mechanisms in poplar remain unknown. Here, we characterized PcXTH22-mediated Al tolerance in Populus cathayana. The PcXTH22OE lines showed negligible injury, whereas the pcxth22 mutants suffered severe damage, confirming the role of PcXTH22 as a positive Al regulator. The pcxth22 lines showed increased xyloglucan endotransglucosylase/hydrolase (XET/XTH) activities and xyloglucan content under Al toxicity. This offered more Al³ ⁺ binding sites and res

Dual regulation of the receptor-like kinase BIR1 involves site-directed transcript cleavage and 5'-leader-mediated translational control.

In Arabidopsis, BRASSINOSTEROID INSENSITIVE1-ASSOCIATED RECEPTOR KINASE 1 (BAK1)-INTERACTING RECEPTOR-LIKE KINASE 1 (BIR1) is a negative regulator of plant immunity and cell death. BIR1 was earlier described as a target of epigenetic and post-transcriptional degradation. During virus infections, degradome analysis of BIR1 transcripts mapped predominant mRNA cleavage sites at the 5'-untranslated leader region (site A) and the protein-coding sequence (sites B and C). Here, we identified another virus-associated cleavage site (D) within the BIR1 coding region and investigated the contribution of

Chloroplasts alter their morphology and accumulate at the pathogen interface during infection by Phytophthora infestans.

Upon immune activation, chloroplasts switch off photosynthesis, produce antimicrobial compounds and associate with the nucleus through tubular extensions called stromules. Although it is well established that chloroplasts alter their position in response to light, little is known about the dynamics of chloroplast movement in response to pathogen attack. Here, we report that during infection with the Irish potato famine pathogen Phytophthora infestans, chloroplasts accumulate at the pathogen interface, associating with the specialized membrane that engulfs the pathogen haustorium. The chemical

Obtusifoliol 14α-demethylase OsCYP51G1 is involved in phytosterol synthesis and affects pollen and seed development.

As structural components of biological membranes, phytosterols are essential not only for a variety of cellular functions but are also precursors for brassinosteroid (BR) biosynthesis. Plant CYP51 is the oldest and most conserved obtusifoliol 14α-demethylase in eukaryotes and is an essential component of the sterol biosynthesis pathway. However, little is known about rice (Oryza sativa L.) CYP51G1. In this study, we showed that rice OsCYP51G1 shared high homology with obtusifoliol 14α-demethylase and OsCYP51G1 was strongly expressed in most of rice organs. Subcellular localization analysis ind

Proteolytic Processing of SERK3/BAK1 Regulates Plant Immunity, Development, and Cell Death.

Plants have evolved many receptor-like kinases (RLKs) to sense extrinsic and intrinsic cues. The signaling pathways mediated by multiple Leucine-rich repeat (LRR) RLK (LRR-RLK) receptors require ligand-induced receptor-coreceptor heterodimerization and transphosphorylation with BRI1-ASSOCIATED RECEPTOR KINASE1 (BAK1)/SOMATIC EMBRYOGENESIS RECEPTOR KINASES family LRR-RLKs. Here we reveal an additional layer of regulation of BAK1 via a Ca2+-dependent proteolytic cleavage process that is conserved in Arabidopsis (Arabidopsis thaliana), Nicotiana benthamiana, and Saccharomyces cerevisiae The prote

An early auxin-responsive Aux/IAA gene from wheat (Triticum aestivum) is induced by epibrassinolide and differentially regulated by light and calcium.

The plant hormone auxin plays a central role in regulating many aspects of plant growth and development. This largely occurs as a consequence of changes in gene expression. The Aux/IAA genes are best characterized among the early auxin-responsive genes, which encode short-lived transcriptional repressors. In most plants examined, including Arabidopsis, soybean, and rice, the Aux/IAA genes constitute a large gene family. By screening the available databases, at least 15 expressed sequence tags (ESTs) have been identified from wheat (Triticum aestivum), which exhibit high sequence identity with

Abscisic acid interplay with phytohormones, plant growth regulators and signaling molecules in orchestrating an elaborate crosstalk for stress resilience in plants.

