Pancreatic spasmolytic polypeptide (PSP) is a peptide that is isolated from the porcine pancreas and that affects intestinal motility and growth of intestinal tumour cells in vitro. We cloned a rat gene that is expressed primarily in the sublingual gland and named the predicted 503 amino-acid protein SLAMP (sublingual acinar membrane protein). SLAMP has 63% homology with human ERGIC-53-like protein, a member of the family of animal L-type lectins. Using a cDNA probe for SLAMP mRNA and rabbit antisera against SLAMP, we examined the expression and localization of SLAMP in major rat organs and tissues. With both Northern and Western blot analyses, abundant expression of SLAMP was demonstrated predominantly in the sublingual gland, with single sizes of the mRNA and protein 1.

The expression, localization, and regulation of aquaporin 5 (AQP5), a member of the water channel family of proteins, was investigated in tissues of the rat gastrointestinal tract. Reverse transcriptase--polymerase chain reaction (RT--PCR) detected AQP5 mRNA in the lower stomach and duodenum. A 52-year-old man underwent esophagogastroduodenoscopy during a routine health checkup, which revealed a 15 mm semipedunculated lesion in the duodenal bulb. The initial biopsy revealed Brunner's gland hyperplasia with gastric foveolar metaplasia (GFM). A second biopsy performed 6 months later revealed signet-ring cell carcinoma (SRCC) beneath a thick overlying layer of GFM, and endoscopic mucosal resection was thus performed. Expression of epidermal growth factor receptor (EGFR) was studied immunohistochemically in 111 cases of human duodenal bulb biopsy. The results showed that the expression of EGFR was strong on the luminal side of epithelial cell membrane, moderate in the basolateral membrane of epithilial cells and the cells of muscularis mucosae, low in the cytoplasm of the epithelial cells, and lower in the mesenchyma and Brunner's gland. BACKGROUND: The pathogenesis of non-ampullary duodenal epithelial tumors (NADETs) is not fully understood. NADETs that express gastric-type mucin phenotypes (G-NADETs) are noteworthy because of their high malignancy. Gastric foveolar metaplasia, from which G-NADETs originate, protects the duodenal mucosa from gastric acidity.