The increasing challenges, posed by plant biotic and abiotic stress, significantly impact agroecosystems, particularly in the context of rapid global climate change. Phytohormones function as critical chemical messengers, enabling plants to endure these stresses through a complex regulatory framework, thereby playing an essential role in plant survival. Recent research has increasingly underscored the significance of abscisic acid (ABA), in facilitating a prominent role under adverse conditions within plants. ABA has been shown to engage with major phytohormones, such as gibberellins (GAs), et

Sunburn mitigation in dragon fruit (Hylocereus spp.): unravelling genotype-specific physiological and biochemical responses.

INTRODUCTION: Sunburn is a major abiotic stress affecting dragon fruit (Hylocereus spp.), impairing tissue integrity, disrupting physiological functions, and significantly reducing yield. Developing effective mitigation strategies is critical for sustaining productivity under high radiation conditions. METHODS: This study evaluated the efficacy of eleven treatments combining kaolin (5%) with shade net, seaweed extract (0.5%), and various biostimulants in red (H. costaricensis 'CHESH-D1') and white (H. undatus 'CHESH-W1') genotypes. Treatments included T1 (water spray control), T2 (kaolin 5% +

Transcriptome reveals endogenous hormone signal transduction and cell wall remodeling mediating compatibility in apple-pear intergeneric distant hybridization.

Distant hybridization is critical for fruit tree germplasm innovation yet difficult to achieve in woody fruit trees. Successful distant hybridization between apple and pear is rare among fruit trees, but the molecular mechanism of their compatibility remains unclear. Revealing this biological process is vital for the innovation of fruit tree cultivation. This study performed distant hybridization experiments using cultivated apple as female parents and multiple pear cultivars as male parents, and obtained compatible hybrids from the cross of 'YYH' apple × 'YH' pear. A bacterial effector triggers dual-layer plant immunity through interaction with cell-surface and intracellular proteins.

Plants deploy a two-layered immune system, comprising pattern-triggered immunity (PTI) and effector-triggered immunity (ETI), to defend against pathogens. Although PTI and ETI pathways converge on similar downstream responses, they are activated by distinct molecules at the cell surface and in the cytoplasm, respectively. Here, we report that the harpin-like effector RipW from Ralstonia solanacearum localizes to both the apoplastic and intracellular compartments, where it acts as a dual elicitor capable of activating PTI-like and ETI-like responses. We show that RipW triggers PTI-like response

A novel plant immune inducer esculetin activates systemic acquired resistance against Ralstonia solanacearum via the SA signaling pathway.

BACKGROUND: Ralstonia solanacearum poses a severe threat to global agriculture due to its broad host range, high dispersal capacity, and limited chemical control options. Plant immune inducers provide an effective strategy for controlling bacterial wilt disease. RESULTS: This study demonstrated a natural-derived compound esculetin (ES) serves as a novel plant immune inducer against tobacco bacterial wilt. Foliar application of ES exhibits considerable control effect on tobacco bacterial wilt, with control efficacy is high as 45.41%-68.69%, significantly higher than positive control treatment-

Regulatory Mechanism of the GmMYB14 Transcription Factor on Auxin-Related Proteins in Soybean.

In a previous study, GmMYB14 overexpressing (GmMYB14-OX) transgenic soybean plants displayed a semi-dwarfism and compact phenotype, which was regulated by the brassinosteroid (BR) pathway. However, the phenotype of GmMYB14-OX plants could be partly rescued after spraying them with exogenous BR. This indicates that other hormones, in addition to BR, also play a role in regulating the architecture of GmMYB14-OX plants. We observed a significant decrease in the content of endogenous indole-3-acetic acid (IAA) in transgenic soybean lines (OX9 and OX12) compared to wild type (WT) plants. The plant

Changes in endogenous phytohormones regulated by microRNA-target mRNAs contribute to the development of Dwarf Autotetraploid Chinese Cabbage (Brassica rapa L. ssp. pekinensis).

Polyploidization is considered as the major force that drives plant species evolution and biodiversity. The leaves of Chinese cabbage, an important vegetable crop valued for its nutritional quality, constitute the main edible organ. In this study, we found that autotetraploid Chinese cabbage (Brassica rapa ssp. pekinensis) generated from a doubled haploid (DH) line via isolated microspore culture exhibits a dwarf phenotype, along with thick leaves and delayed flowering. Abscisic acid (ABA) and brassinosteroid (BR) levels were significantly lower in autotetraploids compared to DHs. Comparative
